The aim of this study was to investigate effects of onion, red onion, or quercetin on plasma antioxidant vitamin, lipid peroxidation, and leukocyte DNA damage in rats fed a high fat-cholesterol diet. Forty SD male rats were assigned to normal control, high fat-cholesterol diet (HF), or HF+5% onion powder, HF+5% red onion powder, or HF+0.0l% quercetin. The HF diet resulted in significantly higher plasma lipid peroxidation which decreased with onion, red onion, or quercetin supplementation. Leukocyte DNA damage induced by HF diet decreased significantly in rats fed onion and red onion, while quercetin supplementation had no effect on preventing leukocyte DNA damage. $H_2O_2$ induced leukocyte DNA damage exhibited a highly significant negative correlation with plasma retinol and tocopherols. These results suggest that onion or red onion powder exerts a protective effect with regard to DNA damage in rats fed HF diet. However, 0.01% quercetin in pure form might not be effective at preventing DNA damage.
The purpose of this study was to investigate the effect of vitamin E supplementation on the lipid peroxidation and activities of antioxidative enzymes in the pancreas of diabetic KK mice. KK mice were fed high ft diet containing 20% corn oil(wt/wt), and sacrificed at 2 months of diabetes. A hish vitamin E diet consisted of the high fat diet supplemented with an excessive amount of 이-$\alpha$-tocopheryl acetate (2080IU/kg diet). The incidence of diabetes mellitus was 61% when mice were fed the high fat diet, but was 44% when mice were fed the high vitamin E diet, Vitamin E supplementation fhus seems to have the effect of decreasing of decreasing the onset of diaetes. In the diabetic group, we found increases of MDA (malondialdehyde) and antioxidative enzyme activities. Treatment with vitamin E did not modify the level of fasting blood glucose. However, MDA and antiosicative enzyme activities in diabetic mice were decreased by the high vitamin E diet. Increased levels of lipid peroxidation products suggests the occurrence of oxidative damage in the pancreas of diabetic mice. The increased level of antiosicative enzyme activities could be due to an adaptive response to conditions of increased peroxidative stress. Significant normalization on catalase activity was noted in vitamin E supplemented animals.

This study investigated the effects of cell cultured Acanthopanax senticosus extract (ASE) supplementation and swimming exercise on lipid profiles and carnitine concentrations in C57BL/6J mice fed high fat diets. Male C57BL/6J mice (n=50), aged 4 weeks, were divided into 5 groups based on exercise and/or ASE supplementation (0.5 g/kg of body weight): normal diet (N-C), high fat diet (H-C), high fat diet non-supplement & exercise (H-NSE), high fat diet supplement & no exercise (H-SNE), high fat diet supplement & exercis (H-SE). Liver nonesterified carnitine (NEC) was significantly higher in the H-SNE group than in the H-C group, and liver total carnitine (TCNE) levels were significantly higher in the H-SNE group than in the H-NSE and H-SE groups. Liver and muscle carnitine palmitoyltransferase-I (CPT-I) mRNA levels tended to be higher with ASE supplementation and/or exercise. These results suggest that supplementation with ASE and/or exercise might have a role in improving lipid oxidation.
The purpose of this study was to investigate the effects of ${\alpha}$-lipoic acid on body weight and lipid profiles in Sprague-Dawley rats fed a high fat diet (HFD). After 4 weeks of feeding, rats on the HFD were divided into three groups by randomized block design; the first group received the high-fat-diet (n = 10), and the second group received the HFD administered with 0.25% ${\alpha}$-lipoic acid (0.25LA), and the third group received the high-fat diet with 0.5% ${\alpha}$-lipoic acid (0.5LA). The high fat diet with ${\alpha}$-lipoic acid supplemented groups had significantly inhibited body weight gain, compared to that in the HFD group (P < 0.05). Organ weights of rats were also significantly reduced in liver, kidney, spleen, and visible fat tissues in rats supplemented with ${\alpha}$-lipoic acid (P < 0.05). Significant differences in plasma lipid profiles, such as total lipids, total cholesterol, triglycerides, low-density lipoprotein, and high-density lipoprotein, were observed between the HFD and 0.5LA groups. The atherogenic index and the plasma high density lipoprotein-cholesterol/total cholesterol ratio improved significantly with ${\alpha}$-lipoic acid supplementation in a dose-dependent manner (P < 0.05). Total hepatic cholesterol and total lipid concentration decreased significantly in high fat fed rats supplemented with ${\alpha}$-lipoic acid in a dose-dependent manner (P < 0.05), whereas liver triglyceride content was not affected. In conclusion, ${\alpha}$-lipoic acid supplementation had a positive effect on weight gain and plasma and liver lipid profiles in rats.
Objectives: Samjunghwan has been known to be effective for the treatment of obesity. To show the effectiveness of Samjunghwan in a more scientific way, Samjunghwan extract was prepared and evaluated in high-fat diet rats by measuring the changes of body weight and lipid metabolism as described below. Methods: 245 g of crushed Samjunghwan was extracted with methyl alcohol. The extract was evaporated under reduced pressure to give 21.8 g. For 30 days, the control group rats were given a high fat diet, while the test group rats were given a high fat diet plus Samjunghwan extract. The normal group rats were given a normal diet. 50 mg of Samjunghwan extract per 1 kg of body weight was added to the diet for the test group rats. Results: The control group rats on a high fat diet gained weight by about 27-28% as compared to the normal group, whereas the test group rats on a high fat diet plus Samjunghwan extract lost weight about 6-8% as compared to the control group. A significant increase of liver weight caused by a high fat diet was also inhibited by the same Samjunghwan extract administration. Similar inhibitory effects on the food intake and on the epididymal adipose tissue weight were observed in the high fat diet rats by the administration of Samjunghwan extract. Serum and liver total lipid levels in the control group on a high fat diet increased significantly as compared to the normal group, whereas their serum and liver levels increased less on a high fat diet plus Samjunghwan extract administered test group than the control group. Impressively, serum leptin levels in the test group decreased almost to the level of the normal group, which was well in accordance with the decreased fat contents in the test group rats. Furthermore, the activities of hepatic acetyl-CoA carboxylase and fatty acid synthetase were increased in the control group, while their activities in the test group on a high fat diet plus Samjunghwan extract decreased nearly to the levels of normal group rats on a normal diet. Conclusions: These results showed that the obesity caused by a high fat diet was effectively inhibited the administration of Samjunghwan extract. Our results also showed that the abnormal lipid metabolism caused by a high fat diet was effectively cured by the administration of Samjunghwan extract.

The purposes of this study is to investigate the effect of Rhus chinensis gall extract on liver function, plasma lipid composition and antioxidant system in the obese rats with high fat diet for seven weeks. Thirty two male rats of Sprague Dawely strain were divided into four groups. they are normal group (basal diet), control group(high fat diet), III group(high fat diet with Rhus chinensis gall extract 25mg/kg body weight per day) and IV group(high fat diet with Rhus chinensis gall extract 250mg/kg body weight per day). Rats in III group and IV group were administered with Rhus chinensis gall extract accordingly. Weight gains showed a tendency to decrease in rat with Rhus chinensis gall extract group but showed no differences among treatment groups. Food intake and Food efficiency ratio were not significantly different among treatment groups. Plasma total cholesterol showed a tendency to decrease in Rhus chinensis gall extract group. HDL cholesterol showed a tendency to decrease in 25mg/kg of Rhus chinensis gall extract group. However, in the 250mg/kg of Rhus chinensis gall extract group, these values showed no significant difference compared to the control group (p<0.05). GPT activities showed no significant difference among treatment groups. GOT activities showed a tendency to decrease in the groups of Rhus chinensis gall extract groups. Lipid peroxide level was significantly higher in control group than those of normal group. In Rhus chinensis gall extract groups, lipid peroxide level showed a tendency to decrease, but glutathione peroxidase activity was progressively increased. Results of our research in this paper show that Rhus chinensis gall extract might improve liver function, antioxidant system and plasma lipid composition in rats with high fat diet.
Objectives: The objective of the present study was to investigate the lipid and the antioxidant regulatory potential of a multigrain diet in laboratory animals with reference to lipid profiles, tissue lipid peroxidation and antioxidant status. Methods: Two types of diets, with or without addition of cholesterol, were used in the study - a commercial diet and a formulated multigrain diet (with Sorghum vulgare, Avena sativa, Pennisetum typhoideum, Oryza sativa, Eleusine coracana and Zea mays grains). After a 10-week period of feeding the diets to albino rats the plasma, liver and fecal lipid profiles and the hepatic and renal antioxidant status of the animals that were fed the commercial and the formulated diets (with and without cholesterol addition) were assessed. Results: The commercial diet supplemented with cholesterol elevated the levels of plasma total lipids, total cholesterol, triglycerides, low-density lipoprotein cholesterol (LDL-C), and very low-density lipoprotein cholesterol (VLDL-C), as well as the atherogenic index (AI). The high-density lipoprotein cholesterol (HDL-C) content and the antioxidant profiles (total ascorbic acid, superoxide dismutase, catalase, glutathione peroxidase reduced glutathione) declined along with increases in lipid peroxidation. The formulated diet (with and without addition of cholesterol) was found to be more efficient than the commercial diet in controlling plasma, hepatic and fecal lipid profiles, as well as hepatic and renal lipid peroxidation and antioxidant status, than of the hypercholesteremic animals. Conclusion: The multigrain diet used in the present study is effective in countering the hyperlipidemia and oxidative stress caused by high cholesterol intake.
This study examined the antioxidant activity and lipid composition of rats fed a high fat-high cholesterol diet containing Salvia plebeia water extract. Three levels (50 mg/kg, 200 mg/kg, 500 mg/kg) of Salvia plebeia water extract were administered to seven weeks old male Sprague Dawley rats for four weeks. The activity of glutathione peroxidase in serum was high in all groups that received the Salvia plebeia water extract (p<0.05). The glutathione reductase activity was high (p<0.05) in groups that received 50 mg/kg and 500 mg/kg of Salvia plebeia water extract. When rats received Salvia plebeia water extract, the content of MDA (malondialdehyde) in the serum and liver was low in all groups. In addition, the serum total lipid, triglyceride, total cholesterol and LDL-cholesterol contents decreased significantly (p<0.05). Total cholesterol and triglyceride levels in the liver were significantly lower in the group that received the Salvia plebeia water extract than in the control group (p<0.05). Salvia plebeia water extract improves lipid metabolism and it almost meets with the increase of antioxidant activity and peroxide formation reduction. Based on these results, supplemented of a high fat-hypercholesterol diet with Salvia plebeia can increase antioxidant activity and depress peroxide formation in serum, as well as liver cholesterol and triglycerides.
Peroxidative stimuli mediated by high polyunsaturated fatty acid administration in rats induced in vivo lipid peroxidation and resulted in increased urinary excretion of a number of lipophilic aldehydes and related carbonyl compounds. These secondary lipid peroxiation products, measured as 2, 4-dinitrophenylhydrazine deritives, were detected and identified by the newly developed HPLC method. The identified urinary lipophilic nonpolar aldehydes and related carbonyl compounds were butanal, butan-2-one, pentan-2-one, hexanal, hex-2-enal, hepta-2, 4-dienal, hept-2-enal, octanal, and oct-2-enal. Lipophilic polar aldehydes such as 4-hydroxyhex-2-enal and 4-hydroxyoct-2-enal were also identified. A polyunsaturated fatty acid diet containing n-3 fatty acids generally caused high levels of urinary excretion of lipophilic aldehydes and related carbonyl compounds in rats than a normal diet. Significantly increased secondary lipid peroxidation products were hexanal, hepta-2, 4-dienal, octanal, 4-hydroxyhex-2-exal, 4-hydroxyoct-2-enal, and a number of unidentified compunds.
Objectives : Mori Folium is used for hyperlipidemia or diabetes. The study was designed to test the anti-obesity effect of Mori Folium on body weight, hepatic fat accumulation and serum lipid level. Methods : The extract from Mori Folium was made by the pharmacy department of Kyung-hee oriental medical hospital. Normal diet, high-fat diet, 30% reduced diet and Mori Folium groups were set. The normal group was administered normal rat food, but the other three groups were administered high fat food. We measured body weight once a week. After 3 weeks experiment, hepatic lipid accumulations were measured. Also we compared serum lipid levels among the 4 groups after 3 weeks. Results : Mori Folium had no effect on body weight, but the 30% reduced diet had an inhibitory effect on body weight gain. Mori Folium had more inhibitory effect on hepatic fat accumulation than a 30% reduced diet. Mori Folium also has more inhibitory effect on serum triglyceride than 30% reduced diet. Conclusions : Mori Folium has anti-obesity effect on hepatic lipid accumulaion and serum triglyceride level in high fat diet induced rats.
