Recently, Jersey cattle was introduced and produced by embryo transfer to Korea. This study was conducted to investigate the differences of milk compositions between Jersey and Holstein cows and the relationship between days in milk (DIM) and milk compositions during early lactation. Data were collected from twelve lactating cows from Department of Animal Resources Development at National Institute of Animal Science. Cows in parity 1 were used, and calved at spring from April to March of 2017. All cows were housed in two sections within a free-stall barn, which divided into six from each breed, and received a basal total mixed ration. Milk samples of each cow were collected at 3 DIM and 30 DIM for analyzing the milk compositions, including fatty acids (FA), amino acids and minerals. Total solids, citrate, and milk urea nitrogen level were differed between the breeds (p < 0.05). As DIM went from 3 to 30, milk protein, total solids, and somatic cell count decreased (p < 0.05), but lactose increased in all breed milk (p < 0.05). Citrate and free fatty acid (FFA) elevated in Jersey milk (p < 0.05), whereas reduced in Holstein milk (p < 0.05). Proportions of some individual FA varied from the breeds. Myristic (C14:0), palmitic (C16:0), and arachidonic acid (C20:4) in milk from all cows were higher at 3 DIM than at 30 DIM (p < 0.05). Also, stearic (C18:0) and oleic acid (C18:1) were lower at 3 DIM than at 30 DIM (p < 0.05), and the C18:1 to C18:0 ratio was significantly differed in DIM × breed interactions (p < 0.05). The contents of the individual amino acids did not differ from the breeds. Calcium, phosphorous, magnesium, and zinc (Zn) contents was significantly increased in Holstein milk than Jersey milk at 3 DIM. Also, K and Zn concentrations were higher in Holstein milk than in Jersey milk at 30 DIM (p < 0.05). It was concluded that Jersey cows would produce more effective milk in processing dairy products and more proper energy status compared with Holstein cows in early lactation under the same environmental and nutritional conditions.

This study was carried out to estimate the effects of environmental factors on the activities and rectal temperatures(heat tolerance) of Holstein dairy cows in a summer season. An activity-meter(Alpro system$^{\circledR}$) was used to record activities of 77 cows for 24 hours. The feeding standards for milking and dry cow were formulated with concentrates and corn silages based on the NRC(1998). Cows to check temperature were kept in stanchions and temperature was checked in rectum. Ambient temperature was $29.0^{\circ}C$ between 1 p.m. and 3 p.m. of the day. The rectal temperatures were affected by parities and status of milking or dry(p<0.01). The activities recorded between 9 a.m. and 12 a.m., and between 1 p.m. and 4 p.m. were not effected by the status of lactating and dry. The rectal temperature of lactating cow was 39.0${\pm}0.03^{\circ}C$, it was higher than dry cow(38.6${\pm}0.04^{\circ}C$). A higher activity of cow under third parity at 1 p.m. to 4 p.m. was observed comparing with fourth and above. The activities were tended to decrease with an increase of parity. Rectal temperature was negatively correlated with milk compositions, which were fat, protein, solid-not-fat and total solid except lactose.
Objective: To examine the effects of Rhodobacter sphaeroides (R. sphaeroides) supplementation as a direct-fed microbial (DFM) on rumen fermentation in dairy cows and on coenzyme Q10 (CoQ10) transition into milk, an in vitro rumen simulation batch culture and an in vivo dairy cow experiment were conducted. Methods: The characteristics of in vitro ruminal fermentation were investigated using rumen fluids from six cannulated Holstein dairy cows at 2 h post-afternoon feeding. A control treatment was included in the experiments based on a typified total mixed ration (TMR) for lactating dairy cows, which was identical to the one used in the in vivo study, plus R. sphaeroides at 0.1%, 0.3%, and 0.5% TMR dry matter. The in vivo study employed six ruminally cannulated lactating Holstein cows randomly allotted to either the control TMR (C-TMR) treatment or to a diet supplemented with a 0.5% R. sphaeroides culture (S-TMR, dry matter basis) ad libitum. The presence of R. sphaeroides was verified using denaturing gradient gel electrophoresis (DGGE) applied to the bacterial samples obtained from the in vivo study. The concentration of CoQ10 in milk and in the supernatant from the in vitro study was determined using high performance liquid chromatography. Results: The results of the in vitro batch culture and DGGE showed that the concentration of CoQ10 significantly increased after 2 h of R. sphaeroides supplementation above 0.1%. When supplemented to the diet of lactating cows at the level of 0.5%, R. sphaeroides did not present any adverse effect on dry matter intake and milk yield. However, the concentration of CoQ10 in milk dramatically increased, with treated cows producing 70.9% more CoQ10 than control cows. Conclusion: The CoQ10 concentration in milk increased via the use of a novel DFM, and R. sphaeroides might be used for producing value-added milk and dairy products in the future.
This study reports the detection and genotyping of the first clinical case of bovine anemia due to Theileria orientalis group (BATOG) in non-grazed dairy cow in upper South Korea. Blood and serum tests revealed anemia and hyperbilirubinemia from animal showing clinical symptoms, and later confirmed as piroplasmosis-positive. Follow-up surveillance on the herd revealed 2 asymptomatic cows with anemia. The three animals were confirmed theileriosis-positive and genotyping revealed the clinical and one of the asymptomatic cases have Chitose, while the other has Ikeda genotype. Clinical BATOG cases were rarely reported worldwide, and asymptomatic animals left untreated could serve as parasite reservoir.
Objective: We determined the impact of maintaining pasture-based dairy cows and calves together over 100 days on cow milk production, cow and calf behavior, and calf liveweight and carcass quality. Methods: Six Holstein-Friesian cows and their male calves were monitored for 106±8.6 days. Cows were temporarily separated twice a day for milking with calves remaining in the paddock. Cow and calf behaviors were recorded via scan sampling at 6 different timepoints, for the first 7 days and twice a week thereafter. Calves were weighed weekly and immediately processed for meat quality and rumen development analysis at 106±8.6 days. Daily cow milk yields were collected from enrollment until 109±8.6 days (3 days post-weaning). Results: The average daily gain of calves was 1.4±0.73 kg/d, with an average carcass dressing percentage of 59%. Calves had the greatest frequency of observed close proximity to cow and suckling in the first two weeks and decreased with experiment duration. During separation for milking, cow vocalizations and attempts to return to their calf decreased over time. Reticulorumen weight was on target for calf age, but as a proportion of total stomach weight was lower than industry averages of calves the same age due to the larger abomasum. Cows produced an average of 12±7.6 kg of milk yield per day over the 3-days before the calves were weaned and increased to mean of 31±8.3 kg/d the 3 days after weaning, indicating a consumption of close to 20 kg per calf per day. Conclusion: The impact of a pasture-based cow-calf rearing system on cow and calf behavior and the potential for high levels of calf liveweight gain when provided ad-libitum milk and feed were determined. Further research is required to determine the practicality of replicating such systems with large herds and impact on reared calves post-weaning.
Effects of recombinant bovine somatotropln(${\gamma}$BST) on hematologie and blood chemical values were investigated in twenty-five multiparous Holstein dairy cows. Recombinant BST was administered by two different routes ; intramusculary(12.5mg and 25mg/day) and subcutaneously(500mg and 750mg) in sustained-release vehicle every 2 weeks beginning 4 weeks postpartum and continuing for 7 months. Whole blood and serum samples were collected 0, 1, 2, 3, 5, and 7 months after beginning of treatments from control and ${\gamma}$BST-administered groups. Hematologic values including RBC, PCV, HB, MCH, MCHC, WBC and differential counts of treatment groups receiving ${\gamma}$BST were similiar to those of control group. Blood chemical values observed were total protein, albumin, A/G ratio, glucose, cholesterol, Ca, Pi, Ca/pi ratio, total bilirubin, creatinine, BUN, alkaline phosphatase, lactate dehydrogenase, alanine aminotransferase and aspartate aminotransferase. There were no significant differences in blood chemical values of cows administered with ${\gamma}$BST from those of control. Although some blood chemical values were fluctuated at a certain observation period, they were remained within the normal physiological ranges. It is concluded from the observations of these experiments that the dose and dosage froms of ${\gamma}$BST employed in this work might not affect hematologic and blood chemical values in dairy cows under the normal sanitary condition and adequate nutritional balance.
The purpose of this study was to isolate cellulolytic and hemicellulolytic anaerobic bacteria inhabiting from gut of ruminants and investigate their hydrolytic enzyme activities. Extracellular CMCase activities of H-strains isolated from the rumen of a Holstein dairy cow were higher than those of D- and DC- strains from the rumen and large intestine of Korean spotted deer. Most isolated bacteria utilized more efficiently Dehority's artificial medium containing starch, glucose and cellobiose (DAS) than those in Dehority's artificial medium containing cellulose only (DAC). The results of biochemical reactions and sugar fermentation indicated that the isolated bacteria belong to one of bacterial strains of Peptostreptococcus spp., Bifidobacterium spp., Prevotela ruminicola/buccae, Clostridium beijer/butyricum and Streptococcus intermedis which are not highly cellulolytic. Activities of Avicelase, xylanase, β-D-glucosidase, α-L-arabinofuranosidase and β-xylosidase of the isolated anaerobic bacteria in DAS were higher than those in DAC. In conclusion, the results indicated the higher enzyme activities of the isolated strains cultured in DAS medium were mainly caused by their specific carbohydrate utilization for enzyme production and growth rate. The highly cellulolytic bacteria were not isolated in the present experiment. Thus further research is required to investigate characteristics of gut bacteria from Korean spotted deer.
The objective of this study was to investigate the effects of abnormal ovarian cycles after superovulation treatment of Holstein Donor Cows. CIDRs were inserted into the vaginas of twenty two head of Holstein cows, regardless of estrous cycle. Superovulation was induced using folliclar stimulating hormone (FSH). For artificial insemination, donor cows were injected with $PGF_2{\alpha}$ and estrus was checked about 48 hours after the injection. Then they were treated with 4 straws of semen 3 times, with 12-hour intervals. Embryos were collected by a non-surgical method 7 days after the first artificial insemination. The cows were considered to have resumed ovarian cyclicity on the day of ovulation if followed by regular ovarian cycles. Seventy two point seven percentage of the cows(16/22) had normal resumption of ovarian cyclicity(resumption within 40 days after superovulation), and 27.3%(6/22) had delayed resumption(resumption did not occur until>40 days after superovulation). Delayed resumption Type II(first ovulation did not occur until ${\geq}40$ days after superovulation, i.e. delayed first ovulation 13.6%) were the most common types of delayed resumptions. The mean numbers of total ova from < 10 and $10{\leq}$ of corpora lutea(CL) was $7.8{\pm}1.8$ and $12.7{\pm}2.7$, respectively. The number of transferable embryos differed between < 10 and $10{\leq}$ CL was $5.4{\pm}1.3$ and $8.1{\pm}3.4$, respectively. Four point five percentage of the cows(1/22) did not resumption their ovarian cyclicity until 60 days after superovulation treatment. Diverse researches on the superovulation treatment method that is suitable for high-producing Holstein donor cows would contribute to preventing ovarian cyclicity disorder, as well as to the early multiplication of cows with superior genes by increasing the utilization value of donor cows.
Pathogen infections or mastitis inflammations usually develop differently on each udder of lactating cow. Although healthy udders will be attacked by the mastitis pathogens or the pathogens from blood in a long term, they would not be always inflamed. Somatic cell counts (SCC) in milk, which is utilized as an index of mastitis diagnosis, and the relation among SCC and milk constituents will have to be examined on each udder individually. Twelve cows of a Holstein cow herd in Nasu Research Station, which were suffering clinical or non-clinical mastitis, were selected, and SCC and milk constituents on each udder milk were measured. The effects of mastitis infection on udder milk components were relatively small except lactose content on udder milks of non-clinical mastitis (SCC< $10.0{\times}10^5$ per ml milk). On udder milks of clinical mastitis, however, high negative correlations were recognized between SCC and milk components. On different sampling days, high contents of fat and protein corresponded to that of total solids.
The objective of this study was to compare the accuracy analysis and the effect of field application of a newly developed automatic heat detector in dairy cows. From 2009 to 2010, we used 48 Holstein cows (mounting cows : 38 heads, standing cows : 10 heads) raised in experimental barn of National Livestock Research Institute (RDA) for the accuracy analysis of automatic heat detector, and 14 Holstein cows raised in three commercial dairy farms of Cheonan and Pochun area for comparison of the effect of field application. The accuracy of response in cows attached with automatic heat detector was 86.8% (33/38) displayed on board when mounting activity observed, and 100% (10/10) when standing activity observed, and on average, 90.0% (43/48) displayed on board. The accuracy of automatic heat detector in on-farm test was 85.7% (12/14), and conception rate was 75.0% (9/12).
