A Study on the Plate for Deep Discharge in Lead Acid Battery (납축전지의 심방전용 극판에 관한 연구)

  • Positive plate was composed of lead hydroxide via reaction between lead oxide and $H_2O$ and lead sulfate was formed of the reaction of lead hydroxide with sulfuric acid. And its density is $3.8g/cm^3$, $4.0g/cm^3$, $4.2g/cm^3$ and $4.4g/cm^3$ by controlling volume of refined water. Curing of positive plate is done for low ($45^{\circ}C$, 40hr, over 95% of relative humidity) & high ($80^{\circ}C$, 40hr, over 95% of relative humidity) temperature, which created 3BS & 4BS active materials. Experimental result of DOD with 100% life cycle test shows that it was not related to the density of active materials but to the low & high temperature aging of active materials. The test makes us to understand that the crystallization which is made by curing of active materials is a more of a main factor than density of active materials under the deep cycle using circumstances. The active materials which were from the high temperature curing are better for deep cycle performance.

Development and Application of a Novel Mammalian Cell Culture System for the Biocompatibility and Toxicity of Polymer Films and Metal Plate Biomaterials (고분자필름과 금속막 의료소재에 대한 생체적합성 및 독성 평가를 위한 새로운 세포배양시스템의 개발 및 적용)

  • Biomaterials including polymer, metal, ceramic, and composite have been widely applied for medical uses as medical fibers, artificial blood vessels, artificial joints, implants, soft tissue, and plastic surgery materials owing to their physicochemical properties. However, the biocompatibility and toxicity for film- and plate-form biomaterials is difficult to measure in mammalian cells because there is no appropriate incubation system. To solve these problems, we developed a novel mammalian cell culture system consisting of a silicone ring, top panel, and bottom panel and we applied two polymer films (PF) and one metal plate (MP). This system was based on the principal of sandwiching a test sample between the top panel and the bottom panel. Following the assembly of the culture system, SK-MEL-2 cells were seeded onto Styela Clava Tunic (SCT)-PF, NaHCO3-added SCT (SCTN)-PF, and magnesium MP (MMP) and incubated at 37℃ for 24 hr and 48 hr. An MTT assay revealed that cell viability was maintained at a normal level in the SCT-PF culture group at 24 or 48 hr, although it rapidly decreased in the SCTN-PF culture group at 48 hr. Furthermore, the cell viability in the MMP culture group was very similar to that of the control group after incubation for 24 hr and 48 hr. Together, these results suggest the sandwich-type mammalian culture system developed here has the potential for the evaluation of the biocompatibility and toxicity of cells against PF- and MP-form biomaterials.

  • The formation and transition of 2212 and 2223 phase have been studied in BPSCCO system. The 2212 phase formed in early sintering state reacts on $Ca_2PbO_4,\;Ca_2CuO_3$ and CuO during sintering process and thus produces the 2223 phase. A long sintering period is need to fabricate the superconductor with large volume fraction of 2223 phase. Also, the thin plate-like grains composed of Bi, Sr, Ca and Cu contribute to 2223 phase formation. Though the sample has lower volume fraction of 2223 phase, the critical temperature is measured highly in case of the grain grown to plate-like shape. In this work, the critical temperature of the sample sintered for 264 hr in air was measured 108 K. Microstructure of the sample was varied with condition of heat treatment after sintering process and the sample annealed with $500^{\circ}C$ for 5 hr showed excellent charateristics of 2223 phase formation.

Expression of Wee1 Gene in the Ascidian, Halocynthia roretzi Embryo

  • It is suggested that FGF/Ras/MEK/Erk signaling plays crucial roles in specification and cell division of the mesodermal precursor cells in ascidian embryos. To investigate how the number of cell division in tissue precursor cells is determined, we have characterized Wee1 homolog, Hr-Wee1 of the ascidian Halocynthia roretzi. We found that the Hr-Wee1 mRNA is expressed both maternally and zygotically. Maternal transcript is localized to the cytoplasm in the animal cells, while zygotic expression is seen in cells of the endoderm lineage from 32-cell to 110-cell stages. Zygotic in situ signal is detected in the A-line neural plate cells of neurulae, and in epidermal cells of the head region of tailbud embryos. Embryos treated with MEK signaling inhibitor showed a similar pattern to normal embryos in expression of Hr-Wee1. Therefore, it is likely that MEK signaling does not affect the maternal and zygotic expression of Hr-Wee1.

  • Solar cell and nanofiltration membrane were utilized in a system of enzymatic hydrogen production through light-sensitized photoanode, which resembles photoelectrochemical(PEC) configuration. Solar cell uses no additional light energy to increase energy for electrons to reduce protons and for holes to oxidize water to oxygen, and nanofiltration membrane replaces a salt bridge successfully with increased ion transport capability. With this system configuration, optimized amount of enzyme(10.98 unit), and an anodized tubular $TiO_2$ electrode($5^{\circ}C$/1 hr in 0.5 wt% HF-$650^{\circ}C$/5 hr) hydrogen evolved at a rate of ca. $43\;{\mu}mol/(cm^2{\times}hr)$ in a cathodic compartment and oxygen generated at a rate of ca. $20\;{\mu}mol/(cm^2{\times}hr)$ in an anodic compartment. The stoichiometric evolution of gases indicated that water was splitted in the system.

  • In the tungsten industry, molybdenum wire which used as the center supporter for coil shape tungsten wire was removed. Nitric acid dissolution method which used prevalently up to the present, takes nitric acid as major component and use noxious material such as sulfuric acid and hydrochloric acid remove molybdenum wire which used as center supporter by dissolve selectively within the range of no damage on tungsten wire. Mixed acid waste water occurred to the process were difficult to be decomposed by the conventional methords. This mixed acid waste water was treated by ozone, and It was obtained using possible by-product through the treatment waste water. For the three reactors with the same volume ; Blank reactor, Disturbance plate reactor, Packed-bed reactor ; the results were as follows : For the blank reactor COD removal efficiency in the pH = 4 (HRT : 6hr) was 28.5%, COD removal efficiency in the pH = 7 (HRT : 6hr) was 28.6%, and COD removal efficiency in the pH = 10 (HRT : 6hr) was 27.8%. For the disturbance plate reactor COD removal efficiency in the pH = 4 (HRT : 6Min.) was 86.5%, COD removal efficiency in the pH =7 (HRT : 6Min.) was 84.4%, and COD removal efficiency in the pH = 10 (HRT : 60Min.) was 86.8%. For the packed-bed reactor COD removal efficiency in the pH = 4 (HRT : 40Min.) was 76.0%, COD removal efficiency in the pH = 7 (HRT : 40Min.) was 81.3%, and COD removal efficiency in the pH = 10 (HRT : 40Min.) was 84.6%. After O3 treatment using possible by-product(Na2SO4) was 150g/ℓ.

THE EFFECT OF KOREAN RED GINSENG SAPONIN ON THE ALKALINE PHOSPHATASE ACTIVITY OF RAT OSTEOBLASTIC CELL(ROS17/2.8) IN CULTURE (한국 홍삼사포닌이 배양중인 쥐 조골세포의 염기성 인산분해효소 활성도에 미치는 영향)

  • Using the Korean red ginseng saponin, which is known to world-wide and thd effects of it have been investigated by many reserachers for years. Ginseng saponin, one of the major components of Korea ginseng root, has many various biologic effects, such as cytotoxic effect, tumorcidal activity, protein biosynthesis and membrane modifying effect. The purpose of this study was to evaluate effects of ginseng saponin on the alkaline phosphatase activity of ROS cells in culture. After ROS cells were seeded into a 96-well plate, 96-well plate cultured until confluence was obtained. To evaluate cytotoxic effect of total saponin in cultured ROS cells, the plates were added to each total saponin concentration (0-1mg/ml). After 48hr., cells were counted by stain with 0.2% trypan blue at randomly selected field microscopically. Also, to evaluate alkaline phosphatase(ALP) activity of total saponin in cultured ROS cell, the plate was added to each total saponin concentration (0-1mg/ml) and ALP activity was assayed. To evaluate time-course of ALP activity, $31.25{\mu}g/ml$ of saponin added to 96-well plate. After culture of 6, 12, 24 and 48hr., ALP activity test was performed. To evaluate effect of cycloheximide in ALP activity, 96-well plate was added to saponin and cycloheximide. In control group, the plate was added saponin only. The results were as follows. 1. After the various concentration of total saponin was added in the medium, 500 and $1000{\mu}g/ml$ of total saponin showed cytotoxic effect of ROS(P<0.005). 2. In contrast to control group, 7.6, 15.6, 31.25, 62.5 and $250{\mu}g/ml$ of total saponin increased ALP activity significantly. 3. Otherwise, 500 and $1000{\mu}g/ml$ of total saponin decreased ALP activity significantly(P<0.005). 4. As the time span increases, $31.25{\mu}g/ml$ of total saponin increased ALP activity. 5. Cycloheximide decreased saponin-indueced ALP actitity in ROS(P<0.005). These results suggest that Ginseng total saponin stimulates the ALP activity of rat osteoblastic cells.

Investigation of the Three-Dimensional Turbulent Flow Fields of the Gas Swirl Burner with a Cone Type Baffle Plate(II)

  • This paper presents three-dimensional mean velocities, turbulent intensities and Reynolds shear stresses measured in the Y-Z plane of the gas swirl burner with a cone type baffle plate by using an X-type hot-wire probe. This experiments is carried out at the flow rate of 450ℓ/min which is equivalent to the combustion air flow rate necessary to heat release 15,000 kcal/hr in a gas furnace. Mean velocities and turbulent intensities etc. show that their maximum values exist around the narrow slits situated radially on the edge of and in front of a burner. According to downstream regions, they have a peculiar shape like a starfish because the flows going out of the narrow slits and the swirl vanes of an inclined baffle plate diffuse and develop into inward and outward of a burner. The rotational flow due to the inclined flow velocity going out of swirl vanes of a cone type baffle plate seems to decrease the magnitudes of mean velocities V and W respectively by about 30% smaller than those of mean velocity U. The turbulent intensities have large values of 50%∼210% within the range of 0.5

  • Fluosilicate salts based hydration heat reducer(SWP-HR), used in this study, is composed of fluosilicate salts, soluble silica, aromatic polymer condensate and nitrate salt based inorganic compound with latent heat property. Effects of SWP-HR addition on the hydration heat and anti-crack property of cement mortar were investigated. Adiabatic hydration temperature and drying shrinkage length of SWP-HR added cement mortar had a tendency to decrease compared to those of cement mortar without SWP-HR addition. Also, it was confirmed through crack pattern experiment of plate-form specimen for elucidating crack-reducing characteristic that anti-crack property of SWP-HR added cement mortar was improved.

Development and Application Performance of a Granular Herbicide Applicator Attached to Rice Transplanter (승용 이앙기 부착형 제초입제 살포기 개발과 살포성능)

  • A herbicide applicator attached to a rice transplanter was developed to perform herbicide application and transplanting simultaneously. The prototype machine was composed of herbicide tank, discharge device, spinner, scattering plate and power drive. The application rate of the machine could be adjusted from 10 to 30 kg/㏊ and the application swath was 1.8m. According to the field test, application uniformity showed the range of 13.6~43.9% in terms of CV(coefficient of variation) depending on the spinner speed, application height and shape of diffuser. The best uniformity could be achieved with the spinner speed of 30.8m/s and application height of 20cm. Field efficiency of the prototype was 4.7hr/㏊. Transplanting machine has field efficiency of 4.6hr/㏊ in Korea. By attaching the herbicide applicator, field efficiency became lowed only by 0.1hr/㏊, which was counted for filling time of herbicide. Simultaneous operation of transplanting and herbicide application had a of labor saying. The weed control efficacy was measured to be 96% in field when AC140 + Stomp and Londax + YRC was applied at the 65th day after transplanting. Nonanmae was treated by manual application to be compared to.
