Isolation and Identification of Marine Bacteria with High Removal Efficiencies for Nitrogen-Phosphate In Gwangyang bay (광양만 해역에서의 고효율 질소-인 제거 해양박테리아 탐색 및 분리)

  • 371 strains of marine bacteria were isolated from Gwangyang bay in Korea. The dominant species were identified as Pseudomonas aeruginosa, Aeromonas hydrophila, P. fluorescens, P. paucimobilis, Chryseomonas luteola and P. vescularis. To screen marine bacteria capable of removing nutrients and organics, marine bacteria was inoculated in 10 mL of marine broth 2216 (DIFCO) with $NH_3-N$ (100 mg/L), ${NO_3}^{-}-N$ (100 mg/L), and ${PO_4}^{-3}-P$ (10 mg/L) with 1.0% (v/v), and incubated for 12 h. Results from the screening test, showed that the removal efficiencies for $COD_{Cr}$, ammonia niterogen, nitrate nitrogen, and phosphate were over 25% for 16 strains, 15% for 9 strains, 50% for 63 strains, and 15% for 80 strains, respectively. Aeromonas hydrophila, Chryseomonas indologenes, Pseudomonas diminuta, Vibrio parahaemolyticus were selected for nutrients removal experiments. For the batch test, 4 species of marine bacteria were inoculated in modified marine broth containing with nutrients($COD_{Cr}$ 250 mg/L, $NH_3-N$ 40 mg/L, ${NO_3}^{-}-N$ 40 mg/L, ${PO_4}^{3-}-P$ 10 mg/L, respectively), incubated for 10 hr and the removal efficiencies were measured.

  • For the research of the natural marine antioxidant, several bacteria were isolated from the coast of jin-Hae in Korea. Among the marine bacteria studied, strain HJ-14, a gram-negative, motile, strait rod, aerobic, and $Na^{+}$ required bacterium showed high activity of 1,1-diphenyl-2-picrylhydrazyl radical scav- enging. The morphological, physiological, and biochemical characteristics of the strain HJ-14 were similar to those of the Alteromonas macleodii ATCC $27126^{T}$ . Thus, it was tentatively identified as Alteromonas sp. HJ-14. The compositions of major fatty acids in cell membrane of Alteromonas sp. HJ-14 were $C_{ 14:0}$ , $ C_{15:0}$ , $C_{16:0}$ and $C_{17:1}$ $_{w8c}$ , which also suggest that it is affiliated with Alteromonas sp. The optimum culture conditions for production of antioxidant materials with Alteromonas sp. HJ-14 were at $25$~$37^{\circ}C$ and pH 6~8. The optimum conditions for the production of antioxidant for carbon, inorganic nitrogen, and sodium chloride sources were 2.5%(w/v) dextrin, 0.5%(w/v) ammonium sulfate, and 2~6%(w/v) sodium chloride, respectively. The hydroxyl radical scavenging ability of Alteromonas sp. HJ-14 broth was 90.03%, which is higher than ascor-bic acid(83.28%) and lower than butylated hydroxyanisole(95.46%) and $\alpha$-tocopherol(97.17%).

Characteristics of Phytophthora capsici Causing Pepper Phytophthora Blight Resistant to Metalaxyl (Metalaxyl에 대한 저항성 고추 역병균의 특성)

  • Isolation frequency of resistant isolates of Phytophthora capsici to metalaxyl was reported to be 38.9% through the resistance monitoring for metalaxyl in P. capsici causing pepper Phytophthora blight. Metalaxyl was very effective to mycelium growth, while not to zoosporangium germination and zoospore release. $EC_{50}$ values of metalaxyl in the inhibition of mycelium growth were 0.204, 0.151, 0.379, and $0.215\;{\mu}g\;mL^{-1}$ against each isolate sensitive to the fungicide as P. capsici 06-119, 06-143, P08-7, and P08-31, respectively, whilst those were 5.242, 5.724, 6.621, and $5.377\;{\mu}g\;mL^{-1}$ in P. capsici 06-125, 06-155, P08-50, and P08-60. For the field fitness, several factors, which were mycelium growth, zoosporangium germination, zoospore release, virulence to pepper plants, and the zoosporangium and the oospore production, were investigated with 4 sensitive isolates and 4 resistant isolates. Between 2 groups differentiated by the sensitivity of metalaxyl, there was no significance in mycelium growth, zoosporangium germination, zoospore release, and virulence to pepper plants. However, the zoosporangium and the oospore production in each resistant isolate, which were related to survival of P. capsici in fields, were superior to those of sensitive isolates. Based on results of this study, it was suggested that the increase of the percentage of resistant isolates to metalaxyl resulted from the high capacities of the zoosporangium and the oospore production.

Isolation of a Fibrinolytic Bacterium from Cheongkukjang and Characterization of Its Bioactivity (청국장으로부터 Fibrin 분해 세균의 분리 및 이를 이용한 발효 청국장의 생리활성)

  • In this study, we have isolated and identified a proteolytic bacterium from conventional Cheongkukjang. We also characterized several bioactivity of Cheongkukjang, which was fermented by an isolated strain. One strain out of about $10^4$ strains obtained from Cheongkukjang showed relatively high proteolytic activity was selected and named as a Bacillus subtilis LSH805 strain. White soy-bean Cheongkukjang possessed less odor and more viscous substance than black soy-bean Cheongkukjang. Cheongkukjang showed fibrinolytic activity, and about 1,500 mg fibrin was degraded after 20 h incubation. Although nitric oxide (NO) assays of soy-bean and Cheongkukjang were almost the same, their activities were significantly higher than that of no treatment. Activity of water fraction of Cheongkukjang was somewhat higher than that of soy-bean. Fibrinolytic and NO assays of Cheongkukjang suggest that Cheongkukjang, which was fermented by an isolated strain may be a useful candidate for natural fibrinolytic and macrophage-stimulating agents.

Prophylactic Effects of Bifidobacterium longum HY8001 against Escherichia coli O157:H7 and Salmonella typhimurium DT104 Enteric Infection and Evaluation of Vero Cytotxin Neutralizing Effects (Bifidobacterium longum HY8001 균주의 Escherichia coli O157:H7과 Salmonella typhimurium DT104 장곤 내 감염 예방효과 및 Vero cytotoxin 중화효과)

  • Prophylactic effects of Bifidobacterium longum HY8001, Korean isolate, against Escherichia coli O157:H7 and Salmonella typhimurium DT104 enteric infection were examined at four groups of specific pathogen free(SPF)-ICR mouse for each pathogen. B. longum HY8001+B. typhimurium DT104+B. longum HY8001(BL+ST+BL) group and B. longum HY8001+E. coli O157:H7+B. longum HY8001(BL+E+BL) group were fed with B. longum HY8001 before and after E. coli O157:H7 or s. typhimurium DT104 challenge, while B. longum HY8001+S. typhimurium DT104(BL+ST) and B. longum HY8001+e. coli O157:H7(BL+E) groups were fed with B. longum HY8001 only before E. coli O157:H7 or S. typhimurium DT104 challenge. E. coli O157:H7(E) and S. typhimurium DT104(ST) groups were challenged with each pathogen without B. longum HY8001 administration and control groups were administered with phosphate buffered solution(PBS). After the oral administration with B. longum HY8001(109cfu), th emice were challenged with E. coli O157:H7(2$\times$1010cfu) or S. typhimurium DT104(108cfu) and the mortality rate and the fecal shedding of challenged pathogen were also examined define the reactivity of the B. longum HY8001. Production of toxin neutralizing substance(s) of B. longum HY8001 was determined by cell cytotoxicity assay using Vero cells. Fecal shedding of th eS. typhimurium DT104 was significantly decreased in BL+ST+BL group fed with B. longum HY8--1 before and after challenge(p<0.05), while the fecal shedding s of S. typhimurium DT104 in BL+ST and St groups remained more than 106cfu. the protective effect of the B. longum HY8001 against E. coli O157:H7 was significantly high only in BL+E+BL group fed with b. longum Hy8001 before and after E. coli O157:H7 challenge from the result of fecal E. coli O157:H7 isolation rate, mortality rate, and intestinal contents culture to detect E. coli O157:H7. the mortality rate of the BL+e and E groups. The cytopathic effect (CPE) of the Vero cytotoxin (Shiga like toxin I & II) in Vero cell was neutralized in B. longum HY8001 culture supernatant added wells which indicate the presence of soluble Vero cytotxin neutralizing substance(s) in B. longum HY8001 culture suprnatant.

Isolation and Characterization of Kimchi Lactic Acid Bacteria Showing Anti-Helicobacter pylori Activity (Helicobacter pylori 억제능 김치 유산균의 분리와 특성 규명)

  • One bacterium, which showed strong antagonistic activity against H. pylori KCCM 41756, was isolated from kimchi. The strain NO1 was designated as Lactobacillus plantarum NO1 based on Gram staining, biochemical properties, and 16S rRNA gene sequencing. The culture medium $(2{\sim}4{\mu}g/ml)$ of Lb. plantarum NO1 reduced $(40{\sim}60%)$ the urease activity of H. pylori KCCM 41756. Lb. plantarum NO1 inhibited the binding of H. pylori to human gastric cancer cell line, AGS cells, by more than 33%. Lb. plantarum NO1 exhibited high viability (maintained initial viable cell count of $10^9CFU/ml$) in 0.05 M sodium phosphate buffer (pH 3.0) for 2 h, in artificial gastricjuice for 2 h and in 0.3%, 0.5% oxgall for 24 h. Hemolysis phenomena did not observed when Lb. plantarum NO1 was incubated in the blood agar media. We concluded that Lb. plantarum NO1 can be a good candidate as a probiotic, harboring anti-H. pylori activity.

A Cost-Efficient Job Scheduling Algorithm in Cloud Resource Broker with Scalable VM Allocation Scheme (클라우드 자원 브로커에서 확장성 있는 가상 머신 할당 기법을 이용한 비용 적응형 작업 스케쥴링 알고리즘)

  • Cloud service users request dedicated virtual computing resource from the cloud service provider to process jobs in independent environment from other users. To optimize this process with automated method, in this paper we proposed a framework for workflow scheduling in the cloud environment, in which the core component is the middleware called broker mediating the interaction between users and cloud service providers. To process jobs in on-demand and virtualized resources from cloud service providers, many papers propose scheduling algorithms that allocate jobs to virtual machines which are dedicated to one machine one job. With this method, the isolation of being processed jobs is guaranteed, but we can't use each resource to its fullest computing capacity with high efficiency in resource utilization. This paper therefore proposed a cost-efficient job scheduling algorithm which maximizes the utilization of managed resources with increasing the degree of multiprogramming to reduce the number of needed virtual machines; consequently we can save the cost for processing requests. We also consider the performance degradation in proposed scheme with thrashing and context switching. By evaluating the experimental results, we have shown that the proposed scheme has better cost-performance feature compared to an existing scheme.

Natural Oleosomes Loading Emulsion Technology -Loading Oleosomes as Delivery Systems for Improved Cosmetic Efficacies- (천연 Oleosomes를 이용한 에멀젼 기술)

  • Natural safflower oleosomes are small ($1{\sim}3{\mu}m$) spherical shaped "reservoir", inside which the seed stores triglycerides for use as a future energy source. The surface of the oleosome is covered with a high molecular weight ($20{\sim}24$ KDa) oleosin protein which has been demonstrated to have emulsification properties. Traditionally, oleosomes from oil bearing seeds such as safflower were simply crushed to liberate the oil within. Our patented DermaSphere technology allows for the isolation of oleosomes in the intact state. Once isolated, these materials can be used in skin care formulations to deliver the emolliency, occlusivity, and anti-oxidant effects typically associated with safflower oil. However, because of the presence of the emulsifying oleosin protein covering the spherical oil body, oleosomes have self-emulsification property as well as can emulsify other oil phase in typical oil-in-water (O/W) emulsion. The oleosomes can literally serve as the entire non-active portion of the oil phase of a typical skin care product. Most importantly, natural oleosomes can be loaded with other oil-soluble active materials and can therefore be used as delivery systems for improved cosmetic efficacies. Oleosomes can be loaded with various actives, such as fragrances, vitamins, inset repellents, and UV chromophores. The loaded oleosomes can be utilized to either protect the active ingredients within the formulation itself or to allow for control release of those actives over time.

A report on 57 unrecorded bacterial species in Korea in the classes Betaproteobacteria and Gammaproteobacteria

  • In an investigation of indigenous prokaryotic species in Korea, a total of 57 bacterial strains assigned to the classes Betaproteobacteria and Gammaproteobacteria were isolated from diverse environments. Samples were collected from fresh water, natural caves, soil, paddy fields, lakes, sea water, jeotgal (fermented seafood), salt flats, soil from abandoned mines, plant roots, digestive organs of both Japanese crested ibis (Nipponia nippon) and Burmese python (Python molurus bivittatus) and tidal flats. From the high 16S rRNA gene sequence similarity (>98.7%) and formation of robust phylogenetic clades with closely related species, it was determined that each strain belonged to an independent and predefined bacterial species within either the Betaproteobacteria or Gammaproteobacteria. There is no official report or publication that describes these 57 proteobacterial species in Korea. Overall, in the class Betaproteobacteria there were 16 species in 12 genera of 4 families in the order Burkholderiales and two species in two genera of one family in the order Neisseriales. Within the class Gammaproteobacteia, there were five species in four genera of four families in the order Alteromonadales, 12 species in 11 genera of one family in the order Enterobacteriales, four species in four genera of three families in the order Oceanospirillales, 11 species in four genera of two families in the order Pseudomonadales, two species in the order Vibrionales and five species in five genera of one family in the order Xanthomonadales. Isolation and Identification of Influenza Viruses from Busan, during 2000-2001 (2000-2001년 부산지역 호흡기 바이러스 발생 양상 비교)

  • Respiratory viruses were isolated from patients with acute respiratory infections in Busan during 2000-2001 and characterized for their antigenic properties. In 2000, 39 out of 43 isolated viruses were identified as influenza viruses and the others were adenoviruses. Among the isolated influenza viruses,23 were type A influenza viruses and 16 were type B influenza viruses. As a result of antigenic characterization, the influenza viruses were determined to A/Sydney/05/97(H3N2)-like, A/Beijing/262/95(H1N1)-like, and B/Harbin07/94-like viruses and serotypes of the isolated adenoviruses were type 1, 2, and 5. In 2001, 56 viruses were isolated and all of the viruses were identified as influenza viruses. They were A/panama/253/99(H3N2)-like and A/Newcaledonia/2007/99(H1Nl)-like viruses when determined by their antigenic properties. The sex distribution of the patients is as follows, 14 males (32.56%),23 females (67.44%) in 2000, and 23 males (41.07%), 33 females (58.93%) in 2001. Occurrence rate was found to be higher in female patients in both years. Age distribution of patients, in 2000, 48.84% of infection occurred in 0 to 1 year old while in 2002, 33.93% occurred among 11-20 year olds. In 2000, occurrence rate was found to be high in January and again in April and various types of viruses were isolated. These results may be useful for vaccine development and establishment of reliable epidemic data.
