Microbiome of Haemaphysalis longicornis Tick in Korea

  • Ticks can transmit pathogenic bacteria, protozoa, and viruses to humans and animals. In this study, we investigated the microbiomes of Haemaphysalis longicornis according to sex and life stages. The Shannon index was significantly higher for nymphs than adult ticks. Principal coordinates analysis showed that the microbiome composition of female adult and male adult ticks were different. Notably, Coxiella-like bacterium (AB001519), known as a tick symbiont, was found in all nymphs and female adult ticks, but only one out of 4 male adult ticks had Coxiella-like bacterium (AB001519). In addition, Rickettsia rickettsii, Coxiella burnetii, and Anaplasma bovis were detected in this study.

Screening and Molecular Cloning of a Protective Antigen from the Midgut of Haemaphysalis longicornis

  • Vaccination is considered a promising alternative for controlling tick infestations. Haemaphysalis longicornis midgut proteins separated by SDS-PAGE and transferred to polyvinylidene difluoride (PVDF) membrane were screened for protective value against bites. The western blot demonstrated the immunogenicity of 92 kDa protein (P92). The analysis of the P92 amino acid sequence by LC-MS/MS indicated that it was a H. longicornis paramyosin (Hl-Pmy). The full lenghth cDNA of Hl-Pmy was obtained by rapid amplification of cDNA ends (RACE) which consisted of 2,783 bp with a 161 bp 3' untranslated region. Sequence alignment of tick paramyosin (Pmy) showed that Hl-Pmy shared a high level of conservation among ticks. Comparison with the protective epitope sequence of other invertebrate Pmy, it was calculated that the protective epitope of Hl-Pmy was a peptide (LEEAEGSSETVVEMNKKRDTE) named LEE, which was close to the N-terminal of Hl-Pmy protein. The secondary structure analysis suggested that LEE had non-helical segments within an ${\alpha}$-helical structure. These results provide the basis for developing a vaccine against biting H. longicornis ticks.

Acaricidal and Repellent Activities of Cnidium officinale-derived Compounds Against Haemaphysalis longicornis (Acari: Ixodidae) (작은소피참진드기에 대한 다양한 식물체 및 천궁유래물질의 살비 및 기피 효과)

  • This study investigated the acaricidal and repellent activities of various plant extracts and Cnidium officinale-derived materials against Haemaphysalis longicornis. Among the 24 methanolic plant extracts, C. officinale showed the highest acaricidal activity (93.3%) against H. longicornis. Platycodon grandiflorus, Phytolacca americana, Magnolia denudata, Houttuynia cordata, C. officinale, and Pueraria montana showed significant differences in their repellent activity. Consequently, the activity of solvent-fractionated materials and the derived material of the C. officinale, was investigated and a 90% acaricidal effect was observed three days after hexane fraction treatment, indicating that the repellent response was statistically significant. Butylidenephthalide, which is a C. officinale-derived compound, showed 90% acaricidal activity seven days after treatment. Additionally, all H. longicornis moved to the untreated area in the t-tube olfactometer experiment, confirming the repellent activity of butylidenephthalide. Therefore, the results of this study suggest the use of C. officinale crude extract and butylidenephthalide as potent acaricidals and repellents against H. longicornis, respectively.

  • Scanning electron microscope (Model; SEM ISI-DS-130) was used to observe the fine surface structures of the predominant cattle tick Haemaphysalis longicornis Neumann 1901 (Acarina: Ixodidae) in Korea and some SEM microphotographs were presented in this pictorial monograph. Brief descriptions of the main contents are consisted of followings; L Mean body size is 2.43mm in unfed males and 2. 58mm in unfed females, the eyes are absent. 2. The scutums are inornated but there are some short-setae on the surface of the body. 3. The festoons on the dorso-marginal part of the body are present and segmented with 11 sectors. 4. The cornua is well-developed and the bypostome dentition is usually 5/5 in both sexes but rarely 4/4 also seen. 5. There is no dorsal projection on the anteriointernal margin of the palpal article 2, howevever, there are well developed dorso-internal setae with setal fossa on it. 6. The joints of the legs (Coxatrochanter and trochanter coxa, etc) are well developed.

Distribution of ticks carrying Severe Fever with Thrombocytopenia Syndrome Virus (SFTSV) around Jiri walking trails of Jeollanam-do, Korea (지리산둘레길에서 채집한 참진드기의 분포와 참진드기에서의 중증열성혈소판감소증후군바이러스 검출)

  • Severe fever with thrombocytopenia syndrome (SFTS) is an emerging disease characterized by fever and thrombocytopenia. Haemaphysalis longicornis ticks comprise the major population of ticks in the environment and have been considered as the main vector for SFTS virus (SFTSV). Here we investigated the distribution of ticks carrying SFTSV collected from the environment using the dragging or sweeping methods during April~October 2015 in Jeollanam-do, Korea. Sampling was taken from Songjeong, Omi, Bangkwang, Sandong areas in Jiri walking trails. Among the total 3,869 ticks collected, 3,823 ticks (98.8%) were H. longicornis, 41 (1.1%) were Amblyomma testudinarium, and 5 (0.1%) were Ixodes nipponensis. Classification results by regional groups of H. longicornis indicated that 1,613 ticks were collected in Sandong, 1,190 ticks in Omi, 603 ticks in Bangkwang, and 417 ticks in Songjeong. In monthly distributional studies of H. longicornis based on the developmental stages, nymph (325 ticks) was collected from May to October, 94% of larvae from April to June, and 94% of adult from June to August. These results showed the different dominant stage of ticks according to seasons. However, no SFTSV-specific gene was detected in 3,823 ticks of H. longicornis, 41 of A. testudinarium and 5 of I. nipponensis.

  • 진드기는 생물학적(生物學的) 여러 분야(分野)의 연구대상(硏究對象)으로 또는 진드기 매개전염병(媒介傳染病)의 역학(疫學)과 방제(防除)를 위(爲)한 연구재료(硏究材料)로 공시(供試)되는데, 이를 위(爲)하여는 실험작군(實驗作群)에 의(依)한 집단생산(集團生産)이 요구(要求)된다. 그러나 이러한 실험작군(實驗作群)에 있어서 동일(同一)한 가토(家兎)를 실험용(實驗用) 숙주(宿主)로 반복(反復)하여 사용(使用)했을 경우(境遇), 진드기의 흡혈성취도(吸血成就度)가 저하(低下)되는 현상(現象)을 경험(經驗)하게 된다. 이러한 현상(現象)에 흥미(興味)를 가지고, 이를 실험용(實驗用) 가토(家兎)에 대(對)한 Haemaphysalis longicornis 미포혈(未飽血) 자충(雌蟲)의 반복흡혈(反復吸血)에 있어서의 흡혈성취도(吸血成就度) 변화(變化)를 조사(調査)해 보고자 몇가지 실험(實驗)이 설계(設計) 진행(遂行)된 바, 결과(結果)는 다음과 같이 요약(要約)된다. 1. 최초(最初)(1차(一次)) 흡혈시도(吸血試圖)에 있어서 공시(供試)된 진드기의 흡혈성취도(吸血成就度)는 실험용(實驗用) 숙주(宿主)인 가토(家兎)의 월령별(月齡別) 비교군간(比較群間)에 유의성(有意性) 있는 차이(差異)가 인정(認定)되지 아니 하였다. 2. 성공적(成功的) 흡혈율(吸血率)은 최초(最初)(1차(一次)) 흡혈시도(吸血試圖)에서 82.29%를 보였으나, 반복흡혈시도(反復吸血試圖) 차수(次數)가 증가(增加)함에 따라, 76.04%(2차(二次)), 53.13%(3차(三次)), 50%(4차(四次)), 46.87%(5차(五次))로 점차적(漸次的)으로 저하(低下)되는 성적(成績)을 보였다. 3. 평균흡혈기간(平均吸血期間)은 최초(最初)(1차(一次))흡혈시도(吸血試圖)에서 9.34일(日)을 보였으나, 반복흡혈시도(反復吸血試圖) 차수(次數)가 증가(增加)함에 따라, 9.74일(日)(2차(二次)), 10.31(日)(3차(三次)), 10.42(日)(4차(四次)), 10.47(日)(5차(五次))로 점차(漸次) 지연(遲延) 되었으며, 평균흡혈속도(平均吸血速度)는 0.1070에서 0.1027, 0.0970, 0.0960, 0.0955로 점차(漸次) 감속화(減速化) 되었다. 4. 평균(平均) 흡혈체중(吸血體重)에 있어서는 최초(最初)(1차(一次)) 흡혈시도(吸血試圖)에서 253.3mg의 성적(成績)을 보였으나, 흡혈시도(吸血試圖)가 반복(反復)됨에 따라, 242.58mg(2차(二次)), 190.73mg(3차(三次)), 187.85mg(4차(四次)), 184.18mg(5차(五次))으로 점차(漸次) 경량화(輕量化)되는 경향(傾向)을 나타냈다. 5. 결론적(結論的)으로 실험용(實驗用) 가토(家兎)에 대(對)한 Haemaphysalis longicornis 진드기의 흡혈성취도(吸血成就度)는, 반복흡혈시도(反復吸血試圖) 차수(次數)가 증가(增加)함에 따라, 숙주체내(宿主體內)에 형성(形成) 누적(累積)되는 진드기에 대(對)한 획득면역(獲得免疫)의 영향(影響)을 받아 점차(漸次) 저하(低下)되며, 따라서 H. longicornis 진드기의 집단생산(集團生産)을 위(爲)한 실험작군(實驗作群)이나 혈류보존(血統保存)을 위(爲)한 계대흡혈(繼代吸血)에 있어서 동일(同一) 가토(家兎)를 2회이상(回以上) 반복사용(反復使用)하는 것은 바람직한 일이 아닌 것으로 사료(思料)된다.

Identification of Tick Species Collected from Wild Boars and Habitats of Wild Boars and Domestic Pigs in the Republic of Korea

  • Tick is one of the most important arthropods in the transmission of vector-borne diseases. In this study, we investigated the abundance and species of ticks associated with swine and their habitats to assess the risk of spread of tick-borne diseases in host species, such as wild boars. Ticks were collected from 24 grazing or traditionally reared domestic pig farms and 8 habitats of wild boars in 8 provinces and 1 city in the Republic of Korea, by using the dragging and flagging methods. Ticks were also collected directly from 49 wild boars by using fine forceps. A total of 9,846 hard ticks were collected, including 4,977 Haemaphysalis longicornis, 4,313 Haemaphysalis flava, 508 Ixodes nipponensis, 1 Ixodes turdus, and 47 Amblyomma testudinarium. A total of 240 hard ticks were collected from 49 wild boars, including 109 H. flava, 84 H. longicornis, and 47 A. testudinarium. A total of 578 hard ticks were collected from areas around domestic pig farms. Only 2 hard tick species, 546 H. longicornis and 32 H. flava, were collected from these areas. A total of 9,028 hard ticks were collected from wild boars of 8 habitats, including 4,347 H. longicornis, 4,172 H. flava, 508 I. nipponensis, and 1 I. turdus. A. testudinarium was collected only from wild boars, and I. nipponensis and I. turdus were collected only from the habitats of wild boars.

Acaricidal and antimicrobial toxicities of Cyanachum paniculatum root oils and these components against Haemaphysalis longicornis and human intestinal bacteria (산해박 뿌리에서 추출한 정유 및 구성성분의 인간 장내미생물에 대한 항균활성 및 작은소피참진드기에 대한 살비활성)

  • Anaerobic growth-inhibiting and acaricidal activities of 2'-hydroxy-5'-methoxyacetophenone derived from Cyanachum paniculatum oil and its derivatives against five intestinal bacteria (Bifidobacterium bifidum, B. longum, Clostridium pefringens, Escherichia coli and Lactobacillus casei) and Haemaphysalis longicornis were examined. In the packet test against the larvae of H. longicornis, none of the C. paniculatum oil exhibited acaricidal activity, while the C. paniculatum oil showed only antimicrobial activity against five intestinal bacteria in the disc diffusion method. Based on the inhibition zones and MIC values, 2',4'-dimethoxyacetophenone, 2',5'-dimethoxyacetophenone, 2'-hydroxy-4'-methoxyacetophenone, 2'-hydroxy-5'-methoxyacetophenone, 2'-methoxyacetophenone, and 4'-methoxyacetophenone, containing a methyl group on the acetophenone skeleton, possessed growthinhibiting activities against C. perfringens and E. coli. However, acetophenone, 2'-hydroxyacetophenone, 4'-hydroxyacetophenone, 2',4'-hydroxyacetophenone and 2',5'-hydroxyacetophenone, which contained a hydroxyl group on the acetophenone skeleton, had no growth-inhibiting activity against intestinal bacteria. These results indicated that 2'-hydroxy-5'-methoxyacetophenone and its derivatives could potentially be developed as natural antimicrobial agents to specific control of C. perfringens and E. coli.

Distribution of Tick Vectors of Severe Fever with Thrombocytopenia Syndrome Virus (SFTSV) Collected from Four Environments in Jeju (제주지역 4개 환경에서 채집한 중증열성혈소판감소증후군 매개 참진드기 분포)

  • The distribution of ticks and severe fever with thrombocytopenia syndrome virus (SFTSV) pathogens were investigated by collecting ticks from March to November 2018 in four environments (grass fields, copses, mountain roads, and tombs) in Jeju. Three thousand and ninety ticks were collected using a tick trap, and 1,569 ticks were collected using the flagging method. Of the 4,659 ticks collected, Haemaphysalis longicornis and Haemaphysalis flava accounted for 4,440 ticks (95.2%) and 219 ticks (4.7%), respectively. Nine hundred and fifty, 883, 847, and 410 ticks were collected from grass fields, copses, mountain roads, and tombs, respectively, using tick traps, whereas 704, 472, 197, and 196 ticks were collected from copses, mountain roads, tombs, and grass fields, respectively, using the flagging method. The largest fraction of ticks (2,978) was collected from April to August, and most were collected in May and June. Adult ticks comprised 94 percent of the total ticks from June to August. SFTSV was not detected in the 4,440 H. longicornis ticks or the 219 H. flava ticks collected in this study.

  • Severe tick infestation was found in a hare in a suburban area of Nanchang, Jiangxi Province, China. We sampled ticks and identified them based on their morphologic characteristics. Three species, Ixodes sinensis, which is commonly found in China and can experimentally transmit Borreliia burgdorferi, Rhipicephalus haemaphysaloides, and Haemaphysalis longicornis which can transmit Lyme disease were detected with an optical microscope and a stereomicroscope. Risk of spreading ticks from suburban to urban areas exists due to human transportation and travel between the infested and non-infested areas around Nanchang.
