Effects of Raw Materials and Bulking Agents on the Thermophilic Composting Process

  • Three typical biological solid wastes, namely, animal manure, garbage, and sewage sludge, were compared with regard to the composting process and the changes in microbial community structure. The effects of different bulking agents such as rice straw, vermiculite, sawdust, and waste paper were compared in manure compost. The differences in the microbial community were characterized by the quinone profile method. The highest mass reduction was found in garbage composting (56.8%), compared with manure and sludge (25% and 20.2%, respectively). A quinone content of $305.2\;{\mu}mol/kg$ was observed in the late stage of garbage composting, although the diversity index of the quinone profile was 9.7, lower than that in manure composting. The predominant quinone species was found to be MK-7, which corresponds to Gram-positive bacteria with a low G+C content, such as Bacillus. The predominance of MK-7 was especially found in the garbage and sludge composting process, and the increase in quinones with partially saturated long side-chains was shown in the late composting process of manure, which corresponded to the proliferation of Actinobacteria. The effects of different bulking agents on the composting process was much smaller than the effects of different raw materials. High organic matter content in the raw materials resulted in a higher microbial biomass and activity, which was connected to the high mass reduction rate.

  • Recently severity of ecological adaptation and climatic change due to global warming grows larger. According to the fourth report of IPCC in 2007, emission quantity of the earth greenhouse gas(GHGs) generated by activity of mankind increased with 80% since 1970. And it is forecasted that worldwide greenhouse gas will be increased with 25~90%(corresponding to $CO_2$) between 2000 and 2030. This increment of greenhouse gas($CO_2$) is expected to raise average temperature of the earth with the maximum $6.4^{\circ}C$, and sea surface with 59cm in 2090. Like this, destruction of environment by greenhouse gas is regarded as universal problem threatening the existence, not only the problem of one nation. Consequently, systematic correspondence to the global warming at the aspect of energy consumption is also needed in Korea. From the analysis result of 'Statistics of Energy Consumption' published by Green Korea in 2007, energy consumption increment of domestic universities was higher as many as 3.7 times than 22.5% of the whole energy consumption increment in our country. This says to be the direct example which shows that universities are huge sources of greenhouse gas emission. New constructing and enlarging buildings of each universities within campus are the most major reason for such a large increment of energy consumption in universities. The opinion that the possibility of causing energy waste and efficiency reduction is raised by increased buildings of universities has been propounded. That is, universities should make concrete goal and the plan for reducing emission of green house gas against climatic change, and should practice. Accordingly, there is the meaning that 2 aspects of environment-friendly design characteristics, that is application of energy utilizing technology, material usage of energy efficiency-side and environment-side, and introduction of natural element in the environmental aspect, were analyzed for facilities of university campus designed in environment-friendly point of view from initial stage of plan, and direction of environment-friendly design of university facilities in the future was groped in order to grasp environment-friendly design tendency of internal and external University facilities based on this analysis of this paper.

Extraction Characteristics, Antioxidative Effect and Preparation of Collagen Gel of Skate Skin Extracts (홍어껍질 추출물의 추출특성과 유지 산화억제 효과 및 콜라겐겔 제조)

  • To enhance the reutilization of waste skate skin for the functional food resources, the investigations of extraction characteristics, antioxidative activity of skate skin water extracts on the oxidation of three cooking oils were carried out, and rheological properties, storage safety and sensory evaluation of collagen gel from skate skin were performed. Aromatic and phenolic compounds contents of $50^{\circ}C$ extracts were higher by 49.4% and 32.7%, respectively, than those of $25^{\circ}C$ extracts. Reducing power of extract at $50^{\circ}C$ was higher by 52.74% than that of $25^{\circ}C$ extract, but was 14.9% of ascorbic acid and 27.8% of BHT. Electron donating ability was corresponded to reducing power and phenolic compounds contents. Antioxidative effect of extracts on cooking oil was higher at $50^{\circ}C$ extract than $25^{\circ}C$ extract, and its order was on corn seed oil, soybean oil and olive oil. Antioxidative effect of $50^{\circ}C$ extract showed 38.27~96.83% and 49.53~75.31% of those of ascorbic acid and BHT, respectively, over three cooking oil. The optimum extraction condition for collagen gellation was $100^{\circ}C$, 2 hours extraction under 2.5 folds hydrolysis, and gel strength was lowered above 50% by 10% seasoning.

Influence of Various Biochars on the Survival, Growth, and Oxidative DNA Damage in the Earthworm Eisenia Fetida

  • BACKGROUND: Biochar (BC) has a great potential for enhancing soil fertility and carbon sequestration while facilitating beneficial waste disposition. Therefore, it is essential to assess and mitigate any inadvertent consequences associated with soil biochar amendment. Earthworm activity is very vital in the soil system, yet there are a limited number of studies that have examined their impact resulting from biochar application to soil. METHODS AND RESULTS: In this study, the survival, growth, reproductive tests, and oxidative DNA damage tests (measured by 8-hydroxydeoxyguanosine (8-OHdG) and catalase (CAT) activities) to assess the potential toxicity to earthworm Eisenia fetida in artificial soil amended with BCs were investigated. The BCs derived from perilla meal, sesame meal, and pumpkin seed were pyrolyzed at 300 and $550^{\circ}C$, and then amended with soil at a rate of 5%. All the earthworms survived, but lost weight compared to control soil after 28 day incubation period. Moreover, the BC-amended soils did not significantly affect the cocoon numbers of earthworms. Slightly higher concentrations of 8-OHdG and CAT were observed in earthworms present in BC-treated soil than those in control soil. Furthermore, the 8-OHdG concentrations in the soil amended with BC produced at $550^{\circ}C$ were greater than those at $300^{\circ}C$, and it slightly decreased as the incubation time increased. CONCLUSION: These observations could be due to higher contents of toxic metal(loid)s and also higher pH in BCs pyrolyzed at $550^{\circ}C$ than $300^{\circ}C$. While BC is efficiently being used in agricultural fields, this study suggests that it is required to assess the unintended negative impacts of BC on soil ecosystems.

Simultaneous Separation and Determination of $^{l4}C\;and\;^3H$ in Spent Resins from PWR Nuclear Power Plants (가압경수로형 원전에서 발생된 폐수지의 $^{14}C$$^3H$ 동시 분리 및 측정)

  • In this work $^{14}C\;and\;^3H$ distribution characteristics of spent resins from nuclear power plants(NPPs), pressurized water reactors(PWRs), was investigated. It was found that the recovery percent of $^{14}C$ by the wet oxidation-acid stripping was $81%{\sim}100%$ for the added activity range of $^{14}C,\;0.72\;Bq{\sim}460\;Bq$, and it was not affected by the kinds of stripping acids, 3N-HCl, $3\;N-HNO_3\;and\;3\;N-H_2SO_4$. And the recovery percent of $^3H$ by distillation using the same apparatus was $81%{\sim}101%$ for the added activity range of $^3H,\;0.60\;Bq{\sim}435\;Bq$. Among the tested stripping acids, 3\;N-HCl, $3\;N-HNO_3\;and\;3\;N-H_2SO_4$, only the trapped $^3H$ solution by distillation in $3\;N-H_2SO_4$ was compatible with the 3H scintillator, Ultimagold XR. Neither of the $^{14}C\;and\;^3H$ trapping solutions from the spent ion exchange resin samples by the wet oxidation-3 $N-H_2SO_4$ stripping contained gamma nuclides. However, some gamma nuclides, $^{60}Co,\;^{134}Cs,\;^{137}Cs\;and\;^{54}Mn$, were found in the trapped $^3H$ solutions of the spent resins by the wet oxidation-3 N-HCl stripping. It was the same for the $^3H$ trapping solutions of the spent resins by Sample Oxidizer(PACKARD MODEL 307). Meanwhile only two nuclides, $^{134}Cs,\;and\;^{134}Cs$, were found in the $^{14}C$ trapping solutions of the spent resins by Sample Oxidizer(PACKARD MODEL 307). It was found that most of the $^{14}C$ in the spent resins existed as inorganic carbon form, more than about 70% of the total $^{14}C$ content. Among the analyzed 30 spent ion exchange resin samples, the average concentration of $^{14}C$ and $^3C$ for the high radioactive samples, 8 samples, was $19000\;Bq/g{\pm}41000\;Bq/g,\;670\;Bq/g{\pm}460\;Bq/g$ and that for the low radioactive samples, 22 samples, was $4.2\;Bq/g{\pm}4.3\;Bq/g,\;6.0\;Bq/g{\pm}5.3\;Bq/g$, respectively. And the average $^{14}C/^3H$ ratio for the high radioactive samples, was higher, 28, than that of low radioactive samples, 0.70. Some linear relationship trend was found between the activity concentrations of $^{14}C\;and\;^3H$.

Effect of Temperature on Soil Microbial Biomass, Enzyme Activities, and PLFA content during Incubation Period of Soil Treated with Organic Materials (유기물원 항온배양 온도가 토양미생물체량과 효소활성 및 PLFA함량에 미치는 영향)

  • This study was carried out to evaluate the effect of temperature on soil microbial biomass, enzyme activities, and PLFA content in the volcanic(VAS) and the non-volcanic ash soil(NVAS). The soils were treated with organic materials such as organic fertilizer pelleted(OFPL), organic fertilizer powdered(OFPD), pig manure compost(PMC), and food waste compost(FWC). Two grams of organic materials were well mixed with 30g of dried volcanic and non-volcanic ash soil(< 2 mm) with 50% of soil moisture content. And the soils were incubated at 10, 20, $30^{\circ}C$ in incubator. Soils were analysed on the incubation times as followed; soil pH, total nitrogen, organic matter(at 75, 150, 270 days), microbial biomass C and PLFA (at 75, 270 days), microbial biomass N and soil enzyme(at 150, 270 days). pH values of soils treated with PMC and FWC had no changes on soil type, and incubation temperature. However, the pH was increased with temperature in the soils treated with OFPL. The changes in NVAS was higher than in VAS. Soil microbial biomass C content were high in the condition of high temperature and organic fertilizers treatment in VAS. But the contents were gradually decreased with incubation period in both NVAS and VAS. Soil microbial biomass N was high in NVAS treated with organic fertilizers and in VBS treated with PMC and FWC. PLFA content was higher in NVBS than in VBS at 75 days but showed high in VBS at 270 days. Urease activity of NVBS treated with OFPL showed $10^{\circ}C$ (75.0)> $20^{\circ}C$ (16.3)>$30^{\circ}C$ ($4.6ug\;NH{_4-}N\;g^{-1}\;2h^{-1}$) at 150 days. It were decreased gradually high temperature and time passes. And it showed high at $10^{\circ}C$ in VBS. Glucosidase activity was higher in NVBS than in VBS. Correlation coefficient of between soil microbial biomass C and microbial activity indicators showed that PLFA was high significantly at $r^2=0.91$ in NVBS and ${\beta}-glucosidase$ was $r^2=0.83$ in VBS. Soil microbial activities showed differences in the relative sensitivities of soil type and soil temperature.

Degradation of Fat, Oil, and Grease (FOGs) by Lipase-Producing Bacterium Pseudomonas sp. Strain D2D3

  • Biodegradation of fat, oil, and grease (FOGs) plays an Important role in wastewater management and water pollution control. However, many industrial food-processing and food restaurants generate FOG-containing waste waters for which there Is no acceptable technology for their pretreatment. To solve these problems, this study evaluated the feasibility of effective FOG-degrading microorganisms on the biodegradation of olive oil and FOG-containing wastewater. Twenty-two strains capable of degrading FOGs were isolated from five FOG-contaminated sites for the evaluation of their FOG degradation capabilities. Among twenty-two strains tested, the lipase-producing Pseudomonas sp. strain D2D3 was selected for actual FOG wastewater treatment. Its biodegradability was performed at 3$0^{\circ}C$ and pH 8. The extent of FOG removal efficiency was varied for each FOG tested, being the highest for olive oil and animal fat (94.5% and 94.4%), and the lowest for safflower oil (62%). The addition of organic nitrogen sources such as yeast extract, soytone, and peptone enhanced the removal efficiency of FOGs, but the addition of the inorganic nitrogen nutrients such as $NH_4$Cl and $(NH_4)_2SO_4$ did not increase. The $KH_2PO_4$ sources in 0.25% to 0.5% concentrations showed more than 90% degradability. As a result, the main pathway for the oxidation of fatty acids results in the removal of two carbon atoms as acetyl-CoA with each reaction sequence: $\beta$-oxidation. Its lipase activity showed 38.5 U/g DCW using the optimal media after 9 h. Real wastewater and FOGs were used for determining the removal efficiency by using Pseudomonas sp. strain D2D3 bioadditive. The degradation by Pseudomonas sp. strain D2D3 was 41% higher than that of the naturally occurring bacteria. This result indicated that the use of isolated Pseudomonas sp. strain D2D3 in a bioaugmentating grease trap or other processes might possibly be sufficient to acclimate biological processes for degrading FOGs.

Development of Functional Hanji Added Citrus Peel(I) - Hanji added Korean citrus peel - (감귤박을 첨가한 기능성 한지제조 기술개발(제1보) - 한국산 감귤박 첨가 한지 -)

  • This study was carried out to develop a new application field and obtain the basic data of citrus peel as waste in Jeju island and traditional Hanji for producing functional Hanji. The results measuring physical and optical properties, water vapor permeance and antibacterial activity are as follows. It was revealed that apparent density go as down but bulk raise up in the structural view of Hanji with increasing of the addition various Korean citrus peel (citrus unshiu, cheonggyun and hanrabong peel, and citrus unshiu peel powder) percentages, and that the density of Hanji added citrus unshiu peel was higher, but bulk was lower in compared with Hanji added other kinds of peel. Those Hanji added citrus unshiu peel, cheonggyun peel, hanrabong peel and citrus unshiu powder were very great not only in the strength (breaking length, burst index, tear index and folding endurance) but also in water vapor permeant rate in comparison with Hanji. The pHs of Hanji were neutrality (7 to 8). The brightness of the Hanji added various citrus peel percentages was low in compared to Hanji, and the 40% addition of hanrabong peel was the lowest. When 40% hanrabong peel was added to Hanji, it was very yellow in the color degree. When cheonggyun peel was added to Hanji manufacture, water vapor permeant rate was highly effective. It is known that vacant space of intrafiber was reduced by image analysis of Hanji and the additions of peel of citrus unshiu, cheonggyun and hanrabong were distributed equally in the interior of Hanji. The antibacterial activity of Hanji added citrus unshiu peel is more than 98%. After all, it would be able to increase utilization of Hanji, extensively. Namely, production of high quality Hanji added functional materials is expected for new valuable industry of citrus peel and Hanji.

Shear Strength of Interface between Natural Aggregate Concrete and Recycled Aggregate Concrete (천연골재 콘크리트와 순환골재 콘크리트 접합면의 전단강도)

  • Concrete recycling is becoming mandatory rather selective due to depletion of constructional materials and increase of concrete waste. Studies on recycling concrete are conducted in various point of view for long time. However, standard or guideline of many countries for the application of recycled aggregate concrete(RAC) has restrictions such as low replacement rate of coarse aggregate and no fine aggregate allowed due to inferior material properties of recycled aggregate. This study intends to figure out the feasibility of casting natural aggregate concrete(NAC) and RAC separately in a structural member. In making RAC, replacement rate of coarse aggregate was 50, 100% in RAC and treatment of interface of two concretes is introduced. RAC treatment of recycled aggregate or inclusion of additives was not done as it can increase embodied energy of concrete work. Double-shear test with uniformly distributed loading was adopted to evaluate shear strength at the interface of two concretes. After curing it was hard to distinguish interface of two concretes. Experimental result revealed that specimen with higher replacement rate showed higher shear-to-compressive strength ratio, which is possibly attributed to coarse aggregate size and roughness of sheared section. Further study on the effect of various parameters is required and subsequent research activity is on-going.

The Characteristic of Selective Attachment and Bioleaching for Pyrite Using Indigenous Acidophilic Bacteria at $42^{\circ}C$ ($42^{\circ}C$에서 토착호산성박테리아의 황철석 표면에 대한 선택적 부착과 용출 특성)

  • The bioleaching experiment under $42^{\circ}C$ was effectively carried out to leach the more valuable element ions from the pyrite in the Gangyang mine waste. Bacteria can survive at this temperature, as indigenous acidophilic bacteria were collected in the Hatchobaru acidic hot spring, in Japan. To enhance the bacterial activity, yeast extract was added to the pyrite-leaching medium. The indigenous acidophilic bacteria appeared to be rod-shaped in the growth-medium which contained elemental sulfur and yeast extract. The rod-shaped bacteria ($0.7\times2.6\;{\mu}m$, $0.6\times7\;{\mu}m$, $0.8\times5\;{\mu}m$ and $0.7\times8.4\;{\mu}m$) were attached to the pyrite surface. The colonies of the rod-shaped bacteria were selectively attached to the surroundings of a hexagonal cavity and the inner wall of the hexagonal cavity, which developed on a pyrite surface. Filament-shaped bacteria ranging from $4.92\;{\mu}m$ to $10.0\;{\mu}m$ in length were subsequently attached to the surrounding cracks and inner wall of the cracks on the pyrite surface. In the XRD analysis, the intensity of (111), (311), (222) and (320) plane on the bacteria pyrite sample relatively decreased in plane on the control pyrite sample, whereas the intensity of (200), (210) and (211) increased in these samples. The microbiological leaching content of Fe ions was found to be 3.4 times higher than that of the chemical leaching content. As for the Zn, microbiological leaching content, it was 2 times higher than the chemical leaching content. The results of XRD analysis for the bioleaching of pyrite indicated that the indigenous acidophilic bacteria are selectively attacked on the pyrite specific plane. It is expected that the more valuable element ions can be leached out from the mine waste, if the temperature is increased in future bioleaching experiments.
