This study was performed to investigate the relationship between Heat shock protein 70 (HSP70) gene polymorphism and in vitro fertilization(IVF) embryo development in the pigs. The single strand conformation polymorphism(SSCP) genotypes from HSP70 K1, K3 and K4 PCR products were detected different patterns. In cleavage rate of oocyte fertilized in vitro, HSP70 K1-AA genotype($73.1\%$) and K1-AB genotype($62.3\%$) showed significantly higher oocyte cleavage rate than HSP70 K1-BB genotype($49.3\%$)(p<0.05). And HSP70 K3-AA genotype ($72.4\%$) and K3-AB($62.2\%$) also showed significantly higher oocyte cleavage rate than HSP70 K3-BB genotype($49.1\%$)(p<0.05). The IVF embryo development of 2-cell stage according to HSP70 genotypes of sperm and pig breeds also showed a significant difference. The number of embryos developed to 2-cell stage in Landrace(28.8) and Duroc(29.8) were significantly higher than in Yorkshire(10.9)(p<0.05). And also HSP70 K4-AB genotype group(29.6) higher than HSP70 K4-AA genotype group(10.6)(p<0.05). However, the number of embryos developed to blastocyst stage did not showed significant differences among breeds as well as HSP70 genotypes. These resrults suggest that in vitro development in porcine early embryos may be affected by HSP70 genotypes and breeds.

This study aimed to evaluate the effects of Astragali radix on brain edema of intracerebral hemorrhage(ICH)-induced rats. Brain edema following ICH was induced via the stereotaxic intrastriatal injection of bacterial collagenase type VII in Sprague-Dawley rats. Ethanol extract of Astragli radix was treated once a day for 3 days. Then brain hematoma volume and edema were examined. Immunohistochemistry was processed for iNOS, c-Fos, Bax, and HSP72 expressions in the brain sections and each immuno-labeling were calculated with image analysis. Ethanol extract of Astragli radix reduced hematoma volume(not significantly) and brain edema(significantly) ICH induced rats. Ethanol extract of Astragli radix reduced iNOS expressions, c-Fos, Bax and HSP72 positive cells significantly and reduced apoptotic bodies and swollen neurons in ICH induced rat brain. These results suggest that Astragli radix plays an inhibitory role in the hemorrhagic, inflammatory and apoptotic events induced by ICH. And it is supposed that neuroprotective effect of Astragli radix reveals by anti-apoptosis mechanism.

In Oriental medicine daeseungki-tang is one of the prescription that is used clinically for constipation of paralytics. The objective of the study was to observe the effect of daeseungki-tang on apoptotic neuronal cell death. In the present study, middle cerebral artery occlusion(MCAO) rats were treated with daeseungi-tang for 5 days and the edema percentage of cerebral hemisphere of MCAO rats were investigated primary. Secondary, appearances of Bax, Bcl-2,-factors that is related to apoptotic neuronal cell death - and HSP72 in the brain of MCAO rats were investigated via immunohistochemistry. Daeseungki-tang significantly decreased edema percentage of the cerebral hemisphere of MCAO rats. Daeseungki-tang significantly decreased Bax positive cells, but did not change the apperances of Bcl-2 positive cells in the penumbra of the cerebral cortex and the caudoputamen of MCAO rats. Daeseungki-tang significantly decreased HSP72 positive cells in the penumbra of the cerebral cortex, but not in the caudoputamen of MCAO rats. Based on the present results, it can be suggested that treatment with daeseungki-tang may decrease edema of the cerebral hemisphere and restrain apoptotic neuronal cell death in the penumbra of the cerebral cortex.

This study aimed to evaluate the effect of purgation therapy with Natrii sulfas, a therapy for stroke patients with constipation in the oriental medicine, on the ischemic brain damage of the rats. The ischemic brain damage was induced by the middle cerebral artery occlusion (MCAO), Natrii sulafas was administered once after the MCAO. After 48 hours, expressions of Bax, Bcl-2, c-Fos, and HSP72 on the brain tissues were observed by immunohistochemistrical methods or technique. Purgation therapy with Natrii sulfas attenuated the excess of Bax expression caused by the ischemic brain damage. It was significant statistically in the penumbra of cerebral cortex, but not in the caudate putamen, of the MCAO rats. Purgation therapy with Natrii sulfas did not attenuate the excess of Bcl-2 expression caused by the ischemic brain damage. Purgation therapy with Natrii sulfas did not attenuate the excess of c-Fos expression caused by the ischemic brain damage. Purgation therapy with Natrii sulfas attenuated the excess of HSP72 expression caused by the ischemic brain damage. It was significant statistically in the penumbra of cerebral cortex, but not in the caudate putamen, of the MCAO rats. These results suggest that purgation therapy with Natrii sulfas has a neuroprotective effect on the ischemic brain damage and an anti-apoptotic effect.

Objective : Daehwangmokdanpi-Tang (DWT) has been frequently used as a remedy for antiinflamation. To evaluate effect of acute pancreatitis by DWT, we examined the effects of DWT on the cholecystokinin-octapeptide (CCK)-induced acute pancreatitis (AP) in rats. Methods : Male Wistar rats weighing 200 to 250 g were divided into three groups. Normal untreated group, in treatment with DWT group; DWT was administered orally, followed by $75\;{\mu}g/kg$ CCK subcutaneously three times, after 1, 3 and 5 h. This whole procedure was repeated for 5 days. In treatment with saline group, the protocol was the same as in treatment group with DWT. Results : The author determined the pancreatic weight/body weight ratio, the levels of pancreatic HSP (heat shock protein)60 and HSP72 and the secretion of pro-inflammatory cytokines. Repeated CCK treatment resulted in the typical laboratory and morphological changes of experimentally induced pancreatitis. DWT was significantly decreased the pancreatic weight/body weight ratio in CCK-induced AP. Futhermore, The author demonstrated that DWT increased HSP60 and HSP72 compared with CCK-induced AP. Additionally, the secretion of $IL-1{\beta}\;and\;TNF-{\alpha}$ and the levels of amylase and lipase were lower than that saline. Conclusions : These results suggested that DWT may has a protective effect against CCK-induced AP.
Although reoxygenation is the best way to salvage hypoxic tissues, reduced oxygen species (ROS) generated during reoxygenation are blown to cause further tissue injuries and the induction of heat shock proteins (HSPs). The present study was undertaken to determine any causal relationship between the severity of hypoxia and the opposite outcomes, either beneficial or detrimental, of the subsequent reoxygenation by measuring the HSP72. To this aim, one group (6 male cats, $2.5{\sim}3.5\;kg$) was subjected to a 5-min episode of hypoventilation (H, ventilation rate: 5/min) for the induction of slight hypoxia and the other group (6 male cats, $2.4{\sim}3.7\;kg$) was subjected to a 5-min episode of apnea (A) for severe hypoxia. Each 3 animals from both groups received a 10-min episode of ventilation with $(95%\;O_2\;(0)$, whereas the remainder did not. After these procedures, all animals were allowed to be ventilated within physiological range for 1, 4, or 8 hours (1H, 1HO, 4H, 4HO, 8H, 8HO, 1A, 1AO, 4A, 4AO, 8A and 8AO groups). Control animals did not receive any manipulation. The arterial blood $pCO_2$ was significantly higher just after apnea than hypoventilation, while $pCO_2$ and pH were significantly lower just after apnea than hypoventilation. Western blot analysis revealed that the magnitude of HSP72 synthesis is larger in 1H, 4H and 8H groups than in 1HO, 4H and 8HO groups, respectively. In contrast, 1AO, 4AO and 8AO groups more induced HSP72 than 1A, 4A and 8A groups, respectively. These results suggest that the reoxygenation is beneficial after slight hypoxia but detrimental after severe hypoxia.
Purpose: Henoch-$Sch{\ddot{o}}nlein$ purpura (HSP) is a small-vessel vasculitic disease that most often affects the skin. Abdominal symptoms precede the typical purpuric rash of HSP in 14~36%. It is a challenge to diagnose HSP in the absence of a rash, because there are no biologic tests that can identify HSP with certainty, so we tried to find out the characteristic features of HSP gastroenteropathy without purpura before diagnosis. Methods: This study included 82 children with HSP who had been admitted or visited outward of the Department of Pediatrics, Pusan National University Hospital from 1995 to 2000. The cases that the onset of purpura preceded or coincided that of abdominal pain were defined as purpura-positive group. The cases that the onset of abdominal pain preceded purpura more than 1 week and purpura was not presented till diagnosed as HSP gastroenteropathy were defined as purpura-negative group. We compared and analyzed the clinical features of the two groups by reviewing the medical records retrospectively. To ensure the diagnosis of HSP gastroenteropathy, we conducted upper GI series, abdominal ultrasonogram, abdominal CT, endoscopy and/or skin biopsy. Results: The number of cases of purpura-positive group and purpura-negative group were 72 and 10, respectively. There is no difference between two groups in the incidence of clinical symptoms and laboratory findings. Children with HSP gastroenteropathy had characteristic erosive or ulcerative lesions in the stomach or duodenum on esophagogastroduodenoscopy, or mural thickening of the small bowel on abdominal ultrasonogram, CT or upper GI series. Skin biopsy revealed leukocytoclastic vasculitis in 3 of them, although biopsy specimen was taken from any areas of normal- appearing skin. In purpura-negative group, 9 patients improved by steroid therapy. Conclusion: In purpura-negative group, there is no diagnostic feature on the laboratory findings and clinical features. Therefore, to diagnose HSP gastroenteropathy in patients with abdominal pain in the absence of the characteristic rash, careful observation of clinical features and laboratory data, and prompt application of available diagnostic tools such as gastrointestinal endoscopy, radiologic study and skin biopsy are recommended. Early use of corticosteroid may reduce the suffering in these patients.

These results suggest that PS may has a inhibitory effect against CCK-induced AP.

Objectives : The aim of this study was to investigate the hepatoprotective effect of Soshiho-tang (SSH) in mouse primary liver cells against hydrogen peroxide ($H_2O_2$)-induced oxidative stress. We also elucidated the molecular mechanism of hepatoprotective effect by SSH. Methods : Cell viability, level of ALT, AST and LDH, intracellular ROS level, mRNA expression and activity of antioxidant enzymes were used to evaluate hepatoprotection of SSH against $H_2O_2$. Target gene expressions were analyzed by real-time PCR. Results : Pre-treatment with SSH for 1 hour prevented cytotoxicity against $H_2O_2$. $H_2O_2$-induced ROS level decreased under SSH pre-treatment. mRNA expression of GPx and SOD increased in SSH-treated cells. In addition, HSP72 and HSP40 gene expression were elevated under SSH-treatment. Conclusions : These results indicate that SSH protects mouse primary liver cells from $H_2O_2$-induced oxidative injury. This hepatoprotective activity of SSH is mediated by decreasing intracellular ROS and increasing antioxidant enzyme expression (GPx and SOD) and stress response protein (HSP72 and HSP40).
