Characterization and Cytotoxic Activities of Nonadecanoic Acid Produced by Streptomyces scabiei subsp. chosunensis M0137 (KCTC 9927)

  • The substance 0116p, which exhibits cytotoxicity against human macrophage cell line THP-1, was isolated from a mycelial extract of Streptomyces scabiei subsp. chosunensis M0137. The cytotoxic substance was purified by Diaion-HP2O adsorption, solvent extraction, Sephadex LH-20 column chromatography, and silica-gel column chromatography. The molecular formula is C$\_$19/H$\_$38/O$\_$2/ (MW301.10) based on elemental and spectrometric analysis. It was identified as nonadecanoic acid by NMR spectral data. It exhibits cytotoxic activities in various human cancer cell lines, including A549, SK-OV-3, SK-MEL-2 and HCT-15. In addition, 0116p also inhibits IL-12 production in lipopolysaccharide-activated macrophages.

  • In this paper, a design for the 900MHz bandwidth RFID reader antenna with circular polarization is proposed and experimentally investigated. The 900MHz bandwidth RFID reader antenna is proposed as a simple radiator with loading capacitor to generate LHCP(Left Hand Circular Polarization). Also, the design and fabrication of antenna has the returnloss of -32.28dB at the center frequency of 1010MHz and Bandwidth of 12.5% at 905Hz~1030MHz. Proposed LHCP antenna of maximum gain is 6dBi and satisfy axial ratio based on 2. From the measured result, axial ratios based on 2 are observed at the operating frequencies. The proposed antenna is suitable for RFID applications in wireless communications.

The Effects of Ikwitang Extract on Indomethacin-induced Gastric Mucosal Lesions (익위탕(益胃湯)의 Indomethacin유발 위점막 손상에 대한 효과)

  • This study was carried out to investigate the effects of Ikwitang extract on indomethacin-induced gastric mucosal lesions in mice. Experimental groups were classified into a non-treated group(NM group), a non-administered group(CON group), the Misoprostol-administered group(MI group) and the Ikwitang-extract-administered group(IW group). This study examined the morphological change, distribution of mast cells, mucous surface cells, acid mucose secreted cells, and apoptic cells, BrdU, COX-1, Hsp70, $NF-{\kappa}B\;p50$, PKC, COX-2 and IL-12B of gastric mucosa. Results : The results of this study were as follows: 1. In the Misoprostol-administered group and the Ikwitang-extract-administered group, the hemorrhagic erosion of gastric mucous and infiltrated mast cells decreased. 2. Mucous surface cells and acid mucose secreted cells abserved in the Misoprostol-administered group and the Ikwitang-extract administered group. 3. The distribution of apoptic cells, Hsp70, $NF-{\kappa}B\;p50$, PKC, COX-2, and IL-12B increased in the Control group, but decreased in the Misoprostol-administered group and Ikwitang-extract-administered group. 4. Cells proliferation of gastric mucosa and the COX-l positive cells decreased in the Control group, but increased in the Misoprostol-administered group and the Ikwitang-extract -administered group. The above results suggest the lkwitang extract had beneficial effects on indomethacin induced gastric mucosal lesions.

Effects of combined therapy with thalidomide and glucantime on leishmaniasis induced by Leishmania major in BALB/c mice

  • For treating Leishmania major infection in BALB/c mice, we used thalidomide in conjunction with glucantime. Groups of mice were challenged with $5{\time}10^3$ metacyclic promastigotes of L. major subcutaneously. A week after the challenge, drug treatment was started and continued for 12 days. Thalidomide was orally administrated 30 mg/kg/day and glucantime was administrated intraperitoneally (200 mg/kg/day). It was shown that the combined therapy is more effective than single therapies with each one of the drugs since the foot pad swelling in the group of mice received thalidomide and glucantime was significantly decreased (0.9 :t 0.2 mm) compared to mice treated with either glucantime, thalidomide, or carrier alone (1.2 :t 0.25, 1.4 :t 0.3, and 1.7 :t 0.27 mm, respectively). Cytokine study showed that the effect of thalidomide was not dependent on IL-12; however, it up-regulated IFN-yand down-regulated IL-10 production. Conclusively, thalidomide seems promising as a conjunctive therapy with antimony in murine model of visceral leishmaniasis.

Inhibitory Effect on TNF-${\alpha}$-Induced IL-8 Production in the HT29 Cell of Constituents from the Leaf and Stem of Weigela subsessilis

  • Twelve compounds were isolated from the MeOH extract of the leaf and stem of the Korean endemic plant Weigela subsessilis L. H. Bailey. Their chemical structures were elucidated on the basis of physicochemical and spectroscopic data and by comparison with those of published literatures. These compounds were identified as three sterols, ${\beta}$-sitosterol acetate (2), ${\beta}$-sitosterol (3), daucosterol (11), eight triterpenoids, squalene (1), ursolic acid (4), ilekudinol A (5), corosolic acid (6), ilekudinol B (7), esculentic acid (8), pomolic acid (9), asiatic acid (10), and one iridoid glycoside, alboside I (12). This is the first report pertaining to the isolation of these compounds from Weigela subsessilis L. H. Bailey. In addition, three compounds 7, 9, and 12 were found to display a strong inhibitory effect on the production of IL-8 in the HT29 cells stimulated by TNF-${\alpha}$.

The Characteristics of Ga-doped ZnO Transparent Thin Films by using Multilayer (다층박막을 이용한 Ga-doped ZnO 투명전도막의 특성)

  • With development of electronic products the demands for miniaturization and weight-lightening have increased until a recent date. Accordingly, The effort to substitute glass substrates was widely made. However, polymer substrates have weak point that substrates were damaged at high temperature. In this paper, we deposited transparent conductive film at low temperature. And we inserted Au thin film between oxide to compensate for deteriorated electrical characteristics. Ga-doped ZnO(GZO) multilayer coatings were deposited on glass substrate by DC sputtering. The optimization of deposition conditions of both AZO and Au layers were performed to obtain better electrical and optical characteristics in advance. We presumed that the properties of multilayer were affected by the deposition process of both GZO and Au layers. The best multilayer coating exhibited the resistivity of $2.72{\times}10^{-3}\;{\Omega}-cm$ and transmittance of 77 %. From these results, we can confirm a possibility of the application as transparent conductive electrodes.

Ionic Additives to Increase Electrochemical Utilization of Sulfur Cathode for Li-S Batteries

  • The high theoretical specific capacity of lithium-sulfur (Li-S) batteries makes them a more promising energy storage system than conventional lithium-ion batteries (LIBs). However, the slow kinetics of the electrochemical conversion reaction seriously hinders the utilization of Li-S as an active battery material and has prevented the successful application of Li-S cells. Therefore, exploration of alternatives that can overcome the sluggish electrochemical reaction is necessary to increase the performance of Li-S batteries. In this work, an ionic liquid (IL) is proposed as a functional additive to promote the electrochemical reactivity of the Li-S cell. The sluggish electrochemical reaction is mainly caused by precipitation of low-order polysulfide (l-PS) onto the positive electrode, so the IL is adopted as a solubilizer to remove the precipitated l-PS from the positive electrode to promote additional electron transfer reactions. The ILs effectively dissolve l-PS and greatly improve the electrochemical performance by allowing greater utilization of l-PS, which results in a higher initial specific capacity, together with a moderate retention rate. The results presented here confirmed that the use of an IL as an additive is quite effective at enhancing the overall performance of the Li-S cell and this understanding will enable the construction of highly efficient Li-S batteries.

Immunomodulatory and anti-metastatic activities of polysaccharide isolated from red cabbage (적양배추에서 분리한 다당의 면역 및 항전이 활성)

  • In this study, we examined the immunostimulating characteristics of a hot water extract (RCW) and crude polysaccharides (RCP) of red cabbage. RCW and RCP did not show any cytotoxicity in B16BL6 cells and macrophages. Although the sugar compositions of RCW and RCP were similar, the uronic acid content of RCP was higher than that of RCW RCP significantly increased the production of various cytokines and NO, whereas RCW did not affect the production of cytokines and NO. In an ex vivo assay of natural killer (NK) cell activity, intravenous (i.v.) administration of RCP significantly augmented NK cytotoxicity against Yac-1 tumor cells at 3 days after RCP treatment. In an experimental lung metastasis model using B16BL6 melanoma cells, i.v. administration of RCP at a dose of $1,000{\mu}g$ per mouse significantly inhibited 47.3% of lung metastasis. These results suggest that crude polysaccharide isolated from red cabbage is a promising food ingredient for the prevention of tumor metastasis.

Screening of Lactobacilli Derived from Chicken Feces and Partial Characterization of Lactobacillus acidophilus A12 as Animal Probiotics

  • This study was performed to screen and select Lactobacillus strains from chicken feces for probiotic use in animals. Of these strains, strain AU had the highest immunostimulatory effect. Therefore, strain A12 was characterized as a potential probiotic. Strain A12 was tentatively identified as Lactobacillus acidophilus A12, using the API 50 CHL kit based on a 99.9% homology. L. acidophilus A12 was highly resistant to artificial gastric juice (pH 2.5) and bile acid (oxgall). Based on results from the API ZYM kit, leucine arylamidase, crystine arylamidase, acid phosphatase, naphthol-AS-BI-phosphohydrolase, ${\alpha}$-galactosidase, ${\beta}$-galactosidase, ${\alpha}$-glucosidase, ${\beta}$-glucosidase, and N-acetyl-${\beta}$-glucosamidase were produced by strain A12. L. acidophilus A12 showed resistance to several antibiotics (nisin, gentamicin, and erythromycin). The amount of interleukin $(IL)-1{\alpha}$ in $20{\times}$ concentrated supernatant from L. acidophilus A12 was approximately 156pg/ml. With regard to antioxidant activity, L. acidophilus A12 supernatant showed 60.6% DPPH radical scavenging activity. These results demonstrate the potential use of L. acidophilus A12 as health-promoting probiotics.

Effects of Coptidis Rhizoma on the Change of Interleukin-6 and $TNF-{\alpha}$ Level induced by LPS I.C.V. Injection in Mice (황연(黃連)이 Lipopolysaccharide 뇌실 주입으로 유발된 생쥐의 IL-6와 $TNF-{\alpha}$ 변화에 미치는 영향)

  • Objective: This study was conducted to investigate the effects of Coptidis Rhizoma on the plasma IL-6 and $TNF-{\alpha}$ level in mice by intracerebroventricular(I.C.V.) injection of Lipopolysaccharide (LPS). Method: 6 mice were assigned to each of the Normal group, the Control group, and the individual Experimental groups. In the Normal group only saline was administered intragastrically, and in the Control group LPS was injected intracerebroventricularly 1 hr after intragastric administration of saline. In the Experimental groups Coptidis Rhizoma(0.5g/kg, 1.0g/kg, 3.0g/kg) was administered intragastrically to mice 1 hr prior to LPS (100ng/mouse) I.C.V. Injection. To measure the plasma IL-6 and $TNF-{\alpha}$ level of mice, their blood samples were collected from retro-orbital venous plexus, immediately centrifuged at $4^{\circ}C$, and plasma was removed and stored frozen at $-83^{\circ}C$ for later determination of plasma IL-6 and $TNF-{\alpha}$. Result: 1. LPS I.C.V. Injection increased plasma IL-6 level significantly in a dose-dependent manner compared with Normal group. (P<0.01) The plasma IL-6 concentration reached a significant maximal level about 1 hr after LPS(100ng/mouse) I.C.V. Injection.(P<0.001) 2. Both the 0.5g/kg(Sample A) and 1.0g/kg(Sample B) groups to which Coptidis Rhizoma was administered intragastrically 1 hr prior to LPS(100ng/mouse) I.C.V. Injection showed insignificant lower plama IL-6 level in 1 hr than Control group(P>0.05), and 3.0g/kg group(Sample C) conversely showed higher plama IL-6 level than Control group. 3. LPS I.C.V. Injection increased plasma $TNF-{\alpha}$ level significantly in a dose-dependent manner compared with Normal group.(P<0.05) The plasma $TNF-{\alpha}$ concentration reached a significant maximal level about 1 hr after LPS(100ng/mouse) I.C.V. Injection.(P<0.001) 4. All Sample groups(0.5g/kg, 1.0g/kg, and 3.0g/kg) to which Coptidis Rhizoma was administered intragastrically with each constituent-dose 1 hr prior to LPS(100ng/mouse) I.C.V. Injection showed significant lower $TNF-{\alpha}$ plama level in 1 hr than Control group.(P<0.001) These data revealed that Coptidis Rhizoma might have anti inflammatory effect by reducing the plasma $TNF-{\alpha}$ level in a dose dependent manner in mice LPS I.C.V. Injection.
