Effect of Subfractions of Carthamus tinctorius L. Semen on the Lipid Peroxidation and Oxygen Free Radical Scavenging Enzyme Activities in CCl_4$-induced Hepatotoxic Rats (홍화자 분획물의 사염화탄소 유발 간손상 흰쥐에서 지질과산화와 oxygen free radical 제거 효소 활성도에 미치는 영향)

  • Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl4-induced hepatotoxicity. The hepatoprotective effect of the subfractions of butanol fraction has been evaluated by analyzing oxygen free radical scavenging enzyme activities and histopathological examinations. In BS-5 subfraction treated group, the activity of superoxide dismutase has been significantly increased as compared with that of CCl4, treated rats. Antioxidant activity has been evaluated by the examination of the scavenging activity against 1,1-diphenyl-2-picrylhydrazyl radical. BS-5 subfraction has shown strong antioxidant activities. The histopathological examination showed that the treatment of BS-5 subfraction has relieved the ballooning degeneration of hepatocytes which had been generated by CCl4. It appears that the protective effect of BS-5 subfraction would be mediated of the attenuation of lipid peroxidation by acting as a free radical scavenger, which were based on the increase of superoxide dismutase activity.

Effect of Oral Administration of Processed Sulphur on Hepatotoxicity (법제 유황 경구투여가 간독성에 미치는 영향)

  • This study was to evaluate the effects of oral administration of Processed Sulphur on Hepatotoxicity. Processed Sulphur was administered orally to rats for 28 days. We measured the body and liver weight index, heamtological and biomedical parameters. We also observed the histopathological changes of liver in rats. No significant differences in body weight, liver weight index, heamtological and biomedical parameters and histopathological changes of hepatocyte between control and Processed Sulphur fed group were found. Our data indicate that hepatotoxicity was not caused by oral medication of Processed Sulphur up to 60mg/200g/day for 28 days in rats. Therefore, Processed Sulphur appears to be safe and non-toxic in these studies and a no-observed adverse effect level (NOAEL) in rats is established at 60mg/200g/day. The data could provide satisfactory preclinical evidence of safety to launch clinical trial on standardized formulation of mineral extracts.

  • In order to clarify the morphological changes in the mammary glands of cows affected with coliform (Escherichia coli) mastitis, histopathological observations were undertaken on the mastitis of the lactating rabbits which was experimentally induced with E. coli or its endotoxin isolated from cases of acute and chronic matitis in dairy cattle. In the bacterial suspension-infused groups the affected quarters of udder showed cloudy swelling, hyperemia and hemorrhage to local necrosis and firmness. The microscopic findings of early stage of the mastitis were appearance of large numbers of heterophils in the glandular lumina and ducts accompanied by degeneration, necrosis and desquamation of epithelial cells, and also infiltration of heterophils, hemorrhage and edema in the interstitial tissue, and destruction of alveoli. Later, proliferation of firoblasts, plasma cells, lymphocytes, eosinophils and histiocytes appeared in the glandular tissue and necrotic foci of glandular tissue were surrounded by highly proliferated connective tissue. Granuloma-like inflammatory changes could be observed in the glandular tissue on the 7th days after infusion. The inflammatory response in the group infused with E. coli strain isolated from the natural case of acute mastitis was rapid and severe as compared with that of chronic mastitis. In the endotoxin-infused group the morphological changes were similiar to those of the bacterial suspension-infused groups.

Studies on PVA/Chitosan/Fibroin Blend Sponge Sheets: Preparation and Wound Healing Effects in Rats

  • The preparation of PVA/Chitosan/Fibroin (PCF)-blended sponge sheets and wound healing effects of these sheets in rats were investigated. We excised the skin off rat, including the dermis, approximately 2${\times}$2 cm in size. The wound was coveted with PCF-blended spongy sheets. The spongy sheets absorbed the exudate, and gained flexibility and softness. Histopathological inspection of the wound 12 days later showed the increase of a vascular ingrowth and the absence of inflammatory cells. Regeneration of the skin around the wound was faster than that of the control. We also tested wound healing effects of PVA, Chitosan and Fibroini alone of in various combinations. Wound healing was accelerated in the order of PVA/chitosan/Fibroin (PCF)-blended sponge>Chitosan/Fibroin (CF)-blended sponge$\geq$Fibroin (F) sponge>PVA/Chitosan-blended sponge (PC)>Chitosan (C) sponge.

  • Presented here are the results for clinical study on 65 cases of simple goiter the author treated during the period of 4 years and 10 months from Jan. 1963 through Oct. 1967 at the Seoul National University Hospital and Han-Il Hospital. It had been customary to classify simple goiter, according to the gross morphological characteristics of the thyroid gland, as diffuse and nodular type. This paper attempts to investigate the histopathological changes and treatment of simple goiter respectively. Sixty five cases of simple goiter were classified into 4 types; namely degenerative type was 20 cases (30.8%), proliferative type was 7 cases (10..8%), colloidal type was 24 cases (36.9%) and cyst formation with degeneration was 14 cases (21.5%), on the basis of histopathological findings. Thus, it is essential that simple goiter should be differentiated from thyroid tumors as soon as practicable. The effectiveness of medical treatment with desicated thyroid and autohemotherapy for certain simple goiter are also discussed.

Unusual or Uncommon Histology of Gastric Cancer

  • This review comprehensively examines the diverse spectrum of gastric cancers, focusing on unusual or uncommon histology that presents significant diagnostic and therapeutic challenges. While the predominant form, tubular adenocarcinoma, is well-characterized, this review focuses on lesser-known variants, including papillary adenocarcinoma, micropapillary carcinoma, adenosquamous carcinoma, squamous cell carcinoma (SCC), hepatoid adenocarcinoma, gastric choriocarcinoma, gastric carcinoma with lymphoid stroma, carcinosarcoma, gastroblastoma, parietal cell carcinoma, oncocytic adenocarcinoma, Paneth cell carcinoma, gastric adenocarcinoma of the fundic gland type, undifferentiated carcinoma, and extremely well-differentiated adenocarcinoma. Although these diseases have different nomenclatures characterized by distinct histopathological features, these phenotypes often overlap, making it difficult to draw clear boundaries. Furthermore, the number of cases was limited, and the unique histopathological nature and potential pathogenic mechanisms were not well defined. This review highlights the importance of understanding these rare variants for accurate diagnosis, effective treatment planning, and improving patient outcomes. This review emphasizes the need for ongoing research and case studies to enhance our knowledge of these uncommon forms of gastric cancer, which will ultimately contribute to more effective treatments and better prognostic assessments. This review aimed to broaden the pathological narrative by acknowledging and addressing the intricacies of all cancer types, regardless of their rarity, to advance patient care and improve prognosis.

Morphological Studies of Developmental Stages of Oculotrema hippopotami (Monogenea: Polystomatidae) Infecting the Eye of Hippopotamus amphibius (Mammalia: Hippopotamidae) Using SEM and EDXA with Notes on Histopathology

  • The present study was performed to observe histopathological effects of Oculotrema hippopotami Stunkard, 1924 infection in the eye of Hippopotamus amphibius, as well as to reveal new details of morphology and structural features of this monogenean and its comparison between 2 age stages of the parasite. This was done using both light and scanning electron microscopy, energy dispersive X-ray analysis (EDXA) and histopathology. The presence of a mixture of different generations (adult and sub-adult) in one host individual is common for Oculotrema Stunkard, 1924 in contrast to Polystoma Zeder, 1800. New metrical and graphical information obtained for adults and sub-adults compared with the previous studies. Here we show the presence of genital papillae in adults, metrical data on the distal part of the vas deferens. SEM micrographs of sperm ejaculatory structures and information about the flattened dorsal side of the body provided for the first time. Histopathological changes, such as necrosis and hemorrhage in host tissues as a result of O. hippopotami attachment structures are described. Structural analysis of different body parts of O. hippopotami of both age groups are also included. We show qualitative differences in the presence of hardening ions (S, P, Ca) in attachment structures (oral and haptor suckers) that increase with the age of the worm. The presence of sub-adults and adults on the same host, together with high levels of infection without high pathogenicity may account for Oculotrema being one of the most successful parasites among the Monogenea.

Histopathological Profile of Benign Colorectal Diseases in Al-Madinah Region of Saudi Arabia

  • Background: Patients with long-standing inflammatory bowel disease (IBD) have an increased risk of developing colorectal cancer (CRC). Colon cancer risk in IBD increases with longer duration and greater anatomic extent of colitis, the presence of primary sclerosing cholangitis, family history of CRC and degree of inflammation of the bowel. This study aimed to characterize the histopathological pattern of benign colorectal diseases among Saudi patients and to highlight age and gender variations of lesions as base line data for future studies to investigate the link between benign/IBD and colorectal cancers in the local population. Materials and Methods: The materials consisted of 684 biopsies, reported as benign (excluding malignancies and polyps) at the Department of Pathology, King Fahad Hospital, Madinah, Saudi Arabia from January 2006 to December 2013. Data collected and entered in MS-Excel and were analyzed using SPSS-20. Results: Of 684 colorectal tissues reviewed, 408 specimens (59.6%) were from male patients and 276 specimens (40.4%) were from females giving a male: female ratio of 1.5:1. Age of the patients ranged from 4 to 75 years with a mean of 39.6 years. The most frequent histologic diagnosis was a chronic non specific proctocolitis followed by ulcerative colitis, accounting respectively for 52.6% and 31.7% of all cases. These were followed by Crohn's disease 22 (3.2%), ischemic bowel disease 20 (2.9%), diverticular disease 14 (2%), eosinophilic colitis 12 (1.7%) and solitary rectal ulcer 12 (1.7%). A minority of 21 patients (3.1%) were cases of acute nonspecific proctocolitis, schistosomiasis, tuberculosis, volvulus and pseudomembranous colitis. Conclusions: These data show that although chronic non specific proctocolitis and ulcerative colitis were the dominant diagnoses, Crohn's disease, ischemic bowel disease and diverticular disease also existed to a lesser extent and should be considered in the differential diagnosis of benign colorectal diseases. This study provides a base line data for future studies which would be taken up to investigate the link between benign/IBD and colorectal cancers in the local population.

Studies on the Sfety of Korean Ginseng Ingested as Food Substance (고려인삼(高麗人蔘)의 식품학적(食品學的) 안전성(安全性)에 관(關)한 연구(硏究))

  • In order to study the safety of ginseng ingested as a food substance, rats were fed ginseng added feed (subgroup 1: 0.625g/kg feed; subgroup 2: 1.25g/kg feed; subgroup 3: 2.5g/kg feed) for periods up to 6 months. Growth rates, blood composition, blood cell counts, and histopathological studies were carried out on both the control and test animals to study the effect of ingested ginseng. Following results were obtained: 1. The growth rates of the test and control animals were similar with the exception of the subgroup 3 in the 2-month fed animals who had a lower weight gain, and the subgroup 2 in the 4-month fed group who had a higher weight gain than the controls. 2. Liver, spleen, kidney, brain, lung and heart weights were similar between the test and the controls. 3. RBC, WBC, hematocrit, and hemoglobin values were similar between the test and control animals. the blood composition determination showed a similar level of alkaline phosphatase, triglyceride, total cholesterol, S-GOT, free cholesterol between the test and controls. However, free fatty acid, total lipid and S-GPT levels decreased while phospholipid, total protein, and HDL-cholesterol levels tended to increase. 4. Fl, F2 generations showed no abnormai values in blood count, organ weight and external appearance. 5. No histopathological differences were observed between the test and control animals even after a long-term administration of ginseng. However, there was evidence that ginseng promotes hematopoiesis in test animals. 6. These results suggest that ginseng is not only safe as a food substance but may indeed strengthen the body and help-prevent diseases of old age.

Uterotrophic Activity of Ethinyl Estradiol by Gavage and Subcutaneous Administration in Immature Female Rats (미성숙 랫드에 있어서 경구 및 피하투여에 의한 Ethinyl estradiol의 자궁증식효과)

  • In association with the international validation program to establish a rodent uterotrophic assay, we conducted preliminary uterotrophic assay proposed by GECD using immature female rats. In the present study, oral and subcutaneous routes were chosen to compare the effects of estrogenic com-pounds in the two dosing regimens. The reference compound ethinyl estradiol (EE) and the antagonist ZM189154(ZM) were administered by gavage or subcutaneously (s.c.) to immature female SD rats from 20 to 22 days of age. For each study, sixty-six female rats were randomly assigned to eleven groups: Untreated control, EE 0,0.01, 0.03, 0.1, 0.3, 1.0,3.0 and 10.0 $\mu\textrm{g}$/kg, EE 3.0 $\mu\textrm{g}$/kg(gavage)/0.3 $\mu\textrm{g}$/kg(s.c) & ZM 0.1 mg/kg, and EE 3.0 $\mu\textrm{g}$/kg(gavage)/0.3 $\mu\textrm{g}$/kg (s.c) & ZM 1.0 mg/kg. There were no treatment-related changes in clinical signs, body weights, food consumption, and necropsy findings in any groups of two studies. The wet and blotted uterus weights increased dose-dependently. Histopathological examination revealed that diameter of uterine duct, height of uterine luminal epithelium. and height oj vaginal epithelium increased dose-dependently. The proliferating cell nuclear antigen (PCNA) immunoreactive cells were increased in number dose-dependently. The estrogenic effects observed in the present studies occurred at $\geq$ 0.3 $\mu\textrm{g}$/kg of oral dose and $\geq$ 0.1 $\mu\textrm{g}$/kg of s.c. dose. An antagonistic effect of ZM against EE was found in both uterus weight and histopathological parameters. From the results obtained, it can be concluded that dose-dependence of the uterotrophic assay using EE and ZM was well demonstrated by gavage and subcutaneous administration and that the estrogenic effects of EE by s.c. dose were higher than those by gavage administration. In addition, blotted uterus weight was more sensitive than wet uterus weight and vaginal epithelial height was found to be the most sensitive parameter among the parameters examined.
