모용(毛茸)이 사과나무 반점낙엽병(斑點落葉病) 발생(發生)과 병원균(病原菌)의 감염(感染)에 미치는 영향(影響) (Effect of Apple Leaf Hair Density on the Occurrence and Infection of Alternaria Leaf Spot Caused by Alternaria mali Roberts)

  • 사과품종별(品種別) 사과나무 반점낙엽병(斑點落葉病)의 발병(發病)과 잎의 모용(毛茸) 발생정도(發生程度)를 조사(調査)하고 사과나무 잎의 부위별(部位別) A. mali의 침입정도(浸入程度)와 모용(毛茸)이 병원균(病原菌)의 침입(浸入)과 감염(感染)에 미치는 영향(影響)을 조사(調査)한 결과(結果)는 다음과 같다. A. mali의 이병정도(罹病程度)에 따른 사과나무 품종(品種)의 분포(分布)는 이병엽율(罹病葉率)이 50.1% 이상(以上)되는 이병성품종(罹病性品種)을 조사(調査)한 82품종(品種) 중(中) 15개품종(個品種)(18.3%) 이었으며 주요(主要) 품종(品種)은 나리탄, 인도(印度), 왕령(王令), 스라크림숀, 스카이스파, 아오리 3호 등(等)이었고 이병엽율(罹病葉率)이 50% 이하(以下)되는 저항성품종(抵抗性品種)이 51개품종(個品種)(62.2%)이었는데 주요품종(主要品種)은 홍옥(紅玉), 무쓰, 모리쓰, 조나골드, 쓰가루, 가라, 골덴, 후지, 스파어리브레이즈 등(等)이었으며, 주요품종(主要品種)들의 엽당병반수(葉當病斑數)도 저항성품종(抵抗性品種)보다 이병성품종(罹病性品種)에서 많았다. 이병성품종군중(罹病性品種群中)에서 이병엽율(罹病葉率)이 6월(月)에 20%, 이상(以上)되어 8월(月)에 70% 이상(以上)되는 품종(品種)이 와인삽, 아오리 3호, 스카이스파, 왕령(王令) 등(等)과 6월(月)에는 5% 이상(以上) 되었다가 8월(月)에 70% 이상(以上)되는 품종(品種)이 라리단, 스타크림숀, 인도(印度) 등(等)으로 구분(區分)되었다. 품종별(品種別) 잎에 모용발생정도(毛茸發生程度)는 이병성(罹病性) 품종군(品種群)에서 보다 저항성품종군(抵抗性品種群)에서 대체(大體)로 많았고 동일(同一) 품종(品種)잎에서 모용무제거부위(毛茸無除去部位)보다 모용제거부위(毛茸除去部位)에서 감염율(感染率)이 높은 것으로 보아 잎의 모용(毛茸)은 품종(品種) 저항성(抵抗性) 요인(要因)으로 추정(推定)되었으며 A. mali의 침입(浸入)과 감염(感染)은 잎의 표면(表面)보다 이면(裏面)에서 용이(容易)하였다.

  • 간척지에서의 제염에 따른 식생의 변화와 수도의 근모형성에 대한 염해의 영향을 알기 위하여 강화도, 남양 및 계화도 간척지를 중심으로 1982년 여름에 본 조사를 하여 다음과 같은 결과를 얻었다. 1. 간척후, 저습지에서는 퉁퉁마디와 칠면초가 최초로 우점종을 형성하는 고등식물이었으며, 제염이 됨에 따라 버들명아주가 출현했고, 또 이것에 이어 매자기와 갈대가 우점종의 자리를 차지하게 되며 매자기와 갈대가 우점종이 되는 염도의 토양에서는 수도재배에 대한 염해의 영향은 문제화되지 않았다. 2. 습지에서는 퉁퉁마디와 칠면초에 이어 사철쑥과 갯개미취가 우점종을 형성하게 되고, 이때에는 단자엽 화본료 잡초들의 이입이 시작되었다. 3. 건조지에서는 나문재와 해홍나물이 최초의 우점 고등식물로 등장하게 되고, 또 갯능쟁이도 있었지만 군락을 형성하지는 않았다. 4. 건조지에의 식물이입은 습지를 중심으로 이미 이입된 식물들이 제염이 진행됨에 따라 서서히 일어나게 된다. 5. 토양염도의 근모형성에 대한 형향은 고염도토양에서 생육된 것일수록 저염도 토양에서 생육된 것에 비하여 근모형성율은 낮추고, 또 근모장도 짧게 하는 것으로 나타났다.

차가버섯 물 추출물의 cisplatin에 의해 유도된 HEI-OC1세포 손상에 대한 항산화효과 (Anti-oxidant Effects of the Water Extracts from the Inonotus Obliquus against Cisplatin- Induced Damage in HEI-OC1 Cells)

  • The medicinal mushroom Inonotus obliquus is a traditional and widely used multi-functional fungus. In this study, we have investigated whether Inonotus obliquus (Chaga mushroom) extracts exerts anti-oxidant effects on cisplatin-induced cytotoxicity in auditory cell line, HEI-OC1 cells. First of all, Chaga extracts has no harmful effects on viability of HEI-OC1 cells in the dose range of $65{\sim}125{\mu}g/m{\ell}$. Moreover, it shows cyto-protective effects on the cells treated with cisplatin-induced cytotoxicity in HEI-OC1 cells and the damage of hair cells arrays of the rat primary organ of Corti explants in the presence of cisplatin. Pretreatment with Chaga extracts inhibited the cell death, reactive oxygen species generation (ROS), lipid peroxidation induced by cisplatin. These effects were associated with the induction of antioxidant enzyme by Chaga extracts. We further investigated the effects of Chaga extracts on expression of antioxidant enzymes such as Cu, Zn superoxide dismutase (SOD 1) and Mn SOD (SOD 2) by RT-PCR. In addition, Chaga extracts shows SOD activity and SOD protein expression in cisplatin treated group induced similar to control group. Taken together, these results indicate that Chaga extracts can prevent cisplatin-induced cytotoxicity by radical-scavenging activity (SOD activity) in HEI-OC1 cells. It might be an effective as antioxidant and further studies on the chemo-preventive mechanisms of Inonotus obliquus are needed.

갈근으로부터 추출한 ${\beta}-sitosterol$이 HEI-OC1 세포의 항산화 효소 활성에 미치는 영향 (Effect of ${\beta}-sitosterol$ from Pueraria thunbergiana on the Antioxidant Enzyme Activities in HEI-OC1 Cells)

  • A mechanism of hair cell damage caused by noise and ototoxic agents is mediated through generation of free radicals and reactive oxygen species(ROS). It is known that most of animals have defense systems of ROS that protect against ROS, and the cochlea of animals also has ROS defense system, which appear efficient in detoxifying ROS generated under normal condition. This system includes several antioxidant enzymes such as superoxide dismutase(SOD), catalase(CAT), glutathione peroxidase (GPX), and glutathione reductase(GR). The radix of Pueraria thunbergiana(P. thunbergiana) is traditionally prescribed to attenuate the clinical manifestation of inner ear dysfunction and various clinical situations including fevers, gastrointestinal disorders, skin problems, migraine headaches, lowering cholesterol, and treating chronic alcoholism in Oriental Medicine. In the present study, to investigate the protection mechanism of ${\beta}-sitosterol$ from P. thunbergiana on cisplatin cytotoxicity toward HEI-OC1, we measured the effects of ${\beta}-sitosterol$ on activities of SOD, CAT, GPX, and GR in cisplatin treated cells. SOD, CAT, GPX, and GR activities were significantly increased in the presence of 0.001-0.1 ${\mu}g/ml$ of ${\beta}-sitosterol$ compared to the control group. These results indicate that ${\beta}-sitosterol$ protects cisplatin-induced HEI-OC1 cell damage through increasing the antioxidant enzyme system such as SOD, CAT, GPX, and GR.

시스플라틴 이독성에서 사물탕의 보호효과 (Protective Effect of Samul against Cisplatin in Primary Rat Organ of Corti Explant)

  • The water extracts of Samultang (Samul) has been used for treatment of ischemic heart and brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extract of Samul rescues cells from oxidative damages in cisplatin-induced ototoxicity. Cisplatin is a widely used chemotherapeutic agent that is also highly ototoxic. This study was designed to investigate the protective effects of Samul on ciplatin-induced ototoxicity in HEI-OC1 auditory cells and organ of Corti explant culture. Cisplatin markedly decreased the viability of HEI-OC1 auditory cells. However, treatment of HEI-OC1 cells with Samul significantly reduced cisplatin-induced cell death and apoptotic characteristics through reduction of intracellular peroxide generation. Cisplatin induced cytotoxicity in isolated and cultured hair cell progenitors from postnatal rat cochleae. These progenitor cells are isolated from the lesser epithelial ridge (LER, or outer spiral sulcus cell) area of pre-plated neonatal rat cochlear segments. However, Samul completely protected the morphological changes of organ of Corti and LER. Taken together, these data suggest that the protective effects of the water extracts of Samul against cisplatin may be mediated by the reduction of intracellular peroxide generation.

Over-expression of OsHsfA7 enhanced salt and drought tolerance in transgenic rice

  • Heat shock proteins play an important role in plant stress tolerance and are mainly regulated by heat shock transcription factors (Hsfs). In this study, we generated transgenic rice over-expressing OsHsfA7 and carried out morphological observation and stress tolerance assays. Transgenic plants exhibited less, shorter lateral roots and root hair. Under salt treatment, over-expressing OsHsfA7 rice showed alleviative appearance of damage symptoms and higher survival rate, leaf electrical conductivity and malondialdehyde content of transgenic plants were lower than those of wild type plants. Meanwhile, transgenic rice seedlings restored normal growth but wild type plants could not be rescued after drought and re-watering treatment. These findings indicate that over-expression of OsHsfA7 gene can increase tolerance to salt and drought stresses in rice seedlings.

  • The flowers of Nyctanthes arbor-tristis Linn. of Oleaceae widely used in Ayurvedic system of medicine for the treatment of diuresis, liver disorder, spleen enlargement sciatica, bitter, stomachic, carminative and tonic to hair. The aim of the present study was to evaluate the alcoholic and aqueous extracts of the flowers of Nyctanthes arbor-tristis for hepatoprotective effect against carbontetrachloride induced liver damage in rats. Administration of alcoholic and aqueous extracts of the leaves of Nyctanthes arbor-tristis protect the liver from toxic effects of carbontetrachloride by reducing the elevated levels of Serum glutamate pyruvate transaminase, Serum glutamate oxaloacetate transaminase, Alkaline phosphatase and serum bilirubin. Results revealed that both the alcoholic and aqueous extracts showed significant hepatoprotective activity by reducing the elevated levels of biochemical parameters at a dose of 200 mg/kg body weight. The results were supported by histopathological studies of liver samples which showed regeneration of hepatocytes by the extracts.

  • Intact mammalian hair and wool fibres are multi-compartmental composite materials consisting of a sulphur-rich outer protective cuticle layer surrounding elongated, highly keratinized, cortex cells. The cortex cells themselves are made up of crystalline, filamentous, low-sulphur a-helical keratin molecules embedded in a matrix of highly cross-linked, globular high-sulphur keratins. It is the structurally organised and highly disulphide cross-linked nature of these materials that provides them with their remarkable mechanical properties. However these mechanical properties are sensitive to environmental conditions such as water content, temperature and chemical treatment and the importance of their ultra-structural arrangements to overall mechanical properties in different environments is still not fully understood.(omitted)

Doxorubicin 에 의한 급성 심근독성과 L-Glutamate 의 효과에 관한 실험적 연구 (Experimental Study on the Effect of L-Glutamate to Acute Myocardial Toxicity by Doxorubicin)

  • The effect of Glutamate to myocardial toxicity induced by doxorubicin was studied with 20 male rats. 20 rats divided into 4 subgroups, 1st group was taken for normal control group without any treatment, 2nd group was injected with only doxorubicin, 3rd group was injected with L-glutamate and doxorubicin, and 4thd group was injected with only L-glutamate [all injections were done intraperitoneally]. Observations were made to each group on their gross findings, body weight, and electrocardiography, complete blood count and serum level of creatine phosphokinase. The results were as follows; l. In 1st group, we found no changes. 2. In 2nd group, there were many changes which were loss of body weight, dehydration, loss of body hair, diarrhea and death, in addition, elevation of CPK-MB isoenzyme and changes in EKG due to myocardial damage, leukopenia, thrombocytopenia were also found. 3. In 3rd group, there were more toxic effects containing 2 death cases, compared to 2nd group. 4. In 4th group, we found no specific changes except weight gain. These results suggest that L-glutamate which is intermediate of Krebs cycle may worsen the doxorubicin-induced myocardial toxicity.

Mercury poisoning in Eurasian river otter (Lutra lutra)

  • An adult male Eurasian river otter (Lutra lutra) with ataxia and lethargy was rescued. Through the necropsy of this otter with neurological symptoms, a broad range of vascular damage caused by mercury toxicity in several organs, hepatocellular necrosis, and vacuolation in the brain. In mercury examination, liver, kidney, and hair showed values of 0.878 ± 0.027, 1.807 ± 0.049, and 5.712 ± 0.102 ㎍/g, respectively. Compared with certified reference material, it was confirmed that the concentration of mercury were 6.7 times, 13.7 times, and 43.3 times higher, respectively. When the symptoms and diagnosis results were comprehensively reviewed, this otter's demise was revealed due to mercury poisoning. The mercury concentration in the liver does not exceed the lowest observed effect level of 3.4 ㎍/g. However, even at low concentrations, long-term accumulation can cause symptoms including neuropathy, and the possibility that these heavy metals have accumulated in other wild animals cannot be ruled out. It seems that continuous monitoring using sentinel animals is necessary.
