Evaluation on Tongue Coating, Volatile Sulfur Compounds, Salivary Flow Rate and Secretory Immunoglobulin A in Saliva in Patients with Oral Malodor (구취환자의 설태, 휘발성 황화합물, 타액분비율 및 타액내 secretory Immunoglobulin A에 대한 평가)

  • Objectives : The aim of this study was to confirm the characteristics of oral malodor patients by evaluating the differences of salivary flow rate, secretory immunoglobulin A (sIgA) level in saliva between the patient and control groups, and the correlation with the tongue coating, volatile sulfur compound (VSC), salivary flow rate and sIgA level in saliva in the patients group. Methods : Forty-seven patients with oral malodor and twenty healthy volunteers were included in this study. Their tongue coating was assessed with the Winkel tongue coating index, and salivary flow rate, sIgA concentrations in saliva and the level of VSC in oral cavity were measured. Results : There were no significant differences of the salivary flow rate and the sIgA level in saliva between the patient and control groups, but there was a significant relationship between the accumulation of tongue coating and the level of VSC in oral cavity. Conclusions : Our results suggest that tongue coating is closely related to oral malodor, but further studies are needed to confirm the relationship between tongue coating and sIgA level in saliva.

  • Immunoglobulin G4 (IgG4)-related dacryoadenitis and sialoadenitis (IgG4-DS) are part of a multiorgan fibroinflammatory condition of unknown etiology termed IgG4-related disease (IgG4-RD), which has been recognized as a single diagnostic entity for less than 15 years. Histopathologic examination is critical for diagnosis of IgG4-RD. CD4+ T and B cells, including IgG4-expressing plasma cells, constitute the major inflammatory cell populations in IgG4-RD and are thought to cause organ damage and tissue fibrosis. Patients with IgG4-RD who have active, untreated disease exhibit significant increase of IgG4-secreting plasmablasts in the blood. Considerable insight into the immunologic mechanisms of IgG4-RD has been achieved in the last decade using novel molecular biology approaches, including next-generation and single-cell RNA sequencing. Exploring the interactions between CD4+ T cells and B lineage cells is critical for understanding the pathophysiology of IgG4-RD. Establishment of pathogenic T cell clones and identification of antigens specific to these clones constitutes the first steps in determining the pathogenesis of the disease. Herein, the clinical features and mechanistic insights regarding pathogenesis of IgG4-RD were reviewed.

  • lgM-like immunoglobulin was purified from the immune serum of M albus which immunized with bovine Serum albumin(BSA) as an antigen(Ag) and characterized. The Ag-specific antibody activity of the immune serum was increased after the immunization. The purified lgM-like immunoglobulin had a tetrameric structure which had a molecular weight of 800 kD and the monomer of IgM-like Ig had a mass of 199 kD which was composed of two heavy chains (Mol. wt. 70 kD) and iwo light chains (Mol. wt. 29.5 kD). The IgM-like Ig showed hemaggluti nating activity to mammalian RBC slightly.

  • Dietary conjugated linoleic acid(CLA) has been shown to affect immune function. Thus, the objective of this study was to investigate the effects of CLA on the mice that treated prednisone. Mice were randomized into 6 groups and fed diet containing either 0(control, P), 0.5%(CLA1, CP1) or 1.5%(CLA2, CP2) CLA for Sweets. Before 1 week of finishing diet supplement CP1, CP2, and P group treated the prednisone by subcutaneous injection. The levels of serum immunoglobulin A, G, E, gut lumen s-IgA, MLN immunoglobulin A, body weight, mucosal protein was compared. The level of serum IgA in CLA1, CLA2, CP1, and CP2 group increased, while which of P group was decreased. The level of serum IgG in CLA 1 group increased, while which of the other group no differences. Serum IgE level showed no difference and the immunoglobulin production in MLN lymphocyte in CLA 1 group increased. The level of gut lumen s-IgA in P group showed decreased, while which of the other group showed no differences. These results support the view that CLA supplement partially enhance the cell-mediated immunity and overcome the immunosuppressive effect of prednisone.

Stability of Immunoglobulin G(IgG) by Heat Treatment (면역단백질 G(IgG)의 열처리에 대한 안정성)

  • This study was carried out to obtain fundamental data when developing new colostrum component fortified milk products. Residual immunoglobulin G (IgG) activities of both IgG fortified milk products under different pasteurization conditions and colostrum fortified milk powder products under different dissolving temperatures were measured. In the study, residual IgG activities of raw milk and IgG (50 mg and 250 mg) fortified milk products were sharply reduced upon increasing the temperature of heat treatment. After the low temperature long time (LTLT) treatment residual IgG activities of raw milk, IgG 50 mg and 250 mg fortified milk products decreased to 79%, 30% and 21.6%, as compared to those before heat treatment respectively. However, almost no residual IgG activities were detected when IgG fortified milk was heated at 95$^{\circ}C$ for 15 sec. There was no significant change in the residual IgG activities of IgG fortified milk powder products upon different dissolving temperatures (30$^{\circ}C$, 40$^{\circ}C$, 50$^{\circ}C$ and 60$^{\circ}C$).

  • For the investigations of the usefulness of immunoglobulin Y (IgY), we attempted to produce and isolate IgY from egg yolk of white Leghorn hens immunized with fetuin as a model antigen. We used three methods to optimize the recovery of IgY: water dilution, polyethylene glycol, and carrageenan. The daily yield of anti-fetuin IgY from egg yolk was also examined, and it was isolated using a fetuin-affinity column at a yield of IgY of 2.7% from total IgY Furthermore, peroxidase-labeled antifetuin IgY was prepared, and was used to determine the minimum sensitivity against fetuin, which was found to occur to a fetuin concentration of 1 ng/$m\ell$.

Effects on the Laughter Score, Cortisol and Immunoglobulin of Laughter Therapy in Middle Aged Women (중년기 여성의 웃음치료가 웃음지수, Cortisol 및 면역글로불린에 미치는 효과)

  • Purpose: In this study, the effects of laughter therapy on a laughter index, cortisol and immunoglobulin of middle aged women were examined. Methods: A quasi-experimental nonequivalent control group pre/post-test design were used. The participants (n=54) included 27 in the experimental group and 27 in the control group. The data were collected from February through March of 2013. The experimental group participated in laughter therapy for 40 minutes per session, five times a week for a period of 2 weeks. The tool was used to measure pre/post-test laughter index, cortisol and immunoglobulin of middle aged women. Results: The results showed that laughter therapy was effective in increasing the laughter index (p<.001), and immunoglobulin IgA (p<.001), IgG (p<.001), and IgM (p=.010). Also, the stress hormone, cortisol of middle aged women decreased but non-significantly (p=.158). Conclusion: The result of the study indicates that laughter therapy may have an influence on nursing intervention and treatment effects to improve the laughter index and immunoglobulin of middle aged women.

Separation of Immunoglobulin G from Colostrum by Reverse Micelles of Cationic Surfactant (양이온 계면활성제로 형성된 역미셀을 이용한 초유 Immunoglobulin G의 분리)

  • This study was carried out to separate immunoglobulin G(IgG) from colostrum using reverse micellar extraction of cationic surfactant and to suggest suitable extraction conditions. The reconstituted colostrum powder was solubilized into a reverse micellar phase containing CDAB(cetyldimethylethyl ammonium bromide) by mixing equal volume of the aqueous and organic phase with constant stirring. The solubilization of proteins from the aqueous to the organic phase was manipulated by pH and ionic strength of the aqueous phase and concentration of surfactant in the organic phase. Based on the SDS-PAGE and densitometry, about more than 90% of initial IgG was remained in the aqueous phase after reverse micellar extraction. Although the aqueous phase contained lactoferrin and bovine serum albumin as minor components, about 93% of the total protein was IgG. The efficient extraction was achieved by the reaction of sodium phosphate buffer(pH 8) containing 50 mM KCl and organic phase containing 100 mM CDAB. The separation of IgG using reverse micellar extraction was simple, highly efficient and easy to be scaled up.

  • Purificationi of egg yolk immunoglobulin(IgY) was performed to understand the property of egg immunoglobulin. IgY differs from mammalian IgY in the molecular size(larger), isoelectric point(more acidic), and binding ability with mammalian complement and protein A(nonbinding ability). IgY is also known as ${\gamma}$-livetin and exists in egg yolk together with other two water-solubel proteins, ${\alpha}$-livetin(chicken serum albumin) and ${\beta}$-livetin(${\alpha}_2$-glycoprotein) and various lipoproteins(Low density lipoprotein, LDL and High density lipoprotein, HDL) which are the major components of egg yolk. The first step of isolation of IgY is to separate the water-solube proteins from lipoproteins. We report a simple method for separation of water soluble proteins using k-carrageenan and sedimentation. k-carrageenan was found to be effective for removal of yolk lipoprotein as a precipitate. IgY remained supernatant, and was isolated by chromatography on columns of DEAE-Sephacel and G 75 gel filtration chromatography.

Production of Egg Yolk Immunoglobulin and Its Application for Pepper mild mottle virus in Serological Tests (Pepper mild mottle virus에 대한 난황항체의 생산과 혈청학적 진단에의 활용)

  • Egg yolk immunoglobulin (IgY) is much widely used in medical fields, but its use in serology of plant viruses is much limited. We produced an IgY against pepper mild mottle virus (PMMoV) and applied it to several serological tests. Polyclonal antibodies were obtained from the egg yolk of chicken immunized with a total of 2mg of purified PMMoV over 2 months. The titers of antibodies were measured with the ring-test over six months after the first injection. The highest.titers of IgY was 1/2,560 at 2 months after the first injection. Approximately 60-80 mg of IgY were obtained from one egg yolk. Using the IgY, 1ng/ml of purified PMMoV was detected with the indirect ELISA. Gelrite gel double diffusion test, ELISA and tissue immuno-binding assay employing IgY gave similar sensitivity and specificity to those of IgG developed in rabbit. Therefore, the IgY which can be obtained in large quantities from a chicken, might be useful for the antibody production and the serology of plant viruses.
