Suppressive Effects of Fucoxanthin on Degranulation in IgE-antigen complex-stimulated RBL-2H3 Cells

  • The marine carotenoid fucoxanthin can be found in marine brown seaweeds, macroalgae, diatoms, and microalgae, and has remarkable biological properties. Numerous studies have shown that fucoxanthin has considerable potential and promising applications in human health, but the underlying mechanisms involved in its anti-allergic activity are not fully understood. We here investigated the mechanisms by anti-allergic activity of fucoxanthin fraction from Eisenia bicyclis in immunoglobulin E-antigen complex (IgE/DNP-BSA)-stimulated RBL-2H3 mast cells. This study we found that the fucoxanthin inhibits the release of ${\beta}$-hexosaminidase and suppressed not only transcriptional activation of NF-${\kappa}B$, but also phosphorylation of ERK and JNK in IgE/DNP-BSA-treated RBL-2H3 cells. Fucoxanthin may be useful for preventing allergic diseases, including asthma and atopic dermatitis.

A Study of Disassembling Major Indication Terms into Minimum Meaning Units and Linking to Diseases (의미 단위 분해를 통한 주치와 병증 용어 연계 연구)

  • Objectives : Ontology is a good tool to represent the knowledge and has developed for Traditional Korean Medicine(TKM) in Korea Institute of Oriental Medicine. There are a lot of TKM terms, which have a complex meaning, especially major indication terms of medicinal treatment and terms of symptom and disease. These complex meaning terms result in the low linkage between major indication terms of medicinal treatment and terms of symptom and disease in TKM ontology. We studied to enhance the percentage of the linkage among those data in TKM ontology. Methods : We disassembled major indication terms of medicinal treatment into minimum meaning units and then linked them to enhance the percentage of the linkage among medicinal material, formula and disease ontology based on Traditional Korean Medicine. To retain objectivity, several experts of Korean Medicine used a web-based tool that supports users in refining terms and disassembling them into the minimum meaning efficiently. Results : The outcome shows that the percentage of the linkage among medicinal material, formula and disease ontology increased. By linking disassembled major indication terms to symptoms and diseases, the amount of information for medicinal materials and formulas also increased quantitatively in comparison with given formula for diseases in disease ontology. Conclusions : By this study, disassembled data increases the percentage of linkage between diseases and medicinal treatments. And the applicability of TKM ontology is also increased.

A Retrospective Study of Acute Lumbar Sprain Patients on the Effect of Korean Medicine Treatment with Dokhwal-tang (Duhuo-tang) (급성 요추부 염좌로 입원했던 환자군의 독활탕 치료 효과에 대한 후향적 고찰)

  • Objectives This study investigated the effect of korean medicine treatment with Dokhwal-tang (Duhuo-tang) on Lumbal sprain patients by comparing with other herbal medicines. Methods 17 lumbar sprain patients of group treated with Dokhwal-tang (Duhuo-tang) were treated with Dokhwal-tang (Duhuo-tang), acupunture, cupping, oriental physical therapy. 13 lumbar sprain patients of group treated with other herbal medicines were treated with 9 prescriptions of herbal medicine, acupunture, cupping, oriental physical therapy. ODI (Oswestry diability index), VAS (Visual Analogue Scale) were checked to evaluate the effectiveness of the treatment. Results 1) ODI, VAS of patients treated with Dokhwal-tang (Duhuo-tang) were all improved and there were statistical significance (p<0.05). 2) VAS of group treated with Dokhwal-tang (Duhuo-tang) and that of group treated with other herbal medicines were all improved significantly. And treatment period of group treated with Dokhwal-tang (Duhuo-tang) was shorter than that of group treated with other herbal medicines. But there was no statistical significance between VAS of group treated with Dokhwal-tang (Duhuo-tang) and that of group treated with other herbal medicines. 3) In comparing medical history period on group treated with Dokhwal-tang (Duhuo-tang), ODI, VAS of hyper-acute phase group and acute phase group were improved but them of sub-acute phase group were less improved. Conclusions According to the study, korean complex therapy with Dokhwal-tang (Duhuo-tang) might improve acute lumbar sprain.

Effects of the Herbal Medicine in Animal Models for Treatment of Sciatic Nerve Impairment: A Review of Animal Study Reports (좌골신경 손상에 대한 한약물 치료효과에 대한 실험논문 분석 - 국내외 한의계 연구를 중심으로 -)

  • Objectives This study is to review the effect of herbal medicine on treatment of sciatic nerve injury induced animal models reported in domestic & foreign journals. Methods 5 electronic databases (Pubmed, CAJ, RISS, Oasis, Koreantk) were searched with term as sciatic nerve injury, and animal study reports on sciatic nerve impairment with herbal medicine treatment were extracted. Results 31 articles were reviewd. All studies used SD rat. 26 studies used crush injury at the sciatic nerve using haemostatic forcep and 5 studies used sciatic nerve transection. 15 studies used single herb extract and 16 studies used complex herb medicine. Angelica gigantis radix was the most frequently used herb. Each study showed significant changes of improvement indicators from sciatic nerve impairment. Conclusions Various herb medicine are expected to have positive effects on sciatic nerve impairment.

A Case Report on Right Hemiparesis after Left Thalamus Infarction Treated with Boyanghwanoh-tang-gagam and Complex Korean Medicine Treatment (보양환오탕가감방 및 복합한의치료로 호전된 시상 부위 뇌경색 후 우측 편마비 환자 치험 1례)

  • Objectives: This study reports the case of a patient with right hemiparesis after a left thalamic infarction, in which the patient experienced improved clinical symptoms (hemiparesis, constipation) after treatment with a complex Korean medicine treatment. Methods: A 81-year-old male patient diagnosed with acute left thalamic infarction was treated with daily acupuncture and herbal medicine (Boyanghwanoh-tang-gagam) three times a day. Jowiseunggi-tang extract granules were administered to treat constipation by adjusting the dosage according to the reported symptoms. The patient's clinical course was assessed using changes in Modified Barthel Index (MBI) score, Manual Muscle Testing (MMT) grade, and defecation frequency . Results: The patient's MBI score improved from 35 to 69 and was discharged with the ability to walk under supervision. The MMT grade also improved from 3/4 to 5/5 in the upper/lower extremities. Other clinical symptoms, including constipation, also improved, leading to the discontinuation of magnesium medication. Conclusion: These findings suggest that complex Korean medicine treatments, including herbal medicine and acupuncture, may be an effective treatment for post-stroke hemiplegic patients.

  • Purpose : The purpose of this case is to report the improvement after acupuncture & moxibustion complex therapy about Jue zheng (厥證) caused by Shang han(傷寒). Methods : We provided acupuncture & moxibustion complex therapy to a patient who suffered from chills and coldness. We used acupuncture technique and Ge-yan-jin in umbilicus (神厥 隔鹽灸). Results & Conclusions : We observed that acupuncture & moxibustion complex therapy decreased symptoms of Jue zheng (厥證) and improved general condition of a patient who suffered from chills and coldness.

The Clinical Effects of Complex Korean Medicine Treatment in Patients with Cervical Spine Fracture Caused by Traffic Accident: A Report of 2 Cases (교통사고로 발생한 경추 골절 환자에 대한 한방복합치료 효과: 증례보고)

  • The objective of this study is to report the effects of complex Korean Medicine treatment for 2 patients who had cervical spine fracture casued by traffic accident. We used acupuncture, pharmacopuncture, moxibustion and herbal medicine to treat patients with cervical spine fracture. We observed the changes of pain by Numeric Rating Scale(NRS), Neck Disability Index(NDI) and EuroQol-5 Dimension Index(EQ-5D Index). After complex Korean Medicine treatment, we found that neck pain was reduced and quality of life were improved by NRS, NDI and EQ-5D Index. Complex Korean Medicine could be effective for patients with cervical spine fracture caused by traffic accident.

  • The interaction of cadmium (II) ion with protocatechuic acid ($H_3PA$) and ethylenediaminetetraacetic acid (EDTA) were investigated in aqueous solution at different pH. The protocatechuic acid and EDTA/cadmium stochiometries for cadmium (II) binding have been determined by UV-vis spectrophotometric method. The complexation of Cd (II) ion with protocatechuic acid was formed in solution. Among the two potential sites of chelation present in the protocatechuic acid structure, the carboxylic function presents higher complexation power toward Cd (II). 1 : 1 Cd (II)-complex had a maximum absorbance and showed the bathochromic shift of the long-wavelength band of the UV-vis spectra in the alkaline pH when interacted with protocatechuic acid in 0.2 M $NH_3$ - 0.2 M ${NH_4}Cl$ (pH 8.0) buffer. These results suggest that Cd $({H_2}PA)^+$ complex has the optimal condition of chelation in buffer solution at 64.22 ${\mu}M$ protocatechuic acid (A=1.01455).

Compilation of 104 Experimental Theses on the Antitumor and Immuno-activating therapies of Oriental Medicine (한의학의 항종양 면역치료에 관한 연구 -1990년 이후 발표된 실험논문을 중심으로-)

  • This study was done to compile 104 experimental theses which are related to the antitumor and immuno-activating therapies between February 1990 through February 2002. Master's and doctoral theses were dassified by schools, degrees, materials, effects, experimental methods of antitumor and immunoactivity, and results. The following results were obtained from this study : 1. Classifying the theses by the school, 34.6% were presented by Daejeon University, 29.8% by Kyung-hee University and 11.5% by Won-kwang University. Of all theses, 51.0% were aimed for the doctoral degree and 43.3% were for the master's degree. All of three universities have their own cancer centers. 2. Classifying the theses by herb materials, complex prescription accounted for 60.3%, single herb accounted for 24.8% and herbal acupuncture accounted for 14.2%. Considering the key principles of the traditional medicine, complex prescription was much more thoroughly studied than single herb prescription. The results showed that the complex prescription had both antitumor activity and immuno-activating activity, which might reflects on multi-activation mechanisms by complex components. 3. Classifying the theses by the efficacy of herbs examined, in single herb, invigorating spleen and supplementing was 35.5%, expelling toxin and cooling was 29.0%, activating blood flow and removing blood stasis was 12.9%. In herbal acupuncture, invigorating spleen and supplementing was 52.9%, expelling toxin and cooling was 29.4%. In complex prescription, pathogen-free status was 41.9%, strengthening healthy qi to eliminate pathogen was 35.5%, strengthening healthy qi was 22.6%. It is presumed that the antitumor and immunoactivating therapy based on syndrome differentiation is the best way to develop oriental oncology. 4. Classifying the theses by antitumor experiments, cytotoxic effect was 48.1 %, survival time was 48.1 % and change of tumor size was 42.3%. Survival rate was not necessarily correlated with cytotoxicity. These data reflect the characteristic, wholistic nature of the oriental medicine which is based on BRM (biological response modifier). 5. Classifying the theses by immunoactivating experiments, hemolysin titer was 51.0%, hemagglutinin titer was 46.2% and NK cell's activity was 44.2%. In the future studies, an effort to elucidate specific molecular and cellular mechanisms of cytokine production in the body would be crucial. 6. Classifying the theses according to the data in terms of antitumor activity, 50% was evaluated good, 24.0% was excellent, and 15.5% have no effect. In an evaluation of immuno-activating activity, 35.9% was excellent and 18.0% showed a little effect. The index point, as described here, may helps to use experimental data for clinical trials. Changes in index points by varying dosage implicate the importance of oriental medical theory for prescription. 7. In 167 materials, IIP (immunoactivating index point, mean : 3.12±0.07) was significantly higher than AIP(antitumor index point, mean : 2.83±0.07). These data demonstrate that the effect of herb medicine on tumor activity depends more on immunoactivating activity than antitumor activity. This further implies that the development of herbal antitumor drugs must be preceded by the mechanistic understanding of immunoactivating effect. 8. After medline-searching tumor and herb-related articles from NCBI web site, we conclude that most of the studies are primarily focused on biomolecular mechanisms and/or pathways. Henceforth, we need to define the biomolecular mechanisms and/or pathways affected by herbs or complicated prescriptions. 9. Therefore, the most important point of oriental medical oncology is to conned between experimental results and clinical trials. For the public application of herbal therapy to cancer, it is critical to present the data to mass media. 10. To develop the relationship of experimental results and clinical trials, university's cancer clinic must have a long-range plan related to the university laboratories and, at the same time, a regular consortium for this relationship is imperative. 11. After all these efforts, a new type herbal medicine for cancer therapy which is to take care of the long-term administering and safety problem must be developed. Then, it would be expected that anti-tumor herbal acupuncture can improve clinical symptoms and quality of life (QOL) for cancer patients. 12. Finally, oriental medical cancer center must be constructed in NCC (National Cancer Center) or government agency for the development of oriental medical oncology which has international competitive power.

Analysis of the Result of Korean Medicine Treatment for Female Subfertility Using Herbal Medicine, Acupuncture and Moxibustion Treatment

  • Objectives: To report on the result of infertility treatment with Korean medicine in clinical settings. Methods: This study was single arm, retrospective observational study. Analysis of the medical records on thirty four infertile women who were treated by acupuncture, moxibustion, herbal medicine(Jokyungjongoktang-gagambang, Sutaehwan-gagambang). Pregnancy rate(PR) and Live birth rate(LBR) were analyzed as the primary outcomes. Changes of AMH, FSH, LH, $E_2$, $P_4$, endometrial thickness, fat mass, and BMI were also analyzed as the secondary outcomes. Results: A total of 34 patients were treated in the traditional Korean medical clinic. Nine patients were dropped, and 25 subjects completed the final examination at the end of this treatment schedule. Five of 25 subjects conceived during treatment, reaching 20% PR and LBR was also 20%. Except for FSH level, the results of the remaining outcome measures, including LFT, showed no significant changes. No serious adverse event has been reported. Conclusion: Although no significant changes in bio-markers were observed, complex treatment using Korean medicine has shown effectiveness on women's infertility. In addition, the safety of the treatment has been verified.
