Studies on Biological Activity of Wood Extractives (X) - Antifungal Compounds of Hovenia dulcis - (수목추출물의 생리활성에 관한 연구(X) - 헛개나무 목부의 항균활성 물질 -)

  • Antimicrobial activities of plant extractives were investigated to develop a natural fungicide. Two stilbenoids and five flavonoids were isolated from wood extractives of Hovenia dulcis (Rhamnaceae) which had been selected due to its high antifungal activity among the tested species. The chemical structures of isolated compounds were determinded as : 5-hydroxy-7-methoxyflavone, 5,7-dihydroxyflavone (chrysin), 5,7-dihydroxyflavanone (pinocembrin), 3,5,7-trihydroxyflavanone (pinobanksin), 3,4',5,7-tetrahydroxyflavanone (aromadendrin), 3-hydroxy-5-methoxystilbene and 3,5-dihydroxystilbene (pinosylvin) on the basis of Mass and NMR spectroscopic data. According to the results of antifungal test, 3-hydroxy-5-methoxystilbene was evaluated as the strongest antifungal compound among the tested compounds and next were pinocembrin and pinosylvin, but those also had high hyphal growth inhibition activities against C. parasitica, T. versicolor, T. palustris and T. viride. However, pinobanksin, 5-hydroxy-7-methoxyflavone, chrysin and aromadendrin showed very low antifungal activity. In this regard, it could inferred that high antifungal activity of wood extractives of H. dulcis were derived from 3-hydroxy-5-methoxystilbene, pinocembrin and pinosylvin, respectively.

Physicochemical properties and physiological activity of bean sprouts extract containing Hovenia dulcis Thunb concentrates

  • Reactive oxygen species are the byproducts of metabolic processes in the body, However, excessive amount may cause side effects such as cancer. Therefore, to reduce the production of these species, but their long-term administration at high doses may induce side effects. Hence, natural materials with antioxidant activities are attracting attention. Two of these natural materials are soybean sprouts and Hovenia dulcis Thunb. fruits, but few studies have evaluated the effects of their combination. Thus, we prepared a soybean sprout extract containing 1.5% H. dulcis Thunb. fruit concentrate (BHM) to develop a functional food material derived from natural products and then confirmed its physicochemical properties and physiological activity. Among the organic acids detected in BHM, malic acid exhibited the highest content of 1,451.03 ppm, and the main free sugars were glucose (645.48 ppm) and fructose (738.11 ppm). Taurine was the most abundant free amino acid at a concentration of 11.95 ppm, followed by those of arginine (10.97 ppm) and glutamic acid (10.16 ppm). Analyses of the mineral components revealed large amounts of Zn and Fe in BHM, and the respective total polyphenol and flavonoid contents in BHM were 957.16 and 601.93 ppm. The DPPH radical and H2O2 scavenging activities and reducing power indicated excellent antioxidant efficacy compared to the positive controls. Furthermore, blood alcohol and acetaldehyde concentrations were measured to confirm the hangover-relieving effects of BHM, with both significantly decreased (p<0.05). BHM displays potential for development as a functional food, and the results of this study may be used as basic data in further research.

Increased Alcohol Decomposition Efficacy of Hoveina dulcis Extract by Carbohydrate-Hydrolyzing Enzymes (당 분해 효소를 이용한 헛개나무 열매 추출물이 알코올 분해에 미치는 영향)

  • In this study, increased alcohol decomposition efficacy (ADH) of Hoveina dulcis extract by Carbohydrate-Hydrolyzing Enzymes was investigated. Carbohydrate decomposition enzymes such as Maxinvert (Invertase), Optidex L-400 (Glucoamylase) and Rohament CL (Cellulase & Pectinase) were added to Hoveina dulcis extract at different concentrations (0.01, 0.05, 0.1, 0.5 and 1%) for 48 hrs, after which samples were taken every 6 hrs for determination of ADH activity. As the enzyme concentration became higher, ADH activity also increased. Especially, the addition of 1% Rohament CL increased enzyme activity to 76% at 30 hrs incubation, after which the increase in activity stopped. In the rat and human body experiment, enzymatic decomposition of Hovenia dulcis extract by addition of 1% Rohament CL was also effective in decreasing serum alcohol concentration and respiration. Especially, in the early stage after alcohol consumption, the efficacy of enzyme treatment of Hovenia dulcis extract was more effective. These results show that if the glycoside forms of active compounds such as flavonols in Hovenia dulcis extract are converted into aglycone forms, alcohol decomposition capability can be enhanced.

  • Antioxidative activities of Methanol and Ethanol extract of branch, bark, leaves, fruits, wood of Hovenia dulcis THUNBER var. koreana Nakai were analyzed for total poly phenol, flavonoid, antioxidant capacity and DPPH free radical scavanging activitiy. The most total polyphenol and flavonoid contents measerd extract from bark and fruits. Extract from fruits and bark show hight on antioxidant capacity and DPPH free radical scavanging activitiy. There was a good correlation between total polyphenol or flavonoid contents and antioxidant activity.

  • The water extracts of Hovenia dulcis Thunb. showed significant hepatoptotective activity against D-galactosamine/LPS and $CCl_4-toxicity$. The extracts significantly inhibited the elevation of serum ALT, AST and LDH levels. The concentration of blood alcohol were decreased from 2hr after injected in rats with Hovenia dulcis extracts.

  • The ADH-like and ALDH-like activities in the Hovenia extracts were studied. The Hovenia extracts have an high effects on the alcohol and acetealdehyde degradation. The ADH-like and ALDH-like activities in the Hovenia extract were $1.14\;unit/{\mu}{\ell}$, $0.33\;unit/{\mu}{\ell}$ respectively. The Hovenia extracts were decreased concertration of fusel oil by biochemical function of alcohol hydrolysis. In this study, antihepatotoxic and anticancer activity of Hovenia extracts were inviestigated. The ethanol extracts have a highest GST activity. Furthermore the growth of SNU-398 cancer cells were inhibited by the addition of Hovenia extracts. The culture media mixture to Hovenia extract of $1\;mg/m{\ell}$ was not observed connective cell and adhere to flat of liver cancer cell.

Effect of Amur Long-tailed Goral on the Germination of Hovenia dulcis Thunb. (산양이 헛개나무 종자의 발아에 미치는 영향)

  • Amur's long-tailed goral(Naemorhedus Caudatus) winter scats inhabitants of Wolaksan National Park were surveyed between 2006 and 2009. The scats included seeds of Hovenia dulcis and we confirmed germination of seed from the scats in the spring. 600 dulcis seeds goral scats were collected in the same location and period and 600 normal seeds were also observed in order to study the effect of goral on germination of dulcis. All of the seed(totally 1,200) were cultured in the same condition. As a result, germination rates of seeds from the goral scats and normal seeds were 32.5% and 0.8% respectively. So we confirmed that dulcis seeds in goral habitats germinate approximately 40 times more than without goral.
