The present study was performed to evaluate the effect of medicinal plant extract on relieving hangovers in mice administered alcohol. The animals were divided into three groups. Each group was treated with fermented plant extract, non-fermented plant extract, or water 30 min after consuming ethanol (2 mL/kg). A locomotor activity test showed that all groups had decreased motor activity until 40 min after plant extract administration. The mice treated with water had lower motor activity until 100 min post-administration. However, the group treated with non-fermented plant extract showed increased motor activity 40 min post-administration, and the higher activity level was maintained until 120 min post-administration. The animals treated with fermented plant extract had a level of motor activity between those of the groups treated with water or non-fermented plant extract. Blood was collected from each mouse 120 min post-administration and aldehyde concentration was measured. The group treated with non-fermented plant extract had a significantly higher (p < 0.05) aldehyde concentration than the other groups. These results demonstrate that the non-fermented medicinal plant extract helped alleviate hangovers 40 min after administration by reducing aldehyde concentrations in the blood.
본 연구에서는 식용 가능한 오디를 용매별로 추출하여 항산화 효과 및 숙취 해소 활성능을 알아보고자 하였다. 오디Fig. 1. Contents of ADH ability of each solvent fraction of mulberry fruit extract. 추출물에 들어있는 총 페놀 함량을 정량하였으며, 오디 추출물의 전자공여능, SOD 유사 활성, TBARS 아질산염 소거능 등을 통하여 오디의 항산화 활성을 측정하였고, 알코올 분해능은 alcohol dehydrogenase activity assay 방법을 이용하여 조사하였다. 오디 추출물의 총 페놀 화합물은 60% methanol 추출물에서 가장 높게 나타났다. 전자공여능(EDA)은 오디의 40% ethanol 추출물에서 BHT와 유사한 활성을 나타났고, SOD 유사 활성은 BHT보다 오디 추출물이 낮은 활성을 나타내었다. 아질산염 소거능은 오디의 60% ethanol과 40% methanol 추출물이 각각 98%, 97%로 ascorbic acid와 상응하는 결과를 보였다. 알코올 분해능에서는 오디의 60% ethanol 추출물이 가장 높은 활성을 나타내었다.
This study was conducted to investigate if the supplement formula may improve alcohol metabolism in healthy adult men. In a double-blinded, randomized, crossover study, subjects were administrated yeast extract mixtures (YEM, n=15), Hovenia dulcis mixtures (HDM, n=15), placebo (PLA, n=15), and control (CON, n=15) in an oral dose followed by one week washout periods. At each visit (0, 1, 2, 3, 4 week), subjects drank 450 mL, 20.1 percent alcohol after administered mixtures. Blood was drawn periodically (0, 0.25, 0.5, 1, 2, 4, 6, 15 hours). Fifteen subjects completed the protocol and were included in the analysis. Plasma ethanol concentration was lower in YEM (10 percent) and the HDM (5 percent) groups. The area under the curves (AUC) and $C_{max}$ for plasma ethanol were significantly decreased only in the YEM group, when compared with the CON group. The AUC and $C_{max}$ for plasma acetaldehyde concentration were significantly decreased in the YEM (45 and 54 percent) and the HDM (35 and 53 percent) groups respectively, when compared with PLA (p<0.01). Together, these findings validate that YEM or HDM improved alcohol metabolism and hangover syndromes, leading to reduce alcohol concentration and acetaldehyde concentration without adverse effects.

Hangover due to alcohol consumption causes social and physical problems. There is a growing interest in edible insects worldwide. We have previously published a new technology to make hard mature silkworm, Bombyx mori, into edible form, steamed and freeze-dried mature silkworm larval powder (SMSP). In this study, AIN-76 or SMSP (0.1 and 1 g/kg rat body weight) containing diets in SD rats were pretreated for 2 weeks, and ethanol (3 g/kg rat body weight) was administered as an oral gavage and sacrificed after 3 hours. As a result, blood alcohol and aldehyde levels were significantly decreased in SMSP fed rats. In addition, liver injury markers, alanine aminotransferase (ALT) and alkaline phosphatase (ALP) were significantly decreased in SMSP group compared to ethanol group. $TNF-{\alpha}$, an inflammatory cytokine, and malondialdehyde (MDA), an oxidative stress marker, also showed a dose-dependent decrease in the group receiving SMSP. Conclusively, consumption of SMSP not only reduced hangover induced by ethanol, but also decreased liver damage, oxidative stress, and inflammatory response.
약용식물의 열수 추출물이 in vitro에서 alcohol dehydrogenase (ADH)와 aldehyde dehydrogenase (ALDH)의 활성에 미치는 영향을 확인하였다. 약용식물에 20배의 증류수를 넣고 $80^{\circ}C$에서 8시간 추출하여 얻어진 추출액을 시료로 사용하였다. 50종의 약용식물 중에서 마늘과 육계 추출물이 숙취해소 천연소재로서의 활용 가능성이 가장 높게 나타났다. 마늘 추출물은 ADH에 비해 ALDH의 활성을 2배 이상 촉진시킴으로써 acetaldehyde의 분해가 잘 되게 하였다. 육계 추출물은 ALDH의 활성에 비해 ADH의 활성을 획기적으로 저해함으로써 acetaldehyde의 생성을 크게 억제하였다. 육계 추출물은 농도에 비례하여 ADH와 ALDH의 활성을 저해하였으며, $45.33{\mu}g/mL$의 농도에서 ADH의 활성을 52.8% 저해하였고 ALDH의 활성을 11.0% 저해하였다.
Objectives : Alcohol hangover is a common phenomenon which basically occurs after heavy drinking. Moreover, heavy alcohol consumption leads to acute and chronic diseases. We investigated the effect of herbal mixture (SJ) on alcohol metabolism in serum or/and liver. Methods : 5W-old Sprague Dawley rats were used for the study. To overnight fasted rats, 0.9% saline or SJ extract was administrated per os before alcohol treatment. Then, 40% alcohol was orally administrated to all rats in 30 mins. Ethanol, acetaldehyde concentrations, alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH) activities were measured by assay kits. Aspartate aminotransferase (AST) and alanine transaminase (ALT) were measured by analyzer. Results : Ethanol and acetaldehyde concentrations lowered in SJ groups compared with CON group. Especially, acetaldehyde concentration significantly decreased in SJ groups compared with CON group. AST and ALT levels tended to increase in SJ groups compared with CON group but there was no significant difference between CON group and SJ groups. ADH activity in serum was higher in SJ groups than CON group but no significant difference in liver. ALDH activity in both serum and liver showed significantly increased in SJ groups compared with CON group. Conclusions : Treatment of SJ extract showed not only reducing concentration of alcohol and acetaldehyde but also increasing activities of ADH and ALDH. These results suggest SJ may influence in alcohol metabolism via control of metabolic enzyme activities and metabolite. Therefore, SJ, herbal mixture, might have a function of preventing hangover after drinking alcohol.

Excessive alcohol consumption can have serious negative consequences on health, including addiction, liver damage, and other long-term effects. The causes of hangovers include dehydration, alcohol and alcohol metabolite toxicity, and nutrient deficiency due to absorption disorders. Additionally, alcohol consumption can slow reaction times, making it more difficult to rapidly respond to situations that require quick thinking. Exposure to a large amount of ethanol can also negatively affect a person's righting reflex and balance. In this study, we evaluated the potential of lactic acid bacteria (LAB) to alleviate alcohol-induced effects and behavioral responses. Two LAB strains isolated from kimchi, Levilactobacillus brevis WiKim0168 and Leuconostoc mesenteroides WiKim0172, were selected for their ethanol tolerance and potential to alleviate hangover symptoms. Enzyme activity assays for alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were then conducted to evaluate the role of these bacteria in alcohol metabolism. Through in vitro and in vivo studies, these strains were assessed for their ability to reduce blood alcohol concentrations and protect against alcohol-induced liver damage. The results indicated that these LAB strains possess significant ethanol tolerance and elevate ADH and ALDH activities. LAB administration remarkably reduced blood alcohol levels in rats after excessive alcohol consumption. Moreover, the LAB strains showed hepatoprotective effects and enhanced behavioral outcomes, highlighting their potential as probiotics for counteracting the adverse effects of alcohol consumption. These findings support the development of functional foods incorporating LAB strains that can mediate behavioral improvements following alcohol intake.
숙취해소효과가 우수한 제품을 개발하기 위하여, 약용식물 25종의 열수추출물에 대하여 in vitro에서 ADH와 ALDH의 저해 및 활성작용이 우수한 약용식물 12종을 선발, 일정 비율로 혼합한 약용식물 추출액에 대하여 발효를 행한 후 in vitro 및 in vivo실험을 통하여 숙취해소효과를 검토하였다, 그 결과, 감초 -0.3%, 갈근 -3.2%, 구기자 -1.7%, 맥문동 4.8%, 속새 8.3%, 오가피 3.5%, 및 지구자 -2.1%를 보여 지구자를 비롯한 7종의 추출물이 90%이상의 강한 저해활성을 보였고, ALDH활성에서는 오가피가 166.3%로 활성이 가장 촉진되었고, 뒤이어 구기자, 속새 및 백복령도 1.5배 이상의 활성을 촉진하였으며 이 외에 맥문동, 인진쑥, 작약, 차전초, 지구자, 홍삼 및 흑두에서도 효소활성을 촉진하는 것으로 나타나는 등 갈근을 포함하여 약 10여종의 약용식물에서 130%이상의 활성을 증가시키는 결과를 보여주었다. 12종으로 조성 혼합된 약용식물 추출물에서는 ADH활성이 -20.22%인데 비하여 추출발효액은 -62.63%정도 ADH저해활성이 높았으며, ALDH활성은 추출물에서는 173.20%이었으며, 추출발효액은 280.17%로 ALDH활성이 추출액보다 약 1.6배가 증가됨을 볼 수 있었다. 또한 흰쥐를 이용하여 약용식물 추출발효물의 숙취해소효과를 보기 위해 알코올 투여 후 시간에 따른 혈중 알코올과 아세트알데히드의 농도를 측정한 결과, 알코올 섭취 후 3시간 경과시 추출 발효물 400 mg/kg 처리군이 시판 숙취개선음료 처리군보다 24.1% 정도 우수한 효능을 보였고, 추출발효물 공급 3시간 경과 시부터 7시간까지 혈중 아세트알데히드의 감소폭은 추출 발효물 400 mg/kg 처리군에서 -65.1%로 시판 숙취개선음료를 공급한 군에서 -57.5%를 나타냈다.
발효에 의한 헛개열매의 기능성 상승 정도를 검토하고자, 헛개열매열수추출물을 발효시킨 후 급성 및 만성 알코올 투여 간손상 동물모델을 통하여 체중감량 억제, 알코올 분해 및 간기능 개선 효능을 검증하였다. 급성 알코올 투여 동물모델에서 헛개열매발효군(ET-FHWE)은 알코올대조군(ET)에 비하여 혈청 알코올 농도가 유의적으로 감소되었고, 특히 알코올 투여 3시간 후의 알코올 농도는 헛개열매추출액발효물에 의해 46.1%, 헛개열매열수추출물에 의해 19.1% 감소된 것으로 나타났다. 또한 알코올 투여에 의해 증가된 혈청 아세트알데하이드 농도는 헛개열매추출액발효물에 의해 알코올 투여 3시간 후에는 48.7%, 5시간 후에는 39%로 알코올대조군(ET)보다 유의적으로 감소하였고, 이는 헛개열매열수추출물은 발효에 의해 알코올 및 아세트알데하이드 분해능이 증가하는 것으로 사료되었다. 만성 알코올 투여 간손상 동물모델 실험에서 알코올 투여에 의해 상승된 혈청 알코올 농도는 헛개열매열수추출물과 헛개열매추출액발효물 투여에 의해 각각 31%, 41% 유의적으로 감소하였다. 혈청 아세트알데하이드 농도와 ${\gamma}$-GTP 활성도는 헛개열매열수추출물과 헛개열매추출액발효물 투여에 의해 알코올대조군(ET)보다 유의적으로 감소되었으며, 장기간 알코올 투여에 의한 체중 감소 억제 및 간조직 지질수준의 유의적 감소를 나타내었다. 또한 헛개열매추출액발효물은 장기간의 알코올 투여로 인해 감소된 혈당을 유의적으로 증가시키는 것으로 나타났다. 본 연구 결과, 급성 알코올 투여 동물모델에서 헛개열매열수추출물은 발효에 의해 알코올 및 아세트알데하이드 분해능이 증진되었고, 만성 알코올 투여 모델을 통한 실험에서는 발효에 의해 헛개열매의 간기능 개선효능이 유지됨과 동시에 일부 효능(혈청 지질 및 혈당 수준 개선능)은 증가하는 것으로 나타났다. 따라서 헛개열매추출액발효물은 급성 및 만성 알코올성 간손상 억제에 있어서 헛개열매열수추출물보다 더욱 강력한 기능성 물질로 활용될 수 있을 것으로 기대된다.
항산화 작용에 효과가 있다고 알려진 복분자, 울금, 산수유 추출물과 알코올분해 효소 작용을 촉진시킬 수 있는 천연미네랄수(HSMW)를 첨가한 복합물(MIX)이 항산화 활성 및 알코올을 투여한 쥐의 혈중 알코올 농도와 체내 알코올 대사에 어떤 영향을 미치는지 검토하였다. 약용식물 추출물에서 복분자, 산수유의 총 페놀 및 플라보노이드 함량이 유의적으로 높게 정량되었으며, DPPH radical scavenging과 SOD 활성에서는 MIX가 유의적으로 높아 항산화 효과를 나타내었다. 혈중 알코올 농도를 측정한 결과에서 10% HSMW 투여군이 대조군의 농도보다 유의적인 차이를 나타내었다. MIX와 CP 투여군은 모든 시간 경과 후에 대조군의 농도보다 유의적인 차이를 나타내었으며 두 군 간의 유의적인 차이는 나타나지 않았지만 고농도가 저농도에 비해 5~10% 더 감소하였다. MIX와 CP 투여군의 간 균질액의 ADH 활성은 정상군에 비해 유의적인 차이를 볼 수 없었으며, ALDH 활성은 MIX와 CP 투여군이 유의적으로 높았다. 또한 혈청 중 간 기능 지표 효소인 GOT, GPT 활성은 MIX 투여군이 정상군과 비슷한 활성을 보여 간 손상이 적었음을 알 수 있었다. 따라서 MIX에 함유된 미네랄수(HSMW)인 Zn, Ca, Mg, Mn 등이 알코올 분해 효소 활성을 조절함과 동시에 약용식물 추출물이 항산화 작용을 하여 음주로 인해 발생하는 숙취 증상과 산화적 스트레스를 효과적으로 완화시킬 수 있으며, 체내 알코올 대사에 직 간접적으로 작용해 간 기능의 저하를 방지하는 기능성 숙취 음료로 사용될 것이라고 사료되어진다.
