Comparative Properties of Red Ginseng Prepared with Different Cultivation Years of Fresh Ginseng Produced in Punggi Region (풍기지역 연근별 수삼의 홍삼가공 특성 비교)

  • Quality properties of red ginseng prepared with different cultivation years of fresh ginseng produced in Punggi region were investigated. Fresh ginseng cultivated for 4, 5, or 6 years was steamed for 3.5 hr and dried for 24 hr at $60{\sim}65^{\circ}C$ and subsequently for $3{\sim}4$ days at $40^{\circ}C$ under commercial conditions. Compared to 6 years-old roots, the five years-old roots showed similar or some lower quality properties in terms of color, appearance, diameter, and inside quality, but higher ones in length and yield of prepared red ginseng. In particular, the levels of ginsenoside $Rg_3$ and $Rh_2$, which are known as specific components in red ginseng, were the highest in 5 years-old roots. The result shows that fresh ginseng of 5 years-old roots produced in Punggi region can be utilized as a raw material for the manufacture of high-quality red ginseng.

USLE/RUSLE Factors for National Scale Soil Loss Estimation Based on the Digital Detailed Soil Map (수치 정밀토양에 기초한 전국 토양유실량의 평가를 위한 USLE/RUSLE 인자의 산정)

  • Factors of universal soil loss equation, USLE, and its revised version, RUSLE for Korean soils were reevaluated to estimate the national scale of soil loss based on digital soil maps. Rainfall erosivity factor, R, of 158 locations of cities and counties were spacially interpolated by the inverse distance weight method. Soil erodibility factor, K, of 1321 soil phases of 390 soil series were calculated using the data of soil survey and agri-environmental quality monitoring. Topographic factor, LS, was estimated using soil map of 1:25,000 scale with soil phase and land use type. Cover management factor, C, of major crops and support practice factor, P, were summarized by analyzing the data of lysimeter and field experiments for 27 years (1975-2001) in the National Institute of Agricultural Science and Technology. R factor varied between 2322 and 6408 MJ mm $ha^{-1}$ $yr^{-1}$ $hr^{-1}$ and the average value was 4276 MJ mm $ha^{-1}$ $yr^{-1}$ $hr^{-1}$. The average K value was evaluated as 0.027 MT hr $MJ^{-1}$ $mm^{-1}$. The highest K factor was found in paddy rice fields, 0.034 MT hr $MJ^{-1}$ $mm^{-1}$, and K factors in upland fields, grassland, and forest were 0.026, 0.019, and 0.020 MT hr $MJ^{-1}$ $mm^{-1}$, respectively. C factors of upland crops ranged from 0.06 to 0.45 and that of grassland was 0.003. P factor varied between 0.01 and 0.85.

Prognostic Value of Vascular Endothelial Growth Factor Expression in Patients with Prostate Cancer: a Systematic Review with Meta-analysis

  • Background: The vascular endothelial growth factor (VEGF) mediates vasculogenesis and angiogenesis through promoting endothelial cell growth, migration and mitosis, and has involvement in cancer pathogenesis, progression and metastasis. However, the prognostic value of VEGF in patients with prostate cancer remains controversial. Objectives: The aim of our study was to evaluate the prognostic value of VEGF in prostate cancer, and summarise the results of related research on VEGF. Methods: In accordance with an established search strategy, 11 studies with 1,529 patients were included in our meta-analysis. The correlation of VEGF-expression with overall survival and progression-free survival was evaluated by hazard ratio, either given or calculated. Results: The studies were categorized by introduction of the author, demographic data in each study, prostate cancer-relatived information, VEGF cut-off value, VEGF subtype, methods of hazard ratio (HR) estimation and its 95% confidence interval (CI). High VEGF-expression in prostate cancer is a poor prognostic factor with statistical significance for OS (HR=2.32, 95%CI: 1.40-3.24). However, high VEGF-expression showed no effect on poor PFS (HR=1.30, 95%CI: 0.88-1.72). Using Begg's, Egger's test and funnel plots, we confirmed lack of publication bias in our analysis. Conclusion: VEGF might be regarded as a prognostic maker for prostate cancer, as supported by our meta-analysis. To achieve a more definitive conclusion enabling the clinical use of VEGF in prostate cancer, we need more high-quality interventional original studies following agreed research approaches or standards.

  • We suggested the new method of thermal screen in Bridgman-Stockbarger method to control the polygrain, the internal cavities and solid-liquid (SL) interface. $CaF_2$ single crystal of 6 inch was grown perfectly when we adopted to use a graphite pipe and a ceramic warmer in the conditions of growth rate 2 mm/hr, vertical temperature of $14^{\circ}C$ for freezing and temperature of $1324^{\circ}C$ at conical tip of crucible. The light scattering phenomena occurred by internal cavities were controlled as decreasing the freezing rate to 2 mm/hr and/or as adopting the rotation of melt (7 rpm).

Changes in Ginsenosides Composition of Ginseng Leaf and Stem after an Ultrasonication Process (초음파 처리에 의한 인삼 잎과 줄기의 진세노사이드 성분 변화)

  • The purpose of this study is to develop a new preparation process of ginseng leaf and stem extracts having high concentrations of ginsenoside Rg2, Rg3, Rg5, Rh1, a special component of red and black ginseng. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of ginseng (Panax ginseng) leaf and stem were processed under several treatment conditions including ultrasonication treatments. The content of total saponin reached their heights at 17 hr (UGL-17) of ultrasonication treatment, followed by 16 hr (UGL-16) and 7 hr (UGL-7) of ultrasonication treatment at $100^{\circ}C$. UGL-17 findings show that the ginseng leaf and stem that had been processed with ultrasonication for 17 hours peaked in the level of Rg2, Rg3 and Rh1. In addition, UGL-16 contained ginsenoside Rg5 at high concentrations. It is thought that such results provide basic information in preparing ginseng leaf and stem extracts with functionality enhanced.

Effect of Thawing Temperature on the Properties of the Danish Pastry Made from Frozen Dough (데니쉬 페이스트리 냉동생지의 해동온도가 품질특성에 미치는 영향)

  • Frozen doughs for danish pastries were made by straight dough method to study the effect of thawing temperature on the quality of pastries. Danish pastry doughs were immediately frozen at -40 $^{\circ}C$ quickly after make-up and stored for 6 weeks at -20$^{\circ}C$ and baked after thawing and fermentation a week. Thawing temperature and fermentation time were controled to 3 hr at 5$^{\circ}C$, 2 hr at 10$^{\circ}C$, and 1.5 hr at 20$^{\circ}C$, respectively. The number of survival yeast cells in dough and the volume, the moisture content, the texture of danish pastries were investigated. Survival yeast cells showed the highest number at thawing temperature of 5 $^{\circ}C$. The volume of danish pastries decreased significantly according to the increase of frozen storage time and danish pastries thawed at 5$^{\circ}C$ revealed the largest volume. In moisture contents of danish pastry, there was no significantly difference without regard to the thawing temperature but showed the highest moisture content at thawing temperature of 20$^{\circ}C$. Danish pastries thawed at 5$^{\circ}C$ accomplished the best texture in the resulting pastry.

Determination of Bumetanide in Human Plasma by a Validated HPLC Method and Its Application to Single-dose Pharmacokinetics (사람 혈장 중 부메타니드의 HPLC 분석법 검증 및 단일 용량 투여에 의한 약물동태 연구)

  • A high-performance liquid chromatographic method was employed for the determination of bumetanide in human plasma. After addition of internal standard (IS, naproxen) and acidification of the plasma with 1 M hydrochloric acid, the drug and IS were extracted into dichloromethane. The organic phase was back-extracted into 1 M sodium bicarbonate solution and 50 ${\mu}l$ of the aqueous phase was injected onto a reversed-phase C18 column with a mobile phase consisting of methanol: water: glacial acetic acid = 65 : 35 : 1. The samples were detected utilizing a fluorescence detector (excitation wavelength 235 nm, emission wavelength 405 nm). The method was specific and validated with a lower limit of 5 ng/mL. Intra- and inter-day precision and accuracy were acceptable for all quality control samples including the lower limit of quantification. The applicability of the method was demonstrated by analysis of plasma after oral administration of a single 2 mg dose to 24 healthy subjects. From the plasma bumetanide concentration vs. time curves, the mean AUC was $246.5{\pm}73.8\;ng{\cdot}hr/mL$ and $C_{max}$ of $132.1{\pm}40.9$ ng/mL reached 1.2 hr after administration. The mean biological half-life of burnet ani de was $1.1{\pm}0.2$hr. Based on the results, this simple and validated assay method could readily be used in any pharmacokinetic or bioequivalence studies using humans.

Validation of a Simple HPLC Method for Determination of Nicardipine in Human Plasma and Its Application to Single-dose Pharmacokinetics (인체 혈장 중 니카르디핀의 정량을 위한 HPLC 분석법 검증 및 단일 용량 투여에 의한 약물동태 연구)

  • A simple HPLC method with ultraviolet detection of nicardipine in human plasma was developed and validated. After drug extraction with solid phase extraction (SPE) method, chromatographic separation of nicardipine in plasma was achieved at $30^{\circ}C$ with a $C_{18}$ column and acetonitrile-0.02% phosphate buffer mixture (with 0.02% triethylamine, final pH 7.0), as mobile phase. Quantitative determination was performed by ultraviolet detection at 254 nm. The method was specific and validated with a limit of quantification of 5 ng/mL. The intra- and inter-day precision and accuracy were acceptable for all quality control samples including the lower limit of quantification. The applicability of the method was demonstrated by analysis of plasma after oral administration of a single 40 mg dose to 8 healthy subjects. From the plasma nicardipine concentration versus time curves, the mean $AUC_{t}$, was $134.04{\pm}59.72\;ng\;hr/mL$ and $C_{max}$ of $108.65{\pm}69.17\;ng/mL$ reached 1.5 hr after administration. The mean biological half-life of nicardipine was $3.93{\pm}0.82\;hr$. Based on the results, this simple and validated assay method could readily be used in any pharmacokinetic or bioequivalence studies using human.

The Risk of Breast Cancer in Women in Their 40s by Economic Activity (경제활동에 따른 40대 여성의 유방암 발생 위험도)

  • In South Korea, female individuals in their forties show a high rate of incidence, with approximately 13% of the patients being <40 years. This statistic is more than twice as high as that in Western countries. It is therefore necessary to identify the risk factors for breast cancer incidence by age and economic activity participation status. Women aged 30 to 59-whether breast cancer patients or those in the control group and having no breast cancer-were appraised from the sample cohort database. The data were analyzed using the statistical software R36.2. To identify the factors affecting breast cancer incidence, the degree of association was determined with HR and 95% CI by means of cox regression analysis. As for the socio-demographic variables, the older the individual, the higher the risk of breast cancer incidence becomes. As for the economic activity variables, those who were dependents (unemployed) and who had higher income (medium and high) were at higher risk of breast cancer incidence, which was statistically significant. The income-adjusted HR (model 1) for breast cancer development associated with the economic activity was 1.452 (95% CI, 1.19-1.77). The body mass index and alcohol intake-adjusted HR (model 2) was 1.431 (95% CI, 1.18-1.74). One needs to pay attention to policy plans regarding women's quality of life, as well as to the risk of breast cancer incidence by their economic activity. In other words, policies need to give post care, instead of focus on early detection and cancer treatment.

Rechlorination for residual chlorine concentration equalization in distribution system (급배수시스템에서 잔류염소 농도 균등화를 위한 재염소 처리)

  • Three water treatment plants(WTPs) in Jeju island whose source water have different characteristics from those of the mainland of Korea were investigated. Coefficients of bulk water decay($k_b$) of free chlorine at $5^{\circ}C$ for ES, GJ, NW WTPs were $-0.003hr^{-1}$, $-0.002hr^{-1}$ and $-0.001hr^{-1}$ respectively based on bottle tests. To simulate the free chlorine variations in the distribution system using EPANET, ES WTP was chosen. Free chlorine concentrations of several sites were less than the drinking water quality standards(i.e., 0.1 mg/L); E5(0.03 mg/L), E6(0.02 mg/L), W21(0.02 mg/L) and W25(0.03 mg/L). To maintain more than 0.1 mg/L of free chlorine in the distribution system, at least 1.9 mg/L of chlorine was needed at the WTP, which suggested rechlorination was needed to supply palatable tap water to customers. Two sites, one that diverged into E5 and E6 in the east-line and another located before E21 in the west-line were selected for the appropriate rechlorination locations. The recommended rechlorination dosages were 0.42 mg/L for the east and 0.27 mg/L for the west. The simulated results indicated that the free chlorine could be reduced to 0.4 mg/L at the WTP with rechlorination, and taps with excessive free chlorine could be more stabilized(i.e., 0.1~0.4 mg/L).
