The Content Analysis of Amino Acids Including GABA of Chlorella protothecoides under Mixtrophic Culture (혼합영양 배양에서 Chlorella protothecoides의 GABA를 포함한 아미노산 함량 분석)

  • Chlorella is quantitatively and qualitatively high in protein with balanced essential amino acid profiles, vitamins and minerals. ${\gamma}-Aminobutyric$ acid (GABA) is broadly distributed in nature and fulfills multi-physiological functions including effect such as a health-promoting functional compound. To improve the GABA production, Chlorella protothecoides were grown through the modified mixtrophic culture medium containing 2L of sterilized bristol medium with 0.01% urea and 4.0% glucose in a 5L fermenter. The results showed that nineteen kinds of amino acid including GABA at C. protothecoides sample were analyzed using high performance liquid chromatography (HPLC). Glutamic acid in total concentration (%) of amino acid is the most abundant amino acid (33.10%), followed by alanine (20.48%) and GABA (17.48%). Three amino acids including GABA were responsible for more than 70% total concentration in C. protothecoides sample including eight essential and nine non-essential amino acids: aspartic acid, asparagine, serine, glutamine, histidine, glycine, threonine, arginine, tyrosine, valine, methionine, tryptophan, phenylalanine, isoleucine, leucine, lysine. As a result of this experiment, it is expected that Chlorella will be developed to a critical product having high value as, GABA, functional food materials.

  • Effect of pine pollen and defatted pine pollen on lipid accumulation and metabolism in serum and liver were investigated with rats fed high fat-supplemented diets for 6 weeks. Concentrations of total lipid and total cholesterol in serum significantly decreased in the experimental groups (treated with pine pollen and defatted pine pollen) compared with the control group. Concentration of triglyceride in serum was remarkably lower than that of control group. This was similar to that of normal group. Concentration of glucose in serum was significantly lower in the experimental group. It was observed from photographs of hepatic tissue in rats that pine pollen inhibits the lipid accumulation induced by high fat diets.

  • We tested the hypothesis that alpine plants have special physiological and biochemical mechanisms in addition to their structural adaptation in order to survive under extreme conditions. The photosynthetic organs of Pinus pumila were used to examine the seasonal changes in sugar concentration, antioxidative enzyme, and lipid peroxidation. The concentrations of sucrose, glucose, fructose and reducing sugar were the highest in the leaves in April. But sugar contents in buds and inner barks did not respond sensitively on temperature change. Meanwhile superoxide dismutase (SOD) activity responded sensitively on the change of temperature and SOD in all tissues maintained high activity in April. Meanwhile anthocyanin content increased rapidly in June but the increase of anthocyanin content was not enough to prevent their tissues from the damage by the exposure of high temperature or other stress. In conclusion, under low temperature condition, P. pumila increased the concentration of soluble sugars and SOD activity in their tissues in order to overcome extreme environmental condition. But in summer, these stress defense system against high temperature might be disturbed slightly. This results in the increase of malondialdehyde (MDA) contents in three tissues by lipid peroxidation.

  • AMP-activated protein kinase (AMPK) maintains energy homeostasis in skeletal muscle. Nonetheless, its functional role on protein synthesis with different nutrient availability has not been elucidated. Therefore, the purpose of this study is to examine the effect of AMPK activity on protein content in C2C12 myotubes incubated with low (5 mM; LG) or high (25 mM; HG) glucose media. LG stimulated (p<0.05) AMPK and acetyl CoA carboxylase (ACC) activity compare to those in HG group. Total protein content was higher in myotubes cultured with HG than in those cultured with LG and further increased by AICAR (5-amino-1-${\beta}$-D-ribofuranosyl-imidazole-4-carboxamide). Myotubes cultured with HG showed 7.5% lower UbFL (Ubiquitin Firefly Luciferase)-to-SV40 (Simian virus40) ratio compared to those in LG. Glucose level did not change the phosphorylation level of mammalian target of rapamycin (mTOR). Interestingly, administration of AICAR significantly increased phosphorylation level of mTOR in myotubes cultured with LG but not in those with HG. Overall, this data indicate that AMPK activity and protein turnover are finely regulated in response to different glucose concentration.

  • In order to investigate and generalize the effects of carbon and nitrogen sources on the growth of and lipid production in Chlorella sp. 227, several nutritional combinations consisting of different carbon and nitrogen sources and concentrations were given to the media for cultivation of Chlorella sp. 227, respectively. The growth rate and lipid content were affected largely by concentration rather than by sources. The maximum specific growth was negatively affected by low concentrations of carbon and nitrogen. There is a maximum allowable inorganic carbon concentration (less than 500~1,000 mM bicarbonate) in autotrophic culture, but the maximum lipid content per gram dry cell weight (g DCW) was little affected by the concentration of inorganic carbon within the concentration. The lipid content per g DCW was increased when the microalga was cultured with the addition of glucose and bicarbonate (mixotrophic) at a fixed nitrogen concentration and with the lowest nitrogen concentration (0.2 mM), relatively. Considering that lipid contents per g DCW increased in those conditions, it suggests that a high ratio of carbon to nitrogen in culture media promotes lipid accumulation in the cells. Interestingly, a significant increase of the oleic acid amount to total fatty acids was observed in those conditions. These results showed the possibility to induce lipid production of high quality and content per g DCW by modifying the cultivation conditions.

Effects of Hagocho (Prunella vulgaris L.), Gamgook (Chrysanthemum indicum L.) and Galgeun (Pueraria Radix) on Plasma Lipid Compositin and Histological Consideration in Hyperlipidemic Rat (하고초, 감국 및 갈근 증류액이 비만쥐의 지질구성 및 각종 장기의 해부조직학적 소견에 미치는 영향)

  • Effects of prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix on plasma lipid composition and histological consideration were investigated in hyperlipidemic rats. Concentration of plasma ${\beta},-lipoprotein$ showed a tendence to decrease in prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix treatment groups. However these values were showed not significantly different from control group. Concentration of plasma FFA in prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix treatment groups showed a lower values compared to control group and concentration of plasma FFA of prunella vulgaris L. and pueraria Radix treatment group showed a lower values than prunella vulgaris L. and chrysanthemum indicum L. treatment group. Concentration of plasma glucose and triglyceride showed a tendence to decrease in prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix treatment groups. However plasma glucose values showed not significantly different from control group. Plasma total cholesterol concentration showed a low and HDL-cholesterol concentration showed a high in prunella vulgaris L. and pueraria Radix treatment group. However LDL-cholesterol concentrations were not significantly different in treatment groups. Histological consideration of heart, liver and kidney in prunella vulgaris L. and pueraria Radix treatment groups showed a good features in fat accumulation condition than those of other treatment groups. However in the condition of high fat accumulation in tissues, heart, liver and kidney were showed a slight congestion and a bashed cell nucleus.

  • Recombinant Escherichia coli BLR(DE3) harboring the hemA gene from Rhodobacter capsulatus under the control of a constitutive promoter, which we constructed previously, was used for the extracellular production of 5-aminolevulinic acid (ALA). The effects of several factors on ALA production were investigated in flask culture. ALA production by the recombinant E. coli was more efficient at $30^{\circ}C$ than $37^{\circ}C$. The glycine concentration had an important effect on cell growth. Glycine and succinic acid concentration of 5-10 and 10-20 g/L, respectively, resulted in high ALA production. In addition, the partial replacement of succinic acid by sodium glutamate increased the ALA production. The ALA production was inhibited by the presence of glucose in the medium. Using the optimal conditions, an ALA concentration of 8.2 g/L was achieved in jar fermentation without an added inducer or ALA dehydratase inhibitor; this is the highest reported concentration.

  • A Pseudomonas fluorescens was selected from mushrooms and studied in both batch and fed-batch cultures in order to get maximal biomass concentration. P. fluorescens is an aerobic bacterium and antagonistic to Pseudomonas tolaasii which causes blotch disease on the mushroom cap. P fluarescens and P. tolaasii were identified by Gram staining, gelatin liquefaction, oxidase test, etc. and were characterized by pigment production, temperature sensitivity, salt tolerance and rapid pitting test, etc., Celts of P. fluorescens well in medium containing 30g/L of glucose, whereas the growth was inhibited at the glucose levels at higher than 30g/L. The highest values of specific growth rate and productivity were obtained when using 10g/1 of yeast extract. Optimum concentrations of $NH_4Cl$ and ${(NH_4Cl)}_2SO_4$ for culture were found to be 1.0g/L and 0.1g/L respectively. Optimum concentration of $MgSO_4{\cdot}7H_2O$ used as a sulfursource was 1.0g/L. It was also found that the cell concentrations reached the maximum level when grown on the medium containing 1.0g/L of $KH_2PO_4$ and 0.1g/L of $CaCl_2$. Also, the optimum culture conditions were $30^{\circ}C$ and pH 6.0. Cultivation of P. fluarescens at high dissolved oxygen (DO) concentration led to a decrease of bacterial productivity in batch culture. Maximum productivity was achieved at 40% DO concentration.

Physico-chemical properties of late-incubation egg amniotic fluid and a potential in ovo feed supplement

  • Objective: This study explored the physico-chemical properties of late-incubation egg amniotic fluid and a potential in ovo feed (IOF) supplement. Methods: Amniotic fluid was collected from broiler breeders (Ross 308, 51 weeks and Cobb 500, 35 weeks) on day 17 after incubation. A mixture of high-quality soy protein supplement - Hamlet Protein AviStart (HPA) was serially diluted in MilliQ water to obtain solutions ranging from 150 to 9.375 mg/mL. The mixtures were heat-treated (0, 30, 60 minutes) in a waterbath ($80^{\circ}C$) and then centrifuged to obtain supernatants. The amniotic fluid and HPA supernatants were analysed for their physico-chemical properties. Results: Only viscosity and $K^+$ were significantly (p<0.05) different in both strains. Of all essential amino acids, leucine and lysine were in the highest concentration in both strains. The osmolality, viscosity and $pCO_2$ of the supernatants decreased (p<0.05) with decreasing HPA concentration. Heat treatment significantly (p<0.05) affected osmolality, pH, and $pCO_2$, of the supernatants. The interactions between HPA concentration and heat treatment were significant with regards to osmolality (p<0.01), pH (p<0.01), $pCO_2$ (p<0.05), glucose (p<0.05), lactate (p<0.01) and acid-base status (p<0.01) of HPA solutions. The $Ca^{2+}$, $K^+$, glucose, and lactate increased with increasing concentration of HPA solution. The protein content of HPA solutions decreased (p<0.05) with reduced HPA solution concentrations. The supernatant from 150 mg/mL HPA solution was richest in glutamic acid, aspartic acid, arginine and lysine. Amino acids concentrations were reduced (p<0.05) with each serial dilution but increased with longer heating. Conclusion: The values obtained in the primary solution (highest concentration) are close to the profiles of high-protein ingredients. This supplement, as a solution, hence, may be suitable for use as an IOF supplement and should be tested for this potential.

A Study on Fatty Liver Syndrome in Periparturient Holstein Cows (분만전후 젖소의 지방간증에 관한 연구)

  • This study was carried out to investigate the Preventive and treatment effects of mixed compound of vitamin E and selenium, and ursodeoxycholic acid on the pre-parturient and post-parturient fatty liver of high. yielding dairy cows. Thirty pregnant dairy cows(2 weeks preparturition) were divided into clinical(13cows) and subclinical(17 cows) groups based upon blood chemical values, blood serum protein levels and clinical symptoms. The clinical group was subdivided into treated clinical group(6 cows) and non-treated clinical group(7 cows). The cows(treated clinical group) with fatty liver diagnosed before parturition were dosed with mixed compound of vitamin I(1head) and selenium(10mg/head) intramuscularly once before and after parturition, respectively. These cows(treated clinical group) were also given ursodeoxycholic acid(50g/head) intramuscularly 3 times before parturition. The cows(treated clinical group) with fatty liver diagnosed after parturition were also given the same compounds as dosed to the above cows at same dose rate once or 3 times, respectively after parturition. Blood samples were collected from the cows on 4th, 7th and 2nd day of preparturition and on 1st, 7th, 14th and 20th day of postparturition. Blood chemical values(non-esterified fatty acid : NEFA) concentration, serum aspartate aminotransferase activity, serum ${\gamma}$-glutamyl transpeptidase activity, bilirubin concentration, glucose level) were checked and clinical symptoms of the cows were also observed. 1. The cows in non-treated clinical group showed severe depression, anorexia and diarrhea from prepartum period, and retained palcenta, endometritis, ketosis, abomasal displacement and mastitis after parturition. Cows in treated clinical group showed moderate depression, anorexia, diarrhea during preparturient period, but they were relieved within a week from parturition. Cows in subclinical group showed mild depression, anorexia and diarrhea after parturition, but these symptoms were not observed in prepartum period. 2. NEFA concentration, serum aspartate aminotransferase activity, serum ${\gamma}$-glutamyl transpeptidase activity and bilirubin concentration observed In the cows of treated clinical group were significantly lower than those of the cows non-treated clinical group. The blood glucose concentration of the cows in treated clinical group were higher than those of the cows in non-treated clinical group. These results indicate that the preparturient and postparturient fatty liver of high-yielding dairy cows could be reduced by dosing the animals with appropriate amounts of mixed compound of vitamin I and selenium, and ursodeoxycholic acid at proper times of the preparturition and postparturition of the dairy cows.
