Protection of Green Leafy Vegetable Extracts Against Oxidation of Human Low Density Lipoprotein

  • Oxidation of low density lipoprotein (LDL) is regarded to play an important role in the development of atherosclerosis. In the present study, salad vegetables with a remarkable DPPH radical-scavenging activity were extracted with methanol, and the methanol extracts were evaluated for the inhibition of $Cu^{2+}$-induced oxidation of human LDL. Separately, the amount of total phenolics was determined colorimetrically using Folin-Ciocalteu reagent. The vegetable extracts, expressing a strong inhibition of LDL oxidation ($IC_{50}$ values, <$100\;{\mu}g/mL$), were from angelica, dandelion, mustard leaf, and water spinach, which contained relatively high level of polyphenol content. Noteworthy, a highly positive correlation was observed between inhibition of LDL oxidation and amount of total polyphenol (p<0.01). Based on these results, it is suggested that salad vegetables, especially angelica, dandelion, and mustard leaf, may be used as easily accessible sources of natural antioxidants, especially in anti-atherosclerosis.

  • Here we report a new chemical inhibitor against HIV-1 with a novel structure and mode of action. The inhibitor, designated as A1836, inhibited HIV-1 replication and virus production with a 50% inhibitory concentration ($IC_{50}$) of $2.0{\mu}M$ in an MT-4 cell-based and cytopathic protection antiviral assay, while its 50% cytotoxic concentration ($CC_{50}$) was much higher than $50{\mu}M$. Examination of the effect of A1836 on in vitro HIV-1 reverse transcriptase (RT) and integrase showed that neither were molecular targets of A1836. The characterization and re-infection assay of the HIV-1 virions generated in the presence of A1836 showed that the synthesis of early RT products in the cells infected with the virions was inhibited dose-dependently, due in part to abnormal protein formation within the virions, thus resulting in an impaired infectivity. These results suggest that A1836 might be a novel candidate for the development of a new type of HIV-1 inhibitor.

Antioxidant and Oxidative DNA Damage Protection Potential of Methanol Extract of Red Tea Stem

  • This study was undertaken to determine free radical scavenging capacity and oxidative DNA damage protecting activity of methanol extract of red tea stem. The extract was subjected to assess their antioxidant potential using various in vitro systems such as $DPPH^{\bullet}$, $ABTS^{{\bullet}+}$, super oxide and nitric oxide free radicals and it exhibited $IC_{50}$ values of $68.88{\pm}1.1$, $12.08{\pm}0.65$, $404.38{\pm}1.6$, $93.6{\pm}2.7{\mu}g/mL$ respectively. Red tea extract also showed ferric reducing ability (FRAP) with 2606.85 mmol Fe (II)/g of extract. Furthermore, Methanol extract of red tea stem showed significant DNA damage protecting activity in concentration dependent manner against $H_2O_2+UV$ induced photolysis on pUC19 plasmid DNA. Results of this study showed that the methanol extract of Red Tea stem has strong antioxidant potential along oxidative DNA damage protecting capacity that would be the significant sources of natural antioxidants, which might be helpful in preventing the progress of various oxidative stress generated diseases. Further study is necessary for isolation and characterization of the active antioxidants, which may serve as a potential source of natural antioxidant.

  • The high efficiency power management IC(PMIC) for Moblie application is proposed in this paper. PMIC is controlled with PWM control method in order to have high power efficiency at high current level. The saw-tooth generator is made to have 1.2 MHz oscillation frequency and full range of output swing from ground to supply voltage(VDD:3.3V). The comparator is designed with two stage OP amplifier. And the error amplifier has 70dB DC gain and $64^{\circ}$ phase margin. DC-DC converter, based on Voltage-mode PWM control circuits, achieved the high efficiency near 95% at 100mA output current. DC-DC converter is designed with LDO in stand-by mode which fewer than 1mA for high efficiency.

  • This study was performed to investigate the effects of fermented extract of Pinus densiflora (FPD) on oxygen radicals and $H_2O_2$-induced damage. The results are as follows: 1. The 1.1-diphenyl-2-picrylhydrazyl (DPPH) free radicals were considerably reduced by FPD and $IC_{50}$ value was showed the concentration of 20 ㎍/㎖. 2. The cytotoxicity did not observe by FPD treatment in A548 cells. 3. The $H_2O_2$-induced cell damage was recovered by FPD pretreatment in A549 cells. These results suggest that FPD, as a natural antioxidant, has scavenging effect of free radicals and protection effect from $H_2O_2$-induced cytotoxicity.

  • This paper describes functional circuits of DC-DC converter IC incorporated with variable frequency PFM technique. In case of output voltage of DC-DC converter is reached setting value or output current is low then PFM switching frequency is slow down. In this work a PFM signal generator, a PFM Frequency Control Circuit, an output voltage detector and an over current protection circuits are designed. This device has been designed at a $0.35[{\mu}m]$, double poly, double metal 12[V] CMOS process.

  • The corrosion failure of electronic devices has been a major reliability concern lately. This failure is an ongoing concern because of miniaturization of integrated circuits (IC) and the increased use of polymers in electronic packaging. Recently, plasma-polymerized cyclohexane films were considered as a possible candidate for a interlayer dielectric for multilever metallization of ultra large scale integrated (ULSI) semiconductor devices. In this paper the protective ability of above films as a function of deposition temperature and RF power in an 3.5 wt.% NaCl solution were examined by polarization measurement. The film was characterized by FTIR spectroscopy and contact angle measurement. The protective efficiency of the film increased with increasing deposition temperature and RF power, which induced the higher degree of cross-linking and hydrophobicity of the films.

Antithrombotic and Antiallergic Activities of Rhaponticin from Rhei Rhizoma Are Activated by Human Intestinal Bacteria

  • To evaluate the antithrombotic and antiallergic properties of rhaponticin extracted from Rhei Rhizoma, the in vitro and ex vivo inhibitory activities of rhaponticin and its metabolite, rhapontigenin, were measured. These compounds inhibited in vitro ADP- and collagen-induced platelet aggregation. Rhapontigenin was more potent, with $IC_{50}$ values of 4 and $70{\;}{\mu}g/ml$, respectively. In ex vivo ADP- and collagen-induced rat platelet aggregation, these compounds also exhibited a potent inhibitory effect. The antiplatelet aggregation effects of rhaponticin and rhapontigenin were more potent than those of aspirin. Rhapontigenin showed significant protection from death due to pulmonary thrombosis in mice. Rhapontigenin also showed the strongest inhibitory activity against $\beta-hexosaminidase$ release induced by DNP-BSA. These compounds inhibited PCA reaction in mice. Rhapontigenin intraperitoneally administered showed the strongest inhibitory activity and significantly inhibited PCA at doses of 25 and 50 mg/kg, with inhibitory activities of 48 and 85%, respectively. The inhibitory activity of orally administered rhaponticin was stronger than that of intraperitoneally administered rhaponticin. These results suggest that rhaponticin, in the rhizome of Rhei Rhizoma, is a prodrug that has extensive antiallergic and antithrombotic properties.

Protection Effect of Undariia pinnatifida sporophylls-derived Fucoidan in Rat Lung Injury from X-ray Irradiation (미역포자엽추출 푸코이단의 X-선 조사 흰쥐 폐장상해에 대한 방오기능 평가)

  • The radioprotect effects of fractinated fucoidan, derived from Undaria pinnatifida sporophylls, were examined in lung injured rats treated with partial body irradiation. The right lung of Sprague-Dawley rats, eight separately grouped by including radiation only control group (ROG) and fucoidan+radiation treated group (FRG), were treated with various fraction of Undaria-derived fucoidan every other day by intraperitoneal injection 6 days prior to irradiation and during monitoring at 24 hours, 48 hours, 2 weeks, 4 weeks and 12-weeks post-irradiation of 6 Mev/2 Gy. The rats were euthanized at completion of the treatment. Tissue samples from the injured lung, fixed in formaldehyde using lung perfusion prior to extraction, were H/E stained for histological analysis using optical microscopy. Interstitial congestion (IC), hemorrhage in alveolar sac (HM), number of macrophage (MP) and alveolar wall thickness (AWT) as a measure of early indication of fibrosis were measured. AWTs in 24 hrs post-irradiation rats increased in comparison with $2.49{\pm}0.06{\mu}m$ of control group by $3.67{\pm}0.09{\mu}m$, $3.21{\pm}0.08{\mu}m$ (p=0.013), $2.98{\pm}0.08{\mu}m$ (p=0.00) in ROG, F1-fucoidan+radiation, and F3-fucoidan+radiation group, respectively. AWT of the ROG was further increased by $4.30{\pm}0.13{\mu}m$ in the 12 weeks post-irradiation group, but AWT on average was $2.56{\pm}0.05{\mu}m$ in the FRG. MP in the 24 hrs post-irradiation group markedly increased in comparison with $2.6{\pm}0.34/0.14\;mm^2$ of the control group by $8.0{\pm}1.48/0.14\;mm^2$ in the ROG, but it was only $3.6{\pm}0.48/0.14\;mm^2$ in F3-FRG. MP in the 12 weeks post-irradiation group was $7.2{\pm}1.28/0.14\;mm^2$ in ROG, but it was $2.8{\pm}0.37/0.14\;mm^2$ or $2.4{\pm}0.4/0.14\;mm^2$ (p<0.05) in the FRG. In addition, increased IC and HM in ROG were relatively smaller in FRG of the 24 hr and 12 weeks post-irradiation rats. In conclusion, Undaria pinnatifida sporophylls-derived fucoidan fractions exhibited radioprotectant activity on injured rat lung subjected to X-ray irradiation. However, success was variable according to the fractions and also time of injection post injury.

Characterization of Aerosol Composition, Concentration, and Sources in Bukhansan National Park, Korea (북한산국립공원 내 초미세먼지 농도 및 화학적 특성)

  • To improve understanding of the physico-chemical characteristics of aerosols in the national park and comparing the air pollution between national park and the urban area nearby national park, the aerosol characterization study was conducted in Bukhansan National Park, Seoul, from July through September 2017. Semi-continuous measurements of $PM_{2.5}$ using PILS (Particle Into Liquid System) coupled with IC (Ion Chromatography) and TOC (Total Organic Carbon) analyzer allowed quantification of concentrations of major ionic species($Cl^-$, $SO_4{^{2-}}$, $NO_3{^-}$, $Na^+$, $NH_4{^+}$, $K^+$, $Mg{^{2+}}$ and $Ca{^{2+}}$) and water soluble organic carbon (WSOC) with 30-minute time resolution. The total mass concentration of $PM_{2.5}$ was measured by T640 (Teledyne) with 5-minute time resolution. The black carbon (BC) and ozone were measured with a minute time resolution. The timeline of aerosol chemical compositions reveals a strong influence from urban area (Seoul) at the site in Bukhansan National Park. Inorganic aerosol composition was observed to be dominated by ammoniated sulfate at most times with ranging from $0.1{\sim}32.6{\mu}g/m^3$ (6.5~76.1% of total mass of $PM_{2.5}$). The concentration of ammonium nitrate, a potential indicator of the presence of local source, ranged from below detection limits to $20{\mu}g/m^3$ and was observed to be highest during times of maximum local urban (Seoul) impact. The total mass of $PM_{2.5}$ in Bukhansan National Park was observed to be 10~23% lower than the total mass of $PM_{2.5}$ in urban area (Gireum-dong and Bulgwang-dong, Seoul). In general, ozone concentration in Bukhansan National Park was observed to be similar or higher than urban sites in Seoul, suggesting additional biogenic VOCs with $NO_x$ from vehicle emission were to be precursors for ozone formation in Bukhansan National Park.
