A Case Report of Three Patients with Menopausal Disorder Managed with Jaeumganghwa-tang (Herbal Extract in National Health Insurance): Focusing on Kupperman's Index, Menopause Rating Scale (자음강화탕(보험제제)으로 호전된 갱년기 장애 3례에 대한 증례 보고 : Kupperman's Index, Menopause Rating Scale을 중심으로)

  • Objectives: This case report describes the management of three patients with menopausal disorder treated with Jaeumganghwa-tang, an herbal extract in national health insurance. Methods: Three patients suffering from Menopause disorder were hospitalized and treated with herbal extract Jaeumganghwa-tang for 4 weeks. This study was measured by Kupperman's Index, Menopause Rating Scale and EuroQol-Five Dimensions. Results: The symptoms of menopausal disorder have improved after treating with Jaeumganghwa-tang. Also the Kupperman's Index and Menopause Rating Scale have decreased, while the EuroQol-Five Dimensions has increased. Conclusions: As seen in these three cases, Jaeumganghwa-tang in herbal extract form has effects on the management of menopause disorder.

Effects of the Ethanol-extract of Allium wageki on the Fructose-induced Hypertensive Rats (쪽파 에탄올 추출물이 과당-유도 고혈압 흰쥐에 미치는 영향)

  • The effects of the ethanol (EtOH)-extract of Allium wageki were studied on the changes of body weight (BW), systolic blood pressure (SBP), triacylglycerol (TG), cholesterol, and insulin levels in Sprague-Dawley rats in which high fructose feeding elicited hyperinsulinemia, hypertriglyceridemia, and hypertension. SBP and BW were measured for 7 weeks on the fructose diet, fructose/EtOH-extract, vehicle- treated, or vehicle/EtOH-treated rats. The EtOH-extract of Allium wageki blocked the increases of SBP ($119.4{\pm}3.7\;mmHg$ in fructose/EtOH-extract of Allium wageki; $141{\pm}4.3 \;mmHg$ in fructose diet, p<0.01) and BW ($368{\pm}11.4\;g$ in fructose/EtOH-extract of Allium wageki; $402{\pm}15.7\;g$ in fructose diet, p<0.05) by high fructose-diet. The increases of TG, total cholesterol, and insulin levels were also blocked by administration of EtOH-extract of Allium wageki in fructose-induced hypertensive rats. These results suggest that EtOH-extract of Allium wageki has protect effects on the increases of TG, total cholesterol, insulin level, and blood pressure in fructose-induced hypertensive rats.

Effects of Rhus verniciflua Stokes (RVS) Extract on Diet-induced Obesity in C57BL/6 mouse (옻나무 추출물이 식이에 의한 비만유도 C57BL/6 mouse에 미치는 영향)

  • We investigated the effects of Rhus verniciflua Stokes (RVS) on high fat diet- induced obesity in C57BL/6 that received ad libitum exposure to a high fat diet for 8 weeks. Male C57BL/6 mice weighing $20.1{\pm}0.95\;g$ were divided into four groups and the body weight and food intake were weighed for 8 weeks. After the administration of RVS extract, the liver, subcutaneous and retroperitoneal adipose tissue in the mice were weighed and common blood count in whole blood and biochemical parameters in plasma were measured. It was founded that body weight increase was significantly suppressed by the oral administration of RVS extract in group with high fat diet. Weight of subcutaneous adipose tissue and concentration of triglyceride in plasma from mice which were administrated RVS extract were lower than obesity control group. These results suggest that RVS extract may effect on inhibition of diet-induced obesity.

Quantitative Analysis of Marker Substances of Paeonia lactiflora by Solid Fermentation (작약의 고체발효에 따른 지표성분의 함량분석)

  • The purpose of this study was investigation of quantitative analysis of marker substances in Paeonia lactiflora extracts by solid fermentation. High performance liquid chromatography (HPLC) for the determination of albiflorin and paeoniflorin in P. lactiflora extracts by solid fermentation, the separation method was performed on C18 column ($250\;mm\;{\times}\;4.6\;mm$, $5\;{\mu}m$, RS tech) using gradient solvent mixtures of water-acetonitrile with photodiode array detector (230nm). The flow rate was 1.0 ml/min. Retention time of albiflorin and paeoniflorin was about 28.88, 31.92 min and linearity of calibration was showed good result(r2 = 0.9998, 0.9996), respectively. Content of albiflorin was $0.090\;{\pm}\;0.03%$ in P. lactiflora extract(control), $0.102\;{\pm}\;0.00%$ in P. lactiflora extract fermented with Paecilomyces japonica, $0.056\;{\pm}\;0.01%$ in P. lactiflora extract fermented with Ganoderma lucidum, $0.093\;{\pm}\;0.00%$ in P. lactiflora extract fermented with honey and $0.046\;{\pm}\;0.00%$ in P. lactiflora extract fermented with Nuruk. Content of paeoniflorin was $4.506\;{\pm}\;0.13%$ in control, $2.599\;{\pm}\;0.04%$ in P. lactiflora extract fermented with Paecilomyces japonica, $1.222\;{\pm}\;0.03%$ in P. lactiflora extract fermented with Ganoderma lucidum, $2.750\;{\pm}\;0.05%$ in P. lactiflora extract fermented with honey and $0.847\;{\pm}\;0.00%$ in P. lactiflora extract fermented with Nuruk, respectively. Content of the marker substances did not increase in all fermentation experiment group.

A Study on Quality Evaluation of Ojeok-san Extract Powders Distributed in Korea (오적산 보험 엑스산제의 품질평가 연구)

  • Objectives : This study was performed to analyze the quality of Ojeok-san extract powders distributed in Korea. Methods : HPLC analyses of Ojeok-san decoction produced in laboratory and 8 kinds of Ojeok-san extract powders were done. Also, quantity analyses of paeoniflorin in Paeonia lactiflora, Glycyrrhizin in Glycyrrhiza uralensis, alkaloid in Ephedra sinica, Hesperidin in Citrus unshiu Markovich, and cinnamic acid in Cinnamomum cassia Blume were performed to investigate the accurate content of 6 kinds of Ojeok-san extract powders. Results : In the pattern analysis, the peaks were compared. The content of components in herbal extract powders was 14.7 to 15.8% of that of decoction in average. The range was measured from 8.5 to 25.7%. The pattern comparison showed significant differences among the pharmaceutical companies' products. As for the quantity analysis, one of herbal extract powder contained insufficient amount of Paeonia lactiflora components and one of herbal extract powder was in short of Ephedra sinica components. All of the others contained more than standard component(over 90%). Conclusions : Because herbal extract powders are also medicine, standardized manufacturing methods and consistent quality management are necessary. Therefore exaction and operation of national standards and various researches to improve the quality management of herbal extract powders is urgent.

Inhibitory Effects of the Seeds of Cornus officinalis on AGEs Formation and AGEs-induced Protein Cross-linking (산수유 씨의 최종당화산물의 형성 및 교차결합에 미치는 효과)

  • An 80% EtOH extract and the solvent fractions of the seeds of Cornus officinalis were evaluated for their inhibitory activities against advanced glycation end products (AGEs) formation and AGEs-induced protein cross-linking in vitro. In vitro assay for AGEs-bovine serum albumin (BSA) formation showed that the 80% EtOH extract, n-hexane, EtOAc, n-BuOH and water fractions significantly inhibited AGEs formation with observed $IC_{50}$ values of 1.13, 17.64, 1.52, 1.24 and $3.27{\mu}g/ml$, respectively. In indirect AGEs-ELISA assay, the 800% EtOH extract, EtOAc and n-BuOH fractions exhibited more potent inhibitory activity on AGEs-BSA formation than aminoguanidine, a well know AGEs inhibitor. Furthermore, the 80% EtOH extract and all the solvent fractions inhibited concentration-dependently AGE-BSA cross-linking to collagen. The 80% EtOH extract, EtOAc, n-BuOH and water fractions also had a breaking activity against preformed AGE-BSA cross-linking concentration dependently. Thus these results suggest that the 80% EtOH extract and fractions of the seeds of C. officinalis could be an inhibitor as well as breaker of AGE-BSA cross-linking.

  • In previous studies we reported that the levels of RBC, hemoglobulin and hematocrit in SAM Rl and SAM P6 were increased significantly from 7 day after oral administration of the pilose antler extract, 5g/kg/day, and were lasted during the study. Therefore, this study was performed to elucidate mechanism of erythropoietic action by the extract administration. SAM Rl and SAM P6 were chosen as experimental animals. At age of 12 weeks, pilose antler extract were given 0.3 and 5 g/kg/day (p.o.) each for 0, 7 and 14 days in both animals. Complete blood cells (CBC) such as WBC, lymphocytes, monocytes, granulocytes, RBC, hemoglobulin, and hematocrit were counted. And plasma concentration of erythropoietin (EPO) which is the major regulator of erythropoiesis was measured using $\^$125/I-antierythropoietin IgG. Ferritin concentration in plasma was also analyzed.

Inhibitory Effect of Hirudo on Neuroinflammation in LPS-stimulated Microglial Cells (Lipopolysccharides에 의해 활성화된 미세아교세포에서 수질(水蛭) 추출물의 NF-kB 억제를 통한 뇌신경염증 억제 효과)

  • Today, environmental pollution has been found to be one of the causes of various diseases, including brain and nervous system diseases. In particular, neurodegenerative diseases have been found to be caused by hyperactivation of immune system cells such as microglia. Preventive and therapeutic measures are needed to suppress them. Hirudo is known as a traditional herbal medicine, based on its multiple biological activities such as anti-eczema and anti-coagulation. In the present study, the anti-neuroinflammatory potential of hirudo extract was investigated in lipopolysccharide (LPS)-stimulated BV2 microglial cells and in mice. Hirudo extract significantly inhibited LPS-stimulated nitric oxide (NO) production and cytokine (IL-1Ra, KC, MCP-5, and RANTES) expression in a dose-dependent manner without causing cytotoxicity. Pretreatment with hirudo extract suppressed LPS-induced NF-κB p65 nuclear translocation. Moreover, hirudo extract reduced LPS-stimulated microglial acitivation and improved memory impairments. The results demonstrated that hirudo extract exerts anti-neuroinflammation activities, partly through inhibition of the NF-κB signaling pathway. These findings suggest that hirudo extract might have therapeutic potential with respect to neuroinflammation and neurodegenerative diseases.

Comparative Study on Biological Activities and Ingredient Contents of Different Solvent Extracts of Samchulkunbi-tang (삼출건비탕의 추출용매에 따른 성분 함량 및 효능 비교 연구)

  • In order to investigate anti-inflammatory, anti-allergic and anti-obesity activities of Samchulkunbi-tang (SCT; Shen zhu jian pi-tang) water and 70% ethanol (EtOH) extracts, in vitro inhibitory activities against nitric oxide (NO), prostaglandin $E_2$ $PGE_2$), interleukin (IL)-6 and tumor necrosis factor (TNF)-${\alpha}$ production in lipopolysaccharide-stimulated RAW 264.7 cells, and macrophage-derived chemokine (MDC/CCL22) and regulated on activation of normal T-cell-expressed and -secreted (RANTES/CCL5) production in TNF-${\alpha}$/interferon-${\gamma}$-stimulated HaCaT and BEAS-2B cells as well as glycerol-3-phosphate dehydrogenase (GPDH) activity and leptin production in 3T3-L1 cells were determined. A HPLC was used for quantification of the seven marker components (albiflorin, paeoniflorin, liquiritin, naringin, hesperidin, poncirin and glycyrrhizin) of SCT water and 70% EtOH extracts. SCT showed inhibitory effects against MDC and RANTES production in HaCaT cells, as well as RANTES production in BEAS-2B cells. In addition, SCT reduced not only NO, $PGE_2$, IL-6 and TNF-${\alpha}$ production in RAW 264.7 cells, but also GPDH activity and leptin production in 3T3-L1 cells. Furthermore, the biological activities and the contents of six compounds (except paeoniflorin) were higher in 70% EtOH extract than water extract. These results suggest that SCT has anti-inflammatory, anti-allergic and anti-obesity activities. These efficacies of 70% EtOH extract are relatively higher than that of water extract.

Neuroprotective effects of some herbal medicine plant extract against ischemia·reperfusion-induced cell death in SK-N-SH neuronal cells (허혈·재관류 유도성 신경세포사멸에 대하여 신경보호효과를 가지는 약용식물 추출물의 검색)

  • Objectives : The purpose of the study is to determine the neuroprotective effects of the water and 80% EtOH extract of some herbal medicine plant on ischemia reperfusion-induced cell death in SK-N-SH human brain neuronal cells. Methods : SK-N-SH cells were treated with 3mM sodium azide and 10 mM 2-deoxy-D-glucose for 45 min, ptior to the addition of different concentrations of herbal medicine plant extract (0, 10, 25, 50, 100, 250, 500, 1000 ${\mu}g/ml$) for 2 hr and then reperfused with growth medium, incubated for 24 h. Cell viability was determined by WST-1 assay, and ATP/ADP levels were measured by ADP/ATP ratio assay kit. Results : Herbal medicine plant extract significantly inhibited decreasing the cell viability in ischemia-induced SK-N-SH cells. Also increased the ratio of ADP/ATP in ischemia-induced neuronal cells. Conclusions : Our results suggest that herbal medicine plant extract has a neuroprotective property via increasing the energy levels in neuronal cells, suggesting that extract may has a therapeutic potential in the treatment of ischemic brain injury. The exact component and mechanism remains for the future study.
