Contents of $\beta$-Glucan in Various Cereals and Its Functional Properties

  • A soluble dietary fiber, $\beta$-glucan, contained in oat and barley has nutritional benefits such as hypocholesterolemic effects and influences blood glucose regulation. The contents of $\beta$-glucan in both cereals range from 3 to 7% with the exception of a certain barley genotype which contains up to 16% $\beta$-glucan. $\beta$-Glucan is distributed mainly in the cell walls of endosperm and the distal (bran) portion of kernel. Various procedures have been developed for increasing the extraction yield of $\beta$-glucan. Oat gum prepared with weak alkali extractionand alcohol proecipitation following protein removal usually contains 80% $\beta$-glucan.The most commonly used method for $\beta$-glucan quantitiation is an enzymatic procedure combining lichenase plus $\beta$-glucosidase followed by measuring the amount of glucos released by glucose oxidase-peroxidase treatment. The increase in foam-and emulsion-stabilizing capacity of $\beta$-glucan is due to the increase in viscosity of the aqueous phase. Therefore, $\beta$-glucan shows great potentials as a thickener and a stabilizer.

Effect of Chicory Root Extract on Cholesterol Metabolism in Rats

  • Effects of water-soluble extract from roasted-chicory root on the cholesterol metabolism in rats fed cholesterol diet were investigated. Sprague-Dawley rats received a hypercholesterolemic diets without (control group) or with 5.0% water-soluble extract from roasted chicory root for 2 weeks. Roasted chicory extract group showed significantly higher body weight gain and food intake compared with the control group. The concentrations of total cholesterol and LDL+VLDL cholesterol in serum were significantly lower in rats fed roasted chicory extract diet. However, HDL-cholesterol concentration, and atherogenic index were not significantly affected by the dietary roasted chicory extract. Fecal net weight, fecal cholesterol, and bile acid excretion were significantly higher in the chicory extract group. The results suggest that the hypocholesterolemic effect in rats fed roasted chicory extract may be caused by an alteration in the absorption of cholesterol by an increase in the fecal excretion of cholesterol and bile acid.

A Preliminary Study on Hypocholesterolemic and Hypoglycemic Activities of Some Medicinal Plants

  • Total cholesterol level in mice with hypercholesterolemia was determined after intraperitoneal administration of the methanolic extract of some medicinal plants. From the data obtained, it was suggested that the methanolic extract of Elaeagnus crispa, Ixeris dentata, Prunus davidiana, Eriobotrya japonice, Aralia elata and Phragmites communis produced a significant hypocholesterotemic effect. In the case of the extract of Saussurea diamantiaca, on the other band, the total cholesterol level was markedly increased. The methanolic extract of Ixeris dentata, Prunus davidiana and Phragmites communes also decreased the level of blood glucose in alloxan-diabetic male albino mice while that of Eriobotrya japonica, Allium tuberosum, Houttuynia cordata and Eucommia japonica did not produce this effect.

  • Mechanisms for the hypocholesterolemic effetcs of $\beta$-glucan remain unclear. Rats were divided into 3 groups; normal control group, atherogenic group(oral administration of cholesterol 40 mg/kg/day and vitamin $D_2$320,000 IU/kg/day),${\beta}$-glucan treatment group(oral administration of atherogenic treatment and ${\beta}$-glucan 0.135 g/kg/day). The ${\beta}$-glucan treatment group showed moderate increases of serum lipids concentration compared with atherogenic group. In histopathological examination, aortas showed no critical lesions. The total fecal neutral sterols and bile acids excreted for 6 days was increased compared with both normal and atherogenic group. These result\ulcorner suggest that mechanisms for the hypocholesterolemic effect of ${\beta}$-glucan on rats were due to the inhibition of cholesterol absorption in the intestinal lumen and acceleration of cholesterol catabolism in the liver.

Isolation and Structure Determination of a Cholesterol Esterase Inhibitor from Ganoderma lucidum

  • Bioassay-guided fractionation of a methanol extract of Ganoderma lucidum gave a pure cholesterol esterase inhibitor. On the basis of spectroscopic analysis and comparison with data from the literature, the structure of this compound was identified as $5{\alpha},8{\alpha}$-epidioxyergosta-6,22-dien-$3{\bet}$-ol (compound I). This compound inhibited cholesterol esterase activity with an $IC_{50}$ value of $42{\mu}M$. Lineweaver-Burk plot analysis revealed that compound I is a noncompetitive inhibitor. The findings of this study suggest that compound I may be the active principle of the hypocholesterolemic effect of Ganoderma lucidum.

Hypocholesterolemic Activity of Bifidobacteria Isolated from a Healthy Korean

  • This study was undertaken to investigate the hypocholesterolemic activity of Bifidobacteria (B. breve K-110, B. breve K-111, and B. infantis K-525) isolated from a healthy Korean. The administration of B. breve K-110 and K-111 with a high cholesterol diet significantly protected the increase of serum total cholesterol and LDL cholesterol relative to that of a high cholesterol diet alone. Such a diet supplemented with 0.5% B. breve K-111 decreased serum total cholesterol and LDL cholesterol to 57 and 55%, respectively. The administration of Bifidobacteria also significantly inhibited the lipid-deposited surface in the aorta. The normalizing activity of serum cholesterol level in cholesterolemic rats was accelerated by Bifidobacteria. The normalizing activity of B. breve K-111 on serum cholesterol level was superior to that of B. breve K-110. These results suggest that Bifidobacteria in the human intestine playa role in the prophylactics of arteriosclerosis.

고들빼기의 고콜레스테롤혈증 개선효과 (Further Study on the Anti-hypercholesterolemic Effect of Ixeris sonchifolia)

  • Effect of various fractions prepared from the methanol extract of Ixeris sonchifolia and flavonoids isolated from the extract on hypercholesterolemic mice were examined. When various fractions $(CHCl_3,\;Interphase,\;EtOAc,\;n-BuOH\;and\;H_2O)$were administered to mice, the Interphase material produced a significant activity. Total cholesterol level in the Interphase material-treated group given doses of 25 and 50mg was decreased significantly by as much as 16% and 30%, respectively. Among the isolated flavonoids, cynaroside caused a significant hypocholesterolemic activity.

Mevinolin Production by Monascus pilosus IFO 480 in Solid State Fermentation of Soymeal

  • Mevinolin, a fungal metabolite, is a potent inhibitor of 3-hydroxy-methyl-3-glutaryl-coenzyme A (HMG-CoA) reductase, the rate-controlling enzyme in cholesterol biosynthesis. In this investigation, the optimum factors for mevinolin production by Monascus pilosus IFO 480 in soymeal fermentation were studied. The highest yield of mevinolin, 2.82 mg mevinolin per g dry weight, without citrinin (a toxic fungal secondary metabolite) was obtained after 21 days of fermentation at $30^{\circ}C$ at 65% moisture content, particle size 0.6-0.9 mm, and initial substrate pH of 6.0. Mevinolin was present in the fermentation substrate predominantly in the hydroxycarboxylate form (open lactone, 92.1-97.3%), which is currently being used as a hypocholesterolemic agent.

식이섬유와 콜레스테롤 대사 (Dietary Fiber and Cholesterol Metabolism)

  • 우리나라 주요 사망원인중의 하나가 되고 있는 혈관질환의 주된 위험인자로 고콜레스테롤 혈증이 지적되고 있다. 콜레스테롤 저하효과가 있는 식이 인자에는 단백질의 종류, 지방의 종류와 양, 식이섬유, 우유, 칼슘, flavonoid 등이 있다. 특히 수용성 식이 섬유의 콜레스테롤 저하효과는 현저하며 동물실험과 인체실험을 통하여 확인되었다. 혈장 콜레스테롤 저하효과가 현저한 식이섬유에는 psyllium husk, pectin, oat fiber, guar gum, sodium alginate 등이 있으며, 농도에 비례하여 그 효과가 커지는 것은 아닌듯하다. 수용성 식이섬유의 콜레스테롤 저하효과에 대한 정확한 기작은 알려져 있지 않으나 몇 가지 가설이 제시되고 있다. 첫째, 식이섬유가 장에서 점성용액을 형성하여 지질 흡수를 저해함으로써 혈장과 간 콜레스테롤 농도를 낮춘다는 것이며, 둘째, 체내 콜레스테롤이 체외로 배설되는 유일한 경로인 담즙산의 소장흡수를 방해하고 변으로의 배설을 증가시켜 체내 콜레스테롤 pool 크기를 감소시킨다는 것이다. 세째, 대장미생물에 의해 생성된 식이섬유의 발효부산물인 short chain fatty acids가 콜레스테롤의 합성을 저해한다는 것이다. 이외에도 여러 가설들이 제시되고 있으며, 이러한 가설들이 서로 상호 배제적인 것은 아니며, 단독으로 혹은 복합적으로 작용하는 것으로 보인다.
