상지에 배양한 상황버섯 균사체로부터 분리된 다당류의 면역증강 효과 (Immune-Enhancing Effects of Polysaccharides Isolated from Phellinus linteus Mycelium on Mori ramulus)

  • 본 연구에서는 상지에 배양한 상황버섯 균사체 유래 다당류의 기능성 식품 소재로의 활용 가능성을 확인하기 위해 조다당류의 분리 및 정제를 통해 분획을 얻었으며, 다양한 면역증강 효과를 확인하였다. 건조된 조다당류를 DEAE-sepharose CL-6B를 이용하여 크로마토그래피한 후 340 nm에서의 흡광도, 단백질, 당 및 uronic acid 함량을 분석한 결과, 증류수로 용출되는 비흡착 분획(PF-1, 분획번호 3~15)과 흡착된 후 NaCl 용액에 의해 용출되는 분획(PF-2, 분획번호 24~33)을 얻었다. 분획물 PF-1 및 PF-2의 유용성분 함량으로 총당 함량은 각각 75.51%, 52.38%, 총단백질 함량은 1.63%, 8.41%, uronic acid 함량은 17.53%, 15.04% 및 ${\beta}-glucan$ 함량은 28.33%, 25.04%로 나타났다. PF-1 및 PF-2에 대해 대식세포주에 처리하여 세포생존율을 확인한 결과 $100{\mu}g/mL$ 농도까지 유의적으로 세포사멸이 나타나지 않아 세포독성이 없음을 확인할 수 있었다. Nitric oxide 생성량은 PF-1 $100{\mu}g/mL$에서는 nitric oxide 생성량이 $23.11{\mu}M$로 나타나 양성대조군으로 사용된 LPS 100 ng/mL 처리군($30.30{\mu}M$)의 약 76%의 효과가 있는 것으로 확인되었으며, cytokine 생성량($TNF-{\alpha}$ 및 IL-6) 또한 무처리군에 비하여 높은 생성량을 나타내었다. Polymerase chain reaction을 통한 면역 관련 유전자 발현분석 결과, iNOS, COX-2, $TNF-{\alpha}$, IL-6에서 PF-1 분획에서 높은 발현량을 나타내어 면역 증강을 목적으로 한 기능성 식품 개발에 활용 가능하다고 판단된다.

우렁쉥이(Halocynthia roretzi) 껍질로부터 분리된 다당류의 면역증강 효과 (Immune-Enhancing Effects of Polysaccharides Isolated from Ascidian (Halocynthia roretzi) Tunic)

  • 본 연구에서는 우렁쉥이 껍질 유래 다당류의 기능성식품 소재로의 활용 가능성을 확인하기 위하여 조다당류의 분리 및 정제를 통해 획분을 얻었으며 다양한 면역증강 효과를 확인하였다. 건조된 조다당류를 DEAE-sepharose CL-6B를 이용하여 크로마토그래피한 후 총당 및 uronic acid 함량을 분석한 결과, 증류수로 용출되는 비흡착 획분(APF-I, fraction No. 11~17)과 흡착된 후 NaCl 용액에 의해 용출되는 획분(APF-II, fraction No. 22~37)을 얻었다. 정제된 APF-I 및 APF-II의 이화학적 특성으로 총당 함량은 각각 66.62%, 27.03%, uronic acid 함량은 47.53%, 15.87%, hexosamine 함량은 16.62%, 46.79% 및 단백질 함량은 2.43%, 4.94%로 나타났다. APF-I 및 APF-II에 대해 RAW 264.7 세포에 처리하여 독성을 평가한 결과 $5{\mu}g/mL$ 농도까지 유의적으로 세포사멸이 나타나지 않아 세포독성이 없음을 확인할 수 있었다. Nitric oxide 생산량은 APF-I이 $5{\mu}g/mL$ 농도에서 $22.23{\mu}m$ 함량을 나타내어 LPS 대비 73.48%로 타 구간에 비해 높은 생성량을 나타내었으며, cytokine 생성량(TNF-${\alpha}$ 및 IL-6) 또한 APF-I $5{\mu}g/mL$ 농도에서 각각 LPS 대비 104%, 100.1%를 나타내어 LPS와 유사한 생성량을 나타내었다. Polymerase chain reaction을 통한 면역관련 유전자 발현 분석 결과, iNOS, COX-2, TNF-${\alpha}$, IL-6에서 APF-I 구간에서 LPS 처리군 보다 높은 발현량을 나타내어 면역증강을 목적으로 한 기능성식품 개발에 활용 가능하다고 사료된다.

The Effect of Angelica gigas Nakai on Immune Enhancement in Cyclophosphamide-induced Immune-suppressed Mice

  • Natural products are important sources for drug development because they have a wide variety of useful biological properties. Angelica gigas Nakai (AGN) has been used as an herbal medicine. The present study was designed to evaluate the immune-enhancing effect of AGN in the cyclophosphamide (CP) induced immune-suppressed mice. As the result, we found that CP induced the reductions of body ratio, spleen weights, hematopoietic parameter and cytokine productions in mice. However, AGN recovered immunosuppression-mediated decreased body ratio, spleen and thymus weights as well as regulation of hematopoietic parameter including white blood cell, lymphocyte, and neutrophil. According to histological study, AGN regenerated on CP-mediated injured spleen. Moreover, AGN increased the CP-induced reduction of cytokine expression in spleen tissue. Collectively, the findings provide experimental evidence that AGN may be a candidate for health-improving herbs.

보중익기탕가미방의 면역기능 증진 효과 (Immune Function-enhancing Effects of Bojungikkitanggami-bang)

  • The immune system acts to protect the host from infectious agents that exist in the environment and from other noxious insults. The immune system has two functional divisions: the innate and the acquired. Both components involve various factors such as cytokines. A number of methodologies exist to assess aspects of immune function. There are large inter-individual variations in many immune functions even among the healthy. Genetics, age, gender, smoking habits, habitual levels of exercise, alcohol consumption, diet, stage in the female menstrual cycle, stress, history of infections and vaccinations, and early life experiences are likely to be important contributors to the observed variation. While it is clear that individuals with immune responses significantly below 'normal' are more susceptible to infectious agents and exhibit increased infectious morbidity and mortality, it is not clear how the variation in immune function among healthy individuals relates to variation in susceptibility to infection. Oriental medicine is an important factor contributing to immune competence. The author investigated the immune enhancement effects of Bojungikkitanggami-bang (BITB). The forced swimming test (FST) has been used as a screening model for new immune enhancement agents. In the present study, the author investigated the effects of BITB on FST and blood biochemical parameters related to fatigue, glucose (Glc); blood urea nitrogen (BUN); lactate dehydrogenase (LDH); creatinine; and total protein (TP). The author found that BITB (1 g/kg) significantly reduced the immobility time in the FST compared to the control. In addition, the contents of Glc, LDH, BUN, TP in the blood serum were increased in BITB (1g/kg)-fed group. Also, the author investigated the effects of BITB on the production of cytokines in human T-cell line, MOLT-4 cells. BITB (1 mg/ml) significantly increased the interferon (IFN)-vproduction compared with media control (about 2.2-fold for IFN-γ) at 24 h. However, BITB has not affect the production of IL-2 and IL-4. In addition, BITB increased the protein expression level of IFN-γ in MOLT-4 cells. Thus, BITB may have therapeutic value in generating or enhancing immune function in a clinical setting.

전통적인 한방 처방 경옥고의 면역 증강 효과 (Immune-enhancing effects of a traditional herbal prescription, Kyung-Ok-Ko)

  • Objectives : A traditional herbal prescription, Kyung-Ok-Ko (KOK), has long been used in oriental medicine as an invigorant for age-related diseases, such as amnesia and stroke. However, the beneficial value of KOK for immune responses is largely unknown. Based on the above mentioned effects of KOK, other previous reports, and its use in traditional medicine, we hypothesized that KOK displays beneficial effects against methotrexate (MTX)-induced immune suppression. Methods : We investigated the effects of KOK (0.6 g/kg/day, oral (p.o.)) on deteriorated immunity caused by MTX (2 mg/kg/day, p.o.) in an immune suppression mouse model. MTX was fed to mice once a day for 7 days. After the immune responses of the mice deteriorated by MTX treatment, KOK in water was fed to the mice once a day for 14 days. We then measured the expression levels of various cytokines, such as T helper cell (Th1, Th2) cytokines, and the number of immune cells, such as spleen T cells, B cells, and macrophages. Results : The data showed that MTX decreased Th1 profiles (interferon $(IFN)-{\gamma}$, interleukin (IL)-2, IL-12) and the number of immune cells, and increased Th2 profiles (IL-4, IL-5, IL-13), which were normalized significantly by post-administration of KOK. However, there was no significant difference in body-weight gain between MTX- and KOK-treated mice. Conclusion : These results indicate that KOK has immune-enhancing functions and reduces immunotoxicity of MTX, suggesting that supplementation with KOK will improve immune responses clinically and be useful for the prevention of immune-related diseases.

Immunomodulatory effects of fermented Platycodon grandiflorum extract through NF-κB signaling in RAW 264.7 cells

  • BACKGROUND/OBJECTIVES: Platycodon grandiflorum (PG), an oriental herbal medicine, has been known to improve liver function, and has both anti-inflammatory and antimicrobial properties. However, little is known about the immune-enhancing effects of PG and its mechanism. In this study, we aimed to investigate whether fermented PG extract (FPGE), which has increased platycodin D content, activates the immune response in a murine macrophage cell line, RAW 264.7. MATERIALS/METHODS: Cell viability was determined by Cell Counting Kit-8 assay and the nitric oxide (NO) levels were measured using Griess reagent. Cytokine messenger RNA levels of were monitored by quantitative reverse transcription polymerase chain reaction. To investigate the molecular mechanisms underlying immunomodulatory actions of FPGE in RAW 264.7 cells, we have conducted luciferase reporter gene assay and western blotting. RESULTS: We found that FPGE treatment induced macrophage cell proliferation in a dose-dependent manner. FPGE also modulated the expression of NO and pro-inflammatory cytokines, such as tumor necrosis factor-α, interleukin (IL)-1β, and IL-6. The activation and phosphorylation levels of nuclear factor kappa B (NF-κB) were increased by FPGE treatment. Moreover, 5-aminoimidazole-4-carboxamide ribonucleotide, an activator of AMP-activated kinase (AMPK), significantly reduced both lipopolysaccharides- and FPGE-induced NF-κB reporter gene activity. CONCLUSIONS: Taken together, our findings suggest that FPGE may be a novel immune-enhancing agent acting via AMPK-NF-κB signaling pathway.

홍조류 유래 네오아가로올리고당의 면역 활성 증강 효과 (Immune Enhancing Activity of Neoagarooligosaccharides from Marine Red Algae)

  • Agar, a heterogeneous polymer of galactose, is the main component of the cell wall of marine red algae. It is well established as a safe, non-digestible carbohydrate in oriental countries. Neoagarooligosaccharides (NAOs) prepared by hydrolyzing agar by microbial β-agarase have been reported to show safety. However, their immunological effects have not been reported yet. Thus, the objective of this study was to investigate immune enhancing effects of neoagarooligosaccharides (NAOs) from marine red algae Gelidium elegans in mice by performing ex vivo experiments. Six-week-old mice were fed ad libitum. NAOs were orally administrated at three different concentrations (100, 500, and 2,500 mg/kg B.W./day) twice a week for four weeks. The group fed with NAOs at 2,500 mg/kg showed the highest proliferation of splenocytes and production levels of cytokines (IL-1β, IL-6, TNF-α) in the ex vivo experiment. In conclusion, NAOs can enhance immune function, increase proliferation of splenocytes, and increase cytokine production by activating macrophages in mice.

Health Promoting Properties of Natural Flavor Substances

  • The study of health promoting and disease preventing compounds in food or by themselves, so called nutraceuticals or functional foods, has become a major field of research in food science. Natural flavor compounds are usually present in food, essential oils, spices, and herbs. These compounds can produce aroma, not only by themselves, but also in combination with other compounds. Today, however, greater interest is being paid to the health promoting properties of natural flavor substances rather than their flavoring properties. In fact, a number of naturally occurring flavor compounds that possess health promoting and disease preventing properties have been extensively studied and identified. The beneficial properties of natural volatile flavor compounds as well as non-volatile substances in spices and herbs discussed in this review include antioxidant, anticarcinogenic, anti-inflammatory, and immune enhancing activities.

동의보감에 수재된 오계(烏鷄)의 선천면역반응 억제에 관한 연구 (Study of Innate Immunity Suppression of Yeonsan Ogye listed on Dong-eui-bo-gam)

  • The aim of the study is to evaluate immune-enhancing effects of Yeonsan Ogye. Various extract of Yeonsan Ogye (200 and 400 mg/kg/daily) was treated orally to Balb/c mice for 1 week, before acute inflammation was induced by LPS. After cytokine (IFN-γ, TNF-α, IL-6, and IL-1β) and immune cells (white blood cell, neutrophil, lymphocyte, and monocyte) level by serum and blood were counted. As a result, Oral treatment of Yeonsan Ogye extract to the Balb/c mice were significantly decreased cytokine level in serum, in comparison with control group. in addition, production of white blood cell and monocyte in blood was decreased and granulocyte was increased respectively, in comparison with control. Our results demonstrated that Yeonsan Ogye extracts seem to have significant immune-enhancing. Thus, Yeonsan ogye may be developed as a raw material for new health food and medicine to ease the symptoms related with inflammatory and immune.

Immuno-Modulatory Effects of Bacteriocin-Producing Pediococcus pentosaceus JWS 939 in Mice

  • Pediococcus pentosaceus JWS 939 (JWS 939) is a nonpathogenic bacteriocin-producing probiotic isolated from the duck intestine. This study assessed the immunomodulatory effects of JWS 939 and compared them with those of Lactobacillus rhamnosus GG (LGG), a well-known immune enhancer. The immune-enhancing effects of JWS 939 were measured by measuring the production of nitric oxide (NO) and cytokines in C57BL/6 mouse peritoneal macrophages. In addition, to assess the immune enhancement abilities of JWS 939, in vivo, a Listeria monocytogenes challenge mice model was used. The results showed that heat-killed JWS 939 induced more NO and interleukin (IL)-$1{\beta}$ production in mouse peritoneal macrophages than in LGG, and that oral administration of viable JWS 939 in mice increased more NO, IL-$1{\beta}$, and tumor necrosis factor (TNF)-${\alpha}$ level than in LGG in serum upon L. monocytogenes challenge. In addition, mice fed with JWS 939 had a longer survival time after lethal challenge with L. monocytogenes, and these effects were stronger than those induced by LGG. Collectively, P. pentosaceus JWS 939 is a remarkable strain that, by releasing bacteriocin and enhancing host immune responses, may have potential as a duck feed additive to suppress pathogens.
