Association between self-assessed gingival bleeding and halitosis, and glycated hemoglobin levels in patients with diabetes (제2형 당뇨병 환자에서 자가 인식한 치은 출혈 및 구취와 당화혈색소 간의 연관성)

  • Objectives: This study aimed to analyze the association between self-assessed periodontal symptoms and glycated hemoglobin levels in patients with type 2 diabetes. Methods: This cross-sectional study involved 156 patients with type 2 diabetes who were aged 50 years or older. Structured questionnaires were used to investigate the self-assessed periodontal symptoms of the patients. The glycated hemoglobin test was performed to evaluate their long-term blood glycemic control. Chi-square test and logistic multiple regression were performed to analyze the factors associated with glycated hemoglobin levels. Results: Compared with patients aged 65 years and above, more patients aged 64 years and below showed poor glycemic control (p=0.020). Further, compared with patients without self-perceived gingival bleeding and halitosis, more patients with these two conditions showed poor glycemic control (p<0.05). Compared with the group of patients without any periodontal symptoms, the group of patients that had at least one periodontal symptom had a higher proportion of patients with poor glycemic control (p<0.001). In the logistic regression model, gingival bleeding and halitosis were the factors most associated with hyperglycemia (p<0.05). Conclusions: The results of our study suggest that gingival bleeding and halitosis can predict hyperglycemia in patients with type 2 diabetes.

Optimization of Enzyme Digestion Conditions for Quantification of Glycated Hemoglobin Using Isotope Dilution Liquid Chromatography-Tandem Mass Spectrometry

  • Glycated hemoglobin (HbA1c) is used as an index of mean glycemia over prolonged periods. This study describes an optimization of enzyme digestion conditions for quantification of non-glycated hemoglobin (HbA0) and HbA1c as diagnostic markers of diabetes mellitus. Both HbA0 and HbA1c were quantitatively determined followed by enzyme digestion using isotope dilution liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) with synthesized N-terminal hexapeptides as standards and synthesized isotope labeled hexapeptides as internal standards. Prior to quantification, each peptide was additionally quantified by amino acid composition analysis using ID-LC-MS/MS via acid hydrolysis. Each parameter was considered strictly as a means to improve digestion efficiency and repeatability. Digestion of hemoglobin was optimized when using 100 mM ammonium acetate (pH 4.2) and a Glu-C-to-HbA1c ratio of 1:50 at $37^{\circ}C$ for 20 h. Quantification was satisfactorily reproducible with a 2.6% relative standard deviation. These conditions were recommended for a primary reference method of HbA1c quantification and for the certification of HbA1c reference material.

The Effect of the Oriental Postpartum Care in Hemoglobin Changes of Maternity (한방산후조리가 산모의 혈색소 변화에 미치는 영향에 대한 연구)

  • Objectives : Anemia is the center of interest in every maternity, both during pregnancy and lactation. The oriental postpartum care has one principle : to invigorate Qi and blood greatly. In this study, we seek to confirm that most sufferers of anemia recover under oriental postpartum care. To assure that this study had significance in non-iron supplement, we made a comparative study between oral iron supplement and non-iron supplement groups in oriental postpartum care. Methods : A comparative study was conducted to discern the difference in hemoglobin change between admission and after 2 weeks under the circumstances of 30 oral iron supplemented and 38 non-iron supplemented women who were admitted to the hospital attached to Woosuk University Oriental Medical College from April to August 2001 for postpartum care. Results : The mean hemoglobin level of after 2 weeks was 11.95 mg% (p<0.001). The mean hemoglobin level after 2 weeks was 11.65 mg% and 12.18 mg% in the iron supplemented and non-iron supplemented women respectively. The difference between the two groups was not significant. Conclusions : Oriental postpartum care overcomes anemia significantly in postpartum maternity. This is confirmed significantly under the non-iron supplement.

Effect of Using Insect Diet on Fecal Properties and Hemoglobin Levels in Companion Canine(2) (곤충사료 급여에 따른 반려견 분변성상과 헤모글로빈 수치에 미치는 영향(2))

  • We investigated the fecal properties and hemoglobin levels in dogs fed with general canine diets and 7 black soldier fly-based companion canine diets. A total of 16 dogs, including 8 poodles (average weight 2.7±0.5 kg) and 8 bichon frises (average weight 2.0±0.5 kg) were used in this study. The changes in fecal properties of all treatments at 0 weeks and 2 weeks showed no significant differences (p>0.05). However, at 4 weeks, compared to the control group, all black soldier fly-based companion canine diets have an effect on fecal properties (p<0.05). Hemoglobin levels in all treatments were within the normal range. Therefore, feeding black soldier fly-based diets to canines affected the fecal properties and the hemoglobin level was not of clinical concern since it was within the desired reference intervals for healthy canines, indicating that the effect of iron supplementation and anemia prevention was not observed.

  • The hemoglobin phenotypes and the gene frequencies of 223 Cheju native horses were studied by starch gel electrophoresis. The results obtained were as follows: 1. In the hemoglobin phenotypes, three phenotypes, HbAA, HbAa and Hbaa, which were controlled by two allelic genes. $Hb^A$ and $Hb^a$, were observed and their frequencies of appearance were 65.47%, 30.04% and 4.48% respectively. 2. The distribution of gene frequency was calculated as 0.805 in $Hb^A$ and 0.195 in $Hb^a$.

Refolding of Acid-Unfolded Globin to Hemoglobin

  • Hemoglobin is oxygen carrier protein within erythrocyte in blood. Apoprotein of this, globin, is synthesized in the cytosol but it's cofactor, heme, is synthesized in the mitochondria. It has not been known very well how globin receives the heme from mitochondria and folds to hemoglobin. In this folding process, the initial structure of globin seems to be very important. A small volume of globin at acid pH was added rapidly into the bulk of an egg phosphatidylcholine $60\%$ liposome, containing hemins, at neutral pH according to the Rapid Dilution method. It was observed that an acid-induced unfolding structure of globin is initially needed to receive hemins from the lipid bilayer of liposomes. Also, this conclusion was confirmed with the absorption spectrum of the refolded globin separated by centrifugation.

Production of methionine γ- lyase in recombinant Citrobacter freundii bearing the hemoglobin gene

  • The production of antileukemic enzyme methionine ${\gamma}$-lyase (MGL) in distinctly related bacteria, Citrobacter freundii and in their recombinants expressing the Vitresocilla hemoglobin (VHb) has been studied. This study concerns the potential of Citrobacter freundii expressing the Vitreoscilla hemoglobin gene (vgb) for the methionine ${\gamma}$- liyase production. Methionine ${\gamma}$- liyase production by Citrobacter freundii and its $vgb^-$ and $vgb^+$ bearing recombinant strain was studied in shake-flasks under 200 rpm agitation, culture medium and $30^{\circ}C$ in a time-course manner. The $vgb^+$ and especially the carbon type had a dramatic effect on methionine ${\gamma}$- liyase production. The $vgb^+$ strain of C. freundii had about 2-fold and 3.1-fold higher levels of MGL than the host and $vgb^-$ strain, respectively.

Acute Variation of Hematological Parameters during 622 km Ultra-Marathon

  • To investigate the effects of strenuous physical exercise on commonly used hematological markers in subjects the intensive long running. Blood samples were obtained from nineteen participants in a 622 km ultra-marathon race before, 300 km and immediately after completion of the 622 km ultra-marathon. Samples were analyzed for total white cell count (WBC) and differential, total red cell count (RBC), hemoglobin, hematocrit, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), red cell distribution width (RDW), platelets, mean platelets volume (MPV), platelets distribution width (PDW). Significant increases were found in WBC, neutrophil and platelets at 622 km compared to the pre-race. RBC, hemoglobin and hematocrit decreased statistically significantly the race at 300 km and 622 km compared to pre-race. A wide range of hematological perturbations occur during 622 km ultra-marathon running but it was physiological changes within a reference range. The 622 km ultra-marathon is less likely to cause clinically significant hematologic changes in athletes.

Adsorption Property of Shrimp Shell Chitosan to Water Soluble Proteins (수용성 단백질에 대한 새우껍질 Chitosan의 흡착 특성)

  • 수산물 가공시 흘러나오는 폐수 중에 함유되어 있는 수용성 단백질을 chitosan에 흡착시키기 위하여 albumin과 hemoglobin, albumin-myoglobin 혼합단백질을 이용하여 chitosan과의 흡착 및 chitosan 제조조건에 따른 흡착효과와 chitosan의 수용성 단백질 흡착에 미치는 인자를 살펴본 결과는 다음과 같다. Chitosan위 탈아세틸화 조건을 60, 70, 80%로 달리하여, albumin, hemoglobin, albumin-myoglobin 혼합용액에 적용했을때 chitosan의 탈아세틸화도가 높을수록 chitosan과 단백질 사이의 흡착률은 높게 나타났다. 초음파 처리에 의하여 chitosan의 분자량이 작을수록 chitosan과 단백질 사이의 흡착률은 높게 나타났다. pH변화에 따른 chitosan과 수용성 단백질의 흡착률은 albumin 및 albumin-myoglobin 혼합용액에서는 pH 4.0에서, hemoglobin용액에서는 pH 7.0에서 흡착률이 높게 나타났다 chitosan과 수용성단백질과의 흡착에서 반응시간은 albumin및 albumin-myoglobin 혼합용액에서는 4시간, hemoglobin용액에서는 3시간까지 흡착률이 증가하였고, 그 이후의 시간이 경화하여도 흡착률의 증가는 거의 보이지 않았다. 수용성 단백질 용액에 NaCl 농도를 0.1M에서 1.0M로 증가시켜 첨가했을때 염의 농도가 높을수록 chitosan과 단백질 사이의 흡착이 잘 일어나지 않았다.

General Pharmacology of PEG-Hemoglobin SB1

  • PEG-hemoglobin SB1 (SB1), which is a hemoglobin-based oxygen carrier, is intended to use as a safe blood substitute against brain ischemia and stroke. The general pharmacological profiles of SB1 were studied. The doses given were 0, 5, 10, 20 ml/kg and drugs were administered intravenously. The animals used for this study were mouse, rat and guinea pig. SB1 showed no effects on general behavior, motor coordination, spontaneous locomotor activity, hexobarbital sleeping time, anticonvulsant activity, analgesic activity, blood pressure and heart rate, left ventricular peak systolic pressure, left ventricular end diastolic pressure, left ventricular developing pressure, double product, heart rate, coronary flow rate, smooth muscle contraction using guinea pig ileum, gastrointestinal transport, gastric secretion, urinary volume and electrolyte excretion at all doses tested except the decrease of body temperature. These findings demonstrated that SB1 possesses no general pharmacological effects at all doses tested.
