Inhibitory Effects of Trichosanthis Radix in the Activity and Proliferation of Th2 T Cells and Eosinophils in vitro : Implications on its Regulatory Roles for Asthma (과루근(瓜蔞根)이 Th2 T 세포와 호산구에 대한 활성 및 증식 억제에 미치는 영향)

  • Objectives : The present study was carried out to investigate the effect of Trichosanthis Radix extract (TRE) on the proliferation and activation of eosinophils which were prepared from lung cells of asthma-induced mice by ovalbumin (OVA) treatment. Methods : C57BL/6 mouse was exposed to OVA three times a week for 6 weeks. The mouse lung tissues were dissected out, chopped and dossiciated with collagenase (1 $\mu$g/ml). Eosinophils were activated by rmIL-3/rmIL-5 co-treatments. The lung cells were treated with TRE, incubated for 48 hr at 37$^{\circ}C$, and analyzed by flow cytometer, ELISA and RT-PCR methods Results : To measure cytotoxicity, mouse lung fibroblast cells (mLFCs) were pretreated with various concentrations of TRE. TRE at 100 $\mu$g/ml, the highest concentration, examined did not have any cytotoxic effects on mLFCs. In FACS analysis, number of granulocyte/lymphocyte, CD3e-/CCR3+, CD3e+/CD69+, CD4+/CD8+ T cells in asthma-induced lung cells were significantly decreased by TRE treatment compared to the control group. But CD4+/CD25+ T cells were not examined significant change in lung cells treated with TRE. In ELISA analysis, production levels of IL-3, IL-5, IL-13 and histamine in asthma-induced lung cells, which were induced by rIL-3 plus rmIL-5 co-treatment, were significantly decreased by TRE treatment. Conclusions : The present data suggested that Trichosanthis Radix on the inhibition of parameters associated with asthma responses in eosinpophils, and thus implicate the possibility for the clinical application of Trichosanthis Radix.

The Effects of Sinapis Semen, Raphani Semen, and mixture decoction on the Asthmatic Mouse Model (백개자, 나복자 및 두 배합 약물의 천식 동물 모델에 대한 효과)

  • Objectives : To clarify the possible effects of Sinapis Semen and Raphani Semen on the development of pulmonary eosinophilic inflammation in a asthmatic mouse model. Methods : BALBav/c mice were sensitized to OVA followed intratracheally and by aerosol allergene challenges. We investigated the effect of Sinapis Semen and Raphani Semen on airway hyperresponsiveness, eosinophiic infitratio, immune cell phenotype, The2 cytokine product, and OVA-spedific IgE production. Results : Total lung cells, eosinophils, and lung leukocytes, OVA specific IgE levels, and Th 2cytokine levels such as IL-5, IL-13, IL-17, TNF-alpha, and eotaxin in BALF were reduced compared with those of OVA sensitized asthma mice (control). The absolute numbers of $CD3^+$, $CD3^+/CD69^+$, $CD3^-/CCR3^+$, $CD4^+$, $CD8^+$, $Gr-1^+/CD11b^+$, $B220^+/CD22^+$, $B220^+/IgE^+$ cells in lung tissiues significantly reduced compared to those of control. Specially total lung cells in BALF and the absolute number of $CD3^+/CD69^+$ and, $B220^+/IgE^+$ cells in lung tissiue effectively reduced in Sinapis Semen plus Raphani Semen compared to those of Sinapis Semen and Raphani Semen. Conclusions : These results indicate that Sinapis Semen plus Raphani Semen has deep inhibitory effects on airway inflammation and hyperresponsiveness in asmatic mouse model and also has effect of suppression of IL-5, IL-13, IL-17, OVA specific IgE production in BALF. The results verified that Sinapis Semen, Raphani Semen, and Sinapis Semen plus Raphani Semen could act as a immunomodulator which possess anti-inflammatory and anti-asthmatic property by modulating the relationship of Th1/Th2 cytokine imbalance.

The Inhibitory Effects of GHJ on Allergic Inflammatory Response in Human Mast Cells (HMC-1) (고삼, 형개, 자초 혼합물(GHJ)의 인간비만세포에서의 항염증 효과)

  • Objectives : This study aimed to evaluate inhibitory effects of GHJ on allergic inflammatory response in human mast cells (HMC-1). Methods : To investigate the inhibitory effect of GHJ (62.5, 125, 250, 500, $1000{\mu}g/mL$), HMC-1 cells were stimulated with phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI). Enzyme-linked immunosorbent assays (ELISAs), RT-PCR and Western blot analysis were investigated using GHJ extract. Results : GHJ inhibited levels of $TNF-{\alpha}$ and IL-6 of $1000{\mu}g/mL$ concentration in ELISA and mRNA expression. GHJ had inhibitory effects in level of MAPKs, $p-I{\kappa}B-{\alpha}$ and p-NF-kB also. GHJ attenuated Compound 48/80-stimulated histamine release. In addition, GHJ inhibited PCA reaction in vivo. Conclusion : This study indicated that GHJ extract can inhibit allergic responses in HMC-1 cell.

Anti-inflammatory and Antioxidant Activities of Ulva lactuca Linnaeus Extract (참갈파래 추출물의 항염증 및 항산화 활성)

  • The anti-inflammatory and antioxidant activities of the Ulva lactuca Linnaeus extracts were measured. As a result of the MTT assay of the extract, it was found that the cell viabilities were higher than 90% at all concentrations, and the cell promotion rate exceeded 100% at a certain concentration. Therefore, in all subsequent experiments, the highest concentration was set at 500 ㎍/ml. In the nitric oxide (NO) assay, one of the anti-inflammatory experiments, the Ulva lactuca Linnaeus extracts exhibited inhibitory effects of 16%, 19%, and 62% at concentrations of 5, 50, and 500 ㎍/ml, respectively, compared with the LPS-only group. In tumor necrosis factor-α (TNF-α) assay, the extracts showed inhibitory effects of 16%, 17%, and 27% at concentrations of 5, 50, and 500 ㎍/ml, respectively, compared with the LPS-only group. In the interleukin-6 (IL-6) assay, the extracts also showed inhibitory effects of 15% and 28% at concentrations of 50 and 500 ㎍/ml, respectively, compared with the LPS-only group. In the total polyphenol content assay, the extracts exhibited 8.02, 9.90, and 23.8 mg GAE/g at concentrations of 0.3, 3, and 30 mg/ml, respectively. In the ABTS assay, the extracts exhibited scavenging activities of 2.6, 12.6, and 77.3% at concentrations of 0.3, 3, and 30 mg/ml, respectively. Therefore, it is considered that Ulva lactuca Linnaeus extracts can be used as anti-inflammatory and antioxidant substances for the development of functional foods.

Effects of Seokgokamibokhapbang(SGBHB) on anti-CD40-and recombinant IL-4-induced cytokine production and immunoglobulin E and histanine release in highly purified mouse B cells (석고가미복합방이 Anti-CD40과 rIL-4로 유도된 생쥐의 B 세포에서 싸이토카인 생성 및 면역글로블린 E에 미치는 효과)

  • In the present study, we exarnined anti-allergic effect of SGBHB in cultured B cells. B cells were prepared from isolated murine splenocytes and activated by co-treatment of anti-CD40 monoclonal antibody and recombinant IL-4 allergens. Anti-allergic effects of SGBHB in activated B cells were determined by measuring B cell surface activated molecules (CD23+ and CD11a+), and expression levels of IL-$1{\beta}$, IL-6, IL-10, TNF-$\alpha$, IgE, and HRF. The major findings are summarized as follows. 1. SGBHB treatment did not produce significant cytotoxic effects on mouse lung fibroblast cells. 2. SGBHB produced significant inhibitory effect on the expression of B cell surface activated molecules (CD23+ and CD11a) in activated B cells. 3. SGBHB treatment significantly inhibited expression levels of IL-$1{\beta}$, IL-6, and TNF-$\alpha$ mRNAs in activated B cells.IL-6 protein levels were significantly decreased by $100{\mu}g/m{\ell}$ of SGBHB treatrrient, and TNF-$\alpha$ protein levels were decreased compared to the control group, but statistically insignificant. 4. SGBHB treatment significantly increased IL-10 at both mRNA and protein levels in activated B cells. 5. SGBHB treatment significantly inhibited levels of IgE production. Thus, the present data suggest that SGBHB has an anti-allergic effect on activated B cells by controlling irnmune responses, and further implicates the possibility on clinical application as a therapeutic agent.

Effects of Chinemys reevesii on Lipopolysaccaride-indused Inflammatory Reactions (구판의 LPS로 유도된 염증 반응 억제 효과)

  • This study is to investigate the effects of Chinemys reevesii (CR) on allergic inflammation mechanism related chronic dermatitis. To investigate the effects of CR, we study inhibitory effect of CR on the levels of pro-inflammatory cytokines released from RAW 264.7 cell stimulated with lipopolysaccaride (LPS), and EoL-1, THP-1, Jutkat cell stimulated with Dermatophagoides pteronyssinus (DP), and LPS induced acute inflammatory BALB/c mouse model. CR reduced the levels of IL-$1{\beta}$ released from RAW 264.7 cell stimulated with LPS at 20 ug/ml, 10 ug/ml concentration. CR significantly reduced the levels of MCP-1 released from EoL-1 cell, IL-6 from THP-1 cell, and IL-4, IL-5, TNF-${\alpha}$ from Jutkat cell stimulated with DP at all the concentration. CR significantly reduced the levels of TNF-${\alpha}$, IL-6, IL-$1{\beta}$, in LPS induced inflammatory BALB/c mouse model, in a dose-dependent manner. These results suggested that CR has suppressive effects on pro-inflammatory cytokines in various inflammation related cell lines through the regulation of immune system. CR could be a therapeutic agent for treatment of chronic inflammatory dermatitis in the future.

Anti-allergic Effects of Samhwangsasim-tang ($S{\bar{a}}nhu{\acute{a}}ngxi{\grave{e}}x{\bar{i}}nt{\bar{a}}ng$) on Ovalbumin-induced Allergic Model in Mice (삼황사심탕(三黃瀉心湯)이 난황 알부민으로 유도된 알레르기 Mouse모델에서 항알레르기 효과)

  • Objectives In this study, we investigated the inhibitory effects of Samhwangsasim-tang (S.H) on the allergic response caused by ovalbumin(OVA) sensitization and challenge in BALB/c mice. Methods The experimental animals were divided into five groups; 1) normal as negative control, 2) OVA-sensitized mice, 3) OVA-sensitized mice treated with 200 mg/kg of S.H 200, 4) OVA-sensitized mice treated with 400 mg/kg of S.H 400, and 5) OVA-sensitized mice treated with 5 mg/kg of Dexamethasone (Dex). Antigen sensitization for allergic mouse model was performed with twice injection of OVA for 2 weeks. After secondary injection, S.H was administrated orally into mice every day for 13 days and the inhibitory effect of S.H on allergic responses was evaluated. Results Treatment of S.H into allergic mice reduced significantly ear edema and infiltration of immune cells in ear tissues induced with OVA challenge in a dose-dependent manner. S.H reduced significantly the serum levels of Total Immunoglobulin(Ig)G and IgE, and particularly inhibited the production of OVA-specific IgE, but not OVA-specific IgG. The serum level of proinflammatory cytokine TNF-${\alpha}$ and Th2-associated cytokine IL-4 also were significantly decreased by S.H adminstration in a dose denpendent manner. S.H attenuated OVA-induced secretion of IFN-${\gamma}$, but not IL-12 which is a cytokine inducing the development of Th1 cells. It also reduced significantly the secretion of IL-4, which is a cytokine inducing the development of Th2 cells, after splenocytes were stimulated with OVA. However the secretion of IL-5 and IL-13 was influenced weakly or a little. Conclusions These results indicate that S.H could reduce the allergic response through inhibition of antigen-specific IgE and Th2-inducing cytokines. It suggest that S.H may be available clinically for the treatment of allergic patients.

Experimental Study on the Anti-allergic Effects of Gamibojungikgi-tang (가미보중익기탕이 알레르기반응에 미치는 영향에 대한 실험적 연구)

  • This experimental research has been done to study the effects of Gmibojungikgi-tang(GBJIKT) on the anti-allergic reaction. We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of GBJIKT on hypersensitivity response to mice. The results obtained from our research are as following: The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand, the survival rates of the other group to which we injected both of the compound 48/80 and GBJIKT are 10%, 10%, 20%, 30%, 20%, and 40% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1(mg/g) of compound 48/80. Time dependency test also shows the 0% survival rates in 5 and 10 minutes. GBJIKT revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. GBJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to picry1 chloride GBJIKT showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood cell. Our research provides the important evidence that GBJIKT is benificial to the prevention and treatment of allergy related diseases.

Inhibitory Effects of Sulfur-Fed Duck Extracts on DSS-induced Colitis in Mouse (DSS(Dextran Sulfate Sodium)로 유도된 마우스의 대장염에 대한 유황오리 추출물의 항염증 효과)

  • Anti-inflammatory effects of sulfur-fed duck extract on colitis induced by 2.5% dextran sulfate sodium (DSS) were examined in male Balb/c mice. Animals were divided into eight groups: normal (0.1 mL of PBS without 2.5% DSS), control (0.1 mL of PBS with 2.5% DSS), SD-H (3 mL/kg of high sulfur-fed duck extract), SD-L (1 mL/kg of low sulfur-fed duck extract), GD-H (3 mL/kg of high general duck extract), GD-L (1 mL/kg of low general duck extract), GC-H (3 mL/kg of high general chicken extract), and GC-L (1 mL/kg of low general chicken extract). Mice were fed PBS or six different doses of extracts (sulfur-fed duck, general duck, and chicken), once daily for 14 days. Colitis was induced from day 7 to 14 via the administration of 2.5% DSS in drinking water. The colon length was significantly shortened in mice compared to the control group. The administration of SD-H, SD-L, and GD-L increased colon length and decreased histological colon injury from DSS-induced colitis. However, chicken extracts did not recover any clinical sign of the colitis. SD-L significantly suppressed not only the concentrations of IL-$1{\beta}$, IL-6, TNF-${\alpha}$, IL-17A, and IL-12 in serum but also the mRNA expressions of IL-6, TNF-${\alpha}$, iNOS and COX-2 in DSS-treated colon tissues (P<0.05). The administration of SD-H suppressed the concentrations of IL-6 in serum and the mRNA expressions of IL-6, iNOS, and COX-2 in colon tissues. Administration of GD-L suppressed the concentrations of IL-6, TNF-${\alpha}$, and IL-17A in serum and the mRNA expressions of IL-6, iNOS, and COX-2 in colon tissues. The inhibitory effects of sulfur-fed duck extracts were effective at a dose of 1 mL/kg. Our results indicate that sulfur-fed duck extracts may possess anti-inflammatory effects on DSS-induced colitis mice.
