BACKGROUND/OBJECTIVES: Activating brown adipose tissue (BAT) and browning of white adipose tissue (WAT) can protect against obesity and obesity-related metabolic conditions. Cryptotanshinone (CT) regulates lipid metabolism and significantly ameliorates insulin resistance. Adenosine-5'-monophosphate (AMP)-activated protein kinase (AMPK), a receptor for cellular energy metabolism, is believed to regulate brown fat activity in humans. MATERIALS/METHODS: The in vivo study included high-fat-fed obese mice administered orally 200/400 mg/kg/d CT. They were evaluated through weight measurement, the intraperitoneal glucose tolerance test (IPGTT), intraperitoneal insulin tolerance test (IPITT), cold stimulation test, serum lipid (total cholesterol, triglycerides, and low-density lipoprotein) measurement, hematoxylin and eosin staining, and immunohistochemistry. Furthermore, the in vitro study investigated primary adipose mesenchymal stem cells (MSCs) with incubation of CT and AMPK agonists (acadesine)/inhibitor (Compound C). Cells were evaluated using Oil Red O staining, Alizarin red staining, flow cytometry, and immunofluorescence staining to identify and observe the osteogenic versus adipogenic differentiation. Quantitative real-time polymerase chain reaction and the Western blot were used to observe related gene expression. RESULTS: In the diet-induced obesity mouse model mice CT suppressed body weight, food intake, glucose levels in the IPGTT and IPTT, serum lipids, the volume of adipose tissue, and increased thermogenesis, uncoupling protein 1, and the AMPK pathway expression. In the in vitro study, CT prevented the formation of lipid droplets from MSCs while activating brown genes and the AMPK pathway. AMPK activator enhanced CT's effects, while the AMPK inhibitor reversed the effects of CT. CONCLUSION: CT promotes adipose tissue browning to increase body thermogenesis and reduce obesity by activating the AMPK pathway. This study provides an experimental foundation for the use of CT in obesity treatment.
Purpose: The purpose of this study was to analyze effects of a community-based case management program for clients with hypertension living in the community. Methods: The research design was a one group pre and post-test design with 30 participants with hypertension who agreed to participate in the 8-12 week case management program provided by case managers from the National Health Insurance Corporation in 2002. Data were collected three times, before and after the case management services, and 6 months later. Outcomes included changes in blood pressure, knowledge of hypertension and daily life practices, including alcohol consumption, smoking, exercise, and medication adherence. Results: Repeated-measures ANOVA and post-hoc tests of means revealed significant differences before and after service for systolic blood pressure, daily life practices (monitoring body weight and BP, low salt and cholesterol and high vegetable diet, and stress-relief practices), and exercise. The goal for medication adherence was attained after service. Significant improvements from baseline to 6 months after service were observed in measures of salt and vegetables in diet. There were no significant differences on hypertension knowledge, alcohol consumption or smoking behavior between before service and after, and at 6 months. Conclusion: The findings provide preliminary evidence that case management intervention can have positive outcomes on BP control, daily life practices, exercise, and medication adherence for clients with hypertension. However, additional interventions are needed to sustain long-term effects.
Allium hookeri is a plant species native to India, Sri Lanka, Myanmar, and China. The plant is widely cultivated in Korea lately as a medicinal food item. This study was conducted to evaluate the effects of Allium hookeri (A. hookeri) on lipid metabolism in Type II diabetic mice (n=8/group, 5 groups). High fat diets with dextrin as a positive control (Dex), leaf (AL), root (AR), and fermented root (FAR) at 3% of diet were fed to all experimental mouse, respectively for 8 weeks. Body weight gain, liver and epididymal fat weights, and excreted fecal lipid levels were measured. Serum and hepatic lipid profiles were analyzed, and fat accumulation in liver was evaluated. In this study, body weight gain and epididymal fat weight were lower in the FAR group, while serum HDL-cholesterol level and excreted fecal total lipid and triglyceride levels were higher in AL or FAR groups. These results suggest that A. hookeri, specially fermented root can be a useful food item to control lipid metabolism in diabetic mice.
This study was performed to investigate the effect of feeding garcinia cambogia extract (HCA) and/or L-carnitine and exercise (swimming) on body weight in rats. Forty-eight male rats (Charles River CD (SD) IGS) of eight weeks old and weighing $323.5\pm2.4g$ were raised for two months with high fat diet ($40\%$ fat as calorie) to induce obesity. After induction of obesity, rats weighing $552.8\pm5.8g$ were blocked into eight groups according to body weight and raised for six weeks with diet containing HCA and/or L-carnitine. Plasma aspartate aminotransferase, alanine aminotransferase, total protein and albumin levels were in normal ranges. Food and calorie intakes of H + C and H + C/H groups were highest among non-exercise (m) groups and exercise (I) groups. Food and calorie intakes of m groups were higher than those of I groups. E groups showed significantly lower body weight gain and calorie efficiency ratio than m groups and H/E group was the lowest among all groups. Perirenal fat pad weights of E groups were lower than m groups, and epididymal fat pad weights of H and H + C groups were lower than CO and C groups among m groups regardless of exercise treatment. Brown adipose tissue weights of I groups were higher than fn groups and highest in H/E group among all groups. Liver citrate lyase activities of H and C groups were lower than CO and H + C groups regardless of exercise treatment. The differences between NE groups and E groups were not found to be significantly. Liver carnitine acyltranferase activity was not significantly different among all groups. Fecal total lipid, triglyceride and total cholesterol excretions were found to be higher in H and H + C uoups compared to CO and C groups both in NE and E animals, those of lipid excretions of NE groups were higher than E groups. In conclusion, HCA was more effective in reduction of body weight and body fat than L-carnitine or HCA + L-carnitine. HCA ingestion with exercise was far more effective than without exercise treatment in reducing body weight and body fat.
The aim of investigated the role of FABP5 in the hepatic lipogenesis and lipid metabolisms. Mice were overexpressed and silenced liver FABP5 using virus particles. Mice were fed a Western-type diet or regular chow for 1week and then sacrificed mouse after 24hr fasted. Liver homogenates were used for protein analysis by Western blot and mRNA levels by RT-PCR. Hepatic and serum lipids were analysed by thin-layer chromatography. Mice fed a Western-type or high saturated fat diet revealed large increases in FABP5 expression. However, FABP5 mRNA levels were drastically reduced under fasted. Hepatic TG was significantly increased FABP5-OEAV mice, but a significantly decreased hepatic free cholesterol under fed. The discovered a substantial decrease in hepatic TG mass with FABP5 silencing. In these data, presented evidence for an important role of FABP5 in hepatic lipogenesis and hepatic TG storage. FABP5 may also be a potential target in the treatment of NAFLD, metabolic syndrome, and obesity. Furthermore, studies to which transcription factors are involved in FABP5 expression and regulation.
Purpose: The atherogenic index of plasma (AIP) is gaining recognition as a superior predictor of coronary artery disease. This study examined the relationship between the AIP and the intake of various food groups in Korean adults, using the baseline data from the Korean Genome and Epidemiology Study-The Health Examinees (KoGES-HEXA) study. Methods: This study included 133,381 adults (46,288 men, 87,093 women) who completed the nutrition survey and biochemical tests of the KoGES-HEXA study. The AIP was calculated as the log (triglycerides/high-density lipoprotein-cholesterol) and was divided into quintiles according to sex for further analysis. Data on food group intake were obtained using the semi-quantitative food frequency questionnaire, and daily food group intake (g/d) was evaluated for each individual. The association between the food group intake and the AIP was examined using the linear regression analysis after adjusting for the confounding variables. Results: In this study population, there was a positive correlation between energy intake and the AIP in men whereas an inverse assocation was shown in women (p < 0.001). In women, a rise in the AIP significantly correlated with an increased percentage of energy intake derived from carbohydrates but decreased percentages of energy from protein and fat (p < 0.001). In men and women, the consumption of grains and kimchi/pickles increased as the AIP rose, while the intake of legumes, nuts, meat and its products, eggs, fish, and milk and dairy products decreased. Among men, individuals in the highest AIP group showed higher intake of wheat products than those in the lowest group of the AIP. In women, the AIP was inversely correlated with potatoes and beverages consumption. Conclusion: This study highlights the importance of a balanced diet, including various protein sources, milk and dairy products, legumes, and nuts, for preventing cardiovascular diseases. Further research into sex-specific dietary patterns is essential for tailoring appropriate dietary recommendations.
The present study was investigated to evaluate on the health improving effect of Kochujang (Fermented Korean hot pepper paste), which was famous as Korean traditional healthy food among foreigners as it was served together with Bibimbap. For comparison, observation was made on the experimental animals (Sprague-Dawley rat) as breeding during 5 weeks, which were divided three groups as into the control groups (N-group) fed with normal feed only and high cholesterol feed only (H-group) and the K-group with combined diet of normal feed and Kochujang extracts (Water soluble complex materials). The most outstanding result was found as the definite fatty liver lesions found from all (100%) of the rats of the H-group at the abdominal opening examination. On the other hand, there was any finding on fatty liver lesions in the N-group as well as the K-group. Therefore, it was more than enough to identify the hypolipidemic effect by visual finding only. Furthermore, from the results of blood plasma analysis from SD-rats fed with Kochujang extract (K-group), more prominent differential findings were found, the value of HDL-cholesterol, which was good for preventing the arteriosclerosis, was more or less higher than that of H-groups. But the level of LDL-cholesterol, which was one of the hazardous agents of hyperlipidemia and arteriosclerosis, was more significantly reduced to the level even lower than that of N-group, based on the statistical analysis, in especial. According to the present results, we could find that the unidentified Korean traditional Kochujang-derived complex components had expressed both the positive health improving effects and hypolipidemic effects on the SD-rat.

The purpose of this study was to compare knowledge on hypertension and blood parameters through blood pressure control of hypertensive patients commuting to a public health center. Hypertensive subjects were classified according to their blood pressures into a well-controlled group (SBP<140 mmHg and DBP<90 mmHg, n=44) and an uncontrolled group ($SBP\geq140mmHg\;or\;DBP\geq90mmHg$, n=41). All subjects were interviewed for clinical characteristics, effort for health maintenance and knowledge on hypertension and nutrition. Biochemical measurement of blood urea nitrogen, creatinine, triglyceride, total cholesterol, HDL-cholesterol and fasting blood glucose (FBG) were obtained from subjects. The knowledge about hypertension and effort for health maintenance were not significantly different between the groups. However, the nutrition knowledge score of the well-controlled group was significantly higher (p<0.05) than that of the uncontrolled group. On the test of nutrition knowledge, 97.7% of the well-controlled group answered correctly to the item, 'Medication is an easy way to control blood pressure'. This result was significantly higher (p<0.05) than that of the uncontrolled group of whom 82.9% answered correctly. Participants in both groups had only a little understanding about the importance of a balanced diet. Blood parameters were in the normal range and there was not a significant difference between the groups. However total cholesterol, triglyceride and fasting blood glucose levels of the uncontrolled group were higher than those of the well-controlled group. Therefore, education programs about the continuous control of hypertension through careful monitoring, medication, and exercise are required. The uncontrolled group was especially in need of strengthening education. Also more effective nutrition education programs about low salt, high fiber, low cholesterol, and balanced diets are required for hypertensive patients commuting to public health center.

Obesity and being overweight are strongly associated with the development of metabolic disease such as diabetes, hypertension, dyslipidemia. High-fat diet (HFD) is one of the most important factors which cause obesity. In this study, C57BL/6 mice were treated with a HFD for 22 weeks in order to induce obesity and hyperglycemia. Twenty-two weeks later, body weight and plasma glucose level of the HFD group were significantly increased, compared with the normal diet (ND) group. Intra-peritoneal glucose tolerance test (IPGTT) showed glucose intolerance in the HFD group compared with the ND group. These results confirmed that a HFD induced obesity and hyperglycemia in C57BL/6 mice. Plasma levels of triglyceride (TG) and total cholesterol (TC) were increased in the HFD group compared with the ND group. Hepatic levels of TG and TC were also increased by a HFD. To investigate the alteration of lipid metabolism in liver, proteins which are related to lipid metabolism were observed. Among lipid synthesis related enzymes, fatty acid synthase (FAS) and glycerol phosphate acyl transferase (GPAT) were significantly increased in the HFD group. Apolipoprotein B (apoB) and microsomal triglyceride transport protein (MTP), which are related to lipid transport, were significantly increased in the HFD group. Interestingly, protein level and phosphorylation of AMP-activated protein kinase (AMPK), which is known as a metabolic regulator, were significantly increased in the HFD group compared with the ND group. In the present study we suggest that HFD may physiologically increase the proteins which are related with lipid synthesis and lipid transport, but that HFD may paradoxically induce the activation of AMPK.

This study was aimed to evaluate the effects garlic such as fresh garlic powder (FGP), red garlic powder (RGP) and black garlic powder (BGP) by lyophilized, on serum lipid metabolism in obese rats induced high fat diet. Food efficiency ratio (FER) of the experimental groups was lower than the control group and it was significant difference. Total lipid content of serum decreased from 7.0 to 20.9% compared to the control group. Especially, triglyceride content decreased from 40.8% (BGP) to 42.1% (RGP) group as compared to the control group. There was no significant difference in HDL-C content between groups fed garlic powder and normal group. LDL-C contents of the experimental groups were lower than the control group, but has not showed significant diggerence compared to control group. Total lipid content was significantly increased in liver compared to the control group, but decreased over 30% in RGP and BGP groups to the control group. The lipid level in feces was increased by feeding periods of garlic powder, while total cholesterol and triglyceride were significantly increased in feces of RGP group. TBARS content in serum and liver of RGP and BGP groups was significantly decreased than the control group. Antioxidant activity of serum was 68.05% in the RGP group, which was significantly higher than the control group. From the above result, we suggested that red and black garlic powder were effective in the improvement of lipid level in obese rats induced high fat diet.
