Plant regeneration depending on explant type was inves-tigated with cotyledon, hypocotyl, and leaf explants of garland chrysanthemum (Chrysanthemum coronarium L.) cultured on MS basal medium supplemented with various concentrations of SAP and NAA combination. Among the three different types of explants, hypocotyl explants grown on MS medium containing $1.0{\mu}M\;NAA,\;1.0{\mu}M\;BA\;and\;6{\mu}M\;AgNO_3$ produced the highest adventitious shoots (4.67 per explant). Hypocotyl explants not only produced more vigorous shoots, which regenerated aster than the cotyledon and leaf explants. An efficient root formation was observed in MS medium containing $3\%$sucrose. The concentration of NAA did not show significant effects on root formation. Results from this experiment suggested that hypocotyl explants were efficient for the regeneration of garland chrysanthemum.
This study was carried out to assess the potential of hypocotyl explants for multiple-propagation by intensive sweetener variety with high yielding in stevia of self-fertilization. Agronomical character of self-fertilization shown a height range 72.0~120.7 cm, branch's 4.9~56.6 in stevia. Selection of high content of Rebaudioside A (RA) from segregating population in mixing variety have a variety, 'DA1' (55.2%) from HPLC analysis. On a developing of multiple-propagation in stevia, hypocotyl explants used to select for reasonable medium under the influence of each plant growth regulators concentrations. In a Mix treatment of plant growth regulators, as Murashige and Skoog medium with 1.5 mg/L BA and 0.5 mg/L Kinetin showed the most favorable. We got the multiple-propagation of 30 shoots from one inoculated hypocotyl explants as the result of these bases. Rooting induced superbly on MS medium with 0.1 mg/L IBA with inoculated shoots. All of the rooting plants were adapted to a normal seedling. This method of multi-propagation is used effectively in Rebaudioside A productivity elevation.

In vitro cultures of watermelon were treated with antimitotic agent colchicine to induce ploidy alterations, particularly the induction of tetraploids. Explants cotyledon, embryonic end of seed, transverse sections of epicotyl and hypocotyl were cultured on MS media supplemented with BA ($1{\mu}M$) and colchicine ($0.01\%,\;0.05\%\;and\;0.1\%$). Explants were subcultured on colchicine free media after 4 and 7 days. Colchicine had negative effect on in vitro regeneration but this exhibited explants related response. However, hypocotyl section of seedlings induced maximum callus on $0.01\%$ colchicine. Shoot proliferation was more in cotyledon explants cultured on colchicine ($0.01\%$) for four days. Maximum root induction and root number were recorded in embryonic end explants. Overall, cotyledon and embryonic end explants, and low colchicine concentration ($0.01\%$) was found optimal in watermelon regeneration.
In vitro shoot regeneration from cotyledon and hypocotyl explants derived from germinating mature Impatiens seeds. The induction of organogenetic tissue was also influenced by the cotyledon and hypocotyl. Multiple shoot induction was higher in hypocotyl than in the cotyledon explant with Thidiazuron and a NAA medium.
Cotyledonary and hypocotyl explants of melon seedlings were cultured on Murashige and Skoog's (MS) medium supplemented with various concentrations of 2,4-Dichlorophenoxyacetic acid (2,4-D) and benzyladenine (B.A).Up to 22% of cotyledonary explants and 7%, of hypocotyl explants, respectively: Produced somatic embryos through intervening two types of calli: bright yellow compact (BYC) callus and pale-yellow compact (PYC) callus. BYC callus was capable of producing somatic embryos at initial culture, but it became necrotic as subrulhues proceeded. In contrast UC callus was incapable of producing somatic embryos during initial culture (first 6 weeks), but it became bright-yellow friable (BYF) callus with forming a few globular embryos after 2 months of subculture, indicating that the callus turned embryogenic. The embryogenic capacity of BYF maintained for over one year when the callus was sucultured at 4-week interval. Upon transfer onto MS basal medium the callus gave rise to numerous somatic embryos and subsequently converted to plantlets. Plantlets were transplanted to potting soil and grown to maturity in the phyotron.
To develop a selection system for regenerating plants from transformed tissues, effects of four antibiotics (kanamycin, hygromycin, carbenicillin, cefotaxime) and herbicide (phosphinotricin) on shoot regeneration from cotyledon and hypocotyl explants of Chinese cabbage (Brassica campestris L. ssp. pekinensis) were studied. For cotyledon, shoot induction was not significantly affected by kanamycin at $1mg{\cdot}L^{-1}$, but the number of shoots formed was significantly reduced at $2mg{\cdot}L^{-1}$, and no shoots were regenerated from any explants at $6mg{\cdot}L^{-1}$ or higher. Hypocotyl explants showed similar result as cotyledon. Kanamycin at $7mg{\cdot}L^{-1}$ may be adequate for selecting Chinese cabbage transformants. Hygromycin at $4mg{\cdot}L^{-1}$ or higher completely inhibited the growth and shoot regeneration of Chinese cabbage explants. Therefore, resistance gene to hygromycin may also be used as a selective marker for Chinese cabbage transformation. Carbenicillin and cefotaxime, the cephalosporin type of antibiotics, had little effect on shoot regeneration of Chinese cabbage explants. Since carbenicillin and cefotaxime have low toxicity to Chinese cabbage, they are suitable for use in tissue culture to eliminate Agrobacterium in transformation experiments after co-cultivation. Shoot regeneration from cotyledon and hypocotyl explants was significantly reduced in presence of $1mg{\cdot}L^{-1}$ phosphinotricin (PPT) and completely inhibited by $2mg{\cdot}L^{-1}$ or higher. PPT, same as antibiotics, may also be used to select transformed cells. Since Chinese cabbage is known to be recalcitrant to in vitro shoot regeneration compared to other Brassica species, even though lower levels of selectable markers result in more transformants but simultaneously allow more untransformed escapes to develop, lower levels of antibiotics and herbicides could be successfully used as a selectable marker to reduce selection pressure.

To establish high frequency shoot formation from two cultivars (Cheongyangjaerae and Myungan) of boxthorn (Lycium chinense Mill.), hypocotyl segments with cotyledons from seedlings were used as explants. High frequency adventitious shoot formation (more than 80%) were obtained from hypocotyl segments with cotyledon on MS medium supplemented with 1.0 mg/L zeatin, when precultured for 3 weeks under dark conditions followed by transfer to light conditions. But there was no shoot induction in the explants cultured without preculture under dark conditions. Roots were induced from the shoots when transferred to rooting medium supplemented with 1.0 mg/L IAA for 4 weeks. Regenerated plantlets were grown to normal mature plants in soil.

Hibiscus cannabinus, Cassia fistula and Abutilon indicum are medicinally important plants in India. Tissue culture studies have done for efficient propagation and for crop improvement in these three plants. Various explants were tried for callus induction, somatic embryogenesis and organagenesis with manipulation in culture media. Calli and somatic embryos were induced from hypocotyl explants in Hibiscus cannabinus, while in Cassia fistula and Abutilon indicum it could be obtained from leaf explants.
The biochemical change during regeneration of perilla callus were investigated by comparing total protein and lipid contents, protein band pattern in SDS-PAGE, and fatty acid composition in the calli cultured for various period(0, 1, 3, 5 and 6 weeks) Calli were induced from cotyledon and hypocotyl explants of peplants of perilla on perilla on MS medium containing BA(0.5 mg/L) and NAA(0.5mg/L). The protein contents reached the peak at 3 weeks after induction of calli, and then was decreased. Total lipid contents was decreased as the culture period increased. The band pattern of polypeptides showed that 30KD and 45KD polypeptides and 22KD and 45KD polypetides were major proteins in the cotyledon and hypocotyl explants, respectively. However increase of culture period only 30KD protein was highly accumulated.
