Root-knot nematodes (Meloidogyne spp.) are major plant pathogens that cause reductions in yield and quality of several solanaceous crops, including pepper (Capsicum spp.). These losses can be averted through planting of resistant cultivars. Plants are defined as resistant when they suppress nematode reproduction. In this study, the resistance degrees of 102 commercial cultivars of chili pepper (Capsicum annuum) to a root-knot nematode, Meloidogyne incognita, were evaluated by comparing the number of egg masses on their roots to those of 'PR huimangchan', a highly susceptible cultivar that exhibited the most egg masses of the chili pepper cultivars evaluated. Among these cultivars, forty-four (43.1%) showed resistance to M. incognita and eighteen (17.6%) were moderately resistant. The other cultivars (39.3%) were determined to be susceptible. For further study, six chili pepper cultivars (i.e., Gangryeokjosenggeon, Shinsegae, Muhanjilju, PR Bulrocho, PR Huimangchan, and Jjang) with different levels of resistance to the nematode were selected. Changes in resistance of the six cultivars under several conditions, such as inoculum concentration, plant growth stage, and cultivation period after transplanting were investigated. We found that an efficient screening method for resistance of chili pepper to M. incognita is to transplant the chili pepper seedlings 7 days before inoculation, to inoculate 28-day-old plants with M. incognita by loading 5,000 eggs per plant into the pot of soil, to cultivate the plants in a greenhouse ($25{\pm}5^{\circ}C$) for 45-60 days, to measure the number of egg masses on roots of the seedlings, and then to determine the resistance response of the plants by comparing the number of egg masses on the roots with a reference-susceptible cultivar 'PR huimangchan'.
This study was carried out to compare the pattern of mutant variation and to evaluate the characteristics of mutants obtained by gamma irradiation in rose 'Kardinal'. Forty four rooted cuttings of 'Kardinal' were irradiated at 70 Gy gamma-ray dose from a $^{60}Co$ source to induce mutants in 2002. The irradiated plants were planted in field, and observed spotting of petal color mutants from 2002 to 2004. Four different kinds of mutant twigs with each different color flower were obtained from the irradiated 'Kardinal' with red petal. After being identified to be a stable mutant from 2004 to 2008, each mutant line propagated by cutting was hydroponic-cultured to evaluate the characteristics in the greenhouse from 2008 to 2009. Four mutant lines obtained from 'Kardinal' with red petal (Red group, 44A, 45B) include KA1 with light pink petal (Red group, 55B-55D), KA2 with pink petal (Red group, 63A-63B), KA3 with deep pink (Red purple, N57A-N57C), and KA4 with orange red (Red group, 43A-43B). Diameters of each flower in four mutant lines were different from 'Kardinal'. The line KA1 was 9.5 cm wide, and it showed the smallest diameter when compared to other mutants. While the line KA2 was the largest one with 12.5 cm 'Kardinal'. Petal number per flower was also variable among the mutants. The line KA2 had 39.8 petals being the largest number among the mutants, while the line KA1 was the lowest one compared to 35.5 petals of 'Kardinal'. Petal color was measured by using colorimeter. Brightness (L) measured at each petal of four mutants increased more than 'Kardinal'. CIE Lab values, a and b decreased more than 'Kardinal' at the petal color of three mutants except the line KA4. Characteristics of shoot, leaf, etc. from four mutants were also different from the ones of 'Kardinal'. The line KA1 was shortest in shoot, node and peduncle length, and lowest in prickle number. The reverse side of leaves was reddish green color in 'Kardinal' as well as the line KA4, but green color in the line KA1, KA2, and KA3.
Red and blue lights are effective wavelengths for photosynthesis in plants. In this study, we determined the effects of various combined ratios of red to blue LEDs on the quality of cherry tomato seedlings prior to transplantation, and their subsequent effects on the yield and quality of tomato fruits after transplanting. Two-week-old cherry tomato seedlings (Solanum lycopersicum cv. 'Cuty') were cultivated under various combined ratios of red (R; peak wavelength 655 nm) to blue (B; 456 nm) LEDs [red:blue = 41:59 (59B), 53:47 (47B), 65:35 (35B), 74:26 (26B), 87:13 (13B), or 100:0 (0B)] and fluorescent lamps and raised for 27 days. The cherry tomato seedlings were subsequently transplanted into a venlo-type greenhouse and cultivated for 75 days. At the seedling stage, the shoot fresh weight of seedlings in all RB combined treatments, except 0B and 59B, was higher than that of the control after 27 days of LED treatment. Shoot dry weight and leaf area also showed trends similar to that of shoot fresh weight. The stem length was significantly higher in 13B, 26B, and 35B treatments compared with the control and other treatments. In particular, the stem length of 26B plants was approximately 3.2 times longer than that of 59B plants. At 37 days after transplanting, the number of nodes was significantly higher in 26B and 47B plants, and the plant height of 26B plants was significantly higher than that of control and 59B plants. Total fruit yield in 26B plants, which was the highest, was approximately 1.6 and 1.8 times higher than that in control and 59B plants, respectively. Thus, the results of this study indicate that various combined ratios of red to blue LEDs directly affected to the growth of cherry tomato seedlings and may also affect parameters of reproductive growth such as fruit yield after transplantation.

This study was conducted to improve the acclimatization rate of in vitro strawberry plantlets through bioreactor culture using the growth retardant flurprimidol. Different concentrations [0 (Control), 0.1, 0.5, 1.0, and $2.0mg{\cdot}L^{-1}$] of flurprimidol were added during bioreactor culture. After six weeks of treatments, various growth characteristics were investigated and in vitro plantlets were acclimated in the greenhouse. The growth rate of treated plantlets was much lower than that of control, and as the treatment concentration increased, the growth rate was much decreased. Shoots of plantlets treated with flurprimidol were shorter (2.2-3.7 cm) than those of control (7.9 cm). The number of roots per treated plant was around 11.6-34.2, compared with 51.8 in the control. Root length was also lower (0.88-3.08 cm) than control (4.36 cm). However, the number of new shoots and leaves increased in all treatments except for $2.0mg{\cdot}L^{-1}$ concentration. The root was partially decayed in $1.0mg{\cdot}L^{-1}$ concentration and was completely decayed in $2.0mg{\cdot}L^{-1}$. The survival rate in $0.1mg{\cdot}L^{-1}$ and $0.5mg{\cdot}L^{-1}$ concentrations was 100% and 23.3% respectively. After four weeks of acclimatization, the plantlets restarted growth, and growth characteristics of shoots and roots recovered to the levels of control, except for fresh weight. Based on our results, a concentration of $0.1mg{\cdot}L^{-1}$ flurprimidol is appropriate for improvement of acclimatization rate of in vitro strawberry plantlets in bioreactor culture.
This study was conducted to develop an efficient mass propagation method for the mature $Prunus$$yedoensis$ Matsumura (43 to 58 years old). Cutting was conducted depending on cutting time, auxin treatments (IBA and NAA treatments mixed with talc powder), and cuttings position on shoots in a plastic house equipped with a fog system without heating. Rooted cuttings were transplanted to a nursery bed, and their growth characteristics were investigated in order to check whether the cuttings are successful or not for roadside tree planting. The average rooting rate was highly significant ($P$ < 0.0001) in all treatments: cutting on June 1st (61.4%) was more than two times greater in rooting rate than that on August 1st (23.6%); IBA 1,000 $mg{\cdot}L^{-1}$ (90.8%) and IBA 500 $mg{\cdot}L^{-1}$ (89.2%) showed much greater rooting rates than those of the other treatments; upper part of the cuttings treated with IBA 1,000 $mg{\cdot}L^{-1}$ showed the highest rooting rate, 96.7%. The interactions among treatments in the average rooting rate were also significant. There were significant differences ($P$ < 0.0001) among the auxin treatments in the survival rate of leafed cuttings transplanted to a nursery bed. The average survival rate was 46.5%, and IBA 1,000 $mg{\cdot}L^{-1}$ treatment was the highest in leafed cuttings 79.2%, but most of leafless cuttings were dead. There were significant differences ($P$ < 0.0001) among the cuttings, grafts, and in the seedlings height, diameter at root collar, the number of roots, branches, and leaves, etc., and the cuttings was the best. We can expect a possibility of mass propagation of improved $P.$$yedoensis$ Matsumura and a high planting survival rate through the transplanting of cuttings to a nursery bed in which the cuttings should be the following conditions: cutting in June to July, use of the upper part of cuttings, IBA treatment, and rooting in August in a cutting-greenhouse equipped with a fog system.
Soil microbial community and soil physiological parameters were investigated by analyzing phospholipid fatty acids extracted from the soils amended with chitosan powder and solution in a cucumber greenhouse. The soils were sampled at 90, 160, 200 days after treatment. Identified fatty acids were analyzed with principal component (PC) analysis. Chitosan powder soils and chitosan solution soils were separated from non-treated control soils by PC1 and PC2 90 days after treatment, respectively. And chitosan powder soils were separated from non-treated control soils by PC2 160 days after treatment. The ratio of fungi to bacteria increased significantly in chitosan solution-amended soils compared with the control soils 90 days after treatment. Microbial groups and physiological parameters were investigated 160 days after treatment: vesicular-arbuscular mycorrhizal fungi (VAM) significantly increased in soils amended with chitosan powder compared with other soils, the ratio of gram negative bacteria to gram positive bacteria and cyclo-fatty acids to precursors were significantly higher and lower in soils amended with chitosan solution and chitosan powder compared with control soils, respectively, and the ratio of fungi to bacteria were significantly lower in control soils compared with chitosan-treated soils. The chitosan powder increased the ratio of aerobic to anaerobic bacteria and lowered the ratio of saturated to unsaturated fatty acids compared with chitosan solution 200 days after soil application. In conclusion, chitosan powder changed the soil microbial community and the effects maintained up to 160 days after soil application. The effect of physiological parameters on the soil microbial community started to appear 160 days after and continued up to 200 days after soil application of chitosan.
Recently, it is difficult to produce uniform scions and rootstocks with high quality in a greenhouse due to weather extremes. The closed transplant production system is useful for producing scions and rootstocks with desirable morphological characteristics by environment control regardless of weather outside. In this study, we investigated transpiration rates and growth of cucumber and tomato scions and rootstocks grown under different light intensity conditions for precise irrigation control in a closed transplant production system. Hanging system to measure continuously the weight of plug tray consisting of seedlings and substrate with load-cell was installed in each growing bed. Using this system, we confirmed initial wilting point of cucumber and tomato seedlings, and conducted subirrigation when moisture content of substrate was not below 50%. The irrigation time of cucumber scions and rootstocks were 7 and 6 days after sowing, respectively. In tomato scions and rootstocks grown under PPF (photosynthetic photon flux) 300 μmol·m-2·s-1, the irrigation time were 5, 8, 11, and 13 days after sowing. Increasing light intensity increased transpiration rates and differences of transpiration rates by light intensity was higher in tomato seedlings. The growth of cucumber and tomato seedlings was promoted by increasing light intensity, especially, hypocotyl elongation and stem thickening was affected by light intensity. Cumulative transpiration rate of plug tray in cucumber and tomato seedlings was increased by increasing light intensity, and daily transpiration rate per seedling was regressed by 1st-order linear equation with high correlation coefficient. Estimation of transpiration rates by weighing continuously plug tray of vegetable seedlings can be useful to control more accurately irrigation schedule in a closed transplant production system.

This study was conducted to establish an efficient screening method to identify cucurbits resistant to powdery mildew. Powdery mildew fungus was obtained from a single lesion of infected cucumber leaf in 2010 at Daejeon. The fungus was identified as Podosphaera xanthii race 1 based on morphological characteristics and resistance responses of four melon differentials. Development of powdery mildew caused by the fungal isolate on 34 commercial cultivars of cucumber was investigated at three plant growth stages in a greenhouse. The degree of resistance of cotyledons of each cultivar to the fungus was not correlated with that of whole plant, but powdery mildew occurrence in the first true leaf was highly correlated with resistance at the level of the whole plant. Based on these results, the first true leaf of cucurbit cultivars can be used for screening of resistance to powdery mildew. In addition, variation of resistance of commercial 12 cucumber and 26 melon cultivars to the powdery mildew fungus due to different growing seasons was tested. In the case of cucumber, the resistance response in some cultivars was influenced by growing season. The resistant cultivars showed higher resistance in the warm season than in the cool season. By contrast, the resistant melon cultivars demonstrated strong resistance in all the tested growing seasons. Interestingly, the tested powdery mildew pathogen, a member of P. xanthii race 1, was not pathogenic on seven cultivars of watermelon (Citrullus lanatus). To follow up on this, diverse race 1 isolates of P. xanthii should be collected and tested.

A new Gomchwi cultivar 'Damogy ' was bred by crossing between Gomchwi (Ligularia fischeri (Ledeb.) Turcz.) and Handaeri-gomchwi (Ligularia fischeri var. spiciformis Nakai). The selection and investigation of growth and yield characteristics were conducted from 2006 to 2011 in field and greenhouse of Highland Agriculture Research Institute, NICS, Rural Development Administration, Korea. On a newly developed cultivar 'Damogy', Color of petiole ear was purple, petiole trichome was exist, light of leaf back was not exist, and density of leaf vein was degree 4. Plant height, leaf length, leaf width and petiole length were 45.9, 16.9, 21.1 and 29.0 cm, respectively in the $3^{rd}$ year in growth characteristics. Plant size was also higher than that of Gondalbi. Bolting and flowering time were Aug. $23^{th}$ and Sept. $10^{th}$, respectively. In contrast, Gondalbi showed quite earlier bolting time more than 38 days compared with 'Damogy' and consequently earlier flowering time more than 26 day. 'Damogy' showed similar leaf number (129) per plant compared to 'Gondalbi' (130). Furthermore, yield was higher 'Damogy' (1,889 g/plant) than in 'Gondalbi' (798 g/plant). 'Damogy' showed higher leaf thickness (0.71 mm) than 'Gonalbi' (0.46 mm), and consequently showed more hardness in leaf characteristics ($28.3kg/cm^2$) compared with 'Gondalbi' ($23.0kg/cm^2$). 'Damogy' showed higher resistance in the susceptibility of powdery mildew disease compared to 'Gondalbi'. 'Damogy' variety was registered plant variety protection right as a No. 89 on December 2015.
A study was conducted to investigate the effect of topophysis, and uniting method of rootstock and scion on rooting and subsequent growth of stenting-propagated cut rose ($Rosa$$hybrida$ Hort.) in an effort to develop an efficient stenting propagation method for domestic rose cultivars. Four cultivars used in this study were two standard type cultivars 'Sweet Yellow' and 'Hanmaum', and two spray type cultivars 'Chelsi' and 'May'. Scions were grafted on cuttings of a rootstock $Rosa$$indica$ 'Major'. The stenting-propagated scion-rootstock unions were planted in rockwool cubes ($50{\times}50{\times}50mm$, Delta, Grodan, Denmark) and were placed in a graft-take chamber for five days before being placed on misted greenhouse beds. The rootstock was removed of all leaves and nodes. Both the base of scions and top of stocks were simultaneously cut at a $45^{\circ}$ angle for grafting. Scions were prepared as single node cuttings, each with a five-leaflet leaf. Three positions of topophysis used were 7-9th (top), 4-6th (middle), and 1st-3rd (bottom) nodes from the stem base. Four uniting materials used were tube, tube + parafilm wrap, tube + clothespin, and clothespin. Rooting and growth were affected by the topophysis and cultivar. The best topophysis for rooting was 7-9th (top) nodes in all cultivars. Topophysis affected percent rooting, and number of roots, length of the longest root, and but not weight, shoot length and graft-take. Rooting and growth were affected by the uniting method and cultivar. Tube uniting method generally showed higher percentage graft-take, percent rooting, and number of roots than other methods. However, rootstock and scion union was not complete in this treatment. On the whole, the greatest rooting and subsequent growth of stenting-propagated plants were found in the tube + clothespin method. Except 'Sweet Yellow', rooting and growth were not adequate in the clothespin method. The results suggested that a tube + clothespin method was the most effective, and this method may be used as a substitute to save labor compared to a tube + parafilm wrap method which is currently being used in commercial nurseries.
