Characterization of Green Mold Contamination caused by Penicillium brevicompactum in Hypsizygus marmoreus (느티만가닥버섯에서 Penicillium brevicompactum에 의해 유발된 곰팡이 오염 특성)

  • Beech mushroom (Hypsizygus marmoreus) is one of the most popular mushrooms in Japan, China, and Korea because of its delicious taste, nutritious value, and unique crunchy texture. In 2018-2019, unusual signs on the mushroom were observed in mushroom farms in Gyeongnam Province. The main signs were the inhibition of mycelial growth and primordial formation on the surface of mushroom media. When green mold caused contamination at early stage, the rate of pinhead formation by the mushroom greatly decreased to 46.5-71.5%. Conidia of the causal green mold were ellipsoidal, dark green, and measured 2.0-3.1㎛ long. The optimum temperature for mycelial growth of the green mold was 25℃. The phylogenetic tree obtained from the internal transcribed spacer (ITS) rDNA sequences showed that the isolated green mold corresponded to Penicillium brevicompactum (100.0%). This is the first report of green mold contamination caused by P. brevicompactum on H. marmoreus.

Effect of LED LightIrradiation on the Mycelial Growth and Fruit Body Development of Hypsizygus Marmoreus (LED 광원이 느티만가닥버섯 균의 균사 생장과 자실체 생육에 미치는 영향)

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated. However, the researches of cultivation and physiological characteristics were not conducted in Korea. In this study, we conducted on artificial cultivation of H. marmoreus and elucidated the effect of light on the mycelial growth and fruit body development using LED light sources with different wavelength; blue (peak wave length 460nm), green(peak wave length 530nm), yellow(peak wave length 590nm), red(peak wave length 630nm), and white as positive control. Mycelial growth of H. marmoreus strains were inhibited about 30~40% in inhibition ratio under the illumination with blue, green, yellow LED light. However, red LED light was not inhibited. Elongation of stipe was effective under the long wave length such as yellow and red light. Abnormal fruit body was produced under the long wavelength and dark. However, development of pileus was effective under the short wavelength such as green and blue light. Also, as a result of cultivation with mixed light for high quality and harvest, many effective numbers and yields of fruiting bodies were obtained in the mixed treatment of blue and white light, and pileus developed well.

Breeding a new variety of white Hypsizygus marmoreus 'Baekmaru' with excellent cultivation stability and storability (재배적 안정성과 저장성이 우수한 백색 느티만가닥버섯 신품종 '백마루' 육성)

  • The purpose of this study was to breed a variety of stable productivity and high storage characteristics of white Hypsizygus marmoreus, which has high demand at domestic and global market due to a low bitter taste and the preference for white species. Accordingly, 'Baekmaru' was bred by hybridizing brown and white cap color species and backcrossing with white species. The 'Baekmaru' variety was bred by crossbreeding the brown and white species and backcrossing the white species. Through repeated cultivation of 'Baekmaru', a variety with a low contamination rate was selected when culturing the spawn for stable cultivation. As a results of demonstration test, the yield of 'Baekmaru' was 14% higher than that of the commercial variety. In addition, among the fruiting body characteristics of 'Baekmaru', diameter and thickness of the pileus were 16.43±15.27mm and 6.46±0.58mm, which were slightly higher than the commercial variety, and the hardness was 2.69±0.89N for the pileus and 3.09±0.89N for the stipe. The shelf life showed less change in thickness and color of pileus compared to commercial variety. The hardness of pileus of 'Baekmaru' was maintained in the range of 3.5 to 4.0N in the 4℃ and 4℃ and 20℃ mixed treatment until 30 days of storage, and was higher than that of the commercial variety. Therefore, it was suggested to be excellent in storability.

Application of electric pulsed power on fruit body production of edible and medicinal mushrooms

  • Effect of pulsed power was investigated on fruit body formation of 10 edible mushrooms, Lentinula edodes, Glifola frondosa, Pholiota nameko, Flammulina velutipes, Hypsizygus marmoreus, Pleurotus ostreatus, Pleurotus eryngii, Pleurotus abalonus, Agrocybe cylindracea and Sparassis crispa. Pulsed power of 100-170 kV was directly charged to the substrate just before fructification. The effect of the pulsed power resulted to promote for 10 edible mushrooms fructification. The treatment especially stimulated the fructification on Pleurotus species.

Genetic variation of local varieties and mutants groups induced by gamma ray in Hypsizigus marmoreus (느티만가닥버섯의 재래종과 감마선 돌연변이체들의 유전적 변이)

  • This research was carried out to analyze the genetic variation of 18 wild strain, 2 breed varieties and 20 mutants of Hypsizygus marmoreus by random amplification of polymorphic DNA(RAPD). Also, 3 strains of Lyophyllum decartes and 1 strain of Lyophyllum shimeji were used. These mushrooms were collected from korea, china, Taiwan and Japan. Spores of H. marmoreus JV-2 strain were irradiated by gamma ray for mutagenesis. 40 kind of primers were used for this reaserch. Number of reaction primer were 31. Electrophorectic patterns of RAPD showed genetic variation. In phylogenetic tree, they were divided into seven group. Discriminative differences were observed between wild strain and mutants in H. marmoreus. These results might suggest that these primers and gamma ray irradiation of spores were useful tools for developing new strain for mushroom.

Skin-whitening effects of hot water extract from domestic edible mushrooms (국내 시판 식용버섯 열수 추출물의 피부미백 기능 연구)

  • Hot water extracts from 16 domestic edible mushrooms including Pleurotus ferulae (Lanzi) X.L. Mao, Lentinula edodes (Berk.) Pegler, and Hypsizygus marmoreus (Peck) H.E. Bigelow, which are commercially available, were used for determining the cosmetic potential of these mushrooms. In this study, we carried out in vitro functional experiments to determine the inhibitory effects of these extracts on L-DOPA oxidation of tyrosinase and melanin synthesis. Based on the results of these experiments, H. marmoreus (Peck) H.E. Bigelow No. 10 and No. 15 were selected for further analysis. We analyzed the melanin synthesis inhibitory activity, TRP1 and MITF expression via real-time PCR, and Fontana Masson staining in artificial skin $Neoderm^{(R)}-ME$. Taken together, we observed that the hot water extract from H. marmoreus (Peck) H.E. Bigelow (No. 15) had better whitening effect than the extracts of other mushrooms. Thus, it can be a potential source of skin-whitening agent.

Analysis of Nutritional Characteristics and Physiological Functionality of Hypsizygus marmoreus (Brown cultivar) (갈색 느티만가닥버섯의 영양특성 및 생리기능성 분석)

  • In order to apply into functional food or medicinal industry, nutritional characteristics and physiological functionality of Hypsizygus marmoreus (brown cultivar) were investigated. Fruiting body of H. marmoreus contained 27.3% of crude protein, 55.8% of total sugar and 11,109.3 mg/100 g dry weight of malic acid. Furthermore, 66.7% of antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity and 37.3% of antigout xanthin oxidase inhibitory activity showed in the water extract from H. marmoreus. The economically ACE inhibitory activity (81.4%) was obtained when the fruiting body of Hysizygus marmoreus was extracted with distilled water of $50^{\circ}C$ for 12 h, even though maximal ACE inhibitory activity (84.4%) was showed the extracts from $70^{\circ}C$ for 12 h.

Selection of High Efficient Enzyme for Protoplasts Isolation from Mushrooms (버섯류의 원형질체 나출을 위한 고효율 효소 선발)

  • This study was carried out to select cell wall degrading enzymes for maximizing protoplast yield from Basidiomycetes. The protoplasts were released from spore suspension, mycelia cultured on cellophane membrane, and homogenized mycelia of Flammulina velutipes using commercial cell wall degrading enzymes. The highest yield of protoplasts was obtained from the homogenized mycelia treated with the enzyme combination of $Glucanex^R$ 200G and cellulase onozuka R-10. The protocol was also available for Pleurotus ostreatus, P. eryngii, and Hypsizygus marmoreus.

Effect of dietary fermented spent mushroom (Hypsizygus marmoreus) substrates on laying hens (느티만가닥버섯 수확후배지 발효사료 급여가 산란계에 미치는 영향)

  • This study was conducted to evaluate the feeding value of the spent mushroom (Hypsizygus marmoreus) substrates (SMS) in laying hens (Hy-Line Brown). The fresh spent mushroom (Hypsizygus marmoreus) substrates collected from the Namkyung farm were fermented with Bacillus subtilis EJ3 for 2 weeks. A total of twenty-four laying hens were fed corn-soy based experimental diets containing 0% (T0), 5% (T1), 10% (T2) and 15%(T3) fermented SMS for 12 weeks. There were no significant differences among the treatments in egg production, egg weight, egg mass, feed conversion and viability during the experimental period. Feed intake was significantly lowered in T0 (118.3 g) than T1 (121.9 g), T2 (120.3 g) and T3 (122.4 g). There were no significant differences among the treatments eggshell breaking strength, thickness and haugh unit, whereas the yolk color of T1, T2 and T3 were significantly heavy than T0. The palatability of boiled meat was significantly better in the T3 laying hens than in the T0 laying hens. In conclusion, fermented SMS can be used as resource of feed in laying hen feed at 5.0-15% level without effect on performance and egg qualify.

Comparison of Endo-, Exo-Cellular Enzyme Activity for New Strains of Hypsizygus marmoreus (느티만가닥버섯의 신품종에 대한 endo-, exo-cellular 효소 활성도의 비교)

  • This study was carried out to investigate the morphological and physiological characteristics of six new cultivars of Hypsizygus marmoreus (Hm) and measure endo-, exo-cellular enzyme-specific activity. The domestic wild stain (Hm3-10) and commercial strain in Japan (Hm1-1) were mated by crossing monokaryon mycelia. We gained 58 strains from one of 400 crosses through the $1^{st}$ cultivation experiment, and selected six strains from one of 58 strains through the $2^{nd}$ cultivation experiment. When six of the selected new strains were grown during several spawn culture periods (60, 70, 80, 90, and 100 days), a spawn culture period of more 80 days was considered to be excellent as being shorter than 19~20 days. Therefore, we determined the period of spawn culture as 80 days. Three strains such as Hm15-3, Hm15-4, and Hm17-5 showed an excellent result. When endo-cellular enzyme activity measured eight strains, we obtained a result of that specific activity of ${\alpha}$-amylase at the highest as 73.9~102.2 unit/mg protein, and chitinase is lower than ${\alpha}$-amylase at 8.1~13.1 unit/mg protein. When exo-cellular enzyme activity measured eight strains, we determined the result of that specific activity of ${\alpha}$-amylase is the highest at 5,292~1,184 unit/mg protein, and CMCase and xylanase were 1,140~245 unit/mg protein, 94~575 unit/mg protein, compared to each other. However, the enzyme activity of ${\beta}$-glucosidase and chitinase is low.
