A Rapid and Simple Method for Construction and Expression of a Synthetic Human Growth Hormone Gene in Escherichia coli

  • A cDNA, encoding the human growth hormone (hGH), was synthesized based on the known 191 amino acid sequence. Its codon usage was optimized for a high level expression in Escherichia coli. Unique restriction sites were incorporated throughout the gene to facilitate mutagenesis in further studies. To minimize an initiation translation problem, a 624-bp cassette that contained a ribosome binding site and a start codon were fused to the hGH-coding sequence that was flanked between the EcoRI and HindIII sites. The whole fragment was synthesized by an overlapped extension of eight long synthetic oligonucleotides. The four-short duplexes of DNA, which were first formed by annealing and filling-in with a Klenow fragment, were assembled to form a complete hGH gene. The hGH was cloned and expressed successfully using a pET17b plasmid that contained the T7 promoter. Recombinant hGH yielded as much as 20% of the total cellular proteins. However, the majority of the protein was in the form of insoluble inclusion bodies. N-terminal amino acid sequencing also showed that the hGH produced in E. coli contained formyl-methionine. This study provides a useful model for synthesis of the gene of interest and production of recombinant proteins in E. coli.

  • Bacteriophage ${\phi}FC1$ is a temperate phage which was identified as a prophage in the Enterococcus faecalis KBL703 chromosome. Phage ${\phi}FC1$ integrates into the host chromosome by site-specific recombination. The phage attachment site P (attP) was localized within the 0.65-kb XhoI-HindIII fragment and the nucleotide sequence of the region was determined. An open reading frame (mj1) which adjoined the phage attachment site encoded a deduced protein related to the site-specific recombinase family. The organization of this region was comparable to other site-specific recombination systems. The molecular weight of the expressed MJ1 in E. coli was in good agreement with the predicted 53,537 Da of the mj1 gene product. Elucidation of the phage-specific integration process in this study would provide useful genetic tools such as a chromosomal integration system.

Metal Effects of Urban Air Particulates on Cytokine Production and DNA Damage

  • Epidemiologic studies have demonstrated an association between short-term exposure to particulate air pollutants and increased mortality. However the biological mechanism underlying these associations have not been fully established and also the chemical and physical characteristics of the pollutant particles are not well understood. The metal constituents of air pollutant particles and their bioavailability are considered to Play an important role as possible mediators of Particle-induced airway injury and inflammation. Sprague-Dawley rat alveolar macrophage cells (NR8383) were exposed to airborne and acid-leached particulate matter (PM). Titanium oxide and nickel subsulfide were used as negative and positive controls. Particle-induced reactive oxygen species formation in cells was detected using the fluorescent probe 2',7'-dichlorofluorescin diacetate. Expression of TNF-$\alpha$ and IL-6 were measured by enzyme-linked immunosorbent assay, and PM-induced DNA double-strand breaks were determined with $\lambda$DNA/Hind III marker. Metals associated with air pollutant particles mediated intracellular oxidant production in alveolar macrophages, and the cytotoxicity and proinflammatory cytokine production induced by PM were associated with oxidative stress. The oxidants produced by air pollutant particles also are likely to induce DNA double-strand breaks. Our findings in alveolar macrophage cells exposed to PM and acid-leached PM support the hypothesis that metal components in urban air pollutants and their bioavailabilities might play an Important role in the induction of the adverse health effects.

  • A strain of Cucumber mosaic virus (CMV) was isolated from a weed, water chickweed (Stellaria aquatica), growing in the pepper field in Chunchon, Korea. This isolate, CMV-Sa, was differentiated from other CMVs based on biological properties and nucleotide sequence analysis of the coat protein (CP) gene. CMV-Sa showed different reactions to all the tested plants, except Capsicum annuum and Cucumis sativus, when compar-ed with those of CMV-Mf (subgroup I) and CMV-PaFM (subgroup II). Remarkably, in Nicotiana tabacum cvs. Samsun, Xanthi-nc and Ky-57, CMV-Sa induced local necrotic ring spots on the inoculated leaves and venal wave pattern and mosaic on the upper leaves. RNA analysis, serology, and RT-PCR of CP gene showed that CMV-Sa belonged to subgroup I of CMV. However, restriction enzyme analysis of the cDNA using AluI, HhaI, HincII, HindIII, HinfI and MspI showed that CMV-Sa was distinct from that of CMV-Mf. Based on comparison of the nucleotide of CP gene and deduced amino acid sequences between other CMV strains, CMV-Sa was closely related to CMV-Mf with 93.7% and 97.2 % identity, respectively.

Nucleotide Sequence of 7.2 kb Mitochondrial Linear Plasmid DNA in Pleurotus ostreatus (Pleurotus ostreatus 미토콘드리아의 7.2 kb 선상 플라스미드 염기서열 분석)

  • Two linear plasmid-like DNAs, 10.2 kb and 7.2 kb were found in the mitochondria of P. ostreatus. They have covalently linked 5'-terminal proteins in both ends. Two continuous fragments of 4.7 kb and 2.3 kb from 7.2 kb DNA were cloned and sequenced. Two long open reading frames (ORF1; 2982 bp, 993 a.a and ORF2; 2703 bp, 900 a.a) and one short open reading frame(ORF3; 771 bp, 256 a.a) were found in the 7.2 kb plasmid. The putative ORF1 and ORF2 have conserved motifs of DNA polymerases and RNA polymerases, respectively, while the ORF3 has homologous regions with phosphatase from Plasmodium, and also with adhesine from Mycoplasma.

  • The intergenic spacer (IGS) region of the ribosomal DNA of species in Fusarium section Liseola was analyzed by amplification and subsequent digestion with several restriction enzymes. The length of the amplified IGS region was about 2.6 Kb in all strains except F.moniliforme 12 Which was about 2.9 Kb. The enzymes, EcoRI, HincII, SalI, HindIII, PstI and SmaI, digested the IGS region and nine haplotypes were identified among 11 strains. In the dendrogram based on PCR-RFLP of IGS region combined the results of section Liseola in this study and section Elegans in previous study, variation in the IGS appears to offer considerable potential to resolve intraspecific relationship as well as interspecies or intersection.

  • Numerical identification was carried out for an isolate of Streptomyces strain producing the extracellular p-lactamase inhibitor. Fifty taxonomic unit characters were tested and the data were analyzed numerically using the TAXON program. The isolate was identified to the major cluster 5 of Streptomyces and it was best matched to Streptomyces omiyaensis which is a synonym of Streptomyces exfoliatus. Therefore, it was concluded that the isolate was identified to be a strain (SMF 19) of Streptomyces exjbliatus.

  • In order to cloning of the nif genes of Enterobacter agglomerans NFB-264, the digested total DNA of the strain was ligated to pBR 322 and transformed into E. coli. Through the negative selection and colony hybridization, the transformants were obtained. The recombinant plasmids, pNEL 10 and pNES 20 were extracted from these transformants. It was known from Southern hybridization that pNEL 10 contained the 12 Mdal foreign DNA fragment hybridized with nif Q-X probe and pNES 20 included the 5 Mdal foreign DNA fragment hybridized with nif NE and nif YK probe.

  • Essential hypertension is considered to be caused by a complicated combination of genetic and environmental factors. Alterations of lipid metabolism in plasma have been reported to be related to an increased risk of essential hypertension. The purpose of this study was to investigate the relationship between two genetic polymorphisms (Pvu II and Hind III RELPs) of the human lipoprotein lipase (LPL) gene and essential hypertension in korean population. In our result the Pvu II RFLP of LPL gene was significantly associated with essential hypertension (P < 0.05). Therefore, we suggest that the Pvu II RFLP of LPL gene may be useful as a genetic marker for essential hypertension in Korean population.

  • In order to investigate the effect of 'Whangryonhaedok-Tang' on the anti-inflammatory, ileum of mice, blood pressure, blood glucose, total cholesterol and antibacterial activity, those pharmacological actions were conducted. The results of this studies were summarized as follows; Rat hind paw edema induced by acetic acid was inhibited. In mice, tone of intestinal smooth muscle was suppressed with the treatment of sample and intestinal contractions induced by $BaCl_2$ was inhibited. Hypertensive effect was not observed in rabbits but was observed by the water extract of 'Whangryonhaedok-Tang' excluded Phellodendri Cortex in rabbits. The increased blood glucose levels in alloxandiabetic rabbits were significantly decreased by the oral administration of the sample I after 6 days. The total cholesterol levels in alloxan-induced rabbits were significantly decreased by the oral administration of the sample I after 9 days. Antibacterial activity was observed. In addition, we thought the pharmacological actions were significantly noted by the sample III (Methanol soluble fraction) because of the purification of effective components.
