  • This study was carried out to establish an improved bioassay system, whole-plant bioassay which is more effective in developing natural herbicides for foliar treatment such as herbicidal essential oils. Two bioassay systems using four weed species (Echinochloa crus-galli, Digitaria sanguinalis, Aeschynomene indica, and Abutilon theophrasti), spraying method and spotting method, were established. Spraying method is applicable if the amount of test compounds is enough, while spotting method is useful for the small amount of test compounds. The initial application rate was desirable at $2,500{\sim}5,000\;{\mu}g\;mL^{-1}$. Herbicidal activities were higher in the NOP treatment when compared to the Tween 20 treatment. To efficiently evaluate volatile compounds such as essential oils, if the compound-treated pots were incubated in dew chamber for about 10hrs, better results were obtained in the degree and stability of herbicidal responses. When the efficiency of bioassay systems established in this study was compared, the spraying method was minimized four times to the conventional method that has beed used for screening of synthetic compounds in KRICT. On the other hand, in the spotting method, screening for development of a natural herbicides was possible even in level of 1/100 test volume and 1/200 amounts of test compound compared to the spraying method.

  • Differential tolerance to protoporphyrinogen oxidase (Protox)-inhibiting herbicides, oxyfluorfen was observed between leaf ages in squash. Physiological responses to oxyfluorfen, including leaf injury, cellular leakage, accumulation of tetrapyrroles, and antioxidative enzymes activity, were investigated in leaf age classes of squash to identify mechanisms of oxyfluorfen tolerance. Leaf 1, 2, and 3 injuries for Joongangaehobak were >10,000, 1,286, and 1.6-fold higher than that of leaf 4, after treatment of oxyfluorfen. On the other hand, leaf 1, 2, and 3 injuries for Sintowjahobak were 725, 366, and >0.6-fold higher than that of leaf 4, after treatment of oxyfluorfen. However, in contrast to oxyfluorfen treatment results, leaf injury of squash leaf 4 treated with paraquat was much smaller than in leaves 1, 2 and 3. Electrolyte leakage from the tissues treated with oxyfluorfen was higher in the youngest leaf (Leaf 4) than in the older leaves 1, 2, and 3. Differential leaf response to oxyfluorfen of squash appears to be due in large part to differences in protoporphyrin IX (Proto IX), Mg-Proto IX, and Mg-Proto IX monomethyl ester accumulation in treated leaves. In contrast, leaf 4 had higher activities of superoxide dismutase, catalase, peroxidase, ascorbate peroxidase, and glutathione reductase than leaf 1 after treatment with oxyfluorfen. However, the induction in antioxidant activity in leaf 4 was not enough to overcome the toxic effects of a Protox inhibitor, oxyfluorfen, so the leaf eventually died.

  • For the development of integrated pest management system by harmonizing biological and chemical control, some experiments wee carried out to select low toxic pesticides against natural enemies. and their residual toxicity were evaluated. Leaf dipping method, body dipping method, and diet treatment method were set up for the toxicity evaluation against Cotesia glomerata adults. We had tested 46 different pesticides (31 insecticides, 11 fungicides, 4 herbicides) at recommending concentration commonly used to control diamond back moth, disease and up-land weeds in chinese cabbage field. Twenty three insecticides, eleven fungicides, and four herbicides were shown to be low toxic to C. glomerata adults in the treatment of body dipping. After insecticide spraying at recommending dose on the chinese cabbage, we examined residual effect of insecticides by introducing natural enemies on different days. Safety interval for the introduction of C. glomerata adults was established according to the residual toxicity of pesticides. Safe insecticides for the introduction of C. glomerata adults at one day after treatment (DAT) were thiacloprid, acephate, chlorfenapyr, clothianidin and at 3 DAT were imidacloprid, deltamethrin, thiamethoxam, dimethylvinphos, emamectin benzoate.

  • Oryzalin is a dinitroaniline herbicide, which disrupts the arrangement of microtubules. Microtubules and microfilaments are cytoskeletal components that are thought to play a role in the sedimentation of statoliths and the formation of cell walls. Statoliths regulate the perception of gravity by columella cells in the root tip. To determine the effect of oryzalin on the gravitropic response, ethylene production in primary roots of maize was investigated. Treatment with 10-4 M oryzalin to the root tip inhibited the growth and gravitropic response of the roots. However, the treatment had no effect on the elongation zone of the roots. An application of 10-4 M oryzalin for 15 hr to the root tip caused root tip swelling. The application of 1-aminocycopropane-1-carboxylic acid (ACC), a precursor of ethylene, to the root tip also inhibited the gravitropic response. To understand the role of oryzalin in the regulation of the growth and gravitropic response of roots, ethylene production in the primary roots of maize was measured following treatment with oryzalin. Oryzalin stimulated ethylene production via the activation of ACC oxidase (ACO) and ACC synthase (ACS), and it increased the expression of ACO and ACS genes. Indole-3-acetic acid (IAA) played a key role in the asymmetric elongation rates observed during gravitropism. The results suggest that oryzalin alters the gravitropic response of maize roots through modification of the arrangement of microtubules. This might reduce the distribution of IAA in the upper and lower sides of the elongation zone and increase ethylene production, thereby inhibiting growth and gravitropic responses.

  • The tolerant and susceptible rice cultivars to oxyfluorfen were selected from 400 rice cultivars in a laboratory, and tested in comparison with barnyardgrass, a typical oxyfluorfen susceptible weed. The responses to oxyfluorfen in the different levels of calli, cells and protoplasts of the rice cultivars were investigated. $I_{50}$ value of the tolerant rice cultivars was about $10^{-4}M$, whereas that of the susceptible rice cultivars and barnyardgrass was about $10^{-6}M$, showing significant difference between the two groups. The growth rate of calli segregated from the tolerant rice cultivars was higher than that from the susceptible rice cultivars and barnyardgrass by treatment of oxyfluorfen to the calli. The growth rate of suspension-cultured cells of the tolerant rice cultivars was higher than that of the susceptible rice cultivars and barnyardgrass after treatment of oxyfluorfen. The viability of protoplasts from the tolerant rice cultivars was higher than that from the susceptible rice cultivars and barnyardgrass after one or two hours of oxyfluorfen treatment. The intactness of protoplasts from the tolerant rice cultivars was also higher than that form the susceptible rice cultivars and barnyardgrass.

  • Recently, the domestic plant disease control policy for fire blight has been implemented partial control in addition to burial control. In this study, an on-site management survey was conducted targeting orchards that implemented partial disease control from 2019 to 2020 in order to find efficient implementation methods for partial disease control. As a result of an investigation into 22 pear and apple orchards in Cheonan and Chungju, 7 orchards were buried. The upper part of the cut infected plants was burned at 16 orchards and covered with plastic vinyl after lime treatment at 6 orchards. The lower stumps of cut infected plants were burned at 7 orchards and covered with plastic vinyl after lime treatment at 15 orchards. There were two orchards where suckers appeared on the stumps even though covers were applied. There was no infection by Erwinia amylovora in the suckers. The conservation condition of lime treatment was good, but warning signs were absent at 6 orchards. Most orchards treated the stumps and surrounding areas with glyphosate-isopropylamine herbicide. The effect of partial control was judged to be safe.

  • The study was conducted during 1975-1980 to evaluate the performance of several herbicides in controling alternate host, Pedicularis spp., of white pine blister rust disease in forest stands. The results obtained were summarized as follows. 1. It was found that Roundup was effective and most suitable herbicide in the fosest stands except that it accompanied some chemical injuries to neighboring plants. 2. As for the reduction density on the plot treated with Roundup was 100% and 65-91% respectively when investigated one and two years after its treatment on Pedicularis spp. 3. Another four chemicals, 2.4-D, Gramoxone, 2.4. 5-T and Para-col killed stems and leaves of Pedicularis spp. at the year treated, but appeared again next spring. 4. Eradication effectiveness in terms of spraying season was proved to be that late August was better than late May or June.

  • This study was conducted to investigate the herbicidal activity against Scirpus planiculmis (SCPPL) which has been reported as resistant biotype on sulfonylurea (SU) herbicides at the west coast reclaimed area first and proliferated continuously at Hwasung and Pyeongtaek in Gyeonggi province, and some Chungnam province. In order to investigate resistance breakability against SCPPL, new rice herbicides containing tefuryltrione [p-hydrophenyl pyruvate dioxygenase (HPPD) inhibitor], pyrimisulfan and triafamone[actolactate synthethase (ALS) inhibitor] were tested. In both lab and field experiments, tefuryltrione GR, 4-HPPD inhibitor showed growth retardation with a bleaching on the young leaves of SCPPL at early time after treatment. However, it restored and regerminated with insufficient control effect. On the other hand, pyrimisulfan+mefenacet SC, triafamone+tefuryltrione GR, and ALS inhibitor showed excellent control effect on SU-resistant SCPPL with growth retardation and necrosis. In particular, pyrimisulfan+mefenacet SC showed excellent controlling effect on SU-resistant SCPPL in regional field experiments.

  • In order to establish labor-saving culture technology in Tartary buckwheat, three cultural practices: hand planting, drill sowing machine and soil cover direct seeding machine, were compared. The highest grain yield was found in soil cover direct seeding plot with a value of 3.4 g per plant. As a result, grain yield may be estimated to be 113kg in soil cover direct seeding and 80kg in hand scattering. In addition, for the weed control, three herbicide treatments: single use of Alachlor, mixture and combination of Alachlor and Paraquat dichloride were conducted. the mixture showed over 90% weed control value, and the highest grain yield was found in the combination treatment. Combine machine was effective to reduce the ratio of grain loss and working hour by enhancing the working efficiency to 15${\sim}$20min/10a.

  • This study was carried out to investigate the toxic action of herbicide, paraquat(1,1'-dimethyl-4,4'-bipyridylium-dichloride), against microorganisms. The toxic effect of paraquat was observed mainly using Escherichia coli(KCTC 1039), as follows; Growth of aerobic microorganisms which comprise 4 strains of bacteria and 2 strains of yeast and 4 strains of mold was inhibited drastically in the presence of 1.0mM paraquat But the growth of anaerobic bacteria was not affected by the chemical. When actively growing cells of E. coli were exposed to the paraquat at the concentration higher than 1.0 mM, they rapidly lost their ability to form colony and clearly formed inhibitory zone by well test More than 50% of the cells were killed by 1.0 mM paraquat treatment, even at immediate addition of paraquat to the medium.
