한국 서해산 가무락조개, Cyclina sinensis의 번식생태 및 기생충에 관한 연구 1. 번식생태 (Reproductive Ecology and Parasite of the Venus Clam, Cyclina sinensis (Gmelin), on the West Coast of Korea 1 Reproductive Ecology)

  • 1999년 4월부터 2000년 3월까지 서해 금강 하구인 내 초도에 서식하고 있는 가무락조개를 대상으로 자원증식 및 적정 관리를 위해 조직학적 방법에 의해 생식소지수, 생식 소발단단계에 따른 생식주기, 군성숙도를 조사하였다. 가무락조개는 자웅이체로서 난생이다. 가무락조개의 생식주기는 초기활성기(2-4월), 후기활성기(3-6월), 완숙기(4-8월), 부분산란기(7-10월초), 퇴화 및 비활성기(9-2월)의 연속적인 5단계로 구분할 수 있었다. 가무락조개의 산란기는 수온과 밀접한 관계를 가지며 수온이 24$^{\circ}C$이상인 7월부터 산란하기 시작하여 9월 중순까지로 산란성기는 7, 8월이었다. 고창산 가무락조개의 군성숙도는 각장 26.0-30.0 mm 사이에서 암, 수 공히 50% 이상이 재생산에 가담하였고, 각장 41.0 mm 이상에서는 100%가 산란에 참여하고 있다. 전 개체의 50%가 재생산에 가담하기 시작하는 개체들의 연령은 만 2세로 추정된다.

백서 간손상에 의한 구기자의 유리자유기 소거능에 관한 연구 (The Study of Free Radical Scavenging Effect of Lycii Fructus by Liver Injury of Rats)

  • In the present study, we investigated the protective effect of the Lycii Fructus water extracts (LFE) against CCl4-induced hepatotoxicity and the mechanism underlying these protective effects in the rats. The pretreatment of LFE has shown to possess a significant protective effect by lowering the serum alanine and aspartate aminoteansferase (AST and ALT) and alkaline phosphatase (ALP). This hepatoprotective action was confirmed by histological observation, In addition, the pretreatment of LFE prevented the elevation of hepatic malondialdehyde (MDA) formation and the depletion of reduced glutathione (GSH) content and catalase activity in the liver of CC1₄-injected rats. The LFE also displayed hydroxide radical scavenging activity in a dose-dependent manner (IC50 = 83.6 μg/ml), as assayed by electron spin resonance (ESR) spin-trapping technique. Moreover, the expression of cytochrome P450 2E1 (CYP2E1) mRNA, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR), was significantly decreased in the liver of LFE-pretreated rats when compared with that in the liver of control group. Based on these results, it was suggested that the hepatoprotective effects of the LFE may be related to antioxidant effects and regulation of CYP2E1 gene expression.

  • 젤란검은 천연재료로써 내열성, 내산성, 내효소성 등이 우수하여 용도가 광범위하나 기계적 강도가 약하다는 단점이 있다. 따라서 본 연구에서는 기계적 성질을 개선하고자 젤란검 스폰지에 PLGA 미립구를 혼합하였다. PLGA 미립구의 다양한 함량의 젤란검 스폰지는 기계적 강도를 알아보고자 압축강도를 측정하였고, MTT 분석, SEM, 생체활성조직학적 평가 및 RT-PCR을 통해 세포의 증식 및 ECM 분비 효과를 확인하였다. 그 결과, PLGA 미립구가 50% 함유된 젤란검 지지체에서 섬유륜세포의 꾸준한 증식과 세포외기질 분비가 우수한 것을 확인할 수 있었다. 이 연구를 통하여 PLGA 미립구가 함유된 젤란검이 디스크조직 재건을 위한 지지체로서 적합함을 알 수 있었으며, 젤란검의 다양한 응용가능성을 제시하였다.

Post-operative Treatment with Cisplatin and Vinorelbine in Chinese Patients with Non-small Cell Lung Cancer: A Clinical Prospective Analysis of 451 Patients

  • Purpose: To determine the efficacy of post-operative chemotherapy with cisplatin plus vinorelbine (NP) in Chinese patients with non-small cell lung cancer (NSCLC). Methods: A total of 451 patients with NSCLCs at stages I, II, and IIIA after surgical resection were treated with cisplatin plus vinorelbine for 4 cycles or volunteers observed between January 2002 and November 2004 and were followed for five years. The therapeutic efficacy was evaluated with reference to overall survival (OS) and disease-free survival (DFS), and adverse effects were also recorded. Potential factors affecting the lengths of OS and DFS were analyzed by multivariate analysis. Results: Most patients (86.7%) completed at least 4 cycles of treatment. Patients with chemotherapy survived significantly longer than those in the observation group (p<0.001). The absolute improvements in the 2 and 5-year OS were 3.8% [hazard ratio (HR) =0.674, 95% confidence interval (CI): 0.554-0.820, P<0.0001] and 13.0% (HR=0.732, 95% CI: 0.579-0.926, P=0.009), respectively. The improvement at 4-year DFS was 2.1% (HR=0.327, 95% CI: 0.214-0.500, P<0.0001). Stratification analysis revealed that older age, histological type, pathological degree, but not the gender and smoking status, are independent factors affecting the length of survival in this population. Many patients (63.3%) had grade 1-III tolerable adverse effects, and there was no treatment-related death. Conclusions: Post-operative chemotherapy with NP regimen is effective and tolerable in Chinese patients with NSCLC.

  • The purpose of this study was investigation to examine the prevention diabetic mellitus and treatment effect on Gastrodia Elata Blume(GEB) dieted at prior and after induced diabetic application. Prior induced diabetic 3 weeks ago application GEB dieted. It is to analysis changes in body weight, blood glucose, SOD, CAT and histopathological findings. For the fingding significantly concentration diabetic rats were divided into 3 different experimental groups and each groups were induced diabetic. Experimental group Ⅰ (STZ-induced diabetic rats; n=10), experimental group Ⅱ (after induced DM and GEB dieted rats; n=10), experimental group Ⅲ (Prior GEB dieted thereafter DM induced; n=10). Prior and After GEB dieted application was that body weight, blood glucose were increase in experimental group Ⅱ, Ⅲ. Specially, group Ⅲ was significantly change than group Ⅱ at 1st weeks. and the level of CAT were significantly decrease in experimental group Ⅱ, Ⅲ than group Ⅰ. but SOD level was increase in experimental group Ⅱ, Ⅲ than group Ⅰ. In histological observation; group Ⅰ showed decrease in the intensity and incidence of vacuolations, cellular infiltration and hypertrophy of in liver and kidney. The Gomori's stain result, group Ⅰ showed disruption ${\beta}$-cell in pancrease.

마늘 배양에 있어서 신초원기 유도 (Induction of Shoot Primordium in Culture of Garlic (Allium sativum L.))

  • 경제적인 채소류 중 향신료뿐만 아니라 약용으로서 이용 가치가 많은 마늘은 영양번식성 작물로 virus에 감염되어 있으므로, 조직배양에 의하여 마늘의 virus-free한 개체의 생산을 목적으로, 배양부위의 상위에 따른 배양효과의 차이와 호르몬 조성과 당의 농도가 조직배양의 생육 분화에 미치는 영향을 조사하였다. 또한 신초원기 유도를 위한 배양조건을 검토하였으며, 배양체의 조직학적 관찰을 행하였다. 치상부위 별로 보통엽 중하부조직이 배지조성에는 NAA 0.02 + BAP 1.0 mg/l 첨가배지가 발육과 분화에 다소 양호하였다. Shoot 분화에는 sucrose 40 g 첨가구에서 BAP의 증가에 따라 shoot형성이 다소 양호하였으며 NAA 0.02 + BAP 0.2 mg/l 첨가구에서는 가근이 유도되었다. 액체 회전 배양에서 신초원기라고 판단되는 조직이 유도되었다.

  • The purpose of this study was to observe the healing process and the distribution of fibronectin in injured condylar cartilage and bone by using LM and SEM. In order to perform this study, 40 male rat, weighing about 250g were selected. Under general anesthesia with Pentobarbital sodium, condylar cartilage and neck bone were resected. Then, the wound was irrigated with saline and closed with 5-0 chromic catgut and 4-0 silk by layer-to-layer suturing. The experimental rats were sacrificed by perfusion with 3% paraformaldehyde at 1st and 4th week after operation. The condylar process and surrounding tissues were cut, demineralized, dehydrated and embedded in paraffin. The histological observation of the specimens in LM level was performed after H-E stain and Azan stain. For localization of fibronectin, immunostaining was achieved by the avidin-biotin complex method. To study the change on condylar surface, the specimens were dehydrated, dried, gold coated and were observed with a scanning electron microscope(Hitachi S-2300). The results were as follows ; 1. The cartilage group and the bone group were repaired with epiphyseal cartilage layer on the cut surface as the normal control group. 2. The cut surface was repaired more quickly in the cartilage group than in the bone group. 3. Chondrocytes, diferentiated during healing, were stained strongly to anti-fibronectin, and fibronectin was supposed to participatein chondrocyte differentiation and cartilagenous matrix formation. 4. Fibronectin was distributed more in the new bone than in the old bone, and the osteoblasts surrounding it were also stained strongly. Fibronectin was supposed to participate in new bone matrix formation. 5. Fibronectin is supposed to be associated with the differentiation, migration and adhesion of chondrocyte and osteoblast and to participate in endochondral bone formation.

자가치아뼈이식재를 이용한 상악동증강술: 일차 보고 (Maxillary Sinus Augmentation Using Autogenous Teeth: Preliminary Report)

  • Purpose: The purpose of this study was to evaluate the effectiveness of autogenous tooth graft materials after maxillary sinus bone grafts. Methods: The study involved 23 implants in 22 patients who visited the Department of Oral and Maxillofacial Surgery and the Department of Periodontics, Chosun University Dental Hospital, in 2008 and received autogenous tooth graft materials for maxillary sinus bone grafts. Results: For eight patients with maxillary bone graft materials prior to implant placement, the healing period averaged five months. For eleven patients with simultaneous maxillary bone graft and implant placement, eight patients received a second surgery, with an average healing time of six months. Three patients had a longer observation period with only a fixture implanted. Three patients who received only a bone graft required more time to implant placement because of the lack of residual bone and also for personal reasons. Only 5 patients had biopsies performed and complications such as infection and dehiscence healed well. The application of autogenous graft materials to the maxillary bone graft sites did not exert any significant effects on the success rates. When a mixture of graft materials was used, the post-surgical bone resorption rate was reduced. Histological analysis showed that new bone formation and remodeling were initiated during the three-to-six month healing period. Bone formation capacity increased continuously up to six months after the maxillary bone graft. Conclusion: According to this analysis, excellent stability and bone-forming capacity were seen in cases where autogenous materials were used alone or mixed with other materials. Autogenous tooth graft materials may be substituted instead of autogenous bones.

카드뮴으로 유발된 생쥐 간독성에 대한 키토산올리고당의 효과 (Effects of Chitosanoligosaccharide on the Mouse Hepatotoxicity Induced by Cadmium)

  • 본 연구는 카드뮴으로 유발된 생쥐 간중독에 대한 키토산올리고당의 효과를 알아보기 위하여 시도되었다. Mouse를 대상으로 카드뮴 (5.0 mg/kg; i.p.) 단독투여군(group Cd), 카드뮴과 키토산올리고당 (0.5% solution) 동시투여군 (group Cd+Chi)으로 구분한 후 간손상 억제효과를 알아보기 위해 간조직 중의 카드뮴 농도와 metallothionein 농도를 비교측정하였다. 또한 간조직의 형태학적 손상을 확인하기 위해 조직학적 관찰을 실시하였다. 간조직의 카드뮴 농도는 Cd군에 비해 Cd+Chi군에서 현저하게 낮게 나타났다. 간조직의 MT 농도는 Cd군이 Cd+Chi군에 비해서 낮게 나타났다. 전자현미경적 관찰 결과, Cd군에서 미토콘드리아는 심한 팽윤현상이 나타났으며, RER의 분절과 리보조옴 탈락이 관찰되었다. 그러나 Cd+Chi군에서는 전자밀도가 높은 다양한 형태의 미토콘드리아가 분포되어 있었으며, 리보조옴이 부착된 채로 전형적인 층판구조를 형성한 RER이 관찰되었다. 이상과 같은 결과로, 키토산올리고당이 생쥐 간조직에 미치는 카드뮴의 독성을 감소시킴을 알 수 있었다.

납으로 유발된 생쥐 간장 독성에 대한 스쿠알렌의 효과 (Effects of Squalene on the Mice Hepatotoxicity Induced by Lead Acetate)

  • 본 연구의 목적은 스쿠알렌이 납 중독에 대해 효과가 있는 지를 관찰하고자 하였다. 건강한 ICR계 생쥐를 사용하였다. 납을 복강 투여한 후 SOD와 간 조직의 미세구조를 관찰하였다. 실험군은 다음과 같다. Group 1은 납을 복강 투여한 후 스쿠알렌을 처치하지 않은 대조군, Group 2는 납을 복강 투여한 후 스쿠알렌을 처치한 군으로 각 실험군당 생쥐 10마리를 사용하였다. 실험 결과는 다음과 같다. SOD 측정 결과 대조군은 정상군에 비해 수치가 훨씬 낮게 나타났지만, 스쿠알렌을 처치한 군에서는 정상군에 비해 낮지만, 대조군 보다 수치가 높게 나타남을 관찰할 수 있었다(P<0.05). 간 조직의 미세구조에의 경우 납 단독 처치군의 경우사립체의 팽대와 rER은 팽대 및 절단된 상태가 관찰되었으나, 스쿠알렌을 처치한 군에서는 납 단독 처치군에 비해 손상 정도가 덜하였다. 본 연구를 통해 스쿠알렌이 납 중독에 의한 간장 조직의 손상 회복에 대해 효과가 있는 것으로 생각되어진다.
