Enhancement of Cosmeceutical Activities of Berberis koreana Bark by High Pressure and Ultrasonification Extraction Processes (초고압 및 초음파 추출공정을 이용한 매자나무 수피의 향장활성 증진)

  • This study was performed to investigate the enhancement of cosmeceutical activities of Berberis koreana bark by different extraction processes. The extracts are WE (water extract at $100^{\circ}C$, control), USE (ultrasonification for 1 hours at $60^{\circ}C$ with water), HPE (high pressure for 5 minutes at $60^{\circ}C$ with water) and USE + HPE (ultrasonification process for 1 hours after high pressure for 5 minutes at $60^{\circ}C$ with water), respectively. The cytotoxicity of the extracts was in the range of 24.02~26.94% at 1.0 mg/ml concentration. The USE + HPE showed the lowest cytotoxicity. Compared to the WE, total phenolic and flavonoid contents in the USE + HPE increased to 121.5% and 154.2%. The USE + HPE showed the highest activity at 1.0 mg/ml concentration in 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging, inhibition activity of xanthine oxidase and superoxide dismutase (SOD)-like test, respectively. Tyrosinase inhibition of WE, USE, HPE and USE + HPE at 1.0 mg/ml concentration was measured as 17.72, 19.62, 22.83 and 24.16%, respectively. Hyaluronidase inhibition activities of the USE + HPE were higher than 20.8%~29.5% of the WE. Our results suggested that the extracts from ultrasonification process after high pressure extraction has relatively high cosmeceutical activities, and that the bark of Berberis koreana could be considered as a candidate of new functional cosmetic agents.

Inhibitory activities on biological enzymes of extracts from Oplismenus undulatifolius (주름조개풀(Oplismenus undulatifolius) 추출물의 생리활성 효소 억제 효과)

  • The phenolic contents which were extracted with water and 70% ethanol from O. undulatifolius were 7.7, 10.1 mg/g, respectively. The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of water and ethanol extracts were 78, 82% at $50{\mu}g/mL$ phenolics, respectively. The 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical cation decolorization activity were 92, 76% at $100{\mu}g/mL$ phenolics. Antioxidant protection factor in water and ethanol extracts at $200{\mu}g/mL$ phenolics were 1.51 and 2.08 PF, respectively. Thiobarbituric acid reactive substance were 84% in water extracts and 99% in ethanol extracts at $50{\mu}g/mL$ phenolics, respectively. The inhibition activity on ${\alpha}-Glucosidase$ was 44% in ethanol extracts at $200{\mu}g/mL$ phenolics. The inhibition activity on ${\alpha}-amylase$ was 37-88% in water extracts at $50-200{\mu}g/mL$ phenolics. The tyrosinase inhibition activity as whitening effect were 82% in ethanol extracts. The elastase inhibition activity were 4, 61% in water and ethanol extracts, respectively. The collagenase inhibition activity of antiwrinkle effect showed an excellent wrinkle improvement effect as 39% in water extracts and 67% in ethanol extracts at $200{\mu}g/mL$ phenolics, respectively. The hyaluronidase inhibition activity as anti-inflammation effect of ethanol extracts was confirmed to 46% of inhibition at $200{\mu}g/mL$ phenolic. The astringent effect of water and ethanol extracts was confirmed to 13, 32% of effect at $200{\mu}g/mL$ phenolic, respectively.

Enhancement of Immuno-modulatory of Centella asiatica L. Urban with Edible Polymer through Nano-encapsulation Process (병풀 추출물의 식용 나노입자화를 통한 면역 활성 증진)

  • Phosphatidylcholine was used to encapsulate aqueous extracts of Centella asiatica, and its biological activity was compared with another aqueous extracts. Nanoparticle of C. asiatica was made by encapsulation to w/o type spherical liposome which of aqueous extracts seized with oil phase as 78.2 nm average diameter. Cytotoxicity of the nanoparticle was measured on human skin fibroblast cells, CCD-986sk, and showed lower cytotoxicity on 1.0 mg/$m{\ell}$ of highest concentration as 28% than that of another extracts. The nanoparticle showed the highest promotion of human B and T cell growth up to 138% and 135%, respectively, compared to the control. and the NK cell growth was promoted up to 8% higher than the control in proportion to secretion of IL-6 and TNF-$\alpha$ from immune cell growth. Also nanoparticle showed highest inhibition activity of hyaluronidase on 1.0 mg/$m{\ell}$ of highest concentration as 60.5%. It seems that because of enhanced biological application of aqueous extracts on cell through nano-encapsulation process.

The Effects of Alpinia katsumadai Extract on Anti-inflammation and Melanogenesis

  • Several solvent extracts from Alpinia katsumudai were prepared and their various anti-inflammatory activities were evaluated. Alpinia katsumadai extract showed high various anti-inflammatory effects among the 8 medicinal plant extracts, Butanol extract from Alpinia katsumndai showed a potent anti-oxidative and free radical scavenging activities, Free radical scavenging effect of butanol fraction of Alpinia katsumadai($IC_{50}$/ : 50$\mu$g/m$\ell$) was higher than butylated hydroxytoluene($IC_{50}$/ : 50$\mu$g/m$\ell$) and ascorbic acid($IC_{50}$/ : 22$\mu$g/m$\ell$). Alpinia katsumadai butanol fraction exhibited relatively high antioxidative activity($IC_{50}$/ : 335$\mu$g/m$\ell$) compared to ascorbic acid. The inhibitory effect of Alpinia katsumadal ethanol extract on elastase exhibited 10 to 78% at 100 to 1000$\mu$g/m$\ell$ concentration , tile $IC_{50}$/ values with 465.7$\mu$g/m$\ell$ for porcine pancreatic elastase(PPE) and 481.9$\mu$g/m$\ell$ for human leukocyte elastase(HLE), respectively The Alpinta katsumadai extract inhibited effectively hyaluronidase activity($IC_{50}$/ 335$\mu$g/m$\ell$), and showed inhibition in vitro on delayed hypersensitivity when it was topically applied. These results suggest that Alpinia katsumadai extract may reduce inflammatory skin trouble. The Alpinia katsumadai extract also showed higher Inhibitory effect of melanin biosynthesis on cultured melanoma cell compared to arbutin and kojic acid.

Biological Activity Investigation, and Phenol Compounds Isolation from Barks of Prunus Sargentii R. (산벚나무 수피추출물의 생리활성 및 페놀성 화합물 분리)

  • Prunus sargentii of Rosaceae familiy, has been reported to have radical scavenging activity and anti-inflammatory effect. On these facts, this research was conducted to evaluate pharmaceutical activities of the bark extracts P. sargentii. Free radical scavenging activities of fraction (Fr) -5${\sim}$10 isolated from P. sargentii was higher than 80% respectively at 10ppm. The superoxide dismutase (SOD)-like activity of Fr-5, 9 were about 97, 84%, respectively at 1,000 ppm. Xanthine oxidase inhibitory effect of Fr-9, 10 were about 75, 78%, respectively at 1,000 ppm. The tyrosinase inhibitory effect related to skin-whitening was 72, 68% in Fr-2, 9 isolated from Prunus sargentii R. at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 98% for Fr-8 at 500 ppm. Isolation of the methanol soluble fraction from P. sargentii yielded two major phenol compounds, (-)-epicatechin and taxifolin. The structure of the compound was certainly determined by chemical analyses, as well as NMR and Mass spectroscopy. The present study was carried out in a search for new cosmetic material from the bark from P. sargentii. and, (-)-epicatechin and taxifolin were isolated as active principles. So P. sargentii R. methanolic extracts may be used for the cosmetic material.

Isolation and Characterization of Elastase Inhibitor from Areca catechu (빈랑으로부터 Elastase 저해물질의 분리 및 특성조사)

  • We have previously screened 150 medicinal plants on the inhibition of elastase and found a significant inhibitory effects of the extracts of Areca catechu L. on the aging and inflammation against the skin tissues. To isolate and identify the compounds having biological activity, we was further purified by each of the solvent fractions, silica gel column chromatography, preparative TLC and reversed-Phase HPLC. Peak in HPLC, which coincided with the inhibitory activity against elastase, was identified as Phenolic substance using various colorimetric methods, UV, and IR. $IC_{50}$/ values of phenolic substance purified from Areca catechu were 26.9 $\mu\textrm{g}$/$m\ell$ for porcine pancreatic elastase (PPE) and 60.8 $\mu\textrm{g}$/$m\ell$ for human neutrophil elastase (HNE). This Phenolic substance showed more potent activity than those of reference compounds, oleanolic acid (76.5 $\mu\textrm{g}$/$m\ell$ for PPE, 219.2 $\mu\textrm{g}$/$m\ell$ for HNE) and ursolic acid (31.0 $\mu\textrm{g}$/$m\ell$ for PPE, 118.6 $\mu\textrm{g}$/$m\ell$ for HNE). According to the Lineweaver-Burk Plots, the inhibition against both PPE and HNE by this phenolic substance was competitive with substrate. Phenolic substance from Areca catechu exhibited high free radical scavenging effect ($SC_{50}$/ : 6 $\mu\textrm{g}$/$m\ell$) and inhibited effectively hyaluronidase activity ($IC_{50}$/: 210 $\mu\textrm{g}$/$m\ell$). These results suggest that the Phenolic substance Purified from Areca catechu showed anti-aging effect by protecting connective tissue proteins.

Anti-Oxidant and Antiinflammatory Effects of Rosa multiflora Root (찔레나무뿌리(Rosa multiflora root)의 항산화 및 항염증효과)

  • Rosa multiflora thunberg belonging to Rosaceae is widely distributed in East Asia including Korea and Japan, and has been reported to have tormentic acid and rosamultin. To develop a new natural anti-inflammatory agent for cosmetics, we investigated the inhibitory effects of inflammation in Rosa multiflora root (R. multiflora root). The biological activity and anti-inflammatory effects were investigated by water, ethanol, methanol and acetone extracts of R. multiflora root. The measurements of polyphenol content from R. multiflora root were highest in water and acetone extracts, at 57.48 ${\pm}$ 0.88 mg/g and 67.05 ${\pm}$ 0.56 mg/g, respectively. The result of DPPH, ABTS and superoxide anion radical scavenging effects showed over 50% efficacy at 50 ${\mu}g/ml$ in ethanol, methanol and acetone extracts. Hyaluronidase inhibition effect showed over 60% efficacy at 500 ${\mu}g/ml$ in ethanol, methanol, and acetone extracts. Nitric oxide radical inhibition effect of R. multiflora root ethanol extracts showed over 30% efficacy at 500 ${\mu}g/ml$. We investigated the effect of R. multiflora root extracts on nitric oxide (NO) production of inducible nitric oxide synthase (iNOS) in LPS-induced RAW 264.7 macrophage cells. The result showed that R. multiflora root extracts have an inhibitory effect on NO production and iNOS expression and also can be used as an anti-inflammatory agent. These antioxidant and anti-inflammatory effects of R. multiflora root show applicant potential application as a functional cosmetic material.

Biological Activities of Extracts from Cornus kousa Fruit (산딸나무(Cornus kousa) 열매 추출물의 생리활성)

  • The extracted phenolic compounds from Cornus kousa fruit for biological activities as functional resources were examined. The phenolic compounds which were extracted with water and 40% ethanol from Cornus kousa fruit were $7.04{\pm}0.27$ and $4.47{\pm}0.18mg/g$, respectively. The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of water and ethanol extracts were 84% and 86% at $50{\mu}g/mL$phenolics, respectively. The 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid radical decolorization activity of water and ethanol extracts were 84 and 95% at $100{\mu}g/mL$ phenolics, respectively. Antioxidant protection factor in water and ethanol extracts at $50{\mu}g/mL$ phenolics were 1.93 and 1.82 PF, respectively. Thiobarbituric acid reactive substance were 69% in water extracts and 89% in ethanol extracts at $150{\mu}g/mL$ phenolics. The inhibition activity on xanthine oxidase in water and ethanol extracts was 34 and 60%, respectively. The inhibition activity on ${\alpha}$-glucosidase was 29% in water extracts and 87% in ethanol extracts. The tyrosinase inhibitory activity was 19% in ethanol extracts. The collagenase inhibition activity of anti-wrinkle effect showed an excellent wrinkle improvement effect as 53% in water extracts and 77% in ethanol extracts at $200{\mu}g/mL$ phenolics. The hyaluronidase inhibition activity as antiinflammation effect of water extracts was confirmed to 34% of inhibition at $200{\mu}g/mL$ phenolic. The results can be expected extracts from Cornus kousa fruit to use as functional resource for antioxidant, antigout, inhibitor of carbohydrate degradation, antiwrinkle activity and antiinflammation activity.

Quality characteristics and biological activity of Yanggaeng with Glechoma hederacea var. longituba Nakai powder (긴병꽃풀의 생리활성과 이를 첨가한 양갱의 품질특성)

  • The objective of this study is to investigate the quality characteristics of Yanggaeng by using the functional properties of Glechoma hederacea (GH). Sample was dried at $50^{\circ}C$ dry oven. The results of the study were as follows : The Phenolic compounds of GH was $12.99{\pm}0.3mg/g$ in water extract (GHWE), $3.14{\pm}0.07mg/g$ in 70% ethanol extract (GHEE). The antioxidant activity of GH was determined in various phenolic concentrations at $50-200{\mu}g/mL$. DPPH activities of GHWE and GHEE were 77.16-78.24% and 73.04-77.00%, respectively. The ABTS were 84.35-99.75% and 83.74-99.55%. The anti-oxidant protection factor (PF) were 1.54-1.62 PF and 1.62-2.09 PF and TBARS were 42.93-94.09% and 91.05-95.19%, respectively. Tyrosinase inhibitory activity of GHEE increased concentration dependently. Hyaluronidase inhibition activity of GHEE and GHWE, showing that there were increasing pattern depending on the increases in the phenolics concentration of GH. In texture, Hardness and springiness were significantly different in the control and 2% groups, but cohesiveness and chewiness did not show any significant difference. In color, L value decreased in proportion to concentration, and a and b values did not change. Sensory characteristics showed that the 1% group had the highest score and the 2% group had the lowest score. Thus, when the GHP Yanggaeng was prepared, in consideration of its sensory characteristics, and at appropriate concentration on below 1%.

Anti-oxidation, anti-inflammation, anti-wrinkle, and pore-tightening effects of phenolic compounds from Aeonium sedifolium leaves (소인제(Aeonium sedifolium) 잎 유래 phenolic 성분의 항산화, 피부주름생성 억제, 항염증 및 모공 수축 효과)

  • The succulent plant Aeonium sedifolium leaves contain several compounds that are of interest for their cosmetic uses on the skin. This study measured the inhibitory effects of enzyme production and antioxidant, astringent effects and skin wrinkles using Aeonium sedifolium leaves (ASL). The total phenolics compounds (TPC) content of ASL under optimal extraction conditions was 34.49 mg/g for hot water extract (ASLW) and 61.64 mg/g for 50% ethanol extract (ASLE). The ASLW and ASLE extracts were freeze-dried, powdered, and used as solids. TPC content, 2,2-diphenyl-1-picrylhydrazy (DPPH) radical scavenging activity, and 2,2'-azinobis (3-ethylben-zothiazoline 6-sulfonate) (ABTS) radical inhibition of the ASL phenolics were tested. The DPPH radical scavenging activities of ASLW and ASLE were tested at a TPC of 100 ㎍/mL. ABTS radical inhibition showed antioxidant activity of 100.00% in ASLW and ASLE, and the antioxidant protection factor of ASLW and ASLE was 1.07 and 1.22, respectively. The thiobarbituric acid-reactive substance (TBARS) inhibitory activity of ASLW and ASLE was 77.00%. The elastase inhibitory activity of ASLE was 69.03%, and collagenase inhibition activity for ASLW and ASLE was 29.82% and 54.76%, respectively. The astringent effect of ASLE was 89.82% at a TPC of 200 ㎍/mL. Thus, we concluded that ASL has the potential as a functional cosmetic ingredient with anti-aging effects on the skin.
