Influence of Level and Source (Inorganic vs Organic) of Zinc Supplementation on Immune Function in Growing Lambs

  • Eighteen lambs were used to determine the effects of zinc (Zn) level and source on Zn status and immune function during both normal conditions and conditions of physiologic stress. Treatments consisted of a basal diet (27.6 mg of Zn/kg), and the basal diet supplemented with 25 mg of Zn/kg, added as either zinc oxide or zinc methionine. The basal diet was a corn-cottonseed hull-isolated soy protein- based diet (14% CP). Lambs were weighed and blood samples taken at 28-d intervals for determination of serum Zn and alkaline phosphatase activity. Weights and serum Zn were similar (p > 0.10) among treatments at all sampling days. To evaluate immune responses and Zn status during conditions of physiologic stress lambs were administered 100 I.U. of adrenocorticotrophin (ACTH) on d 112 and feed was withheld for 48 h. Cortisol levels were elevated (p < .01) 5 h post ACTH injection, but had returned to initial levels after 48 h. Lymphocyte blastogenesis ([$^3H$]-thymidine incorporation) on d 112 (prior to ACTH injection) and 114 was unaffected (p > .10) by dietary treatment. However, blastogenesis in response to pokeweed mitogen was greater (p < .0001), whereas the response to phytohemagglutinin was reduced (p < .01) following ACTH administration and fasting. Antibody response to administration of porcine red blood cells was unaffected (p > .05) by dietary treatment. These results indicate that, given the Zn concentration of the basal diet, there was no enhancement of immune function by supplemental Zn, either before or after lambs were subjected to stress.

Studies on the Function of Peroxidasins in Innate Immune System in C. elegans

  • Peroxidasin is a unique member of peroxidase family in that it has extracellular matrix (ECM) motif as well as peroxidase activity. Peroxidasins are involved in consolidation the extracellular matrix during development and in innate immune defense. C. elegans has two functional peroxidasins, PXN-1 and PXN-2, and PXN-2 is known to contribute to innate immune system. However, it is not clear of PXN-1 function in innate immune system. Therefore, this study is focused on the function of PXN-1 and the relationship between PXN-1 and PXN-2 in innate defense system in C. elegans. When pxn-1 was knocked down by RNAi, the worm turned to be more resistant to pathogens, Staphylococcus aureus and Pseudomonas aeruginosa and the enhanced resistance was abolished in pxn-1pxn-2 double knock down. By contrast, pxn-2 knock down showed stronger susceptibility to the pathogens. These results suggest that pxn-2 can contribute the pathogen resistance and pxn-1 can suppress the pathogen resistance. To confirm the idea, overexpression experiments were performed. Overexpression of pxn-1 showed more susceptible to pathogens compared to the control and double overexpression of pxn-1pxn-2 overcame the susceptibility of pxn-1 overexpression to the pathogens. On the other hand, pxn-2 overexpression made the worm more resistant to the pathogens and the resistance was maintained in pxn-1pxn-2 double overexpression. The comparison of the susceptibilities to the bacterial pathogens in above mentioned constructs suggests that PXN-1 suppress the function of PXN-2 in defense against bacterial pathogens in Caenorhabditis elegans.

  • The present study examined the ex vivo effect of Job's tear on immune function. Seven to eight week old mice(Balb/c) were fed a chow diet ad libitum two different concentrations (50 and 500 mg/kg BW) of water extract of Job's tear were orally administ every other day for two weeks. The results indicated that macrophage activation had occurred in the mice receiving 50 mg/kg B. W. of Job's tear water extract. Overall, using a mouse model, this study demonstrated that Job's tear extract may enhance immune function by regulating the $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ and IL-10 cytokine production capacity of activated macrophages in mice. This study may suggest that supplementation of Job's tear water extracts may enhance the immune function by regulating the enhancing the cytokine production by activated macrophage ex vivo.

신생아 마사지를 통한 모아상호작용이 어머니의 스트레스, 면역기능 및 영아기르기 자신감에 미치는 효과 (Effects of Maternal Massage to Newborn on Stress, Immune Function and Self-confidence of Mothers during the Postpartum Period)

  • Purpose: To investigate the effect of infant massage on stress, immune function and self-confidence in postpartum mothers. Method: From September, 2003 to February, 2004, 65 women who had delivered babies and were enrolled in the postpartum clinic were assigned to the experimental group or control group. In the experimental group, infant massage was given for 20 minute sessions twice a day from 4 days to 14 days after delivery. The instruments were Parental Stress Index, serum cortisol levels, WBCs, lymphocytes, and Self Confidence Scale. Results: Stress scores, WBCs and lymphocytes showed significant differences by the time, but there weren't significant differences according to the group and interactions between the group and the time. Cortisol and self-confidence scores showed significant differences by the time and the group. Conclusions: Applying massage to their newborn baby influenced cortisol levels of postpartum mothers. Therefore stress, cortisol, and immune functions in postpartum mothers should be generally screened and follow up studies are needed for the effective application of massage in postpartum mothers.

  • The nutritional status of host has a profound effect on immune response and its ability to defend aganinst invading pathogen. Almost all nutrient dificiencies impaired host defence, and more than recommended levels of certain nutrient enhance immune response beyond that observed with 'adequate'. But high-fat diets have been associated with various types of immune dysfunction in experimental animal models and humans. Also, high linoleic acid suppressed immune function and growth and metastasis of tumor than saturated fatty acids. The present review focused on the effect of dietary lipid on immune fuction, cytokine production and clinical conditions like infection, autoimmune disease and cancer.

생체영상과 미세가공을 이용한 면역 시스템 연구 (Studying immune system using imaging and microfabrication)

  • Immune system is composed of multiple cells with distinct functions, and immune responses are orchestrated by complex and dynamic cell-cell interactions. Therefore, each cell behavior and function should be understood under right spatio-temporal context. Studying such complexity and dynamics has been challenging with conventional biological tools. Recent development of new technologies such as state of art imaging instruments and microfabrication techniques compatible with biological systems have provided many exciting opportunities to dissect complex and dynamic immune cell interactions; new microscopy techniques enable us to observe stunning dynamics of immune system in real time. Microfabrication permits us to manipulate microenvironments governing molecular/cellular dynamics of immune cells to study detailed mechanisms of phenomena observed by microscopy. Also, microfabrication can be used to engineer microenvironments optimal for specific imaging techniques. In this presentation, I am going to present an example of how these two techniques can be combined to tackle challenging problems in immunology. Obviously, this strategy can readily be applied to many different fields of biology other than immunology.

  • PID controllers with constant gains have been widely used in various control systems due to its powerful performance and easy implementation. But it is difficult to have uniformly good control performance in all operating conditions. In this paper, we propose a nonlinear variable PR controller with immune feedback mechanism. An immune feedback mechanism is based on the functioning of biological T-cells, they include both an active term, which controls response speed. and an inhibitive term, which controls stabilization effect. Therefore, the proposed nonlinear PID controller is based on immune responses of biological. immune feedback mechanism which is the cell mediated immunity and In order to choose the optimal nonlinear PID controller games, we also propose the tuning algorithm of nonlinear function parameter in immune feedback mechanism. To verify performance of the proposed algorithm, the speed control of nonlinear DC motor are performed. Front the simulation results, we have found that the proposed algorithm is more superior to the conventional constant fain PID controller.

Regulation of Intestinal Immune System by Dendritic Cells

  • Innate immune cells survey antigenic materials beneath our body surfaces and provide a front-line response to internal and external danger signals. Dendritic cells (DCs), a subset of innate immune cells, are critical sentinels that perform multiple roles in immune responses, from acting as principal modulators to priming an adaptive immune response through antigen-specific signaling. In the gut, DCs meet exogenous, non-harmful food antigens as well as vast commensal microbes under steady-state conditions. In other instances, they must combat pathogenic microbes to prevent infections. In this review, we focus on the function of intestinal DCs in maintaining intestinal immune homeostasis. Specifically, we describe how intestinal DCs affect IgA production from B cells and influence the generation of unique subsets of T cell.

Emerging roles of neutrophils in immune homeostasis

  • Neutrophils, the most abundant innate immune cells, play essential roles in the innate immune system. As key innate immune cells, neutrophils detect intrusion of pathogens and initiate immune cascades with their functions; swarming (arresting), cytokine production, degranulation, phagocytosis, and projection of neutrophil extracellular trap. Because of their short lifespan and consumption during immune response, neutrophils need to be generated consistently, and generation of newborn neutrophils (granulopoiesis) should fulfill the environmental/systemic demands for training in cases of infection. Accumulating evidence suggests that neutrophils also play important roles in the regulation of adaptive immunity. Neutrophil-mediated immune responses end with apoptosis of the cells, and proper phagocytosis of the apoptotic body (efferocytosis) is crucial for initial and post resolution by producing tolerogenic innate/adaptive immune cells. However, inflammatory cues can impair these cascades, resulting in systemic immune activation; necrotic/pyroptotic neutrophil bodies can aggravate the excessive inflammation, increasing inflammatory macrophage and dendritic cell activation and subsequent TH1/TH17 responses contributing to the regulation of the pathogenesis of autoimmune disease. In this review, we briefly introduce recent studies of neutrophil function as players of immune response.

소화기계암의 수술후 면역기능에 대한 고려홍삼의 효과 (The Effect of Red Ginseng for Postoperative Immune Response in Gastrointestinal Carcinoma)

  • (Backgrounds) This study was performed to evaluate the usefulness of red ginseng ex rant as adjuvant therapeutic agent improving immune function in immune compromizing gas-trointestinal carcinoma patient. (Material and Methods) We were treated 72 patients with two groups after we were undertaken the curative resection for gastrointestinal carcinoma; 1) only chemotherapy and immunotherapy (control group) 2) chemotherapy and immunotherapy with 4500 mg (15 tablets) red ginseng for 6 months (study group). For investigating the immunologic alternations alongside the numerical changes in peripheral blood Iymphocyte and their subsets in the gastrolntestinal carcinoma patients, Iymphocyte surface markers were determined by monoclonal antibodies on the preoperative day, postoperative 1 months, 3 months, 6 months, 12 months and 18 months in 40 controls and 32 red ginseng groups In gastrointestinal carcinoma patients which was recruited at Korea diversity Hospital from March, 1995 to January, 1997. The patient was measured and compared in both groups with the body weight, total protein and albumin, blood hematocrit and hemoglobin, total leukocyte, lymphocyte and lymphocyte subsets count in peripheral blood through planed schedules. (Couclusion) This data suggests that red ginseng may be useful as a adjuvant therapeutic agent for improving the immune function after curative operation for immune compromizing gastrointestinal carcinoma patients.
