A Role of Indole-3-acetic Acid on the Growth and the Accumulation of Tannin in Callus Induced from Gallas of the Sumac (붉나무충영의 배양조직에서 생육 및 tannin 축적에 미치는 indole-3-acetic acid의 역할)

  • These studies were carried out to examine the developmental process of galls caused by aphids in Rhus gavanica and the effects of indole-3-acetic acid (IAA) and ${\alpha}$-naphthalene acetic acid (NAA) on the tannin accumulation in the callus induced from that galls. The results are follows. The development of fist-shaped galls has begun at the beginning of June earlier than the case of finger-shaped galls, and also, the growth of fist-shaped galls has last to September longer than the period of finger-shaped galls. These results indicate that the life cycle and feeding activity of the aphids inhabited in fist-shaped galls were longer and mire active than the case of the aphids inhabited in finger-shaped galls. Tannin contents of fist-shaped galls revealed about 60${\sim}70%$ of total dry weights during the whole growth periods, however, the contents in finger-shaped galls were under 10% at the maximum value. These facts mean that finger-shaped galls seem to be as a habitat of aphids rather than as a major source of tannin such as fist-shaped galls. The growth of callus induced from fist-shaped galls was the most effective in the plot of $10^{-5}$ mole IAA, but the tannin accumulation in callus growth was not even caused in any plots of IAA treatments as wells as in any NAA plots. These results considered that the tannin accumulation in fist-shaped galls may be caused only in specific relation between host plant and life cycle of aphids.

Physiological Changes in Rooting Zone of Dwarf Apple Rootstocks (Malus domestica Borkh.) after Stem Etiolation Treatment (사과 왜성대목들의 줄기 황화처리에 따른 발근 부위의 생리적 변화)

  • This work was conducted to investigate the important rooting factors through comparative analysis of a physiological differentiation after layering treatment using four apple rootstocks of different rooting abilities; M.26, M.9, O.3, and Mo.84. Mo.84 showed the highest rooting rate in from rootstocks, while O.3 was the lowest. Mo.84 also found to have the highest indole-3-acetic acid (IAA) content, although the fluctuation of IAA contents was not consistent with layering treatment. In contrast, abscisic acid (ABA) content of Mo.84 which showed highest rooting was lowest among rootstocks regardless of layering treatment. And ABA contents of all rootstocks were decreased after layering treatment than before layering treatment. O.3 which showed poor rooting rate revealed lowest in boron content. Carbohydrate/nitrogen (C/N) ratio of Mo.84 was the highest in all rootstocks. Therefor, we assumed that he IAA contents in layering treated rootstocks were not seemed to be a major rooting factor, but the changes in ABA contents and boron levels limit rooting in dwarf apple rootstocks.

Effects of the Agar and Auxins Concentrations on Vitrification in Tissue Culture of Gypsophila paniculata L. cv. 'Bristol Fairy' (숙근(宿根)안개초의 조직배양(組織培養)에 있어서 Agar 농도(濃度) 및 Auxins의 농도(濃度)가 Vitrification에 미치는 영향(影響))

  • The present work deals with the effect of agar and auxins concentrations on vitrification in tissue culture of Gypsophila paniculata L. cv. 'Bristol Fairy' in vitro. The results were summarized as follows. 1. Plant growth, that is, plant height, fresh weight and branching were decreased as increasing agar concentration. On the other hand, addition effect of IAA 1.0mg/l+NAA 0.5mg/l and IAA 2.0mg/l+NAA 1.0mg/l on the plant height were increased strikingly. 2. Addition effect of auxins on the days to rooting were little. And the root development showed same tendency as plant growth. 3. The rate of non-vitrified plants were gradually increased as rising agar concentration. But the addition of agar 1.5g/l in the medium resulted in poor growth. 4. From these results, it was found that following media were the most effective for increasing of non-vitrified and good plant growth in Gypsophila paniculata L. tissue culture.

Effect of Presowing Treatment with Growth Regulators on Different Growth and Yield Contributing Parameters in Soybean [Glycine max (L.) Merril] (식물생장과 조절제의 종자처리가 콩의 생장과 수량구성요소에 미치는 영향)

  • To know the effect of presowing seed soaking with some growth substances (kinetin, IAA, ethrel and salicylic acid) on vegetative growth and yield contributing parameters in soybean [Glycine max (L.) Merrill] the investigation was undertaken. The salient features from the study are;-All the treatments showed the enhancement effect on vegetative growth, viz. seediling emergence, germination percentage, plant height, number of leaves and branches per plant as compared with control, whereas salicylic acid delayed the seedling emergence process and lowered the germination percentage.-Nodule initiation, number of nodules and nodule weight were hastened and increased respectively under all the treatments whereas IAA showed an opposite effect on all the parameters at the early stages of nodule development.-Kinetin, IAA and ethrel showed the significant enhancement effect on the levels of biochemical parameters throughout the time of seed development whereas salicylic acid and water showed the tendency similar to that of control.-Yield and its components, viz. number of flowers, pods and seeds per plant were significant increased under all the treatments but they didn't show any significant enhancement effect on number of seeds per pod and pod setting rate. One hundred seed weight was lower under all the treatments, indicating the dilution effect resulting from incapability of increased number and size of source to provide the increased size of sink with assimilates.

Adventitious Shoot Formation and Plant Regeneration from Explants of Solanum nigrum L. (까마중(Solanum nigrum L.)의 유식물 절편체에서 부정아 형성 및 식물체 재분화)

  • In the present study, the effects of plant growth regulators on adventitious shoot and root formation of various explants of $in$ $vitro$ seedlings of $Solanum$ $nigrum$ L. were investigated to determine the optimum conditions for the high-efficiency plant regeneration of this species. The formation of adventitious shoots was higher in leaf explants than in cotyledon, hypocotyl, or epicotyl explants at low concentrations (0.5~2.0 mg $L^{-1}$ ) of 6-benzylaminopurine (BAP). The number of adventitious shoots and the shoot length were also higher in both leaf and cotyledon explants. In particular, 2.0 mg $L^{-1}$ BAP was most effective for stimulating the induction and multiplication of adventitious shoots. In terms of root formation and root development from shoots that were separated from multiple shoots, indole butyric acid (IBA) and indole acetic acid (IAA) were more effective than ${\alpha}$-naphthalene acetic acid (NAA). The percentage of rooting as well as the number of roots per shoot (4.0), root length (7.82 cm), and shoot length (8.76 cm) was highest on MS media supplemented with 0.05 mg $L^{-1}$ IAA. Furthermore, 100% of the regenerated plantlets survived when transplanted to compost soil. These results suggest that leaf explants are the best source for the high-efficiency regeneration of $S.$ $nigrum$ L., and that 2.0 mg $L^{-1}$ BAP and 0.05 mg $L^{-1}$ IAA are the best conditions for shoot and root induction, respectively.

Effect of TIBA on the Brassiolide-induced Gravitropic Response in the Primary Roots of Maize (옥수수 일차뿌리에서 TIBA가 brassinolide에 의해 유도된 굴중성 반응에 미치는 영향)

  • It has been known that brassiolide (BL) increased the positive gravitropic response and ethylene production in maize roots. This study examined the relationship between the BL-induced gravitropic response and ethylene Production. The ethylene production was inhibited to about 90% of the control by the treatment of $10^{-4}$ M aminoethoxyvinylglycine (AVG), the ethylene synthesis inhibitor. However, the gravitropic response did not show any significant changes compared to the control at $10^{-4}$ M AVG. In the case of treatment of AVG with BL, the ethylene production decreased to 60% of the control. However, the gravitropic response increased to the level which was induced by BL. Cobalt ions, another ethylene biosynthesis inhibitor, inhibited ethylene production, but not gravitropic response. When roots were treated with BL and cobalt ions, they showed the inhibition of ethylene production and promotion of gravitropic response. To elucidate the possibility that the effect of BL is related to auxin transport, roots were treated with TIBA (2,3,5-triiodobenzoic acid), an auxin transport inhibitor. Both treatment of TIBA alone and TIBA with BL stimulated ethylene production to about 96% and 132%, respectively. However, gravitropic response was completely inhibited in both treatments. Further, roots treated with BL in the presence of TIBA and IAA showed a negative gravitropic response, which means that IAA accumulates in the upper side of horizontal roots. Root elongation was also stimulated in this treatment. Taken together, these results suggest that BL might affect the differential distribution of internal IAA on roots, causing the regulation of positive gravitropic response.

  • In here, we investigated the quantitative analysis method of indole-3-acetic acid (IAA) and indole-3-butyric acid (IBA) with liquid chromatography-mass/mass spectrometry (LC-MS/MS) or gas chromatography-MS. Two ways of clean-up process were investigated for LC-MS/MS instrumental analysis of IAA, but both a simple dilution and hydrophile-lipophile balance (HLB) solid phase extraction (SPE) were not met the optimal recovery rates for quantitative analysis. On the other hand, the clean-up method for GC-MS was finally optimized through HLB-SPE from 250-folds diluted sample and methylation with trimethylsilyl chloride in methanol for 4 h. The limit of detection for methyl ester of IAA and IBA were both 1.4 mg/L, and recovery rates showed 93-107% from the concentrated liquid fertilizer.

Changes in $GA_3$, IAA and ABA Contents at the Different Growth Stage of Angelica gigas NAKAI (참당귀(當歸)의 생육단계별(生育段階別) $GA_3$, IAA 및 ABA 함량변화(含量變化))

  • This study was carried out to observe the changes in $gibberellin(GA_3)$, indol-3 acetatic acid(IAA) and abscisic acid(ABA) contents known as plant growth regulators in the leaves of Angelica gigas and to get the basic information for the establishment of cultural practice for the inhibition of floral induction. Changes in $GA_3$, IAA and ABA contents were investgated at different growth stage in the young flag leaves of two year old plants.The contents of $GA_3$ showed the highest value by 4. 87mg /kg at early flowering stage, and those were gradually increased from vegetative stage to early flowering stage, but greatly decreased from early flowering stage to fruiting stage. The contents of IAA were also the highest by 2. 27mg /kg at early flowering stage. The contents of ABA were gradually increased along with the development of growth stage. In the fruiting stage those were the highest value by 0.37mg /kg and in early flowering stage the contents were decreased temporarily.

Effects of Some Plant Growth Regulators on Protein Biosynthesis of Carrot Cells (당근 세포의 단백질 생합성에 대한 몇가지 식물생장조절제의 영향)

  • Electrophoretic studies of protein extracts from carrot calluses suspension-cultured on the media containing kinetin, BA, IAA, NAA or $GA_3$ at the levels of $10^{-6},\;10^{-5},\;10^{-4}M$, respectively, were performed to identify polypeptides and proteins regulated by auxin, cytokinin or GA. Fifteen bands of polypeptide(s) were observed in the callus cultured in the control medium devoid of growth regulators, and their molecular weights were $18._4,\;20._2,\;20._0,\;34._9,\;35._7,\;37._4,\;40._3,\;42._2,\;44._1,\;44._4,\;49._3,\;55._0,\;56._6,\;58._1,\;and\;59._9\;KD$, respectively. The synthesis of polypeptide appeared to be promoted in two bands by kinetin, in six bands by BA, in one band by IAA, in two bands by NAA, and in four bands by $GA_3$, while inhibited in five bands by kinetin, in three bands by BA, in four bands by IAA, in three bands by NAA and in three bands by $GA_3$. The polypeptides of $40._3\;KD\;42._2\;KD$ seemed to be regulated by cytokinins, and those of $44._1\;KD,37._4\;KD,\;and\;56._6\;KD$ by auxins. The proteins of three bands with relative mobilities of 0.56, 0.84, and 0.92, respectively, increased in the calluses cultured on the media containing kinetin, IAA, $GA_3$, NAA or BA, compared to the control, but it was difficult to identify the proteins specific for each growth regulator.

Phosphate Solubilization and Plant Growth Promotion by Crop Associated Bacteria (인산용해미생물에 의한 불용성 인의 용해와 식물생장에 미치는 영향)

  • Fourteen bacterial strains were isolated from crop rhizosphere and identified as phosphate solubilizing bacteria (PSB) by 16S rRNA analysis. Only 3 strains exhibited a strong ability to solubilize insoluble phosphate in agar medium containing a hydroxyapatite. The rates of P solubilization by isolates were ranged from 200 and $2300\;mg\;L^{-1}$, which are inversely correlated with pH in culture medium. Furthermore, HPLC analyses reveal the production of organic acid from the culture filtrates of PSB. Among these, strain Acinetobacter sp. released only gluconic acid, Pseudomonas orientalis produced gluconic acid which was subsequently converted into 2-ketogluconic acid, and Enterobacter asburiae released acetic acid and succinic acid. On the other hand, P. orientalis and E. asburiae released $372\;mg\;L^{-1}$ and $191\;mg\;L^{-1}$ of IAA into broth culture, respectively, while Acinetobacter sp. did not produce IAA. Furthermore, in vivo study showed that plant growth promoting effect by bacteria generally seemed to be increased IAA production and phosphate solubilization.
