Effects of Juakium-derivative on Experimental Aplastic Anemia (좌귀음가미방이 실험적 재생불량성빈혈에 미치는 영향)

  • Objectives : Korean traditional medicinal herbs have been used to improve the function of the hematopoietic system. The purpose of this study is to examine the hematopoietic effects of Juakium-derivative on aplastic anemia. Methods : In vitro, after bone marrow cells of mice with aplastic anemia induced by benzene were cultured with Juakium-derivative, we measured the gene expressions of hematopoietic cytokines, the intracellular TPO and SCF expression and the colony number of CFU-GEMM and BFU-E. Results : The results are summarized as follows : 1. The gene expressions of hematopoietic cytokines (TPO, c-mpl, SCF, c-kit, IL-3, EPO, EpoR, GM-CSF) and the TPO and SCF expression of the group treated with Juakium-derivative increased significantly more than those of the control group. 2. The colony number of CFU-GEMM and BFU-E in the rEPO plus rIL-3 plus Juakium-derivative group increased significantly compared with the only rEPO plus rIL-3 group. Conclusion : It was acknowledged that Juakium-derivative has hemato-potentiating effects on aplastic anemia induced by benzene and it is expected that Juakium-derivative can be used clinically for the patient with hematopoietic system disorder. This study could also be used further as important research data for Korean oriental medicine about hemato-potentiating. The exact mechanism explaining our study would need to be studied further.

The Effect of DanSamSaMul-Tang on Hematopoiesis (단삼사물탕이 조혈작용에 미치는 영향)

  • Objectives : Korean traditional medicinal herbs and some formulas have been used to improve the function of the hematopoietic system. The present research is to examine the effect of DanSamSaMul- Tang(DSSMT) as a biological response modifier related to hemato-potentiating function using bone marrow cell as a testing system. Methods : Mice with leukopenia and thrombocytopenia induced by CTX were treated with DSSMT Extracts. Then we observed cytokines mRNA expression level, the number of CFU-GEMM and BFU-E, and the expression of TPO and SCF in bone marrow cells of mice. Results : In the DSSMT group, the gene expressions of hematopoietic cytokines were increased significantly and the number of CFU-GEMM and BFU-E of this group was significantly increased compared to the group treated only with EPO and IL-3. The expressions of TPO and SCF in the group treated with DSSMT were higher than those of the control group. Conclusions : We showed that DSSMT has a good effect on the hematopoietic system. This could also be used as important research data for Korean oriental medicine about hemato-potentiating.

  • Ex vivo hematopoietic cells can treat patients suffering from hematopoietic malignancies using bone marrow transplantation therapies. A initial cell density of $1.5{\times}10^6cells/ml$ and a growth factors of IL-3(5ng/ml), SCF(5ng/ml) and FL(25ng/ml) result in a 3.6-fold expansion of LTC- IC but a unexpansion of total cells.

Effects of Korean Traditional Medicine on Murine Hematopiesis (Regulation of Hematopoietic Cytokine & $CD34^{+}$ cell Expression) (수 종의 한약제제가 조혈작용에 미치는 영향)

  • Objectives : To evaluate the diverse actions of stimulation on the hematopoietic system, 4 formulas (KH I, KH 2, KH 3, KH 4) were studied. Method and Result : RT-PCR was performed to measure the gene expression of hematopoietic cytokines (TPO, GM-CSF, SCF, IL-3). When bone marrow cells were treated with KH 1, 2, 3, 4, the gene expressions of TPO, SCF, IL-3, and GM-CSF were increased. Flow cytometric analysis was performed to measure the expression of CD34+ cell activity. After 72 hrs culture supplemented with KH 1, 2, 3, 4, the percent of CD34+ cell of KH 2, 3, 4 were increased. To measure the expression of colony forming units - granulocyte erythrocytes, macrophages, megakaryocytes (CFU-GEMM) and burst forming unit-erythroid (BFU-E), semisolid clonogenic assay was performed. After 14 days of culture the number of CFU-GEMM and BFU-E of KH I, 2, 3, 4 were significantly increased compared to those of EPO groups (KH 1 P<0.0l, KH 2 P<0.05, KH 3 P<0.001, KH 4 P<0.0l). To determine the intracelluar TPO expression by KH 3, KH 4 in bone marrow cells, intracelluar staining and flow cytometric analysis were performed. After 24 hrs cultures, the TPO expression of the KH 3 and KH 4 treated groups were increased over those of the controlled groups (control : 50%, KH 3 : 87%, KH 4 : 78%). Conclusion : These results suggest that KH I, KH 2, KH 3, KH 4 have hematopoietic effects through increasing the production of hematopoietic cytokines and stimulating the activity of $CD34^{+}$ cells. This study also shows that KH 3 has a more effective hematopoietic effect than KH 1, 2, 4. These results suggest that the formulas (KH I, 2, 3, 4) can be applied to the patients with inappropriate hematopoietic system, and that KH 3 can be the most effective formula among these 4 in treating bone marrow disease in clinics.

  • The transcription factor RUNX1 first came to prominence due to its involvement in the t(8;21) translocation in acute myeloid leukemia (AML). Since this discovery, RUNX1 has been shown to play important roles not only in leukemia but also in the ontogeny of the normal hematopoietic system. Although it is currently still challenging to fully assess the different parameters regulating RUNX1 dosage, it has become clear that the dose of RUNX1 can greatly affect both leukemia and normal hematopoietic development. It is also becoming evident that varying levels of RUNX1 expression can be used as markers of tumor progression not only in the hematopoietic system, but also in non-hematopoietic cancers. Here, we provide an overview of the current knowledge of the effects of RUNX1 dosage in normal development of both hematopoietic and epithelial tissues and their associated cancers.

EXPERIMENTAL STUDY ABOUT ESTROGEN EFFECT OF BONE MARROW AND SPLEEN OF OVARIECTOMIZED RATS (에스트로겐 투여가 난소절제 백서의 골수와 비장에 미치는 효과에 관한 실험적 연구)

  • The most serious problem resulting from estrogen deficiency induce osteo porosis. Recently, they make efforts to inquire a relation between hematopoietic organ and bone loss due to estrogen deficiency. Estrogen have an effect on growth and formation of skeletal system, and inhibit bone resorption under the influence of osteoblast and osteoclast, and basically inhibit the increase of hematopoietic progenitor and immune factor connected with bone resorption and prevent the osteoid formation. The purpose of this article was to observe the change of spleen and effect on hematopoietic function following estrogen administration. In this study, female rats of 150g weight was ovariectomized, after 70 days, experimental group was injected estrogen at interval of a week and sacrificed on 1, 2, 3, 4, 6 weeks. Control group was sacrificed after ovariectomy on 11, 12, 13, 14, 16 weeks without estrogen injection, and normal rats were sacrificed for harvest of spleen and femur. Paraffin sections and H&E stain was performed, and observed under light microscope. The obtained results were as follows. 1. From 11 to 12 weeks at bone marrow of control group, hematopoietic cells were decreased in comparison with normal group, and lipid infiltration was seen, and irregular bone remodelling was seen after 13 weeks. From 14 to 16 weeks, there were more decreased hematopoietic cells and lipid degeneration, and lipid degeneration of hematopoietic cells appeared. 2. All the bone marrow of experimental group, the structure of hematopoietic cells with decreased lipid infiltration was recovered from 2 weeks of estrogen adminstration and maintained to 6 weeks. 3. At spleen of control group, borders of white and red pulp was not well demarcated, and size of white pulp was decreased. 4. At spleen of experimental group, borders between white and red pulp have been well demarcated from 3 weeks of estrogen adminstration relatively, and white pulp was increased with distinct border. From above findings, we could regarded that estrogen deficiency due to ovariectomy influenced on hematopoietic cells of bone marrow and spleen, and histologic recovery of hematopoietic cells were observed after 3 weeks of estrogen adminstration even if it was not reach to normal group.

Enhancing effect of Canavalia gladiata DC semen on the hematopoietic expansion and function of stem cells (도두(刀豆)의 조혈증진 작용에 관한 연구)

  • Objective : Canavalia gladiata DC semen (CGS) have been used to improve hematopoietic activity. In the current study, we investigated whether CGS regulate hemato-potentiating function using hematopoietic stem cells (HSCs) as a testing system. Methods : HSCs isolated from femur in mice with leukopenia and thrombocytopenia induced induced by CTX. Then, Real-time PCR was performed to measure the mRNA expression and hematopoietic related gene (EPO, IL-3, SCF, c-kit, GM-CSF), the phoaphorylation of GATA-1 and STAT-5a/b were observed by ELISA method, and the number of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E), semisolid clonogenic assay was performed. Result : When HSCs were treated with CGS, the expression of hematopoietic related genes (EPO, IL-3, SCF, c-kit, and GM-CSF) were significantly increased at the levels of mRNA as well as production in HSCs. Additionally, CGS enhanced phosphorylation of STAT-1 and signal transducer and activator of transcription-5a/b (STAT-5a/b) in HSCs. Furthermore, CGS significantly enhanced the growth rate of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E) in vitro. Conclusion : These result suggest that CGS has hematopoietic enhancement via hematopoietic cytokine-mediated GATA-1/STAT-5a/b pathway.

Leukemia in Iran: Epidemiology and Morphology Trends

  • Background: Leukemia accounts for 8% of total cancer cases and involves all age groups with different prevalence and incidence rates in Iran and the entire world and causes a significant death toll and heavy expenses for diagnosis and treatment processes. This study was done to evaluate epidemiology and morphology of blood cancer during 2003-2008. Materials and Methods: This cross-sectional study was carried out based on re-analysis of the Cancer Registry Center report of the Health Deputy in Iran during a 6-year period (2003 - 2008). Statistical analysis for incidence time trends and morphology change percentage was performed with joinpoint regression analysis using the software Joinpoint Regression Program. Results: During the studied years a total of 18,353 hematopoietic and reticuloendothelial system cancers were recorded. Chi square test showed significant difference between sex and morphological types of blood cancer (P-value<0.001). Joinpoint analysis showed a significant increasing trend for the adjusted standard incidence rate (ASIR) for both sexes (P-value<0.05). Annual percent changes (APC) for women and men were 18.7 and 19.9, respectively. The most common morphological blood cancers were ALL, ALM, MM and CLL which accounted for 60% of total hematopoietic system cancers. Joinpoint analyze showed a significant decreasing trend for ALM in both sexes (P-value<0.05). Conclusions: Hematopoietic system cancers in Iran demonstrate an increasing trend for incidence rate and decreasing trend for ALL, ALM and CLL morphology.

Acute Radiation Syndrome in an Irradiated Minipig Model for Patients with Radiation Exposure

  • Background: Acute radiation syndrome (ARS) primarily refers to damage to the hematopoietic system, myeloid system, and gastrointestinal (GI) system caused by radiation exposure. Such damage progresses to become life-threatening. In particular, as the syndrome develops very rapidly-within several hours from radiation exposure-prompt and accurate diagnosis and treatment are needed, as is further research into appropriate diagnostic and treatment modalities. Materials and Methods: Minipigs, which display human-like properties, underwent whole-body irradiation at 2 or 4 Gy (doses causing hematopoietic ARS) or at higher doses of 7 or 12 Gy. Changes in the blood cells and clinical symptoms were analyzed and we performed a necropsy when the animals succumbed to ARS. Results and Discussion: The minipig irradiated with 2 Gy showed a decrease in white blood cells, including neutrophils, lymphocytes, and platelets in the early stages. However, the blood cell counts gradually increased and returned to normal values. The minipig irradiated with 4 Gy succumbed due to hematopoietic ARS. In contrast, the minipigs irradiated with 7 or 12 Gy exhibited clinical symptoms of combined GI damage and hematopoietic syndrome. Moreover, a characteristic pattern of platelet changes was observed in the 7 and 12 Gy irradiated minipigs. Conclusion: The changes in the platelet count caused by radiation exposure observed in minipigs, which are hematologically and pathohistologically similar to humans, suggest that they can be used as a novel diagnostic criterion.

Effects of Juakium-derivative on Hematopoietic Effects (좌귀음가미방(左歸飮加味方)이 조혈작용에 미치는 영향)

  • Objectives : Acording to the fact that Juakium has been frequently used in a lot of prescriptions to tonify bone marrow, We examined the hematopoietic effects of Juakium-derivative on aplastic anemia. Methods : After C57BL/6 mice were oral administrated with Juakium-derivative and injected with benzene, we counted the number of WBC, RBC, hemoglobin, platelet, nucleated cells and other erythroid parameters and weighed the spleen. Also we measured the expression of $CD34^+$ cell activity and analyzed the bone marrow tissue and spleen tissue histologically. Results : The results are summerized as follow: 1. The Juakium-derivative plus benzene group showed the improvement in the number of WBC, RBC, hemoglobin. platelet, nucleated cells and other erythroid parameters, compared with the benzene only group. 2. The spleen weight and the percent of $CD34^+$ cell of Juakium-derivative plus benzene group was higher, compared with the benzene only group. 3. The bone marrow tissue and spleen tissue of the Juakium-derivative plus benzene group showed the decrease of the infarcted area compared with the benzene only group. Conclusions : These results suggest that Juakium-derivative has hematopoietic effects on aplastic anemia induced by benzene through increasing the blood cells and stimulating the activity of $CD34^+$ cells. Therefore it is expected that Juakium-derivative can be used clinically to the patient with hematopoietic system disorder.
