The Study on the Effect of Acanthopanax Senticocus Herbal Acupuncture on Metabolic Syndrome in High-fat Diet Fed Mice (가시오가피약침(五加皮藥鍼)이 High-fat Diet로 유발(誘發)된 대사증후군(代謝症候群)에 미치는 영향(影響))

  • Objective : The aim of the study was to investigate the effect of Acanthopanax senticocus(AS) herbal acupuncture on the metabolic syndrome in high-fat diet fed mice. Methods : ICR mice were fed with high-fat diet to induce the metabolic syndrome. During the inducement of the metabolic syndrome, the groups were treated with AS herbal acupuncture with different concentrations(125mg/kg, 250mg/kg and 500mg/kg) to the point of Sinsu(BL23) everyday for 5 weeks. Thereafter, body weight, feed efficiency ratio, blood pressure, blood glucose, insulin level, insulin resistance, oral glucose tolerance test(OGTT), lipid profile(TG, TC, HDL-C, LDL-C, NEFA), mass of liver, histology of white adipose tissue(WAT) and brown adipose tissue(BAT), and expression of GLUT-4 and UCP-1 mRNA were measured. Results : The risk factors of metabolic syndrome such as obesity, non-insulin dependent diabetes mellitus(NIDDM), insulin resistance, hypertension, dyslipidemia were aggravated by high-fat diet for 5-weeks. AS herbal acupuncture inhibited the development of weight gain, hyperglycemia, hyperinsulinemia, insulin resistance, hypertension, dylipidemia and expression of GLUT-4 in WAT and UCP-1 mRNA in BAT, and also improved oral glucose intolerance and distribution of adipose tissue.

The Inhibitory Effects of Soongijeseub-bang(Shunqichushi-fang) on the Obese-mice Caused by High Fat Diet (순기제습방(順氣除濕方)이 고지방식이(高脂肪食餌)로 유발된 비만(肥滿) 백서(白鼠)에 미치는 영향)

  • Objectives : In order to investigate the effects of Soongijeseub-bang(Shunqichushi-fang)(here in after referred to SJB) on the obese gene and obese inhibitory, on C57BL/6 mice were induced by high fat diet. Methods : C57BL/6 mice were divided into three groups(normal, high fat diet with control, high fat diet with SJB extract) and fed for 15 weeks. And observed that, body weight change, final increase of body weight, the weight change of the adipocytes, the level change of ALT, AST, creatinine, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, NEFA, glucose, the expression of ${\beta}3AR$, leptin, and $TNF-{\alpha}$ gene in primary adipocytes and adipocytes tissue. Results : The following results were obtained in this study. 1. SJB 500 mg/kg extract group showed considerable decrease in weight, the final increase of weight and the amount of adipocyte in weight. 2. All experimental group showed that the amount of ALT, AST, total cholesterol, LDL-cholesterol, triglyceride and NEFA were decreased considerably. SJB 500 mg/kg extract group showed the amount of HDL-cholesterol and leptin were increased considerably. 3. All experimental group showed that the revelation of ${\beta}3AR$ in primary adipose cell and 3T3-L1 cell were increased considerably, and that the revelation of leptin in primary adipose cell and 3T3-L1 cell were decreased considerably. SJB $100{\mu}{\ell}/m{\ell}$ extract group showed that the revelation of $TNF-{\alpha}$ were decreased considerably. 4. SJB 500 mg/kg extract group showed that the size of adipocyte in adipocytes tissue were decreased. 5. All experimental group showed that the adipose vacuoles in liver tissue were decreased considerably. Conclusions : Comparison of the results for this study showed that SJB is effective on obesity care and has obese-inhibitory effects in obese mouse induced by high fat diet. So it is respected that the clinical application of SJB can help the treatment of obesity.

Effect of PPARα activator and exercise on angiogenesis of white adipose tissue in high fat diet fed mice (고지방 사료를 섭취한 쥐에서 백색지방조직의 혈관신생에 대한 PPARα activator와 운동의 영향 )

  • It was investigated whether PPARα activator more effectively inhibits angiogenesis of white adipose tissue in exercise mice that ate high fat diet compared to non-exercise mice that ate high fat diet. Male mice were randomly divided into a control group not treated with a PPARα activator fenofibrate and exercise (Con), a group treated with fenofibrate alone (FF), a group treated with exercise alone (Ex), and a group treated with a combination of fenofibrate and exercise (Ex+FF). (Ex+FF). All mice was fed high-fat diet for 8 weeks. The weight of white adipose tissue and the size of white adipocytes decreased in FF, Ex, and Ex+FF compared to Con, and decreased more in Ex+FF Ex+FF compared to FF. In white adipose tissue, the gene expression of MMPs and angiogenic factors decreased in FF, Ex, and Ex+FF compared to Con, and more decreased in Ex+FF compared to FF. On the other hand, gene expression of angiogenic inhibitors increased in FF, Ex and Ex+FF compared to Con, and increased more in Ex+FF compared to FF. Therefore, this study revealed that the combined treatment of fenofibrate and exercise effectively inhibits the angiogenesis of white adipose tissue, reducing the increase in white adipose tissue and suppressing abdominal obesity, rather than the single treatment of fenofibrate.

Bifidobacterium bifidum DS0908 and Bifidobacterium longum DS0950 Culture-Supernatants Ameliorate Obesity-Related Characteristics in Mice with High-Fat Diet-Induced Obesity

  • Probiotic supplements have promising therapeutic effects on chronic diseases. In this study, we demonstrated the anti-obesity effects of two potential probiotics, Bifidobacterium bifidum DS0908 (DS0908) and Bifidobacterium longum DS0950 (DS0950). Treatment with DS0908 and DS0950 postbiotics significantly induced the expression of the brown adipocyte-specific markers UCP1, PPARγ, PGC1α, PRDM16 and beige adipocyte-specific markers CD137, FGF21, P2RX5, and COX2 in C3H10T1/2 mesenchymal stem cells (MSCs). In mice with high-fat diet (HFD)-induced obesity, both potential probiotics and postbiotics noticeably reduced body weight and epididymal fat accumulation without affecting food intake. DS0908 and DS0950 also improved insulin sensitivity and glucose use in mice with HFD-induced obesity. In addition, DS0908 and DS0950 improved the plasma lipid profile, proved by reduced triglyceride, low-density lipoprotein, and cholesterol levels. Furthermore, DS0908 and DS0950 improved mitochondrial respiratory function, confirmed by the high expression of oxidative phosphorylation proteins, during thermogenesis induction in the visceral and epididymal fat in mice with HFD-induced obesity. Notably, the physiological and metabolic changes were more significant after treatment with potential probiotic culture-supernatants than those with the bacterial pellet. Finally, gene knockdown and co-treatment with inhibitor-mediated mechanistic analyses showed that both DS0908 and DS0950 exerted anti-obesity-related effects via the PKA/p38 MAPK signaling activation in C3H10T1/2 MSCs. Our observations suggest that DS0908 and DS0950 could potentially alleviate obesity as dietary supplements.

Anti-obesity effects of herbal extract YY312 in C57BL/6 mice fed a high-fat diet and 3T3-L1 cells (백모근(白茅根) 청피(靑皮) 오수유(吳茱萸) 복합방(複合方)(YY312)의 고지방식이로 유도된 마우스와 3T3-L1 세포에서 항비만 효과)

  • Objectives : The purpose of this study was to determine the anti-obesity effect and molecular mechanism of YY312, a herbal extract composed of Imperatae Rhizoma, Citri Unshius Pericarpium Immaturus, and Evodiae Fructus, on a high-fat diet-induced animal model and on 3T3-L1 cells. Methods : C57BL/6 mice were fed for 6 weeks with a normal diet or a high-fat diet (HFD). Then they orally administered daily with 300 mg/kg YY312 for next 10 weeks. Body weight and food consumption were recorded weekly and daily, respectively. Tissue weights, serum lipid, and glucose levels were analyzed at the end of the study. Additionally, the effects of YY312 on adipocyte differentiation in 3T3-L1 cells were examined. After differentiating 3T3-L1 cells were treated with YY312, Oil-red O staining, RT-PCR, and Western blotting were performed for lipid accumulation, mRNA expression of adipogenesis gene, and AMP-activated protein kinase (AMPK) phosphorylation, respectively. Results : YY312-administered mice showed a significant reduction of body weights and abdominal adipose tissue weights. YY312 also reduced the serum levels of triglycerides and total cholesterol, compared with the HFD group. Treatment with YY312 inhibited lipid accumulation and blocked expression of adipogenic transcription factors and lipogenesis genes, including peroxisome proliferator-activated receptor ${\gamma}$, CCAT/enhancer binding protein ${\alpha}$ and fatty acid synthase. YY312 increased AMPK phosphorylation in 3T3-L1 adipocytes. Conclusions : This study showed that herbal extract YY312 has an anti-obesity effect in vitro and in vivo. Thus, YY312 could be developed as a supplement for reduction of body weight gain induced by an HFD.

Effect of Gardenia Jasminoides on Insulin Resistance Induced by Non-alcoholic Fatty Liver Disease (치자 추출물의 지방대사조절을 통한 비알콜성 지방간 유도 인슐린저항성 개선 효과)

  • Gardenia jasminoides is an herbal medicine that treats obesity and dampness-phlegm. This study aimed to investigate the efficacy of Gardenia jasminoides on insulin resistance induced by Non-alcoholic fatty liver disease (NAFLD). 8-week-old C57BL/6 male mice were divided into three groups: control group (Ctrl), high-fat diet group (HFF), and high fat diet with Gardenia jasminoides extract administration group (GJT). Each 10 mice was allocated to each group (a total of 30 mice). All mice were allowed to eat fat-rich diet freely throughout the experiment. To examine the effect of Gardenia jasminoides, we observed weight changes, lipid blot distributions, PPAR-${\gamma}$, p-IkB, p-JNK in liver tissue, total cholesterol, and glucose levels in serum. Comparing of body weight measurements between 3 groups, in the GJT group, weight gain was significantly suppressed compared to the HFF group. The distribution of lipid blots and positive reaction of PPAR-${\gamma}$ were significantly lower in GJT group. The expression levels of p-$I{\kappa}B$ and p-JNK that plays critical roles in the development of insulin resistance were significantly decreased by GJ treatment. Total cholesterol and glucose levels in serum were also significantly lower in GJT group. Gardenia jasminoides has the effect of improving non - alcoholic fatty liver induced insulin resistance through the regulation of lipid metabolism.

Improvement of menopausal obesity by Schizandra chinensis extract (오미자(五味子) 추출물의 갱년기 비만 개선 효과 연구)

  • Objectives: The purpose of this study was to investigate the anti-obesity effects of the aqueous extract of Schizandra chinensis (SC) in menopausal mice. Methods: To induce menopausal obesity, female mice were ovariectomized (OVX) and fed a high-fat diet (HFD; 60% fat, 28% carbohydrates, 14% protein) for 12 weeks. The mice were divided into 6 groups (n = 8): NOR (sham-operated and vehicle-treated), HFD+OVX (vehicle-treated), E2 (17-beta estradiol 50 ㎍/kg-treated), SC1 (1 mg/kg SC-treated), SC10 (10 mg/kg SC-treated), and SC100 (100 mg/kg SC-treated). Samples were orally administered for 6 weeks, after which all experimental mice were sacrificed. Body weight, feeding efficiency, white adipose tissue weight, adipocyte diameter, and fat vacuoles in liver were analyzed. Results: By treating with SC extract, the body weight and feeding efficiency of mice were significantly decreased. The weight of visceral fat tissues was decreased in the SC10 and SC100 groups. Histopathology showed that fat cell diameters of white adipose tissue were also decreased in the SC10 and SC100 groups. Additionally, SC extract regenerated the hepatocyte damage and decreased the size and number of follicular adipocytes Conclusion: In summary, these results suggest that SC has inhibitory effects against menopausal obesity. Schizandra chinensis may be a potential alternative for obesity among female menopausal diseases.

Dietary supplementation with Korean pine nut oil decreases body fat accumulation and dysregulation of the appetite-suppressing pathway in the hypothalamus of high-fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: Korean pine nut oil (PNO) has been reported to suppress appetite by increasing satiety hormone release. However, previous studies have rendered inconsistent results and there is lack of information on whether dietary Korean PNO affects the expression of satiety hormone receptors and hypothalamic neuropeptides. Therefore, our study sought to evaluate the chronic effects of Korean PNO on the long-term regulation of energy balance. MATERIALS/METHODS: Five-week-old male C57BL/6 mice were fed with control diets containing 10% kcal fat from Korean PNO or soybean oil (SBO) (PC or SC) or high-fat diets (HFDs) containing 35% kcal fat from lard and 10% kcal fat from Korean PNO or SBO (PHFD or SHFD) for 12 weeks. The expression of gastrointestinal satiety hormone receptors, hypothalamic neuropeptides, and genes related to intestinal lipid absorption and adipose lipid metabolism was then measured. RESULTS: There was no difference in the daily food intake between PNO- and SBO-fed mice; however, the PC and PHFD groups accumulated 30% and 18% less fat compared to SC and SHFD, respectively. Korean PNO-fed mice exhibited higher messenger RNA (mRNA) expression of Ghsr (ghrelin receptor) and Agrp (agouti-related peptide) (P < 0.05), which are expressed when energy consumption is low to induce appetite as well as the appetitesuppressing neuropeptides Pomc and Cartpt (P = 0.079 and 0.056, respectively). Korean PNO downregulated jejunal Cd36 and epididymal Lpl mRNA expressions, which could suppress intestinal fatty acid absorption and fat storage in white adipose tissue. Consistent with these findings, Korean PNO-fed mice had higher levels of fecal non-esterified fatty acid excretion. Korean PNO also tended to downregulate jejunal Apoa4 and upregulate epididymal Adrb3 mRNA levels, suggesting that PNO may decrease chylomicron synthesis and induce lipolysis. CONCLUSIONS: In summary, Korean PNO attenuated body fat accumulation, and appeared to prevent HFD-induced dysregulation of the hypothalamic appetite-suppressing pathway.

Synergistic combination effect of anti-obesity in the extracts of Phyllostachys pubescence Mael and Scutellaria baicalensis Georgi (죽엽(竹葉)과 황금(黃芩) 복합물의 항비만 효과)

  • Objectives : Anti-obesity drugs that have been developed so far have limited efficacies and considerable adverse effects affecting tolerability and safety. Therefore, most anti-obesity durgs have been withdrawn. We tried to develop anti-obesity agent by combinations from herbs that are used in food ingredients as well as in traditional medicines. Methods : The 80% (v/v) ethanol extracts from Bamboo (Phyllostachys pubescence) leaf (BL) and Scutellaria baicalensis (SB) and their 1:1 combination (BLSB) was evaluated on high fat diet induced obese mice compared to Omega-3 as a positive control. The mice were divided into six groups (n=5), one group fed a normal diet (ND), and the others fed a high fat diets for eight weeks. Two weeks after starting feeding the diets, the high fat diet groups were orally administered vehicle and Omega-3, BL, SB, and BLSB at dosage of 200 mg/kg/day for six weeks. All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : BLSB group showed significant reductions in body weight gain and fat weights of liver and epididymal adipose tissue compared to BL or SB alone as well as control. Total-cholesterol and LDL-cholesterol levels significantly decreased, and HDL-cholesterol level increased. In liver tissue, macrovesicular steaotisis was remarkably improved and its fat cell size was also significantly decreased. Conclusions : These results suggested that a combination preparation of bamboo leaf and S. baicalensis has anti-obesity effect and have synergistic effect compared to bamboo leaf or S. baicalesis.
