  • The conformation studies of tenecin 3, which has been purified from the hemolymph of the meal worms Tenebrio molitor, was carried out by CD and NMR. This highly Gly-rich protein consisting of 78 amino acid residues shows similar biochemical features such as heat stability, humoral existence, and high contents of Gly and His residues to other insect antifungal proteins.   • The influence of baculovirus Bombyx mori Nuclear Polyhedrosis virus (BmNPV) infection on intermediary metabolic pathways in silkworm Bombyx mori L. was investigated. Studies revealed that NADP-linked malate dehydrogenase activity in hemolymph of infected silkworms at 96 hrs post infection (p.i.) with visible symptoms of infection was enhanced in comparison to healthy larvae of the same age. Also, NADP-dependent MDH activity was significantly lower in fat body cytosol of infected larvae at 96 hrs p.i. when compared to healthy larvae. Similarly, some biometabolic parameters like growth, protein content and cholesterol titer were observed to be influenced by baculovirus infection. While the growth of infected larvae was significantly retardedi protein content was also drastically reduced in both hemolymph and fat body tissues. Cholesterol titers however, was enhanced in infected larvae. The results observed herein point to a significant change in the normal biochemical and biometabolic pathways required for growth and development following BmNPV infection.

Antifungal Effect and activity spectrum of crude antifungal proteins from hemolymph of larvae of Tenebrio molitor in Korea (한국산 갈색거저리로부터 분리된 항진균단백질의 항균효과와 그 작용 범위)

  • Antifungal protein from the hemolymph of larvae of Tenebrio molitor in Korea was partially purified by $C_{18}$ open column chromatography and assayed for the activity spectrum using 3 kinds of yeast and 4 kinds of filamentous fungi. The crude antifungal protein showed static activity for a broad range of fungal species. Weak cidal effects were observed in growing yeast type cells, including Candida and Saccharomyces. The affected cells were changed from ovoid to swollen and spherical form in shape. For filamentous fungi including Aspergillus and Fusarium, the crude antifungal protein affected the spore germination and the hyphal growth but not the viability significantly.

Effects of Storage-protein 2 Derived from Silkworm Hemolymph on Reduction of Aggregation and Cell Death in CHO Cells (CHO 세포에서 누에 혈림프 유래 Storage-protein 2의 세포응집 및 세포사멸 억제 효과)

  • Chinese hamster ovary (CHO) cells have been widely used for production of various recombinant proteins such as cytokines and monoclonal antibodies. The cell aggregation and cell death in CHO cell culture directly affect cell viability, and productivity and quality of products. In this study, we investigated preventing effects of storage-protein 2 (SP2) derived from silkworm hemolymph on cell aggregation and cell death in CHO cell culture producing albuminerythropoietin (Alb-EPO). The viable cell density in the culture supplemented with 2 mg/mL SP2 was 1.71-fold higher than that in control culture. Increased titer of Alb-EPO was also found in the culture with SP2. Morphology of CHO cells in SP2 supplemented cultures did not differ from that of control. In addition, the cell aggregation rate of the SP2 cultures was reduced 20% compared to the control. Finally, we confirmed that the apoptosis was strongly suppressed by addition of SP2 in the cultures. These results clearly demonstrate that SP2 can be served as an effective supplement for enhancing titer of Alb-EPO via reducing cell aggregation and cell death.

Studies on the Effects of various Levels of Protein in the Artificial Diet on Nutritional Physiology of the Silkworm, Bombyx mori (인공사료의 단백질수준이 가잠의 영양생리에 미치는 영향에 관한 연구)

  • Larvae of the silkworm (Bombyx mori L.) were reared during the 5th instar on the four kinds of artificial diets on the basis of the different amounts of soybean meal used as the protein source. In this experiment it was shown that the various levels of protein in the diet affected not only the growth and silk production but the digestibility of the diet. haemolymph protein and uric acid excretion. The results obtained are summarized as follows; 1. By an increase of the level of protein in the diet the apparant digestibility was increased. but the protein digestibility was comparatively decreased. 2. Larval body weight increment was not observed by the 3rd day of the 5th instar, but was increased from the 4th day as the level of protein was increased in the diet. 3. After the 3rd day of the 5th instar, protein content in the hemolymph was increased steeply by an increase of the protein content in the diet. However, the percentage of hemolymph protein to the ingested protein was decreased from the 2nd day of the 5th instar and increased more or less after the 4th day. 4. An increase of the uric acid excretion was observed as the content of protein in the diet was increased but the pattern of the uric acid excretion was different between high and low-protein diet. However, the percentage of the uric acid excretion to the ingested protein and to the hemolymph protein were both decreased steeply after the 2nd day of the 5th instar. 5. It was also evident that the high-protein diet increased the cocoon productivity. 6. It showed that the feed efficiency for body weight increment and silk formation was high by an increase of the level of protein in the diet, but the protein efficiency was not.

  • This experiment was to know properties of Sericin Jam that development, growth of silkgland, content of sericin and composition of amino acid in hemolymph. The characters of Sericin Jam can he seen form this experiment. Hatching ratio was 85% in Sericin Jam and 95% in Jam 120. Especially hatching period of Sericin Jam was longer than Jam 120 and also hatching of Sericin Jam was ununiform. The larval duration of Sericin Jam was 20 days and 23 hours, and Jaw 120 was 21 days and 22 hours. In Sericin Jam, middle silkgland contain a great p arts in silkgalod and posterior silkgand is short and no curves. The period of mounting to emergence was 12 days in Sericin Jam. The period of pupa was 7day. It is property of Sericin Jam that the period is short. Cocoon was very thin and light in Sericin Jam. Weight of cocoon shell of Sericin Jam is 2.7cg. The sericin protein quantity was 100% in cocoon shell of Sericin Jam, about 28% in cocoon shell of Jam 120, however the sericin ratio per Sericin Jam cocoon was 34.6% compare to cocoon shell of Jaw 120 in sericin protein quantity. The amino acids in hemolymph of Sericin Jaw was much hidtidine, lysine, glut amic acid. And the amount of almost amino acids were increased depends upon development at t he 5th instar.

Silk Protein as a Fetal Bovine Serum Substitute for Animal Cell Culture

  • Fetal Bovine Serum (FBS) is an essential substance added to animal cell culture medium. However, its composition is unclear causing problems such as development of an immune response when cultured cells are transplanted into the human body. In this study, silk sericin, silk fibroin, and hemolymph obtained from silkworms were added to the cell culture medium in order to determine if it can replace FBS. After establishment of the cell culture, cell proliferation and expression levels of cell growth-related genes were compared with those of control cells (cells cultured in the medium with 10% FBS). Results showed that the test group treated with silk fibroin extracted from a Korean silkworm variety, Kumokjam could replace 10% FBS. In addition, expression levels of cell growth related genes such as Fibronectin and TGF-β1 increased significantly in cells cultured using silk fibroin, depending on the concentration used in cell adhesion and cell proliferation [24]. To date, no studies have been conducted to find a replacement for FBS. Thus, this study was carried out to develop a substitute for FBS by using silkworm-derived alternatives such as silkworm hemolymph, silk sericin, and silk fibroin, which are cheap and have various physiological effects, cell promoting effects, and can be mass produced.

  • An increased understanding of apoptosis makes anti-apoptosis engineering possible, which is an approach used to inhibit apoptosis for the purpose of therapeutic, or industrial applications in the treatment of the diseases associated with increased apoptosis, or to improve the productivity of animal cell cultures, respectively. Some known anti-apoptosis proteins are the Bcl-2 family, IAP (inhibitor of apoptosis) and Hsps (heat shock proteins), with which anti-apoptosis engineering has progressed. This article reviews anti-apoptosis engineering using known anti-apoptosis compounds, and introduces a 30 K protein, isolated from silkworm hemolymph, as a novel anti-apoptotic protein, that Shows no homology with other known anti-apoptotic proteins. The regulation of apoptosis, using anti-apoptotic proteins and genes originating from the silkworm, Bombyx mori, may provide a new strategy in this field.

  • We identified juvenile hormone binding protein (JHBP) from last instar larval hemollvnph of Hvphantria cunea using gel filtration and non-SDS PAGE. Two kinds of JHBP in hemollnnph were found at two peaks by gel filtration (Sephadex G-100) and also at Rm values of 0.13 and 0.57 by non-SDS PAGE. JHBP was partially purified using anion exchange chromatosraphv, preparative gel filteration, and preparative PAGE. Dextrin coated charcoal (DCC) binding assay was employed to monitor the location of JHBP in chromatographic profile during the purification process. Purity of JHBP was checked by silver staining of 1091 SDS-Polyacrvlamide.

  • Male-specific protein (MSP) is a soluble protein which is accumulated in high amounts In the hemolymph and other organs of adult male wax moth. The MSP was purified from adult male wax moth by gel filtration and reversed Phase column chromatography, and its amino acid sequence was determined. Three internal amino acid sequences of MSP were obtained by the in-gel digestion method using trypsin because of its blocked N-terminus. (omitted)
