Bevacizumab Regulates Cancer Cell Migration by Activation of STAT3

  • There are numerous clinical cases indicating that long-term use of bevacizumab may increase the invasiveness of tumors. However, to date, little is known about underlying molecular mechanisms. Therefore, the purpose of our study was to investigate effects of bevacizumab in four cancer cells lines (WSU-HN6, CAL27, Tca83, and HeLa). It was found to promote migration and invasion in the WSU-HN6 and Tca83 cases, while exerting inhibitory effects in CAL27 and HeLa cells. The signal transducer and activator of transcription (STAT) 3 inhibitors niclosamide and S3I-201 inhibited the STAT3 signal pathway, which is activated by bevacizumab. These inhibitors also substantially blocked bevacizumab-induced migration of WSU-HN6 and Tca83 cells. Bevacizumab upregulated interleukin (IL)-6 and phosphorylated (p)-STAT3 expression time-dependently. Therefore, we propose that bevacizumab has differential effects on the migration of different cancer cell lines and promotes migration via the IL-6/STAT3 signaling pathway.

Oldenlandia diffusa Ameliorates on Atopic Dermatitis in Mice

  • Oldenlandia diffusa (OD), a member of the Rubiaceae family, has been used for treating inflammatory and infectious diseases. However, the anti-atopic effects of OD are not completely understood yet. Thus, the aim of the present study was to elucidate beneficial effects of OD on atopic dermatitis (AD) in vivo. We examined the pharmacological effects of OD on compound 48/80- or histamine-induced scratching behaviors and 2, 4-dinitrochlrobenzene (DNCB)-induced AD-like skin lesions in mice. Additionally, we evaluated regulatory effects of OD on the expression of interleukin (IL)-6 and tumor necrosis factor (TNF)-α in DNCB-induced AD-like skin lesions. Results showed that OD inhibited the scratching behavior and AD symptoms. It also decreased serum levels of IgE and histamine in mice. Moreover, OD significantly ameliorated the expression of IL-6 and TNF-α in AD-like skin lesion. These finding suggest experimental evidence of that OD's potential for treating AD.

Characterization of lactoferrin hydrolysates on inflammatory cytokine expression in Raw264.7 macrophages

  • Lactoferrin is an iron-binding glycoprotein which is present in colostrum, milk, and other body secretions. Lactoferrin activities are associated with inflammatory and immune responses. The aim of this study was to investigate the effect of lactoferrin hydrolysates (LH) on the production of immunomodulatory factors such as inflammatory related cytokines (tumor necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$, interleukin (IL)-6, and interleukin (IL)-13) in Raw264.7 cells, which originated from murine macrophages. The results show that the Raw264.7 cells cultured in 3 types (whole, and above and below 10 kDa) of lactoferrin hydrolysates (LH) did not show any cytotoxicity in the cells. $TNF-{\alpha}$ decreased dose-dependently to 1,500 - 2,000 ng/mL by treatment with the 3 types of LH at 1, 50, $100{\mu}g/mL$, whereas the positive control, lipopolysaccharide (LPS), and negative control produced 2,450 and 1,000 ng/mL of $TNF-{\alpha}$, respectively, in the Raw264.7 cells. The treatment with the 3 types of LH (whole and above and below 10 kDa) at $50{\mu}g/mL$ produced about 20 - 28 ng/mL of $IL-1{\beta}$ at 3, 6, and 9 h, respectively, while the negative control produced 7 ng/mL, and LPS as the positive control produced 48 - 60 ng/mL. $TNF-{\alpha}$ and IL-6 expression was decreased dose-dependently by the 3 types of LH. The mRNA levels of IL-13 were slightly increased dose-dependently by the whole and above 10 kDa LH, but decreased dose-dependently by the below 10 kDa LH in the Raw264.7 cells. The results show that LH had immunomodulating effects on cytokine production in anti- and pro-inflammatory reactions as well as anti-allergic reactions.

생약 추출물의 RAW 264.7 세포를 이용한 면역증강 효과 (Immune Enhancing Effect of Medicinal Herb Extracts on a RAW 264.7 Macrophage Cell Line)

  • 본 연구에서 12종류의 생약재 중 대식세포를 활성화시켜 NO 생성능이 높은 감초, 건지황, 당귀, 도라지, 목천료 5종의 생약재를 선별하였다. 선별된 5종의 생약추출물의 성분을 실험한 결과 단백질, 다당 물질의 함유량이 감초(58.1%), 건지황(90.4%), 당귀(87.4%), 도라지(41.3%), 목천료(85.7%)로 대부분 고분자 물질이 함유되어 있는 것으로 나타났다. 생약추출물이 대식세포를 활성화시켜 면역을 증진시키는 효과를 알아보기 위해 RAW 264.7세포와 T세포를 이용하여 면역활성능 관련 지표를 측정한 결과 5종의 생약재를 RAW 264.7 세포에 처리하였을 때 면역 활성의 지표가 되는 NO, cytokine(TNF-${\alpha}$, IL-$1{\beta}$, IL-6, IL-10)의 생성이 추출물을 처리하지 않은 대조군에 비해 증가되었고, Molt-4 세포에 처리하였을 때 대조군에 비해 세포가 증식되었다. 이와 같은 결과는 고분자 물질인 단백질과 다당으로 이루어진 생약추출물을 섭취 시 외부로부터 침입한 항원이 들어오기 이전에 면역세포를 자극하여 활성을 증가시켜 면역매개물질을 생성하여 인체의 비특이적 면역반응을 증가시킴으로써 항원을 공격, 제거하는 등의 작용을 통해 자연 면역반응에 있어 중요한 역할을 할 수 있을 것으로 보인다.

  • 9 종류의 종자 중 대식세포를 자극하여 NO 생성능이 높은 메밀, 민들레, 봉선화, 해바라기 4종의 종자를 선별하였다. 선별된 4종의 종자 추출물이 RAW 264.7 세포를 활성화시켜 면역을 증진시키는 효과를 알아보기 위해 RAW 264.7 세포와 T세포를 이용하여 면역 활성능 관련 지표를 조사하였다. 4종의 종자를 대식세포에 처리하였을 때 면역 활성의 지표가 되는 NO, cytokine(TNF-${\alpha}$, IL-$1{\beta}$, IL-6, IL-10)의 생성이 추출물을 처리하지 않은 대조군에 비해 증가되었고, Molt-4 세포에 처리하였을 때 대조군에 비해 세포가 증식되었다. 이와 같은 결과는 종자 추출물을 섭취하였을 때 외부로부터의 어떠한 자극이 있기 이전에 체내의 모든 조직에 분포하면서 1차적으로 이물질을 제거하는 대식세포를 자극하여 cytokine 등의 면역매개물질을 생성하여 인체의 비특이적 면역반응을 증가시킴으로써 항원을 공격, 제거하는 등의 작용을 통해 자연 면역반응에 있어 중요한 역할을 할 수 있을 것으로 판단된다.

안지오텐신전환효소 및 사이토카인 혈장 레벨과 재발성 우울증의 연관성 : 예비 연구 (Association of Plasma Levels of Angiotensin-Converting Enzyme and Cytokines with Recurrent Depression : A Preliminary Study)

  • Objectives Angiotensin-converting enzyme (ACE) gene and plasma levels of cytokines, such as tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin-4 (IL-4), interleukin-6 (IL-6), interleukin-10 (IL-10), have previously been determined to be associated with depression. The purpose of this study was to investigate the association of plasma levels of ACE and cytokines with recurrent depression. Methods A total of 52 participants (14 male, 38 female, aged $43.9{\pm}14.0years$) were enrolled after being diagnosed with depression by experienced psychiatrists using the Mini International Neuropsychiatric Interview from the outpatient clinic of the Department of Psychiatry, CHA Bundang Medical Center. The participants completed blood sampling, the Hamilton Depression Rating Scale (HAMD), the Beck Depression Inventory, the Beck Anxiety Inventory, and the Scale for Suicidal Ideation. Results ACE plasma levels are higher in patients with recurrent depression ($27.4{\pm}10.4U/L$) than in patients with newly diagnosed depression ($19.1{\pm}7.7U/L$) (p = 0.004). The levels of cytokines, such as $TNF-{\alpha}$, IL-4, IL-6, IL-10, are not significantly different between the two groups. Additionally, the ACE plasma level is negatively correlated with a reduction in the HAMD over six weeks (r = -0.429, p = 0.046, n = 22). Conclusions The current findings show that plasma ACE levels may be associated with recurrent depression and further suggest that the renin-angiotensin system could play a role in recurrent depression.

LPS로 활성화한 RAW 264.7 세포에서 HK표고버섯균사체의 NF-κB 활성 억제를 통한 항염증 효과 (Anti-inflammatory Efficacy of HK Shiitake Mushroom Mycelium in LPS-treated RAW 264.7 Cells Through Down-regulation of NF-κB Activation)

  • HK표고버섯균사체(HK shiitake mushroom mycelium, HKSMM)는 간 건강 개별 인정 건강기능식품이다. LPS로 활성화된 RAW 264.7 세포에서 HKSMM50 (HKSMM의 50% ethanol 수용액 추출물)의 항염증효과를 연구하였다. AHCC는 positive control로 사용하였다. LPS로 활성화된 RAW 264.7 세포에 HKSMM50 및 AHCC를 처리(0, 20, 100, 500 ㎍/ml)하고 24시간 배양하여 배양물의 염증 관련 인자는 ELISA kits로, 세포에 함유된 iNOS와 COX-2 protein 발현은 Western blotting으로 측정하였다. HKSMM50는 LPS 처리에 비해 농도 의존적으로 NF-κB 함량을 낮추었고, iNOS와 COX-2 protein 발현을 억제하여 NO와 PGE2 함량을 낮추었다. 더불어 HKSMM50는 LPS 처리에 비해 IL-1β, TNF-α, IL-4 및 IL-6의 함량을 낮추었으나 SOD와 CAT의 활성은 증가시켰다. AHCC도 HKSSM50 처리와 비슷한 효과를 나타내었다. 이 결과는 HKSMM50이 LPS로 활성화된 RAW 264.7 세포에서 NF-κB 신호전달을 억제하여 항염증효과를 나타내었으며, HKSMM은 면역기능증진에 도움을 줄 수 있는 건강기능식품원료로 사용할 수 있을 것이다.

Lipopolysaccharide로 활성화된 RAW 264.7 세포에서 적하수오(Polygonum multiflorum) 추출물의 항염증 효과 검증 (Anti-inflammatory effect of Polygonum multiflorum extraction in activated RAW 264.7 cells with lipopolysaccharide)

  • 적하수오 추출물의 추출용매별 항염증 작용을 알아보기 위해 LPS로 자극된 RAW 264.7 세포를 이용하여 실험을 진행 한 결과 열수 추출물 및 70% 에탄올 추출물 모두 $100{\mu}g/mL$에서 60%이상의 NO 생성 억제 율을 보이는 것을 확인 하였다. 또한 사이토 카인들에 대하여 적하수오 추출물은 TNF-${\alpha}$, IL-$1{\beta}$, IL-6, $PGE_2$ 생성량은 농도 의존적으로 감소하였고, 이는 적하수오 추출물이 염증성 사이토 카인의 발현을 억제 하여 염증 발생을 억제 할 수 있다고 할 수 있다. PMW는 $100{\mu}g/mL$의 농도에서 iNOS 단백질 발현량이 89%, COX-2는 54%의 효과를 나타내었으며, PME는 $100{\mu}g/mL$의 농도에서 iNOS 91%, COX-2는 57%의 단백질 발현 저해효과가 있음을 확인 할 결과로 보아 적하수오는 대식세포에서 NO, TNF-${\alpha}$, IL-$1{\beta}$, IL-6, PGE2 iNOS, COX-2 발현을 억제 하며, 특히 적하수오 70% 에탄올 추출물의 경우 저 농도에서부터 효과를 나타내어 우수한 항염증 소재임을 확인 할 수 있었다.

The Effect of Barbaloin on LPS-stimulated Inflammatory Reaction in Mice Peritoneal Macrophages

  • Barbaloin is a major component of Aloe vera, which has been used for a laxative. Also, barbaloin is C-glucoside of aloe emodin anthrone which is founded in Aloe vera. Barbaloin has varieties of pharmacological activity such as inhibitory effects on inflammation, histamine release, cancer and microbial infection. But the effect of barbaloin on lipopolysaccharide (LPS)-stimulated macrophages has not been understood. In this study, we evaluated the effects of barbaloin against LPS-stimulated production of nitric oxide (NO), inflammatory cytokines and MAPKs activation in macrophage. We treated barbaloin (0.1, 1, 10, $100{\mu}M$) in LPS-stimulated mice peritoneal macrophage. Our results showed that barbaloin significantly inhibited production of NO and cytokines of tumor necrosis factor $(TNF)-{\alpha}$, interleukin (IL)-6, interleukin $(IL)-1{\beta}$ in LPS-stimulated peritoneal macrophage. Moreover, barbaloin inhibited the phosphorylation of ERK and JNK in a dose dependent manner. These results indicated that barbaloin could be useful for inflammatory diseases.

Comparison of Interleukin-8 Levels in Long-Distance Runners and Healthy Sedentary Non-Athletic Control Subjects

  • We have previously demonstrated that the level of leukocytes and neutrophils significantly increased immediately and 30 min after exercise. Interleukin-8 (IL-8) is an inflammatory cytokine that acts as a chemokine on neutrophils. In the present study, we evaluated the correlation between the number of neutrophils and leukocytes, and between the number of neutrophils and plasma IL-8 level. Long-distance trained runners (TRs, n = 10) and untrained sedentary control subjects (SEDs, n = 10) ran for one hour at 70% of heart rate reserve. In the TR, the number of neutrophils correlated significantly with the number of leukocytes in the blood. However, there was no correlation between the number of neutrophils and the plasma IL-8 concentration in both groups. Expressions of IL-8 protein and mRNA were markedly higher in the TRs as compared to the SEDs at three time intervals (pre-exercise, immediately after exercise, and post exercise). In conclusion, our results show that 1) the neutrophil level was dependent on the level of leukocytes 2) there was no correlation between the neutrophils count and plasma IL-8 concentration and 3) a higher plasma IL-8 level in athletes may be a unique characteristic of intensive training.
