Melanin Biosynthesis Inhibition Effects of Ginsenoside Rb2 Isolated from Panax ginseng Berry

  • Ginsenoside Rb2 (Gin-Rb2) was purified from the fruit extract of Panax ginseng. Its chemical structure was measured by spectroscopic analysis, including HR-FAB-MS, 1H-NMR, and IR spectroscopy. Gin-Rb2 decreased potent melanogenesis in melan-a cells, with 23.4% at 80 μM without cytotoxicity. Gin-Rb2 also decreased tyrosinase and MITF protein expression in melan-a cells. Furthermore, Gin-Rb2 presented inhibition of the body pigmentation in the zebrafish in vivo system and reduced melanin contents and tyrosinase activity. These results show that Gin-Rb2 isolated from P. ginseng may be an effective skin-whitening agent via the in vitro and in vivo systems.

Suitability of Lactobacillus plantarum SPC-SNU 72-2 as a Probiotic Starter for Sourdough Fermentation

  • In sourdough fermentation, lactic acid bacteria perform important roles in the production of volatile and antimicrobial compounds, and exerting health-promoting effects. In this study, we report the probiotic properties and baking characteristics of Lactobacillus plantarum SPC-SNU 72-2 isolated from kimchi. This strain is safe to use in food fermentation as it does not carry genes for biogenic amine production (i.e., hdc, tdc, and ldc) and shows no β-hemolytic activity against red blood cells. The strain is also stable under simulated human gastrointestinal conditions, showing tolerance to gastric acid and bile salt, and adheres well to colonic epithelial cells. Additionally, this strain prevents pathogen growth and activates mouse peritoneal macrophages by inducing cytokines such as tumor necrosis factor-α, interleukin (IL)-6, and IL-12. Furthermore, the strain possesses good baking properties, providing rich aroma during dough fermentation and contributing to the enhancement of bread texture. Taken together, L. plantarum SPC-SNU 72-2 has the properties of a good starter strain based on the observation that it improves bread flavor and texture while also providing probiotic effects comparable with commercial strains.

Influence of Waterlogging Period on the Growth, Physiological Responses, and Yield of Kimchi Cabbage (침수기간이 배추의 생육, 생리적 반응 및 수량에 미치는 영향)

  • This study was conducted to investigate effects of waterlogging on the growth, physiological responses, and yield of Kimchi cabbage. The growth of Kimchi cabbage with applied optimized air temperature (set to $20^{\circ}C$) was greater than those with high air temperature (set to $30^{\circ}C$) and the growth significantly decreased by severe waterlogging treatment. The net photosynthetic rate of outer leaves on one hour after waterlogging treatment was higher at 72 hours waterlogging treatment in $20^{\circ}C$ and lower at 24 hours of waterlogging treatment in $30^{\circ}C$. The root activity was decreased by the elevation of waterlogging periods in $20^{\circ}C$ treatment and lower by the short of waterlogging periods in $30^{\circ}C$ treatment. The ratio of formality with non-waterlogging treatment was approximately 64% under $20^{\circ}C$ air temperature and that of range was from 16 to 30% under $30^{\circ}C$. The yield under $20^{\circ}C$ showed higher than that under high air temperature. The non-waterlogging treatment in $20^{\circ}C$ had 4,463 kg/10a, which was the greatest among all treatments, while yields of non-waterlogging treatment at $30^{\circ}C$ were significantly low as 1,082 kg/10a. Results suggested that additional drainage work should be needed to overcome waterlogged conditions of open field during heavy rainfall and should be drainage as soon as possible if there are waterlogging.

  • To fine out the anti-inflammatory activities of the C. fragile. and its mechanism were investigated in macrophages induced by Peptidoglycan (PGN). Treatments of macrophages with 100 ug/ml of ethanol extract of Codium fragile (EECF) inhibited PGN-induced IL-6, NO and PGE2 production in a dose-dependent manner as well as expression of iNOS and COX-2. EECF inhibited PGN-induced extracellular signal-regulated kinase (ERK) 1/2, JNK 1/2 and p38 MAPK phosphorylation, which suggests that EECF inhibits IL-6 and NO secretion by blocking MAPKs phosphorylation. These findings may help elucidate the mechanism by which EECF modulates RAW 264.7 cell activation under inflammatory conditions.

Analysis of Psychological and Physiological Changes According to the Experience of the Care Farm of Fire-fighters (소방공무원의 치유농장 체험 활동에 따른 심리·생리적 변화 분석)

  • This study conducted a survey and analysis of saliva cortisol and psychological questionnaire in order to find out the psychological and physiological changes according to the experience of the care farm targeting fire-fighters. In the program to experience care farms, the total cortisol concentration as a stress hormone decreased after participation, and there was a statistically significant difference (z = 2.858, p <.01). As a result of the risk distribution analysis of the level of post-traumatic stress symptoms of firefighting officials who participated in the program to experience care farms, the high-risk group was found to be as high as 31.2%. In addition, the relative explanatory power of the independent variables that influence the loyalty of the care farm experience activity was found to have an effect in the order of program satisfaction, age, and reliability, and among them, program satisfaction was found to be the most influential variable. Therefore, it is believed that the care farm experience activities as the application of the healing program will have a positive effect on the activation of care farms in each region and the relief of stress symptoms of fire-fighters.

Arabinoxylo- and Arabino-Oligosaccharides-Specific α-ʟ-Arabinofuranosidase GH51 Isozymes from the Amylolytic Yeast Saccharomycopsis fibuligera

  • Two genes encoding probable α-ʟ-arabinofuranosidase (E.C. 3.2.1.55) isozymes (ABFs) with 92.3% amino acid sequence identity, ABF51A and ABF51B, were found from chromosomes 3 and 5 of Saccharomycopsis fibuligera KJJ81, an amylolytic yeast isolated from Korean wheat-based nuruk, respectively. Each open reading frame consists of 1,551 nucleotides and encodes a protein of 517 amino acids with the molecular mass of approximately 59 kDa. These isozymes share approximately 49% amino acid sequence identity with eukaryotic ABFs from filamentous fungi. The corresponding genes were cloned, functionally expressed, and purified from Escherichia coli. SfABF51A and SfABF51B showed the highest activities on p-nitrophenyl arabinofuranoside at 40~45℃ and pH 7.0 in sodium phosphate buffer and at 50℃ and pH 6.0 in sodium acetate buffer, respectively. These exoacting enzymes belonging to the glycoside hydrolase (GH) family 51 could hydrolyze arabinoxylo-oligosaccharides (AXOS) and arabino-oligosaccharides (AOS) to produce only ʟ-arabinose, whereas they could hardly degrade any polymeric substrates including arabinans and arabinoxylans. The detailed product analyses revealed that both SfABF51 isozymes can catalyze the versatile hydrolysis of α-(1,2)- and α-(1,3)-ʟ-arabinofuranosidic linkages of AXOS, and α-(1,2)-, α-(1,3)-, and α-(1,5)-linkages of linear and branched AOS. On the contrary, they have much lower activity against the α-(1,2)- and α-(1,3)-double-substituted substrates than the single-substituted ones. These hydrolases could potentially play important roles in the degradation and utilization of hemicellulosic biomass by S. fibuligera.

Analysis of Preference in Plant Fragrances and Psychological Evaluation of Firefighter

  • Background and objective: This study was carried out to investigate and analyze the preference for plant fragrance and the psychological effects of a agro-healing activity in 101 firefighters aged 39.71 ± 10.94. Methods: For the psychological evaluation, post-traumatic stress symptoms, Korean occupational stress scale short form (KOSS-SF), emotional intelligence, and job satisfaction were surveyed on the Likert scale. For the preference for plant fragrances, 20 ㎕ of commercially available essential oils from four plant species (Mentha spp., Lavandula spp., Citrus limon, C. sinensis) were absorbed into each test paper and provided to the participants. Results: The rate of post-traumatic stress symptoms was high at 37.6% of the firefighters surveyed.. C. limon fragrance had the highest preference among the four plant fragrances, followed by C. sinensis > Mentha spp. > Lavandula spp. with statistically significant difference (F = 14.256, p < .001). The lower their age, income, and position, and higher the education level, the higher the preference for plant fragrances. And the administrative group had higher preference for plant fragrances than field group. In addition, as a result of examining the correlation between job satisfaction, preference for plant fragrance, post-traumatic stress, emotional intelligence, and job stress for fire-fighters, the higher the job satisfaction, the higher the fragrance preference and emotional intelligence, and the lower the post-traumatic stress and job stress. Conclusion: Therefore, installing a garden using botanical fragrances at workplaces that firefighters can easily access will improve job satisfaction and relieve stress. It can be concluded that applying the fragrance of plants to the healing farming activities at the fire-fighter's workplace can increase the usefulness of the healing activities.

Antioxidant and Anti-inflammatory Effects of Solvent Fractions from the Peel of the Native Jeju Citrus ‘Hongkyool’ and ‘Pyunkyool’ (제주 재래종 감귤 ‘홍귤’, ‘편귤’ 과피 분획물의 항산화, 항염증 효과)

  • This study investigated the anti-oxidative and anti-inflammatory activity of unripe fruit peel solvent fractions of the native jeju citrus ‘Hongkyool’ (Citrus tachibana Tanaka) and ‘Pyunkyool’ (C. tangerina Hort. ex Tanaka). The total polyphenol content and total flavonoid content were highest in the butanol fraction of both ‘Hongkyool’ (534.4 mg/g, 431.8 mg/g) and ‘Pyunkyool’ (342.9 mg/g, 415.7 mg/g). In both cultivars, the butanol fraction showed the strong antioxidant activity by DPPH radical scavenging and ABTS radical scavenging. The DPPH radical scavenging of the butanol fraction from ‘Hongkyool’ and ‘Pyunkyool’ was 89% and 64% at a concentration of 1 mg/ml, respectively. The ABTS radical scavenging of the butanol fraction from ‘Hongkyool’ and ‘Pyunkyool‘ was 94% and 85% at a concentration of 1 mg/ml, respectively. We investigated the effect of the anti-inflammatory activity of the ethyl acetate fraction from ‘Hongkyool’ and ‘Pyunkyool’ on LPS-induced NO production, IL-6, iNOS, and COX-2 protein expression in Raw 264.7 cells. At concentrations of 50 and 100 μg/ml of the ‘Hongkyool’ ethyl acetate fraction, the anti-inflammatory effect was excellent. These results suggest that ethyl acetate and butanol fractions from ‘Hongkyool’ and ‘Pyunkyool’ could be useful functional materials, with anti-oxidative and anti-inflammatory properties.

Comparative analysis of nitrite scavenging activity and anti-inflammation effects in the fruiting bodies of medicinal mushrooms (약용버섯류 자실체의 아질산염 소거능 및 항염증 효능 분석)

  • This study was carried out to compare the anti-inflammation effects of various fruiting body of Ganoderma species and Cordyceps militaris, Phelinus linteus extracts. We concentrated Ganoderma species and other medicinal mushrooms by extracting with ethanol. And We made it $100{\mu}g/ml$ concentration. As a result of nitrite scavenging activity, in the contrast to the positive control; Ascorbic acid was 25%, ASI 7080 of Ganoderma species was disappeared up to around 40%. And in the contrast to Ascorbic acid was 55%, ASI 7002 was 78.5% that was the highest anti-inflammation effect in the result of "No assay test". The Cordyceps militaris showed 75% and Hericium erinaceus showed 59.7% of anti-inflammation effect. As a result of the fungus yield control test of $TNF-{\alpha}$ through ELISA method to ASI 7002 of Ganoderma species that showed the highest anti-inflammation, it was reduced as same as LPS non-treatment. We extracted RNA from ASI 7002 Ganoderma species 10, 50, $100{\mu}g/ml$ concentration and LPS $10{\mu}g/ml$ of Raw 264.7 cell. And we tested the expression of iNOS, COX-2 and TNF-a that are kinds of inflammation gene after synthesizing RNA with cDNA. Finally we could find that iNOS, COX-2 and TNF-a were all controlled expression in the result of above experiment.

Antioxidant Activity of Pyrus pyrifolia Fruit in Different Cultivars and Parts (국내 육성 배 신품종의 과실 부위별 항산화활성)

  • This study was performed to confirm physiological activities according to parts of new pear cultivars (Gamcheonbae, Manpungbae, Chuwhangbae, Hanareum) and Niitaka pear. The total polyphenol compound contents of pear peel, flesh and core were 178~235, 95~113, 177~229 mg/100 g as tannic acid equivalent, respectively. There were differences in the contents by cultivars, Chuwhangbae and Hanareum cultivars showed high contents. The total flavonoid contents of the pear peel, flesh and core were 29.2~40.2, 24.3~34.3, 26.9~38.8 mg/100 g, respectively and those of Chuwhangbae and Gamcheonbae cultivars showed comparatively high values. The electron-donating ability was high in Chuwhangbae, Gamcheonbae and in the pear peel (29.7~57.7%), core (29.1~38.2%), flesh (7.6~17.7%), in that order. The nitrate scavenging activity was highest in that pear peel (21.0~49.8%), followed by the core (11.8~16.2%) and flesh (7.8~9.7%), but there was little difference by cultivar.
