저 에너지 레이저 파장이 화상쥐 혈청면역물질 농도 변화에 미치는 영향 (The Effects of the Low Power Wavelengths of Helium-Neon Infra- Red Laser on the Changes of the Serum Immunoglobulin Concentration in Burn Rats)

  • This studies were to investigate the effects of low power wavelengths Helium-Neon Infra-Red(He-Ne 1R) laser on the changes of the serum immunoblobulin(Ig) concentrations in bum rats, The thirty Spraque-Dawley adult male rats were assigned to the 5 groups: the experimental groups(3). the bum control group(1) and the control group(1). There was made three degree burn by the 290 mW IR on the back oreach rats, from 3 days after being burned. the experimental laser groups were irradiated low power wavelengths(292Hz- 1168Hz, 4672Hz) He-Ne IR laser far 5 minutes every day during the 7days. The results were as follows; 1. The concentrations of immunoglobulin G(Ig G) in serum of all experimental groups on the treated with the low power wavelengths of the He-Ne IR laser during the 5 minutes far 7 days were significantly lower than those of bum groups, but those 1168 Hz group were significantly higher than those of the 292 Hz and 4672 Hz groups. 2. There were significantly decreased the change of the level of immunoglobulin M(Ig M) in serum of all experimental groups on the treated with 5 minutes laser fer 7 days to the burn control group. and were significantly increased on treated with more higher wavelengths groups to those of lower groups. 3. The concentrations immunoglobulin E(Ig E) in serum of the 292Hz wavelengths groups were significantly lower than those in bum control group. As above results. the changes of the immunoglobulin in serum levels on the healing process have meaningful effected with the low power wavelengths role on the treated with He-Ne IR laser.

Effect of Chromium Dietary Supplementation on the Immune Response and Some Blood Biochemical Parameters of Transport-stressed Lambs

  • Forty-eight Naemi lambs (avg. BW 31.7 kg) were transported by truck for a distance of 1,450 km from Al-Jouf to Riyadh, Saudi Arabia. On arrival day, the lambs were randomly allocated to four groups receiving diets supplemented with 0.0, 0.3, 0.6 and 0.9 ppm organic chromium (Cr). Each group consisted of four separately housed replicates of three lambs each. The animals were fed ad libitum on a grower diet for 84 days. Blood samples were obtained shortly before transportation, upon arrival and at weekly intervals thereafter from all lambs for analysis of plasma and serum. Plasma glucose and serum cortisol, total protein, albumin, urea-N and total cholesterol concentrations were determined. A cursory clinical examination of the lambs, along with rectal temperature, was undertaken at different intervals during the experiment. The lambs were inoculated each with 2 ml i.v. chicken red blood cells (CRBC) on days 0, 21, and 42. Serum total, IgG and IgM antibody titers were determined at weekly intervals post-immunization. An in vivo intradermal hypersensitivity test was carried out on 6 lambs from each group on days 10 and 70. Transportation of the lambs resulted in a significant (p<0.001) elevation of serum cortisol, total protein and albumin levels, as well as increased plasma glucose concentration, with corresponding decrease in total cholesterol, while blood urea-N remained largely unchanged. These constituents returned to normal levels during subsequent weeks, with no significant differences in their concentrations being observed between the Cr-supplemented groups and controls. Rise in rectal temperature after transportation was reduced to a greater extent (p<0.05) in Cr-supplemented versus control lambs. Total, IgG and IgM antibody titers against CRBC rose significantly (p<0.05) during immunizations in all groups, with significantly and linearly higher (p<0.05) total and IgG titers in Cr-supplemented versus control lambs. By contrast, no significant effect due to Cr supplementation was recorded in IgG titers, which increased equally in Cr-fed and control groups. Skin thickness in response to intradermal inoculation of phytohaemagglutinin (PHA) was also significantly (p<0.01) increased as a result of Cr supplementation. These results indicate that dietary Cr supplementation might be useful during stress especially for enhancing immune responses in transport-stressed lambs.

Effects of Fruit By-product Extracts on Blood Characteristics, Antioxidant Activity, and Immune Response in Pigs

  • This experiment was conducted to determine the effects of extracts from fruit by-products on the blood characteristics, antioxidant activities, and immune response to Escherichia coli lipopolysaccharide (LPS) in growing pigs. A total of 96 pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc] with an initial BW of $27.94{\pm}0.92kg$ were used in a 6-week feeding trial. The pigs were randomly placed into one of four treatment groups with six replications (four pigs per replication) per treatment according to their initial BW. Treatments were: 1) CON (basal diet), 2) PRO (CON + 0.5% procyanidin), 3) HES (CON + 0.5% hesperetin), 4) TAN (CON + 0.5% tannin). At the end of the sixth week, five pigs (total 20 pigs, $BW=27.94{\pm}0.92kg$) were selected from each treatment and injected with LPS ($100{\mu}g/kg$ of BW). Blood samples were collected 3 h after LPS injection to assess anti-oxidative and inflammatory responses. After the LPS challenge, the concentration of serum cholesterol decreased with fruit by-product treatment compared with CON (p<0.05). The administration of TAN increased the concentration of blood total protein compared with the CON group 3 h after LPS challenge (p<0.05). The albumin concentration was also higher with PRO treatment compared to HES treatment (p<0.05). The concentration of IgM was increased by fruit by-product supplementation at 0 and 3 h (p<0.05). In addition, IgG concentration was higher in PRO, HES, and TAN treatments compared to CON treatment at 0 h, and IgG concentrations were also higher in the HES group compared to the CON group at 3 h (p<0.05). The concentration of IgA also increased with fruit by-product treatments at 3 h (p<0.05). In conclusion, dietary supplementation with fruit by-products may moderate the immune response after a LPS challenge in growing pigs.

Suppressive Effects of Scutellaria radix Water Extract on $Fc{\varepsilon}RI$ Expression

  • The initiation of immunoglobulin E (IgE)-mediated allergic reactions requires binding of IgE antibody to its high-affinity receptor. Human basophilic KU812F cells express $Fc{\varepsilon}RI$ on the cell surface and act as effector cells in the allergic response. In this study, we investigated the effects of Scutellaria radix extract on the expression of the $Fc{\varepsilon}RI$ in human KU812F cells. Flow cytometric analysis showed that S. radix extract treatment caused a concentration-dependent decrease in $Fc{\varepsilon}RI$ expression on the cell surface. Furthermore, the level of $Fc{\varepsilon}RI$ ${\alpha}$, ${\beta}$, and ${\gamma}$ chain mRNA in KU812F cells was examined by RT-PCR. S. radix extract reduced total cellular $Fc{\varepsilon}RI$ $\alpha$ and ${\gamma}$ chain mRNA expression in a concentration-dependent manner. $Fc{\varepsilon}RI$-mediated histamine release was reduced from $21.75{\pm}1.34\;ng/10^6$ cells in non-treated cells to $16.46{\pm}1.98\;ng/10^6$ cells in S. radix extract treated cells. These results suggested that S. radix extract has the potential to down-regulate of FcRI expression and to inactivate basophils.

Hot environment on reproductive performance, immunoglobulins, vitamin E, and vitamin A status in sows and their progeny under commercial husbandry

  • Hot environments can affect feed intake and lactation, and the subsequent unavailability of important micronutrients to the newborn piglet can impair piglet growth, reduce the viability of newborn piglets and limit their subsequent performance. This work addresses the effects of hot environments (summer season) upon the reproductive performance of sows during gestation and lactation as well as on the serum levels of vitamins and the concentration of immunoglobulins in their litters in comparison with the winter season. Fourteen sows were evaluated over 100 ± 2 days of gestation in each season. The temperature and humidity index (THI) was used as an indirect measure of heat stress during gestation. The reproductive performance, milk yield, and body condition of the sows were recorded. The concentrations of vitamin E and vitamin A in piglets and in sow serum, colostrum, milk and feed were determined by HPLC; immunoglobulins were assessed by an ELISA. The THI index indicated that animals were subject to heat stress only in during the summer. Although the effect was not significant, there were a lower number of piglets at birth and at weaning and the milk yield in summer compared with winter. There was no difference (p > 0.05) in the body condition of sows between seasons. Season had an effect (p < 0.05) on the vitamin A concentration of postpartum sow serum (0.29 ㎍/mL in winter vs 0.21 ㎍/mL in summer) and on the vitamin E concentration before birth (2.00 ㎍/mL in winter vs 0.90 ㎍/mL in summer). Vitamin E in milk was higher (p < 0.05) in winter than in summer (2.23 vs 1.81 ㎍/mL). Serum levels of vitamins A and E in piglets at birth were lower (p < 0.05) in winter than in summer. The concentrations of immunoglobulins (IgG and IgA) in colostrum and milk were similar between seasons (p > 0.05), but the IgA in piglet serum was higher in winter than in summer (p < 0.05). High temperatures produced heat stress in sows, which affected certain aspects of production that can be translated into economic losses for this sector.

Effect of Dietary Genetically Modified ${\beta}$-Carotene Biofortified Rice on Immune in Rats

  • This study aims to examine the effect of Genetically Modified ${\beta}$-Carotene Biofortified Rice rice developed by simultaneous expression technology in NAAS on biological immunity. Accordingly, this study added Genetically Modified ${\beta}$-Carotene Biofortified Rice 25, 50% and general rice 50% as control group into diet and provided rats with the prescribed feeds and then measured the contents of immunoglobulin and cytokine in blood. As a result, male and female IgM, IgE, male IgG1, female IgG2a and TNF-a, IL5 and IL12 showed no significant difference; male IgG2a tended to decrease dependently on the combined concentration of Genetically Modified ${\beta}$-Carotene Biofortified Rice; female IgG1 showed significance with control group, but its association with diet was not found. The higher the dietary mixing ratio, the more the male and female IFN-a and female IL-4 contents, regardless of rice variety, and it was found that female IL6 content decreased significantly, but its association with diet was not found. The risk of beta carotene-enriched rice into environment and human body has not been reported yet. The digestion of Genetically Modified ${\beta}$-Carotene Biofortified Rice can be seen as "safe" as this test result showed no big difference between general rice and Genetically Modified ${\beta}$-Carotene Biofortified Rice, and its usability is full of suggestions.

  • Hybridoma 세포의 세포 성장 속도, monoclo nal antibody의 생산성 및 세포 대사에 미치는 glucose의 영향이 조사되었다. IgG2,를 생산하는 m mouse-mouse hybridona VIII H-8 세포가 모델로 이용되었다. Glucose 농도에 따른 바성장속도의 변화는 기질저해형식 (substrate inhibition type) 의 성장모델로 나타낼 수 있었다. 초기 glucose 농도 4g/f까지는 최대 세포밀도의 증가를 보여 주었다. Glucose는 세포사망속도에 큰 영향을 나타내었고 glucose 농도와 비사망속도 간에는 반비례의 관계가 성립됨을 보였다. Glucose 농도가 증가될수록 세포의 생존율과 monoclonal an tibody의 생산이 증가되었다. Glucose 놓도가 증가될수록 glucose의 비소비속도가 증가되 었고, 초기 glucose 농도의 증가는 lactate의 총괄 비 생산속도의 증대를 가져왔다. 암모늄 이온의 총괄비 생산속도는 초기 Glucose의 농도에 의존하 였지만 암모늄 이온의 전반적인 생성속도는 초기 glucose 농도에 거 의 무관함을 보여 주었다.

Branched-chain Amino Acids are Beneficial to Maintain Growth Performance and Intestinal Immune-related Function in Weaned Piglets Fed Protein Restricted Diet

  • As a novel approach for disease control and prevention, nutritional modulation of the intestinal health has been proved. However, It is still unknown whether branched-chain amino acid (BCAA) is needed to maintain intestinal immune-related function. The objective of this study was to determine whether BCAA supplementation in protein restricted diet affects growth performance, intestinal barrier function and modulates post-weaning gut disorders. One hundred and eight weaned piglets ($7.96{\pm}0.26kg$) were randomly fed one of the three diets including a control diet (21% crude protein [CP], CON), a protein restricted diet (17% CP, PR) and a BCAA diet (BCAA supplementation in the PR diet) for 14 d. The growth performance, plasma amino acid concentrations, small intestinal morphology and intestinal immunoglobulins were tested. First, average daily gain (ADG) (p<0.05) and average daily feed intake (ADFI) (p<0.05) of weaned pigs in PR group were lower, while gain:feed ratio was lower than the CON group (p<0.05). Compared with PR group, BCAA group improved ADG (p<0.05), ADFI (p<0.05) and feed:gain ratio (p<0.05) of piglets. The growth performance data between CON and BCAA groups was not different (p>0.05). The PR and BCAA treatments had a higher (p<0.05) plasma concentration of methionine and threonine than the CON treatment. The level of some essential and functional amino acids (such as arginine, phenylalanine, histidine, glutamine etc.) in plasma of the PR group was lower (p<0.05) than that of the CON group. Compared with CON group, BCAA supplementation significantly increased BCAA concentrations (p<0.01) and decreased urea concentration (p<0.01) in pig plasma indicating that the efficiency of dietary nitrogen utilization was increased. Compared with CON group, the small intestine of piglets fed PR diet showed villous atrophy, increasing of intra-epithelial lymphocytes (IELs) number (p<0.05) and declining of the immunoglobulin concentration, including jejunal immunoglobulin A (IgA) (p = 0.04), secreted IgA (sIgA) (p = 0.03) and immunoglobulin M (p = 0.08), and ileal IgA (p = 0.01) and immunoglobulin G (p = 0.08). The BCAA supplementation increased villous height in the duodenum (p<0.01), reversed the trend of an increasing IELs number. Notably, BCAA supplementation increased levels of jejunal and ileal immunoglobulin mentioned above. In conclusion, BCAA supplementation to protein restricted diet improved intestinal immune defense function by protecting villous morphology and by increasing levels of intestinal immunoglobulins in weaned piglets. Our finding has the important implication that BCAA may be used to reduce the negative effects of a protein restricted diet on growth performance and intestinal immunity in weaned piglets.

진폐증 환자에서의 혈청 면역 글로불린의 변화 (Serum Immunoglobulin levels in Coal Worker's Pneumoconiosis Patients)

  • 연구배경: CWP은 주로 석탄 분진을 흡입함으로써 발생하는 폐 실질의 병변으로 단순 흉부 방사선 소견상 폐 결절의 기준을 1cm로 하여 SP과 PMF으로 구분한다. PMF의 경우 환기 능력의 장애가 심하고 조기사망율이 증가하고 있으나 모든 광부에서 PMF이 발생하는 것은 아니여서 이러한 CWP의 발생에 관여하는 요인을 찾기 위한 많은 연구가 있어왔다. 본 연구의 주목적은 진폐증 환자에서 질환의 심한 정도를 ILO 분류에 따라 SP, PMF으로 구분한 후 감염이나 기타 기저 질환이 없는 상태에서의 혈중 면역 글로블린 농도를 흡연력과 연령에 의한 면역 글로불린의 변화를 고려한 후 진폐증의 심한 정도와 혈청면역 글로불린 농도와의 연관성을 관찰하기 위하여 시행하였다. 방 법: 태백 중앙 병원과 영동 병원의 진폐증 환자중 정밀 검사를 시행하기 위해 병원을 내원한 SP 환자 51명과 입원중이거나 정밀검사를 시행한 PMF 환자 59명을 대상으로 하였고 대조군은 진폐증 환자와 나이, 성별 및 흡연 병력이 적합하면서 진폐에 노출된 병력이 전혀 없는 주변 아파트 경비원과 병원내 지원자 58명을 대상으로 하였다. 면역 글로불린의 검사(IgA, IgG, IgM)는 독일 Behring사에서 제조한 각 각의 Kit를 사용하여 Nephelometer 분석기(Behring Nephelometer : Germany)를 사용하여 측정하여 대조군 57명, SP 51명, PMF 49명의 면역 글로불린을 측정하였다. 결 과: 본 연구에서 대조군의 나이에 따른 혈청 면역 글로불린의 변화는 나이가 증가 할수록 IgG는 의의 있게 증가 하였고 흡연력에 의한 면역 글로불린의 차이는 없었다. 대조군, SP, PMF사이의 혈중 면역 글로불린의 농도간에는 차이가 없었으나 흡연력과 나이의 영향을 고려한 다중회귀 분석에서 IgG의 농도는 대조군과 비교하여 SP 환자군에서 통계학적으로 의미 있게 감소하였고 PMF환자군에서는 감소하는 경향이 있었다. 폐기능 검사의 중증도와 면역 글로불린 농도사이에는 관련이 없었다. 결 론: 흡연력과 연령을 고려한 상태에서 진폐증 환자의 혈청 면역 글로불린의 농도는 대조군과 비교하여 SP 환자군에서 IgG의 농도가 감소하였으며 PMF 환자군에서도 감소하는 경향을 나타내어 진폐증과 IgG사이에 연관성이 있다고 생각되며 폐기능 검사의 중증도와 면역 글로불린과는 관련이 없다고 생각된다. 향후에 CWP의 발생 및 진행 기전을 이해하기 위해서 많은 수의 진폐증 환자를 대상으로 하는 전향적 연구와 함께 염증세포 및 싸이토카인의 역할에 대한 연구가 필요할 것으로 생각된다.

Beevenom 처리가 돼지의 체내 면역반응에 미치는 효과 (Effects of Natural Honeybee (Apis mellifera ligustica) Venom Treatment on the Humoral Immune Response in Pigs)

  • 본 실험은 생봉독 처리가 돼지의 체액성 면역반응에 대한 생봉독의 효과를 조사하기 위하여 실시하였다. 혈중 Immunoglobulin(Ig) G, IgA, IgM의 농도에 미치는 생봉독의 효과를 조사하기 위하여 모돈 3두에서 생산한 자돈 20두(LY ×D)를 생봉독 처리군과 대조군으로 각각 10두씩 배치하였다. 생봉독 처리군은 출생시와 3일령에 교소혈 (GV-1), 해문혈(ST-25) 및 두구혈(CV-8), 6일령에 거세, 단미 창상부위에 생봉 1마리씩 직침 시술하였고, 대조군은 생리식염수 1ml를 동일한 혈위에 주입하였다. 혈중 Ig 농도 측정을 위하여 출생시와 3일, 7일, 14일 및 21일령에 채혈하여 Immunoturbidimetric method로 IgG, IgM, IgA를 측정하였다. 지표항원으로 사용한 돈 콜레라와 위축성비염 백신에 대한 생봉독의 항체 생성효과를 조사하기 위하여 모돈 5두에서 생산된 자돈 40두(LY×D)를 생봉독 처리군과 대조군으로 각각 20두씩 공시하였다. 생봉독 처리군은 출생시에 교소혈(Jiao-Chao, GV-1), 해문혈(Hai-Men, ST-25) 및 두구혈(Du-Kou, CV-8)에, 3일령에 단미 및 거세시술 시 창상부위에 생봉독을 처리하였고, 21일령 이유시에는 교소혈(GV-1)과 백회혈(Bai- Hui, GV-20)에 생봉독을 처리하였다. 지표항원으로 위축성비염 백신은 24일령과 44일령에 접종하였고 돈콜레라 백신은 44일령과 64일령에 각각 접종하였다. 항체가 분석용 혈액은 24, 34, 44, 54 및 74일령에 채혈하여 위축성비염은 시험관응집반응, 돈콜레라는 ELISA법에 의하여 항체가를 조사하였다. IgG 농도는 처리군에서 출생시 339.52, 3일령에 366.48, 7일령에 296.52, 14일령에 242.06, 21일령에는 219.06mg/dl이었고 대조군은 각각 347.10, 333.14, 243.28, 205.18 및 191.58mg/dl 이었다. 처리전(출생시)에는 처리군과 대조군간에 유의차가 없었으나 처리군의 IgG 농도가 대조군에 비하여 3일령 10.3% (P<0.02), 7일령에는 21.9% (P<0.01), 14일령에는 18.0% (P<0.07), 21일령에는 14.3(P<0.07) 더 높게 나타났다. IgA와 IgM의 농도는 전체기간동안 처리군과 대조군간에 유의차가 인정되지 않았다. 돈콜레라 virus에 대한 항체역가는 처리군에서 대조군에 비하여 24일령 때 57.0%(P<0.03), 34일령 때 74.6% (P<0.006), 44일령에 48.6% (P<0.017), 54일령 때 45.0% (P<0.16), 74일령 때 44.4%(P<0.006)가 유의하게 높은 것으로 나타났다. 위축성비염 원인균인 Bordetella bronchiceptica에 대한 항체역가는 처리군이 대조군에 비하여 34일령 때는 39.7%(P<0.002), 44일령 때 31.9% (P<0.02), 54일령 때에 33.4%(P<0.01), 74일령 때에는 57.3%(P<0.007)가 높게 나타나 접종일인 24일령을 제외하고 전체기간에 걸쳐 높은 항체수준을 보였다. 위의 결과를 종합해 볼 때 돼지에 대한 생봉독 처리는 면역기능향상 효과가 있는 것으로 사료된다. 이와 같은 결과를 볼 때 돼지에 대한 생봉독의 처리는 양돈 생산성면에서 생봉독 처리는 돼지의 생존율을 높이고, 증체량이 높았다는 조 등(2005)의 보고를 뒷받침하고 있어 양돈 산업발전에 기여할 것으로 사료된다.
