인진청간탕이 C형 간염 바이러스의 임파구계 세포감염에 미치는 영향 (The Effect of Injinchunggan-tang on Hepatitis C Virus Infection)

  • Objective : The aim of this study was to evaluate the efficacy of Injinchunggan-tang on Hepatitis C virus infection, and to clarify the mechanism of treatment by indentifying the effect of Injinchunggan-tang on cytokine secretion. Methods : In vitro model of HCV infection in MOLT 4 cell was used. The effect of Injinchunggan-tang on the attachment of HCV on MOLT 4 cell was studied by PCR method. The change of cytokine secretion according to Injinchunggan-tang treatment was investigated by ELISA. Results : Injinchunggan-tang inhibited the attachment of HCV on MOLT 4 in the concentration of $10-2{\mu\textrm{g}}/\mu\textrm{\ell}$ and $10-1{\mu\textrm{g}}/\mu\textrm{\ell}$. In cytokine assay, Injinchunggan-tang increased the secretion of IL-4 of mouse splenocytes and PBMC in 48 hour culture as well as the secretion of IL-12 of mouse splenocytes and PBMC in 48 hour culture, whereas it decreased the secretion of $IFN-{\gamma}$ of mouse splenocytes in 24 and 48 hour culture. Conclusion : The results of this study show that Injinchunggan-tang has an inhibitory effect on the attachment on HCV on Mo1t4 Cell, and that it increases the secretion of IL-4 and IL-12 of mouse splenocyte and PBMC.

Immunomodulatory Activity of Pine Needle (Pinus densiflora) Extracts in Macrophages

  • Pinus densiflora belongs to the Pinaceae family which has been widely used for health promoting purposes as folk medicine or as a food. Various curative effects of different parts of the pine have been reported including as a remedy for carcinoma. We examined the effects of pine needle water extracts (PNE) on macrophage function using peritoneal macrophage, pre-osteoclast bone macrophage (Raw 264.7 cell) and brain macrophage (C6 microglia). When peritoneal macrophages were treated with various concentrations of PNE ($1{\sim}100{\mu}g/mL$) for 24 hours, phagocytic activity was significantly increased, whereas it had no effect on tumoricidal activity and NO production. However, the treatment of Raw 264.7 with PNE resulted in the enhancement of NO production at high concentration ($100{\mu}g/mL$). Furthermore, the treatment of C6 with PNE increased the production of NO in a concentration-dependent manner, whereas PNE suppressed NO production in $LPS/IFN-{\gamma}-stimulated$ microglia. These results suggest that PNE has differential immunomodulatory effects on macrophages.

Rhei Rhizoma Extracts Have Antiproliferative Properties and Differential Effects on NO Production in Macrophages

  • Recently, Rhei Rhizoma extracts (RRE) have begun to receive more attention as potential biological response modifiers. In the present study, we studied the antiproliferative effect of RRE on tumor cells and the effect of RRE on macrophage function. A variety of tumor cells and macrophages were treated with RRE at various concentrations. The effect of RRE on cell proliferation was measured by MTT assay and the effect of RRE on the production of nitric oxide (NO) was determined in the macrophage-like cell lines Raw264.7, C6 and peritoneal macrophages (pMQ). RRE inhibited the growth of tumor cells (e.g., B16, HOS). However, the effects of RRE on the production of NO varied with macrophage types. RRE had no effect on C6 cell growth and slightly increased the growth of Raw264.7 cells. In addition, treatment of normal pMQ with RRE enhanced NO production in a concentration-dependent manner, whereas RRE suppressed NO production at $50\;{\mu}g/mL$ in both Raw264.7 and C6 cells. However, RRE suppressed NO production in LPS/IFN-$\gamma$-stimulated C6 cells. Overall, these results suggest that RRE elicits an antiproliferative property and differentially modulates NO production in various macrophages, and have a potential for therapeutic application.

  • This study was carried out to know the immunity responses of Sogyughwalhyel-tanggami(SGHHT) to on Rheumatoid Arthritis in Collagen-induced Arthritis(CIA) Mice. Various experimental were peformed to analyse the immunity effects of SGHHT. The cytotoxicity against mLFCs was not measured. The production of pro-inflammatory cytokines $IL-1{\beta}$, IL-6, $TNF-\alpha$ were reduced in hFLSs. The production of pro-inflammatory cytokines $IFN-\gamma$, IgG3, IgG2b, IgM were reduced. Comparison of the results for this study showed that SGHHT had immunomodulatory effects of suppressing or enhancing. So we expect that SGHHT should be used as a effective drugs for not only rheumatoid arthritis but also another auto-immune disease. Therefore we have to survey continuously in looking for the effective substance and mechanism in the future.

십전대보탕의 면역활성에 관한 기초 실험 연구 문헌 분석 (Analysis of Biological Experiment on Immunoactivity of Sipjeondabo-tang)

  • To investigate scientific evidence for the use of a Korean Medicine (KM), the papers regarding Sipjeondabo-tang (Shiquandabu-tang) frequently used in Korean medical clinics and hospitals were collected and analyzed. The papers were classified as being from domestic or international journals and by the year of publication. The mechanisms of immuno-activity Sipjeondabo-tang (Shiquandabu-tang) were researched. Among 41 papers, 4 were published as Korean papers whereas 7 were as Chinese papers and 40 were as Japanese papers. Sipjeondabo-tang (Shiquandabu-tang) regulated the productions of Th1 cytokines such as interleukin-2 (IL-2) and interferon-${\gamma}$ (IFN-${\gamma}$), and Th2 cytokines such as IL-4, IL-5, IL-6, IL-12 improving the function of immune organ including bone marrow, spleen, liver, thymus and peyer's patch. It also modulated the releases of immunoglobulin-G (IgG) and IgM. Thus, Sipjeondabo-tang (Shiquandabu-tang) balanced the Th1/Th2 immune responses as well as T/B cell. The regulation of immunoactivity could be speculated as an objective and scientific evidence of Sipjeondabo-tang (Shiquandabu-tang).

형방패독산(荊防敗毒散)이 DNFB로 유발된 생쥐의 알레르기접촉피부염에 미치는 영향 (Effects of Hyeongbangpaedok-san (HBPDS) on Allergic Contact Dermatitis (ACD) induced by DNFB in Mice)

  • Objectives The present study was carried out to investigate effects of HBPDS on allergic contact dermatitis (ACD) induced by 2,4-Dinitro-1-fluorobenzene (DNFB) in mice. Methods In this experiment, effects of HBPDS on body weights, skin thicknesses, skin weights, histopathological changes, clinical aspects, erythema index, melanin index, production levels of cytokines in ACD mice were investigated. In addition, effects on proliferation rates, release of ${\beta}$-hexosaminidase and histamine were also investigated in vitro. Results & Conclusions 1) HBPDS inhibited enlargement of skin thickness and weight significantly (P < 0.05). 2) HBPDS treatment prevented spongiosis, edema and immune cell infiltrations. 3) Erythema, desquamation and keratosis were diminished by oral administration of HBPDS. 4) Production levels of TNF-alpha and IFN-gamma in serum were decreased by HBPDS treatment in vivo. 5) More than 200 ${\mu}g/ml$ of HBPDS treatment decreased ${\beta}$-hexosaminidase release and more than 400 ${\mu}g/ml$ of HBPDS treatment also decreased histamine release in vitro.

강황이 난황의 비강내 점적을 통하여 유발된 생쥐의 천식에 미치는 영향 (Effects of Curcuma longa Rhizoma on Asthma induced intra-nasal instillation of Ovalbumin in Mice)

  • Objective : This study was designed to investigate the effects of Curcuma longa Rhizoma(CLR) on asthma. Methods : Detecting splenocyte proliferation rates, cytokines and antigen specific antibody isotypes in bronchoalveolar lavage fluid (BALF). In addition, the present author I also investigated changes in spleen and histopathological changes of lung tissues. Results : Oral administration of CLR lowered spleen weight and splenocyte proliferation rates. In addition, levels of IL-4, IL-17A and Granulocyte/Macrophage Colony-Stimulating Factor(GM-CSF), Th2 driven cytokines, were lowered respectively and IFN-g, and Th1 driven cytokine, were elevated by CLR. Levels of Ovalbumin(OVA) specific IgE and IgGl in BALF were also lowered by oral administration of CLR too. Conclusion : CLR is useful to treat patients with asthma and the mechanisms are related to the in suppression of Th2 skewing reactions.

호장으로부터 분리한 스틸벤류의 Nitric Oxide 저해활성 (Inhibition of Nitric Oxide Production by Stilbenes from Polygonum cuspidatum)

  • Polygonum cuspidatum which is widely distributed in Korea has been used as treatments of dermatitis, gonorrhea, favus athlete's foot, hyperlipidemia and inflammation in traditional medicine. We examined anti-oxidant and antiinflammatory activity by measuring DPPH radical scavenging activity and the inhibition of IFN-${\gamma}$ and LPS-induced NO production in RAW 264.7 murine macrophage cells. We isolated and characterized resveratrol (1), trans-resveratrol 3-O-${\beta}$-Dglucopyranoside (2). Compounds 1 and 2 showed potent activities compared with L-NMMA ($N^G$-monomethyl-L-arginine). These results suggested that the stilbene compounds isolated from Polygonum cuspidatum might be used as antiinflammatory agents.

Immunization of Mice with Recombinant Brucella abortus Organic Hydroperoxide Resistance (Ohr) Protein Protects Against a Virulent Brucella abortus 544 Infection

  • In this study, the Brucella abortus ohr gene coding for an organic hydroperoxide resistance protein (Ohr) was cloned into a maltose fusion protein expression system (pMAL), inserted into Escherichia coli, and purified, and its immunogenicity was evaluated by western blot analysis using Brucella-positive mouse sera. The purified recombinant Ohr (rOhr) was treated with adjuvant and injected intraperitoneally into BALB/c mice. A protective immune response analysis revealed that rOhr induced a significant increase in both the IgG1 and IgG2a titers, and IgG2a reached a higher level than IgG1 after the second and third immunizations. Additionally, immunization with rOhr induced high production of IFN-γ as well as proinflammatory cytokines such as TNF, MCP-1, IL-12p70, and IL-6, but a lesser amount of IL-10, suggesting that rOhr predominantly elicited a cell-mediated immune response. In addition, immunization with rOhr caused a significantly higher degree of protection against a virulent B. abortus infection compared with a positive control group consisting of mice immunized with maltose-binding protein. These findings showed that B. abortus rOhr was able to induce both humoral and cell-mediated immunity in mice, which suggested that this recombinant protein could be a potential vaccine candidate for animal brucellosis.

Prophylactic and Therapeutic Potential of Asp f1 Epitopes in Naive and Sensitized BALB/c Mice

  • Background: The present study examines a hypothesis that short allergen-derived peptides may shift an Aspergillus fumigatus (Afu-) specific TH2 response towards a protective TH1. Five overlapping peptides (P1-P5) derived from Asp f1, a major allergen/antigen of Afu, were evaluated for prophylactic or therapeutic efficacy in BALB/c mice. Methods: To evaluate the prophylactic efficacy, peptides were intranasally administered to naive mice and challenged with Afu-allergens/antigens. For evaluation of therapeutic efficacy, the mice were sensitized with Afu-allergens/antigens followed by intranasal administration of peptides. The groups were compared for the levels of Afu-specific antibodies in sera and splenic cytokines evaluated by ELISA. Eosinophil peroxidase activity was examined in the lung cell suspensions and lung inflammation was assessed by histopathogy. Results: Peptides P1-, P2- and P3 decreased Afu-specific IgE (84.5~98.9%) and IgG antibodies (45.7~71.6%) in comparison with Afu-sensitized mice prophylactically. P1- and P2-treated ABPA mice showed decline in Afu-specific IgE (76.4~88%) and IgG antibodies (15~54%). Increased IgG2a/IgG1 and IFN-${\gamma}$/IL-4 ratios were observed. P1-P3 prophylactically and P1 therapeutically decreased IL-5 levels and eosinophil peroxidase activity. P1 decreased inflammatory cells' infiltration in lung tissue comparable to non-challenged control. Conclusion: Asp f1-derived peptide P1, prophylactically and therapeutically administered to Balb/c mice, is effective in regulating allergic response to allergens/antigens of Afu, and may be explored for immunotherapy of allergic aspergillosis in humans.
