Anticholinesterase activity of Cinnamomum zeylanicum L. leaf extract

  • Cinnamomum zeylanicum (C. zeylanicum) is a tropical evergreen tree of Lauraceae family. It is one of the oldest culinary spices known and used traditionally in many cultures for centuries. In addition to its culinary uses, cinnamon also possesses as a folk remedy of many health disease condition including analgesic, antiseptic, antispasmodic, aphrodisiac, astringent, carminative, haemostatic, insecticidal, and parasiticide and memory enhancing property. This study was aimed to assess the acetylcholinesterase and butyrylcholinesterase inhibitory activity of standardized methanol extract of the C. zeylanicum. Gas chromatography - mass spectrometry (GC-MS) and high performance liquid chromatography (HPLC) analysis were done to identify the presence of eugenol as chemical component and support the neuroprotective activity in the extract. Anticholinesterase inhibitory activity of crude methanol extract of C. zeylanicum leaves and cinnamon oil were evaluated by 96-well microtiter plate assay and thin layer chromatography bioassay detection methods. This study revealed that cinnamon oil ($IC_{50}:45.88{\pm}1.94{\mu}g/ml$) has better anticholinesterase activity than methanol extract ($IC_{50}:77.78{\pm}0.03{\mu}g/ml$). In HPLC analysis, retention time of eugenol in cinnamon oil was found to be 15.81 min which was comparable with the retention time (15.99 min) of the reference standard, eugenol. Seven chemical compounds were identified by GC-MS analysis, in which eugenol as an important phytoconstituents. Thus the phytochemicals from C. zeylanicum methanol leaves extract could be developed as potential source of anticholinesterase activity, with particular benefit in the symptomatic treatment of Alzheimer's disease.

  • 본 논문에서는 4인치 WVGA 광학 보상 복굴절형 (Optical Compensated Bend, OCB) 디스플레이 패널을 제안한다. 개발한 패널은 블랙 데이터 삽입 기능을 가진 저온 폴리 실리콘 (low-temperature poly-Si, LTPS) 집적 게이트 구동 회로를 내장하고 있다. 블랙 데이터 삽입 기능은 4ms의 고속 응답 시간 및 $160^{\circ}$의 광 시야각을 가능하게 한다. 본 연구에서는 상대적으로 적은 소비전력을 가진 밝은 영상에 대해 RGBW 픽 셀 구조를 적용하였다. 패널의 특성은 OCB 광학효율을 극대화하였고, 구동 중 영상의 안정성을 유지하는 목표를 달성 할 수 있었다. 패널 상에 OCB 구동을 위해 필요한 회로를 LTPS를 이용하여 설계함으로써 새로운 외부 구동 IC 개발 없이 고효율 OCB 모드를 구현할 수 있었다.

더덕 추출물과 용매 분획물의 항산화 및 티로시나아제 저해활성 (Antioxidant Activity and Tyrosinase Inhibitory Activities of Codonopsis lanceolata Extract and Solvent Fraction)

  • The purpose of this study was to investigate the antioxidant activity and tyrosinase inhibitory activity of Codonopsis lanceolata 50% ethanol extract, and its solvent fractions (n-hexane, ethyl acetate (EA), n-butanol, water). The main components of the EA fraction were qualitatively analyzed using UPLC Q-ToF/MS. Additionally, a quantitative analysis was performed using UPLC. As a result, the total polyphenol content was 113.36 mg gallic acid/g in the EA fraction, which contained the largest amount of the C. lancolata solvent fractions. Also EA showed the highest antioxidant activity than other fractions. The IC50 of DPPH(2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity was 0.03 mg/mL and the IC50 of ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonate)] radical scavenging activity was 0.049 mg/mL. The EA fraction showed tyrosinase inhibitory activity than other fractions and especially inhibited monophenolase oxidase reaction higher than diphenolase oxidase reaction. The monophenolase oxidase inhibited 55% when the concentration of the EA fraction was 0.25 mg/mL. As a result of Q-ToF/MS analysis, it was confirmed that tangshenoside I and lobetyolin were the main components of EA fraction. Thus, these results suggest that C. lanceolata may be used as a potent source of cosmetic agents.

Identification and Characterization of a Novel Antioxidant Peptide from Bovine Skim Milk Fermented by Lactococcus lactis SL6

  • A novel peptide having free radical scavenging activity was separated, using an on-line high-performance liquid chromatography (HPLC) - ABTS screening method, from bovine skim milk fermented by Lactococcus lactis SL6 (KCTC 11865BP). It was further purified using reverse phase-HPLC (RP-HPLC) and sequenced by RP-HPLC-tandem mass spectrometry. The amino acid sequence of the identified peptide was determined to be Phe-Ser-Asp-Ile-Pro-Asn-Pro-Ile-Gly-Ser-Glu-Asn-Ser-Glu-Lys-Thr-Thr-Met-Pro-Leu-Trp (2,362 Da), which is corresponding to the C-terminal fragment of bovine ${\alpha}_{s1}$-casein (f179-199). The hydroxyl radicals scavenging activity ($IC_{50}$ $28.25{\pm}0.96{\mu}M$) of the peptide chemically synthesized based on the MS/MS data showed a slightly lower than that of the natural antioxidant Trolox ($IC_{50}$ $15.37{\pm}0.52{\mu}M$). Furthermore, derivatives of the antioxidant peptide were synthesized. The antioxidative activity of the derivatives whose all three proline residues replaced by alanine significantly decreased, whereas replacement of two proline residues in N-terminal region did not affect its antioxidative activity, indicating that $3^{rd}$ proline in C-terminal region is critical for the antioxidative activity of the peptide identified in this study. In addition, N-terminal region of the antioxidant peptide did not show its activity, whereas C-terminal region maintained antioxidative activity, suggesting that C-terminal region of the peptide is important for antioxidative activity.

Constituents and the Antitumor Principle of Allium victoriallis var. platyphyllum

  • To search for cytotoxic components from Allium victoriallis , MTT assays on each extract and an isolated component, gitogenin 3-O-lycotetroside, were performed against cancer cell lines. Cytotoxicities of most extract were shown to be comparatively weak, though $IC_50$ values of $CHCl_3$fraction was found to be <31.3-368.4 $\mu\textrm{g}/ml$. From the incubated methanol extract at $36^{\circ}C, eleven kinds of organosulfuric flavours were predictable by CG-MS performance. The most abundant peak was revealed to be 2-vinyl-4H-1,3-dithiin(1) by its mass spectrum. Further, this extract showed significant cytotoxicities toward cancer cell lies. Silica gel column chromatography of the n-butanol fraction led to the isolation of gitogenin 3-O-lycotetroside (3) along with astragalin (4) and kaempferol 3, 4'-di-O-$\beta$-D-glycoside (5). This steroidal saponin exhibited significant cytotoxic activities ($IC_50$, 6.51-36.5 $\mu\textrm{g}/ml$) over several cancer cell lines. When compound 3 was incubated for 24 h with human intestinal bacteria, a major metabolite was produced and then isolated by silica gel column chromatography. By examining parent and prominent ion peak in FAB-MS spectrum of the metabolite, the structure was speculated not to be any of prosapogenins of 3, suggesting that spiroketal ring were labile to the bacterial reaction. These suggest that disulfides produced secondarily are the antitumor principles.

미생물 유래 Dykellic Acid가 담배 녹색배양세포의 생장 및 Superoxide Dismutase 활성에 미치는 영향 (Effects of Dykellic Acid Derived from Microorganism on the Cell Growth and Superoxide Dismutase Activity in Tobacco Photomixotrophic Cultured Cells)

  • 미생물에서 분리한 dykellic acid가 식물세포에 미치는 생리적 영향을 평가하기 위하여, 담배 광혼용영양배양세포(photomixotrophic cultured cells, PM세포)에 dykellic acid를 다양한 농도로 처리하여 세포의 생장과 단백질 함량 및 superoxide diamutase (SOD)활성에 미치는 영향을 조사하였다. 담배 PM세포는 0.7 mg/L 2,4-D, 0.3 mg/L kinetin, 30g/L sucrose, 200 mM NaCl를 함유한 MS배지에서 $25^{\circ}C$, 광조건에서 현탁배양 (100 rpm) 하였다. 계대배양시 화합물을 처리한 후 12일째의 세포생장 억제와 배지의 이온 전도도를 측정한 세포막손상의 결과는 일치하였으며, 화합물은 세포생장을 강하게 억제시켰다 ($IC_{50}$/, 약 20 $\mu$M). Dykellic acid 처리농도가 증가함에 따라 단위세포 무게당 단백질 함량과 SOD 비활성도를 현저히 증가시켰다. 이상의 결과로 dykellic acid는 식물세포 생장을 저해하는 활성을 가지고 있으며, 담배 PM세포는 적은 양으로도 천연물의 생리활성을 평가할 수 있는 유용한 생물소재임이 확인되었다.

  • 능소화를 80% MeOH 용액으로 추출하고, 추출물을 EtOAc, n-BuOH 및 물로 분배, 추출하였다. 이 중 EtOAc 분획을 silica gel 및 octadecylsilica gel(ODS) column chromatography로 정제하여 4종의 화합물을 분리하였다. 각 화합물의 화학구조는 NMR, MS 및 IR 등의 스펙트럼 데이터를 해석하여, linoleic acid의 methyl ester, linolenic acid의 methyl ester, ${\beta}-sitosterol$ 및 daucosterol로 동정하였다. Daucosterol은 $14{\pm}0.04\;{\mu}M$$IC_{50}$ 값으로 FPTase의 활성을 저해하였다.

더위지기 정유로부터 아세틸콜린에스테라제 억제활성 성분의 동정 (Identification of the Component with Anti-acetylcholinesterase Activity from the Essential Oil of Artemisia iwayomogi)

  • AChE 억제제는 알츠하이머 질환 치료에 이용될 수 있으므로, 본 연구는 더위지기 정유로부터 AChE 활성성분을 탐색하기 위하여 수행한 것이다. 더위지기로부터 얻은 정유를 GC-MS로 분석했을 때 camphor가 29.8%, borneol이 28.0%, eucalyptol이 5.81%, coumarin이 5.49%의 피크 면적 비율을 보였다. 이 중 camphor, borneol, coumarin을 사용하여 Anti-AChE 활성 검색을 수행하였다. 그 결과, $0.298mg/m{\ell}$, coumarin은 $0.236mg/m{\ell}$$IC_{50}$ 값을 나타내었다. 이러한 결과는 coumarin이 anti-AChE 활성을 나타내는 활성물질임을 나타내는 것이다.

소목(Caesalpinia sappan)으로부터 분리한 caspase 유도 저해 물질 (A Caspase Inducing Inhibitor Isolated from Caesalpinia sappan)

  • 한약재로부터 U937 세포주를 사용한 caspase-3 유도저해물질을 탐색한 결과 소목(Caesalpiniae sappan)을 선발하였다. 소목의 메탄올 추출물로부터 silica gel column chromatography, HPLC 등을 사용하여 저해물질을 분리하였으며, ESI-MS, $^1H-NMR,\;^{13}C-NMR$ 등의 기기분석을 실시하여 brazilin이 동정되었다. 이 물질은 $IC_{50}\;4.5\;{\mu}g/mL$의 농도로 U937세포주의 etoposide에 의한 caspase-3 유도를 저해하였으며, DPPH 라디칼 소거능도 나타내었다.

In Vitro Inhibitory Effect of Licoricidin on Human Cytochrome P450s

  • Licoricidin isolated from Glycyrrhiza uralensis is known to have anticancer, anti-nephritic, anti-Helicobacter pylori, and antibacterial effects. In this study, a cocktail probe assay and liquid chromatography-tandem mass spectrometry (LC-MS/MS) were used to investigate the modulating effect of licoricidin on cytochrome P450 (CYP) enzymes in human liver microsomes. When licoricidin was incubated at $0-25{\mu}m$ with CYP probes for 60 min at $37^{\circ}C$, it showed potent inhibitory effects on CYP2B6-catalyzed bupropion hydroxylation and CYP2C9-catalyzed diclofenac 4'-hydroxylation with half maximal inhibitory concentration ($IC_{50}$) values of 3.4 and $4.0{\mu}m$, respectively. The inhibition mode of licoricidin was revealed as competitive, dose-dependent, and non-time-dependent, and following the pattern of Lineweaver-Burk plots. The inhibitory effect of licoricidin has been confirmed in human recombinant cDNA-expressed CYP2B6 and 2C9 with $IC_{50}$ values of 4.5 and $0.73{\mu}m$, respectively. In conclusion, this study has shown the potent inhibitory effect of licoricidin on CYP2B6 and CYP2C9 activity could be important for predicting potential herb-drug interactions with substrates that mainly undergo CYP2B- and CYP2C9-mediated metabolism.
