Effects of Temperature and Plant Growth Regulators on Bud Sprouting of Stem Cutting of Hibiscus syriacus 'Honghwarang' (온도(溫度)와 생장조절물질(生長調節物質)이 무궁화(Hibiscus syriacus ''Honghwarang') 삽수의 맹아(萌芽)에 미치는 영향)

  • Various temperatures and plant growth regulators were assessed to investigate the nature of the dormancy and bud sprouting of stem cutting of Hibiscus syriacus 'Honghwarang'. The bud sprouting was promoted as temperature increased. Under $30^{\circ}C$ condition, stem cuttings began sprouting at 4days after cutting and showed 100% sprouting at 6 days after cutting. With GA or BA treatment, the cuttings started sprouting at 6 days after cuttings under $25^{\circ}C$ condition and reached 65% sprouting, which was not different from the results without hormone treatments. However, ABA dipping under below $25^{\circ}C$ condition resulted in low sprouting, whereas under over $25^{\circ}C$ condition they started sprouting at 6 days after cutting and reached 93% at 12 days after cutting, which indicated that ABA is not effective on inhibition of sprouting under high temperature condition.

  • To investigate the effects of topping method on the yield and major agronomic traits of three varieties differed in plant type of Hibiscus manihot the experiments were carried out Mokpo Branch Station of Crop Experiment Station, in 1993. Fresh weight of root, dry weight of culm, dry weight of capsule and dry matter ratio on root were highest in the long culmed variety, and at the plots with topping of growing point in main culm and branches(Treatment No.3). Considering from the above results, optimum topping method seemed to be topping of growing point in main culm and branches in the long culmed variety.

Effect of Feeding Chitosan, Hibiscus Extract and L-Carnitine Mixture on Body Weight and Lipid Metabolism in Rats (키토산, 히비스커스 추출물 및 L-카르니틴 함유 식이가 흰쥐의 체중과 지질대사에 미치는 영향)

  • This study was peformed to investigate effect of feeding experimental mixture containing chitosan, hibiscus extract and L-carnitine on body weight and lipid metabolism in rats. Forty-eight male rats(Charles River CD) of eight weeks old and weighing $336.5{\pm}2.3g$ were raised for five weeks with high fat diet(40% fat as calorie) to induce obesity. After induction of obesity, rats weighing $560.4{\pm}5.6g$ were blocked into four groups according to body weight and raised for eight weeks with diet containing either 0.09%(+1D group), 0.9%(+10D group) or 4.5%(+50D group) of experimental mixture. Aspartate aminotransferase(AST) and alanine aminotransferase(ALT), total protein and albumin were normal levels in plasma. Body weight gain and epididymal fat pad weight were lower in experimental mixture groups than control group However, weights of perirenal fat pad and brown adipose tissue were not significantly different among all groups. There was no significant difference in plasma and hepatic lipid levels among all groups. Liver citrate lyase and camitine acyltransferase activities were not significantly different among all groups, however, citrate lyase activity was tended to be decreased with increasing experimental mixture level in diet. Fecal total lipid and total cholesterol excretions were highest in +50D group, and triglyceride excretion was highest in +1D group. in conclusion, intake of experimental mixture containing chitosan, hibiscus extract and L-camitine was effective in reducing body weight and body fat, and its inhibitory effects might lead to obesity improvement.

The Effect of Plant Growth Regulators on Callus Induction and Shoot Regeneration from the Dwarf Type Variety, Hibiscus syriacus L.var. Ggoma (왜성 변이 품종 '꼬마' 무궁화의 캘러스 유도 및 지상부 형성에 식물생장조절물질이 미치는 영향)

  • 'Ggoma' is a new Hibiscus dwarf type variety developed by gamma irradiation at the Korea Atomic Energy Research Institute (KAERI). This study was conducted to determine the best optimum cultural callus formation and shoot regeneration condition. Sterilized leaf tissues were cultured on MS (Marashige and skoog's) medium containing 3% sucrose, 0.8% agar with different concentration and combination of TDZ, 2, 4-D, KT, BA, and 2iP for 4 weeks in vitro culture. MS medium containing 2,4-D $0.1mg\;l^{-1}$ and BA $0.5mg\;l^{-1}$ were most effective on callus formation and growth. After 4 weeks, callus was transferred on BA (0.5, 1, $1.5mg\;l^{-1}$) and TDZ (0.1, 0.2, $0.3mg\;l^{-1}$) for shoot formation. The best condition for inducing the shoot from callus was BA $1.5mg\;l^{-1}$ and TDZ $0.3mg\;l^{-1}$. This result will be useful for the rapid multiplication of Hibiscus syriacus L.var. Ggoma.

  • This research investigates the effect of the Hibiscus syriacus(HSS) on Alzheimer's disease. The effects of the HSS extract on the behavior in the Morris water maze experiment; the expression of IL-1β, TNF-α, IL-1β mRNA, TNF-α mRNA, CD68/GFAP and RDS; the the infarction area of the hippocampus, and brain tissue injury in Alzheimer's diseased mice induced with M were investigated. The HSS extract group showed a significant inhibitory effect on the memory deficit on the mice with Alzheimer's disease induced by βA in the Morris water maze experiment. The HSS extract group suppressed the over-expression of IL-1β, TNF-α, IL-1β and TNF-α mRNA, CD68/GFAP, RDS in the mice with Alzheimer's disease induced by βA. The HSS extract reduced the infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer's disease induced by βA. This study suggest that HSS may be effective for the prevention and treatment of Alzheimer's disease

Occurrence of Root Rot and Vascular Wilt Diseases in Roselle (Hibiscus sabdariffa L.) in Upper Egypt

  • Roselle (Hibiscus sabdariffa L.) family Malvaceae is an important crop used in food, cosmetics and pharmaceutics industries. Roselle is cultivated mainly in Upper Egypt (Qena and Aswan governorates) producing 94% of total production. Root rot disease of roselle is one of the most important diseases that attack both seedlings and adult plants causing serious losses in crop productivity and quality. The main objective of the present study is to identify and characterize pathogens associated with root rot and wilt symptoms of roselle in Qena, Upper Egypt and evaluate their pathogenicity under greenhouse and field condition. Fusarium oxysporum, Macrophomina phaseolina, Fusarium solani, Fusarium equiseti and Fusarium semitectum were isolated from the natural root rot diseases in roselle. All isolated fungi were morphologically characterized and varied in their pathogenic potentialities. They could attack roselle plants causing damping-off and root rot/wilt diseases in different pathogenicity tests. The highest pathogenicity was caused by F. oxysporum and M. phaseolina followed by F. solani. The least pathogenic fungi were F. equiseti followed by F. semitectum. It obviously noted that Baladi roselle cultivar was more susceptible to infection with all tested fungi than Sobhia 17 under greenhouse and field conditions. This is the first report of fungal pathogens causing root rot and vascular wilt in roselle in Upper Egypt.

  • PCR-mediated recombination describes the process of in vitro chimera formation from related template sequences present in a single PCR amplification. The high levels of genetic redundancy in eukaryotic genomes should make recombination artifacts occur readily. However, few evolutionary biologists adequately consider this phenomenon when studying gene lineages. The cytosolic glyceraldehyde-3-phosphate dehydrogenase gene (GapC), which encodes a NADP-dependent nonphosphorylating glyceraldehyde-3-phosphate dehydrogenase in the cytosol, is a classical lowcopy nuclear gene marker and is commonly used in molecular evolutionary studies. Here, we report on the occurrence of PCR-mediated recombination in the GapC gene family of Hibiscus tiliaceus. The study suggests that recombinant areas appear to be correlated with DNA template secondary structures. Our observations highlight that recombination artifacts should be considered when studying specific and allelic phylogenies. The authors suggest that nested PCR be used to suppress PCRmediated recombination.

Antimicrobial and Antioxidative Effects of Roselle (Hibiscus sabdariffa L.) Flower Extract and Its Fractions on Skin Microorganisms and Oxidation

  • Antimicrobial and antioxidative effects of 14 different herbal flower extracts on skin microorganisms and oxidation were tested in this research. Herbal flower extracts were prepared with 70% ethanol. Among the herbal flower extracts, roselle (Hibiscus sabdariffa L.) flower extract showed the highest antimicrobial activity against Staphylococcus epidermidis as determined by a paper disc method. The seventy % ethanol extract of roselle flower was fractionated by sequential hexane, chloroform, ethyl acetate, n-butanol, and water fractionation. The growth of S. epidermidis, Streptomyces collinus, Streptomyces coeruleoprunus, Salmonella enteritidis, Vibrio parahaemolyticus, and Malassezia pachydermatis was most efficiently inhibited by ethyl acetate fraction of roselle flower extract as determined by a paper disc method and growth inhibition curves. In addition, the ethyl acetate fraction, water fraction and butanol fraction showed free radical scavenging and DNA cleavage inhibition activities. These results demonstrate that roselle flowers hold antimicrobial and antioxidative activities against skin microorganisms and oxidants.

Study on the Anti-inflammatory Effect of Kaempferol and Kaempferol Rhamnosides Isolated from Hibiscus cannabinus L. (양마에서 분리한 Kaempferol 및 그 배당체의 항염증 효과에 관한 연구)

  • In this study, to evaluate the anti-inflammatory effects of kaempferol and its rhamnosides isolated from Hibiscus cannabinus L. leaves, we investigated UVB-induced inhibitory effects on inflammatory reaction by measuring the cytokine as the prostaglandin ($PGE_2$), interleukine-6 (IL-6) and interleukine-8 (IL-8). We investigated the inhibitory effects of kaempferol and its rhamnosides on TARC (thymus and activation-regulated chemokine) and $PGE_2$. Kaempferol and ${\alpha}$-rhamnoisorobin showed inhibition activity of TARC generated to compared to positive control. Kaempferol, ${\alpha}$-rhamnoisorobin and afzelin Inhibited the release of $PGE_2$. Also, only kaempferol significantly inhibited interleukine-6 (IL-6), interleukine-8 (IL-8) among UVB-induced inflammatory cytokine.

Hydroxyhibiscone A, a Novel Human Neutrophil Elastase Inhibitor from Hibiscus syriacus

  • In an ongoing investigation of compounds from natural products that exhibit anti-aging properties, hydroxyhibiscone A (1), a new furanosesquiterpenoid, together with hibiscone D (2), was isolated from the root bark of Hibiscus syriacus. Utilizing UV, IR, NMR, and MS spectroscopic analyses, these chemical structures were revealed. Compounds 1 and 2 were found to posses significant anti-aging properties on the human neutrophil elastase (HNE) assay, exhibiting HNE inhibitory activities with $IC_{50}$ values of 5.2 and 4.6 ${\mu}M$, respectively.
