Insulin has crucial roles in energy metabolism in all mammals but has been less studied in ruminants. An experiment was conducted to investigate the effects of hypoinsulinemia induction on appetite, performance, carcass composition and blood metabolite levels in sheep. Treatments were intravenous injection of four doses of streptozotocin; 0, 25, 50 and 75 mg/kg BW named C, L, M and H, respectively. Twenty male lambs were divided into four treatment groups. Animals in group H could not continue the experiment because of abnormalities. The duration of the experiment was eight consecutive weeks, and injection was performed at the end of week 3. Feed and water intakes were measured weekly and weight changes of animals were recorded and used for calculation of other growth parameters. Blood samples were collected weekly via venipuncture at fasting and 2.5 h post-prandial and analyzed for hormones and blood metabolites. Results showed a marked hypoinsulinemia in group M with significant decrease in fasted and postprandial insulin concentrations and also fasted leptin concentrations vs. the control group C (p<0.05). Group M showed significant increases in blood glucose, triglycerides, cholesterol, total protein, blood urea nitrogen and ketone body levels vs. group C (p<0.05). After injection, animals in group M showed diabetic hyperphagia and enhanced water intake as compared to group C (p<0.05) but, despite increased feed intake, they did not gain more weight than controls (p<0.05), and consequently, their feed conversion ratio was greater. Protein and fat contents of meat and liver were not significantly different among groups (p>0.05). In conclusion, the results suggested a regulatory role of insulin in energy metabolism of ruminants by exerting two opposing effects; central catabolic and peripheral anabolic.
Objective : The present study was carried out to investigate the preventive effect of Sophorae Flos on streptozotocin - induced Diabetes mellitus. Method : Sophorae Flos was given to rats with oral administration. The experimental animals were divided into 3groups : normal group of rats, control group of Streptozotocin-induced diabetic rats, sample group with Sophorae Flos . The effect of Sophorae Flos on Streptozotocin-induced diabetes was observed by measuring the survival rate of rats, weight of rats, FER, blood glucose, each organ weight of rat, total cholesterol, HDL, GOT, GPT & creatinine. Result : Streptozotocin caused hyperglycemia and hypoinsulinemia by a selectively destroying pancreatic ${\beta}-cell$. Sophorae Flos treatment don't protected them from hyperglycemia and hypoinsulinemia. Organ weight liver, kidney, heart & spleen shows no significant changes. Sophorae Flos significantly don't recoverd the increase of several biochemical parameters such as blood glucose, total cholesterol, HDL, GOT, GPT, antioxidant & creatinine is vice versa. Conclusion : Sophorae Flos extract group did not show significant decrease than Streptozotocin control group.

Method : Agrimonia pilosa Ledebour was given to rats with oral administration. The experimental animals were divided into 3groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with Agrimonia pilosa Ledebour. The effect of Agrimonia pilosa Ledebour on STZ-induced diabetes was observed by measuring the survival rate of rats, weight of rats, FER, blood glucose, each organ weight of rat, total cholesterol, HDL, GOT, GPT & creatinine. Result : STZ caused hyperglycemia and hypoinsulinemia by a selectively destroying pancreatic ${\beta}-cell$. Agrimonia pilosa Ledebour treatment don't protected them from hyperglycemia and hypoinsulinemia. Organ weight only liver weight decreased but kidney, heart & spleen shows no significant changes. Agrimonia pilosa Ledebour significantly don't recoverd the increase of several biochemical parameters such as blood glucose, total cholesterol, HDL, GOT, GPT & creatinine is vice versa. Conclusion : Agrimonia pilosa Ledebour extract group did not show significant difference compared with STZ-induced Diabetes Mellitus group.
The antidiabetic effect of Amomum xanthioides(A. xanthioides) extract was investigated. Alloxan caused the hyperglycemia and hypoinsulinemia by a selective destruction of pancreatic ${\beta}$-cell. Pretreatment of mouse with A. xanthiodies extract for 2 days prior to alloxan administration completely protected hyperglycemia induced by alloxan. In addition, administration of A. xanthioides extract to alloxan-induced diabetic mouse significantly abolished hyperglycemia, hypoinsulinemia, and, the reduction of size and number of insulin-secreting cells induced by alloxan. Administration of A. xanthioides extract to alloxan-induced diabetic mouse rapidly increased pancreatic Reg gene expression to 7 days, and then decreased. In alloxan-diabetic mouse. Reg gene expression was increased at 3 days after alloxan injection, and sustained until 24 days. The present results indicate that A. xanthioides extract contains potentially effective components exhibiting both protection and treatment of alloxan-induced diabetes. These results suggested that the antidiabetic effect of A. xanthoides extract may be mediated through the regeneration of pancreatic ${\beta}$-cells.
한약복합처방(SHP)의 약침 및 경구투여가 STZ으로 유발된 흰쥐의 당뇨병과 항산화능에 미치는 영향을 연구한 결과 다음과 같은 결론을 얻었다. 1. In vitro 에서 SHP은 ${\alpha}$-glucosidase 저해 및 DPPH 라디칼 소거 활성을 나타냈으며, t-BHP에 의한 신장 피질 조직에서의 지질 과산화 생성을 억제하였다. 2. SHP은 대조군에 비해 STZ에 의한 혈청 중 insulin 함량 저하를 유의성있게 증가시켰으며, 혈청 중 glucose 함량변화에 있어서도 유의성 있게 감소시켰다. 3. SHP은 대조군에 비해 STZ에 의해 상승된 혈청 중 triglyceride 함량을 유의성있게 감소시켰고, 혈청 중 total cholesterol 함량을 감소시키는 경향을 보였으나 유의성은 없었다. 4. STZ 투여로 인해 혈청 내 지질과산화물의 함량이 증가되었으며 SHP의 약침 및 경구투여로 감소되는 경향을 보였으나 유의성은 없었다. 5. STZ에 의해 증가된 catalase 활성은 SHP에 의해 감소되는 경향을 보였으나 유의성은 없었으며, glutathione 활성 역시 SHP에 의해 감소되는 경향을 보였으나 유의성은 없었다.
CD4+ regulatory T cells (Tregs) are essential for normal immune surveillance, and their dysfunction can lead to the development of autoimmune diseases, such as type-1 diabetes (T1D). T1D is a T cell-mediated autoimmune disease characterized by islet b cell destruction, hypoinsulinemia, and severely altered glucose homeostasis. Tregs play a critical role in the development of T1D and participate in peripheral tolerance. Pluripotent stem cells (PSCs) can be utilized to obtain a renewable source of healthy Tregs to treat T1D as they have the ability to produce almost all cell types in the body, including Tregs. However, the right conditions for the development of antigen (Ag)-specific Tregs from PSCs (i.e., PSC-Tregs) remain undefined, especially molecular mechanisms that direct differentiation of such Tregs. Auto Ag-specific PSC-Tregs can be programmed to be tissue-associated and infiltrate to local inflamed tissue (e.g., islets) to suppress autoimmune responses after adoptive transfer, thereby avoiding potential overall immunosuppression from non-specific Tregs. Developing auto Ag-specific PSC-Tregs can reduce overall immunosuppression after adoptive transfer by accumulating inflamed islets, which drives forward the use of therapeutic PSC-Tregs for cell-based therapies in T1D.

In our previous study (Kim et al, 1991), GLUT 4 protein content correlated negatively with plasma glucose levels in skeletal muscles of STZ-induced diabetic rats. Thus, in this study, to confirm whether expression of GLUT 4 correlate negatively with degree of hyperglycemia, we measured levels of GLUT 4 mRNA in red and white gastrocnemius muscles in STZ-induced mild and severe diabetic rats. Rats were randomly assigned to control, mild, and severe diabetic groups, and the diabetes was induced by intraperitoneal administration of STZ. The experiment was carried out 10 days after STZ administration. Gastrocnemius red and white muscles were used fur the measurement of GLUT 4 expression. Plasma glucose levels of mild and severe diabetic rats were increased compared to control rats (control, mild, and severe diabetes; $6.4{\pm}0.32,\;9.4{\pm}0.68,\;and\;22.0{\pm}0.58$ mmol/L, respectively). Plasma insulin levels of mild and severe diabetic rats were decreased compared to control rats (control, mild, and severe diabetes; $198{\pm}37,\;l14{\pm}14,\;and\;90{\pm}15$ pmol/L, respectively). GLUT 4 mRNA levels of gastrocnemius red muscles in mild and severe diabetic rats were decreased compared to control rats ($64{\pm}1.2%\;and\;71{\pm}2.0%$ of control, respectively), but GLUT 4 mRNA levels in gastrocnemius white muscles were unaltered in diabetic rats. In summary, GLUT 4 mRNA levels were decreased in STZ-induced diabetic rats but did not correlated negatively with degree of hyperglycemia, and this result suggest that the regulatory mechanisms of decreased GLUT 4 mRNA levels are hypoinsulinemia and/or other metabolic factor but not hyperglycemia. And regulation of GLUT 4 expression in STZ-induced diabetes between red and white enriched skeletal muscles may be related to a fiber specific gene regulatory mechanism.
A folk prescription consisting of Alnus hirsuta, Rosa davurica, Acanthopanax senticosus and Panax schinseng has been used in the treatment of diabetes mellitus. The aim of the present investigation was to evaluate the antidiabetic effects of the herb formula extract (HFE) composed of Alnus hirsuta, Rosa davurica, Acanthopanax senticosus and Panax schinseng in the streptozotocin (STZ)-induced diabetic rats. The HFE was mixed in the food supply of the healthy and STZ-induced diabetic male Sprague-Dawley rats, and its effects on the body weight, water and food intake, hyperglycemia, hypolipidemic and islet structure were studied. The treatment of the rats with STZ for 6 weeks resulted in marasmus, polydipsia, polyphagia, hyperglycemia and hypoinsulinemia. In addition, the diabetic rats showed an apparent decrease in the insulin immunoreactivity and the number of ${\beta}$-cells in the pancreas. The addition of the HFE to the rats' food supply significantly lowered the serum glucose and the serum triglycerides level and preserved the normal histological appearance of the pancreatic islets. These results indicate that the HEF have a strong antidiabetic potential along with the significant hypoglycemic and hypolipidemic effects, which may be applicable in the pharmaceutical industry.
In the present study, Radix clematidis extract (RCE) was evaluated to determine if it could protect pancreatic ${\beta}$ cells against multiple low dose streptozotocin (MLDS)-induced diabetes. Injection of mice with MLDS resulted in hyperglycemia and hypoinsulinemia, which was confirmed by immunohistochemical staining. However, the induction of diabetes by MLDS was completely prevented when mice were pre-administrated with RCE. Generation of oxidative stress is implicated in MLDS, a ${\beta}$ cell specific toxin-induced islet cell death. In this context, to elucidate the mechanisms of protective effects in RCE pre-administrated diabetic mice, we investigated the expression of heme oxygenase-1 (HO-1), which is one of the anti-oxidant enzymes. MLDS-induced HO-1 expressions were significantly reduced in MLDS-treated mice. However, the decrease of HO-1 by MLDS were protected by pretreatment of RCE. The molecular mechanism by which RCE inhibits diabetic conditions by MLDS appears to involve inhibition of HO-1 expression. Taken together, these results reveal the possible therapeutic value of RCE for the prevention of type 1 diabetes progression.
