동아프리카 자원식물을 활용하여 개발된 지속가능한 영양보충식품의 영양성분 분석 (Nutrient Analyses of Sustainable Ready-to-Use-Supplemental Food (RUSF) developed with East African Ethnic Plant Resources)

  • The objectives of the study were to analyze the various nutrient contents of ready-to-use-supplemental-food (RUSF) developed with Eastern African ethnic plants and compare them with commercially available supplemental food (CASF) for infants. RUSF was made with three African plants, including Ficus sur, Amaranthus caudatus, and Prosopis juliflorafa and locally available food ingredients in DR Congo and Ethiopia and was cooked at $180^{\circ}C$ for 20 min in a conventional oven. Macronutrients were analyzed by the AOAC method. Contents of 17 amino acids were analysed with amino acid analyzer, and vitamin $B_1%$ niacin, vitamin $B_6$, and vitamin C were analyzed with HPLC. Isomers of vitamin E were analyzed by GC-FID. The price of RUSF was 0.133 USD per 100g. Contents of macronutrients were not significantly different between developed RUSF and CASF. Results of amino acid analysis showed that most amino acid contents were similar or higher in RUSF compared with CASF. However, glutamic acid and lysine contents were less in RUSF than in CASF. Vitamin E and vitamin $B_1$ contents were more than five times higher in RUSF (22.40, 6.26 mg/100 g, respectively) than in CASF (4.00, 0.80 mg/100 g, respectively). Niacin was about three times higher while vitamin $B_6$ was almost 20 times higher in RUSF than in CASF. However, ascorbic acid was significantly lower in RUSF (3.43 mg/100 g) than in CASF (42.08 mg/100 g). Our study found that the developed localized RUSF showed higher nutrient contents in most micronutrients analyzed except for ascorbic acid. Out of 17 amino acids, glutamic acid and lysine contents in RUSF were less than in CASF. Therefore, further study is needed in order to improve these nutrients contents.

Analysis of the Phenolic Content and Antioxidant Activities of Soybean Extracts from Different Regions and Cultivars

  • Phenolic compounds such as isoflavones contribute to the antioxidant properties of soybean. This study investigated differences in the phenolic content and antioxidant activity of ten soybean cultivars grown in two different locations in Korea. We observed a wide range of variation in total isoflavone and phenolic content by location and cultivar. The total isoflavone content of cultivars grown at Pyeongchang (sub-highland) was significantly higher than that in cultivars grown at Gangneung (lowland). In contrast, the total phenolic content of cultivars grown at Gangneung was greater than that of cultivars grown at Pyeongchang. The radical scavenging activity of DPPH was similar to that of the total phenolic content rather than that of the total isoflavone content. These results suggest that antioxidant activity of soybean was associated with phenolic compounds rather than isoflavones. To identify the individual antioxidant components, we used an on-line HPLC-$ABTS^+$-based assay system, ESI/MS, and NMR. The results showed that the strongest antioxidant activity was linked to epicatechin.

Evaluation of Antioxidant Activities and Active Compounds Separated from Water Soluble Extracts of Korean Black Pine Barks

  • Black pine barks from the southern region of Korea were extracted using pressurized hot water and the water soluble extracts were then separated in a stepwise fashion using a variety of solvents, column chromatography (CC), thin layer chromatography (TLC), and high pressure liquid chromatography (HPLC). The antioxidant activities of each fraction and the active compounds were determined based on the radical scavenging activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH), reductive potential of ferric ion, and total phenol contents. A DPPH test showed that the half maximal effective concentration ($EC_{50}$ value : $6.59{\pm}0.31\;{\mu}g/mL$) of the ethyl acetate fraction (ca. 0.67%) was almost the same as that of the control compounds and inversely proportional to the value of the total phenol contents. The cell viability of the water extracts was confirmed by methyl thiazol-2-yl-2,5-diphenyl tetrazolium bromide (MTT) with enzyme linked immune sorbent assay (ELISA). Catechin, epicatechin, quercetin and ferulic acid were isolated from the ethyl acetate fraction as active compounds and identified by nuclear magnetic resonance. The antioxidant activity as value of DPPH of each of the separated compounds was lower than the ethyl acetate fraction, and ferulic acid was the lowest among these compounds.

빨간집모기와 점박이응애에 대한 관동화(Tussilago farfara) 추출물의 살충효과 (Insecticidal Activities of Tussilago farfara Extracts against Culex pipiens pallens and Tetranychus urticae)

  • Aspiring to the new raw materials of insecticides is one of the plant extracts. The material structure of a variety of plant extracts have because the material to the defense of the plant itself, the case of insecticides, using plant extracts, safe, has low toxicity and has the advantage of highly distinctive and fall. Coltsfoot (Tussilago farfara L.) is belonging to the family Compositae, and distributed in all parts of China as medicinal plants and indigenous plants used. Coltsfoot is known that it is effect to respiratory disease and has an antiviral effect. However, the reported insecticidal activity of coltsfoot could be not found. Fortunately, I found insecticidal activities when I was screening the bioassay against several insects with a lot of plant extracts. Using the ethanol extract of the Tussilago farfara, there were insecticidal activities against Culex pipiens pallens and Tetranychus urticae. There were several fractions in ethanol extract of coltsfoot by using various organic solvents. Hexane fraction showed a higher insecticidal activity than any other fraction. It is confirmed that hexane fraction contained pyrethrin by using HPLC analysis. So, it might be suggested that extract of coltsfoot has an insecticidal activity and its effect due to a ingredient of pyrethrin.

Physiological Characteristics and Seedling Growth Patterns of Neem (Azadirachta indica A. Juss) under Different Soil Conditions

  • The study of germination and seedling growth characteristics of Neem under different soil and environment conditions was undertaken. The seed germination started 8 days after sowing in commercial bed soil, whereas, delayed germination was observed in sandy-loam (15 days) and sandy (19 days) soil. The highest germination (73.33%) was observed in commercial bed soil in green house, whereas, the lowest germination was observed in sandy soil (16.67%) and sandy-loam soil (8.33%). The seeds in the open field (sandy soil) also showed poor (10%) germination. The mean number of germination seed/day (GD) and seed germination vigor rate (GV) both were highest in the commercial bed soil with 0.733% and 16.67% respectively in the green house, whereas sandy and sandy-loam soil in green house and open field (sandy soil) all showed much lower GD and GV values. The seedling characteristics of nursery revealed that the seedling grown in the growth chamber in commercial bed soil was significantly higher in all the parameters comparing to others grown in green house and open field. The growth was nearly 7 fold in the chamber compared to that of the green house nursery observed in three months old seedlings. Likewise, HPLC analysis revealed that the green house grown seedling contain higher quantity of pigments compare to the chamber grown seedlings. Among the soils used the commercial soil alone or in combination with sandy and sandy-loam soil in the ratio of 2:1:1 respectively with the temperature of $27{\pm}2^{\circ}C$ showed better for Neem nursery preparation.

Ginkgo Terpene의 Candidate albicans로 인한 관절염에 대한 치료효과 (Therapeutic Anti-inflammatory Effect of Ginkgo Terpene on Arthritis due to Candida albicans)

  • Candida albicans, a polymorphic fungus, causes systemic and local infections. Recent reports show that the fungus is a main etiological agent for the arthritis. For trea tment, antifungal drugs and/or rheumatoid drugs are used, but resistance and side effects limit application of the drugs. In search of new sources for treatment of the fungal arthritis, we choose Egb 761 (extract of Ginkgo leaves 761), one of the most popular over-the-counter herbal medicines. The Egb 761 contains two major ingredients such as terpene and flavonoid. In the present study, we examined if the terpene portion of Egb 761 had anti-inflammatory activity against C.albicans-caused arthritis. The terpene was extracted with combination of methanol and water from the Egb 761, followed by gel-permeation chromatography. Presence of terpene was determined by the Salkowski colorimetric method and HPLC analysis. For an animal model of inflammation induction, mice were given an emulsion form of C.albicans cell wall mixed with Complete Freund's Adjuvant (CFA) by footpad-injection. Results showed that intraperitoneal administration of the water-soluble portion that contained terpene and flavonoid reduced the inflammation. Whereas the terpene had anti-inflammatory activity, flavonoid portion had no such activity, For determination of possible mechanism of the activity, the terpene seemed to be suppression of nitric oxide (NO) production from LPS-treated macrophages. Taken together the Ginkgo terpene may have anti-inflammatory effect against C.albicans-caused arthritis, possibly by blocking NO production.

The Influences of Extremely Low Frequency Magnetic Fields on Drug-Induced Convulsion in Mouse

  • This study investigated the effects of extremely low frequency magnetic fields (ELF-MFs) on the sensitivity of seizure response to bicuculline, picrotoxin and NMDA in mice. The mice were exposed to either a sham or 20 G ELF-MFs for 24 hours. Convulsants were then administered i.p. at various doses. The seizure induction time and duration were measured and lethal dose ($LD_{50$}) and convulsant dose ($CD_{50}$) of the clonic and tonic convulsion were calculated. The analysis of glutamate, glycine, taurine and GABA of mouse brain was accomplished by HPLC. The mice exposed to ELF-MFs showed moderately higher $CD_{50}.{\;}LD_{50}$ and onset time on the bicuculline-induced seizure. However, the ELF-MFs did not influence them in the NMDA and picrotoxin-induced seizures. After the exposure to MFs exposure, the glutamate level was increased and GABA was decreased significantly in NMDA and picrotoxin-induced seizure. The level of glutamate and GABA were not changed by MFs in bicuculline-induced seizure. These results suggest that ELF-MFs may alter the convulsion susceptibility through GABAergic mechanism with the involvement of the level of glutamate and GABA.

미세캡슐내의 캡사이시노이드의 새로운 분석법 (A New Method for Analysis of Capsaicinoids Content in Microcapsule.)

  • 미세캡슐화된 고추올레오레진 capsicum을 사용하여 core material인 올레오레진을 파괴하지 않고, wall material를 제거함으로써 올레오레진 중의 capsaicinoids 함량을 측정하는 방법을 확립하였다. 미세캡슐을 용해할 수 있는 용매로서 dimethyl sulfoxide(DMSO)를 선발하였으며, DMSO에 혼합되는 용매로서 탄수화물을 침전시킬 수 있으면서 올레오레진에 대하여 높은 용해성을 가진 용매인 에탄올을 wall material 침전에 이용하였다. Wall material은 DMSO사용량에 대한 미세캡슐의 사용량의 비율이 $7.5{\sim}10%(w/v)$범위에서 최적이었으며 DMSO 사용량의 8배 이상의 에탄올을 첨가하였을 때 완벽한 추출이 되었다. 또한 동량의 에탄올을 사용하여 추출하였을 때 1회 추출 보다는 2회 반복추출이 완벽하게 capsaicinoids를 용해 할 수 있었다. 가열처리조건은 $55^{\circ}C$, 30분에서 DMSO가 wall materials를 용해시키기에 용이하였고 에탄올 첨가후 capsaicinoids를 추출하기 위한 vortexing time은 40㎐에서 3분 이상의 혼합에서 일정한 값을 보여주었고, 원심분리조건은 21000rpm의 회전속도에서 15분이상이 capsaicinoids함량에 최고치를 나타내었다. 확립된 방법은 고추올레오레진 미세캡슐의 제조 및 저장등의 capsaicinoids의 함량변화측정에 응용이 기대된다.

Anthocyanins in 'Cabernet Gernischet' (Vitis vinifera L. cv.) Aged Red Wine and Their Color in Aqueous Solution Analyzed by Partial Least Square Regression

  • Anthocyanins are considered one of the main color determinants in aged red wine. The anthocyanins in aged red wine made from 'Cabernet Gernischet' (Vitis vinifera L. cv.) grape were investigated by high performance liquid chromatography- electronic spray ionization- mass spectrometry (HPLC-ESI-MS) and their color presented in aqueous solution were evaluated using partial least square regression (PLS). The results showed that there were 37 anthocyanins identified in this wine, including 22 pyranoanthocyanins. The analysis of PLS indicated that different anthocyanins showed distinct color values: malvidin 3-O-(6-O-acetyl)-glucoside-4-vinylguaiacol (Mv3-acet-glu-vg) presented the highest color values, while malvidin 3-O-glucoside (Mv3-glu) showed least. Among the free non-acylated anthocyanins, peonidin 3-O-oglucoside (Pn3-glu) showed the highest color values; the coumarylated anthocyanins presented higher color values than their corresponding acetylated anthocyanins and parent anthocyanins; pyranoanthocyanins presented also higher color values than their original anthocyanins; the color of anthocyanins depended on their structure. This work will be helpful to reveal evolution in aged red wine.

열처리에 따른 돼지감자 Methanol 추출물의 항산화 및 α-glucosidase 저해 효과 (Antioxidant Activity and α-Glucosidase Inhibitory Effect of Jerusalem Artichoke (Helianthus tuberosus) Methanol Extracts by Heat Treatment Conditions)

  • This study investigated the changes of antioxidant activity and ${\alpha}$-glucosidase inhibitory effect of Jerusalem artichoke (Helianthus tuberosus) 100% methanol extracts by various heat treatment. The contents of total phenolic and flavonoid compounds in methanol extract tended to increased gradually with the rise of temperature to 180$^{\circ}C$. The maximum yield of gallic acid (51.52 ${\pm}$ 2.17mg/g extract weight) and quercetin (13.39 ${\pm}$ 0.03mg/g extract weight) were obtained with extraction temperature of 180$^{\circ}C$ for 120min. In addition, the improving extraction efficiency resulted in the increased biological activities, such as electronic donation ability (EDA, 90.36${\pm}$ 0.57%), reducing power (Abs 1.14) and ${\alpha}$-glucosidase inhibitory effect (92.14 ${\pm}$ 1.14%). Overall, the results of this study indicate that the optimum conditions for the extraction process were an extraction temperature at 180$^{\circ}C$ for 120 min, and will provide the basis for future research on the improving extraction yield of phenolic and flavonoid compounds.
