Inhibition of $\gamma$-Irradiation Induced Adhesion Molecules and NO Production by Alginate in Human Endothelial Cells

  • Inflammation is a frequent radiation-induced reaction following therapeutic irradiation. Treatment of human umbilical endothelial cells (HUVEC) with $\gamma$-irradiation ($\gamma$IR) induces the expression of adhesion proteins such as intercellular adhesion molecule-1 (VCAM-1 ), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. Since the upregulation of these proteins on endothelial cell surface has been known to be associated with inflammation, interioring with the expression of adhesion molecules is an important therapeutic target. In the present study, we demonstrate that high mannronic acid-containing alginate (HMA) inhibits $\gamma$IR induced expression of ICAM-1, VCAM-1, and E-selectin on HUVEC in a dose dependent manner. HMA also inhibited $\gamma$IR induced production of Nitric oxide (NO). These data suggest that HMA has therapeutic potential for the treatment of various inflammatory disorder associated with an increase of endothelial leukocyte adhesion molecules.

Indomethacin으로 유발된 생쥐의 위점막 손상에 대한 평진탕의 효과 (Effects of Pyungjintang on Indomethacin-induced Gastric Mucosal Lesions in Mice)

  • Objectives : This study was carried out to investigate the effects of Pyungjintang on indomethacin-induced gastric mucosal lesions of mire. Methods : Experimental mice were classified into not-treated group (NOR group), gastro-inflammation elicitated group (CON group), misoprostol-administered group after gastro-inflammation elicitation (MA group), and Pyungjintang-administered group after gastro-inflammation elicitation (PA group). This study examined the morphological change, distribution of mast cells, mucus surface cells, neutral mucus secreting cells, acid mucus secreting cells, PNA reaction, angiogenesis (MIP-2), COX-1, Hsp70, NF-kB p50, COX-2IL-12B, ICAM-1, BrdU and apoptotic cells of gastric mucosa. Results : 1. The scars of diapedesis, dilatation of right gastric artery and the hemorrhagic erosions of gastric mucosa were reduced in the MA and PA groups. 2. Gastric perforation was observed in the gastro-inflammation elicitated group, but not in the MA and PA groups. 3. The COX-1 positive cellsl, cell proliferation of gastric mucosa, neutral mucus secreting ce31s, acid mucus secreting cells and PNA positive reaction of surface mucus cells were increased in the MA and PA groups. 4. The distribution of apoptotic cells, mast cells, MIP-2, Hsp70, NF-kB p50, COX-2, IL-l2B and ICAM-1 were decreased in the MA and PA groups. Conclusions : Pyungjintang had excellent effects on indomethacin-induced gastric mucosal lesions in mice.

거풍제습탕(祛風除濕湯)의 류마토이드 관절염 치료효과에 대한 실험적 연구 (Study on the Effect of Geopungjeseup-tang in Rheumatoid Arthritis)

  • This study was carried out to know the effect of Geopungjeseup-tang(GPJST) to on Rheumatoid Arthritis by using human fibroblast-like synoviocytes(hFLS). GPJST consists of Coisis Semen, Coccllus Trilobus DC., Lycopi Herba, Arisaematis Rhizoma. The author peformed several experimetal items : that is cytotoxicity of GPJST, mRNA expression of TNF-${\alpha}$, IL-1${\beta}$, ICAM-1, production of NO. The results were obtained as follows : The mRNA expression of pro-inflammatory cytokines TNF-${\alpha}$, IL-1${\beta}$, ICAM-1 were dose-dependently reduced in hFLS. The production of NO was reduced. Talking all these observations into account, GPJST is considered to be effective in treating rheumatoid arthritis. Therefore we have to survey continuously in looking for the effective substance and mechanism in the future.

A Role of Mitogen Activated Protein Kinases and Inflammatory Responses in Gender Differences in Kidney Ischemia Injury

  • It is not known whether gender differences play a role in susceptibility to ischemic acute renal failure. Thus, we examined if there were any differences in susceptibility between male and female mice to kidney ischemic injury, and if so, whether it is due to differences in mitogen activated protein kinases (MAPKs) or inflammatory responses to ischemia. Female mice were protected against kidney ischemia when compared with males. Thirty minutes of bilateral ischemia resulted in marked functional and morphological damages in males, but not in females. The ischemia-induced phosphorylation of c-jun N-terminal stress-activated protein kinases (JNKs) was higher in males than in females. Phosphorylation of extracellular signal-regulated kinases (ERKs) was lower in males than in females. Post- ischemia medullary infiltration of RAW 264.7 cell, a monocyte-macrophage cell, and intercellular adhesion molecule-1 (ICAM-1) were greater in males than in females. In conclusion, males were much more susceptible to ischemia than females. The enhanced propensity to ischemic injury in males was correlated with greater activation of JNKs, greater expression of ICAM-1, and greater trapping of leukocytes in the medulla.

홍삼과 발효홍삼의 항염증 작용 및 항알러지 효과 비교 (The comparative study of anti-allergic and anti-inflammatory effects by fermented red ginseng and red ginseng)

  • 홍삼은 인삼을 증숙하는 과정에서 생겨나는 화학적으로 변화된 생리활성물질들에 의해 인삼과 차별화된 효과를 보여준다. 또한 홍삼은 장내미생물의 생물학적 전환에 의해 변화되어 생체내 약리활성을 보이는데, 이는 개인의 장내 미생물의 분포 차이에 의해 약리작용의 차이를 보일 수 있다. 최근 이 같은 차이를 극복하기 위해 인위적으로 장내미생물에 의해 사전 발효시킨 발효홍삼은 홍삼과 다른 성분량의 차이를 보임으로써 독특한 약리작용을 보인다고 알려져 있다. 이에 본 연구는 홍삼과 발효홍삼의 추출물이 알러지 및 염증반응에 끼치는 영향에 대해 알아보기 위해 비만세포의 탈과립 과정에서 분비되는 $\beta$-hexosaminidase 분비량 비교, 염증매개 세포부착물질인 ICAM-1 발현비교, 생쥐림프구의 증식능, 각질형성세포 증식능을 상호 비교하였다. 실험결과 홍삼은 1, $10\;{\mu}g/ml$에서 $\beta$-hexosaminidase 분비량 억제 및 ICAM-1 발현 억제 조절 효과를 나타내었으며, 발효 홍삼은 모든 농도에서 유의적인 억제 효과를 나타내었다. 그러므로 홍삼과 발효홍삼에서는 모두 항알러지 및 항염증 효과가 있다고 사료된다. 그러나 LPS에 의한 B세포의 증식능에서 홍삼 $1\;{\mu}g/ml$은 증가효과를 발효홍삼에서는 $100\;{\mu}g/ml$에서 억제효과를 나타내었고, ConA에 의한 T세포의 증식능에서는 홍삼 $100\;{\mu}g/ml$에서 증가효과를 발효홍삼 $100\;{\mu}g/ml$에서는 억제효과를 나타냄으로써 림프구 증식능에 관한 실험에서는 두 추출물간에 상반된 효과를 나타내었다. HaCaT세포를 이용한 각질형성세포 증식능에서는 발효홍삼 1, $10\;{\mu}g/ml$에서만 효과를 나타내었다. 이러한 결과를 종합해 볼 때, 홍삼과 발효홍삼에 존재하는 성분량의 변화에 따라 그 효능에서 차이를 나타내고 있는 것으로 사료되며, 홍삼 및 발효홍삼의 고농도 $100\;{\mu}g/ml$에서는 저농도와는 다른 효과를 보이는 경향을 나타내고 있으므로 홍삼 및 발효홍삼의 사용에 있어 적절한 적응증과 주성분의 검증 및 유효 농도에 대한 연구가 더욱 필요하다고 판단된다.

백수오, 우엉, 마 추출물 혼합비율에 따른 혈관부착인자 VCAM-1의 발현억제 효과 (Suppression of VCAM-1 Expression in Human Aortic Smooth Muscle Cells Treated with Ethanol Extracts of Cynanchum wilfordii Radix, Arctium lappa L., and Dioscorea opposita)

  • 본 연구에서는 혈관염증 억제 효능이 입증된 원료인 백수오, 우엉, 마를 혼합하여 혈관염증을 완화시키고 동맥경화의 발생 위험을 줄일 수 있는 소재를 개발하기 위하여, 혼합물이 혈관염증을 가장 효과적으로 억제하는 배합비율을 찾아내고자 하였다. 백수오, 우엉, 마 단독투여 및 다양한 혼합비로 혼합물을 제조하여 인간유래 동맥 평활근 세포에 공급하였다. 세포부착인자인 VCAM-1의 mRNA 발현에 미치는 영향을 비교하여 가장 강한 억제효과를 나타낸 CADM5 (백수오:우엉:마=2:1:1)을 선택하였다. 선택된 혼합물이 혈관세포에서 얼마나 독성을 나타내는지 실험하였고, 백수오, 우엉, 마 추출혼합물이 혈관염증에 관여된 단백질 발현에 미치는 영향을 측정하였다. CADM5처리 결과 염증으로 인하여 증가하였던 ICAM-1과 VCAM-1 단백질의 발현이 감소하였다. 또한 CADM5를 처리한 결과 혈관내피세포에서 산화적 손상 및 염증 방어와 관련이 있는 HO-1과 Nrf-2의 발현이 증가되었다. 따라서 CADM5이 염증에 의해 유도된 ICAM-1 그리고 VCAM-1의 발현을 조절하고 산화스트레스의 방어기전을 활성화 함으로써 동맥경화증을 유발하는 혈관염증의 초기단계를 억제하여 항염증 작용에 효과가 있음을 기대할 수 있다. 비교적 저농도인 32 ㎍/㎖에서 효과적으로 혈관염증 관련 단백질 발현을 조절하였으므로 본 연구를 통해 선택된 CADM5의 혈관염증개선 및 혈관건강개선 소재로서의 개발 가능성을 확인하였다.

Butein, a tetrahydroxychalcone, suppresses pro-inflammatory responses in HaCaT keratinocytes

  • Up-regulation of cell adhesion molecules and proinflammatory cytokines contributes to enhanced monocyte adhesiveness and infiltration into the skin, during the pathogenesis of various inflammatory skin diseases, including atopic dermatitis. In this study, we examined the anti-inflammatory effects of butein, a tetrahydroxychalcone, and its action mechanisms using TNF-α-stimulated keratinocytes. Butein significantly inhibited TNF-α-induced ICAM-I expression and monocyte adhesion in human keratinocyte cell line HaCaT. Butein also decreased TNF-α-induced pro-inflammatory mediators, such as IL-6, IP-10 and MCP-1, in HaCaT cells. Butein decreased TNF-α-induced ROS generation in a dose-dependent manner in HaCaT cells. In addition, treatment of HaCaT cells with butein suppressed TNF-α-induced MAPK activation. Furthermore, butein suppressed TNF-α-induced NF-kappaB activation. Overall, our results indicate that butein has immunomodulatory activities by inhibiting expression of proinflammatory mediators in keratinocytes. Therefore, butein may be used as a therapeutic agent for the treatment of inflammatory skin diseases. [BMB Reports 2015; 48(9): 495-500]

Transcriptional Responses of Human Respiratory Epithelial Cells to Nontypeable Haemophilus influenzae Infection Analyzed by High Density cDNA Microarrays

  • Nontypeable H. influenzae (NTHi), a Gram-negative obligate human pathogen, causes pneumonia, chronic bronchitis, and otitis media, and the respiratory epithelium is the first line of defense that copes with the pathogen. In an effort to identify transcriptional responses of human respiratory epithelial cells to infection with NTHi, we examined its differential gene expression using high density cDNA microarrays. BEAS-2B human bronchial epithelial cells were exposed to NTHi for 3 hand 24 h, and the alteration of mRNA expression was analyzed using microarrays consisting of 8,170 human cDNA clones. The results indicated that approximately 2.6% of the genes present on the microarrays increased in expression over 2-fold and 3.8% of the genes decreased during the 24-h infection period. Upregulated genes included cytokines (granulocyte-macrophage colony stimulating factor 2, granulocyte chemotactic protein 2, IL-6, IL-10, IL-8), transcription factors (Kruppel-like factor 7, CCAAT/enhancer binding protein $\beta$, E2F-1, NF-$\kappa$B, cell surface molecules (CD74, ICAM-1, ICAM-2, HLA class I), as well as those involved in signal transduction and cellular transport. Selected genes were further confirmed by reverse-transcription-PCR. These data expand our knowledge of host cellular responses during NTHi infection and should provide a molecular basis for the study of host-NTHi interaction.
