복분자 미숙과 물추출물의 콜레스테롤 개선 효과 (Cholesterol-lowering Effects of Unripe Black Raspberry Water Extract)

  • 복분자 미숙과 물추출물이 콜레스테롤 개선에 미치는 영향을 측정한 결과는 다음과 같다. 간세포주(HepG2 cells)에서 복분자 미숙과 물추출물은 SREBPs를 증가시킴으로써 혈액 내에 LDL을 LDL receptor를 통해서 세포 안으로 흡수시키고, 콜레스테롤 합성에 관여하는 HMG-CoA reductase 활성을 억제하면서 체내 콜레스테롤을 조절하였다. 그러나 HDL의 생성에 관여하는 유전자(ABCA1, SR-B1)는 변화를 보이지 않았다. 결과적으로 복분자 미숙과 물추출물이 LDL receptor를 통해서 LDL을 억제시키고 체내에서는 콜레스테롤 생합성을 억제하였으며, HDL의 생성에는 관여를 하지 않음을 확인하였다. 또한 ApoB1/ApoA1 ratio 값을 통해서 동맥경화 지표를 확인해 본 결과 유의성 있게 수치가 감소함을 확인하였고, 이는 미숙과 물추출물이 콜레스테롤을 개선하여 동맥경화를 예방할 것으로 기대한다. 또한 대식세포(RAW 264.7 cells)에서 복분자 미숙과 물추출물이 CD 36, SR-A 수용체를 억제시킴으로 인해 세포 내의 ox-LDL 의 흡수를 차단시키고, 세포 안에서는 macrophage에 있는 PPAR-${\gamma}$를 억제시킴으로 인해 LDL 산화가 억제되었다. Adipophilin의 활성이 억제됨에 따라 세포 안에 있는 콜레스테롤 방출을 촉진시킴으로 인해 동맥경화를 완화시킬 수 있다고 사료된다.

Investigation of Hetero - Material - Gate in CNTFETs for Ultra Low Power Circuits

  • An extensive investigation of the influence of gate engineering on the CNTFET switching, high frequency and circuit level performance has been carried out. At device level, the effects of gate engineering on the switching and high frequency characteristics for CNTFET have been theoretically investigated by using a quantum kinetic model. It is revealed that hetero - material - gate CNTFET(HMG - CNTFET) structure can significantly reduce leakage current, enhance control ability of the gate on channel, and is more suitable for use in low power and high frequency circuits. At circuit level, using the HSPICE with look - up table(LUT) based Verilog - A models, the performance parameters of circuits have been calculated and the optimum combinations of ${\Phi}_{M1}/{\Phi}_{M2}/{\Phi}_{M3}$ have been concluded in terms of power consumption, average delay, stability, energy consumption and power - delay product(PDP). We show that, compared to a traditional CNTFET - based circuit, the one based on HMG - CNTFET has a significantly better performance (SNM, energy, PDP). In addition, results also illustrate that HMG - CNTFET circuits have a consistent trend in delay, power, and PDP with respect to the transistor size, indicating that gate engineering of CNTFETs is a promising technology. Our results may be useful for designing and optimizing CNTFET devices and circuits.

Comparison of Jujube Extract with Tangerine Peel Extract in Lowering Plasma Lipids and Activities of Cholesterol Regulating Enzymes in Cholesterol-Fed Rats

  • A potential mechanism through which the jujube extract might produce a cholesterol-lowering effect was compared with that of tangerine peel extract in vivo. Two extracts were prepared using ethanol. Male rats were fed a high cholesterol (1%, w/w) lab chow with jujube extract (1.2%) or tangerine peel extract (6.3%, w/w) for 3 weeks. Activities of hepatic HMG-CoA reductase (289.6$\pm$12.9 and 296.7$\pm$11.6 nmole/min/mg vs. 347.9$\pm$17.5 nmole/min/mg) and ACAT (554.8$\pm$18.2 and 451.7$\pm$19.4 nmole/min/mg vs. 602.6$\pm$21.4 nmole/min/mg) were significantly lowered by both supplements compared to the control group. These two supplements also substantially reduced the concentrations of plasma cholesterol (103.3$\pm$15.9 and 101.6$\pm$19.4 mg/dL vs. 141.6$\pm$18.1 mg/dL) and triglyceride (61.3$\pm$5.5 and 55.5$\pm$3.9 mg/dL vs. 96.0$\pm$4.2 mg/dL). The inhibition of HMG-CoA reductase resulting from the supplementation of jujube or tangerine extracts could count for the reduction in plasma cholesterol. Accordingly, lipid-lowering action of both supplements appears to be similar in high-cholesterol fed rats.

Streptozotocin 유발 당뇨쥐 간조직에서의 3-Hydroxy-3-Methylglutaryl Co Enzyme A Reductase 활성과 혈중지질수준에 미치는 녹차 Catechin의 영향 (Effects of Green Tea Catechin on Liver 3-Hydroxy-3-Methylglutaryl Co Enzyme A Reductase Activity and Serum Lipid Levels in Streptozotocin-induced Diabetic Rats)

  • The purpose of this study was to investigate the effects of green tea catechin on lipid metabolism in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats weighing 150$\pm$10gm were randomly assigned to one normal and three STZ-induced diabetic groups. Diabetic animals were fed catechin free diet(DM-0C group), 0.5% catechin diet(DM-0.5C group) and 1% catechin diet(DM-1C group). Diabetes was experimentally induced by intravenous injection of 55mg/kg body wt of STZ in citrate buffer(pH 4.3) after feeding of three experimental diets for 4 weeks. Animals were sacrificed at the 6th day of diabetic states. Levels of blood glucose were three fold higher in all three STZ-induced diabetic groups than that of the normal group. The levels of plasma insulin were markedly lower in three STZ-induced diabetic groups than that of the normal group. The levels of plasma cortisol were increased in DM-0C group compared with that of the normal group. Triglyceride, total-cholesterol and LDL-cholesterol levels in serum were increased in DM-0C groups compared with the normal group but were not significantly different between catechin diet groups and normal group. Serum HDL-cholesterol levels were reduced in DM-0C and DM-0.5C groups by 38% and 25%, respectively and had similar tendency in the DM-1C group compared with that of control group. Atherogenic index have shown same pattern as the result of total cholesterol. Activities of 3-hydroxy-3-methylglutaryl Co enzyme A(HMG-CoA) reductase were higher in DM-0C groups than those of the normal group but were not significantly different between catechin diet groups and the normal group. It is concluded that dietary catechins can modulate lipid levels of serum and liver HMG-CoA reductase activity in diabetic rats.

죽력(Bambusae Caulis in Liquamen)이 고지방식이를 급여한 흰쥐의 체내 지질대사에 미치는 영향 (Effect of Bambusae Caulis in Liquamen on Lipid Metabolism in Rats Fed High Fat Diet)

  • 재래적인 방법으로 제조된 죽력의 생리활성 효능을 구명하기 위하여 in vitro에서 항산화활성 및 HMG-CoA reductase 저해활성과 in vivo에서 고콜레스테롤혈증 개선효능을 실험하여 다음과 같은 결과를 얻었다 1. In vitro에서, Rancimat로 측정한 항산화활성은 죽력 1.25희석액과 원액은 대조구보다 높았고, HMG-Co A reductase저해활성은 죽력 원액이 57.9$\%$이었고 1.25희석액은 36.0$\%$이 었다. 2. In vivo에서, 6주에서 죽력 투여로 체중증가율은 대조군에 비하여 유의성있게 둔화되었고, 식이효율은 감소되었으나 유의적인 차이는 아니었으며, 간장/체중 비율은 실험군간에 유의성있는 변화를 나타내지 않았다. 죽력 저용량투여군의 총콜레스테롤량은 대조군에 비하여유의적인 감소를 나타내지 않았으나 고용량투여로 대조군보다 약 17$\%$정도 감소되었고, 중성지질량은 죽력 투여로대조군에 비하여 각각 26$\%$, 39$\%$의 유의적인 감소를 나타내었는데, 특히 고용량 투여군은 정상군보다 낮았으며, 인지질량은죽력투여로 대조군에 비하여 증가는 되었으나 유의적인 차이는 아니 었다. LDL-콜레스테를 농도는 고콜레스테롤식이 급여로 정상군보다 약 41$\%$ 정도 증가되었으나 죽력투여로 대조군보다 각각 12$\%$, 20$\%$씩 유의한 감소를 나타내었는데, 특히 고용량 투여군은 정상군의 농도에 근접하였으며, 죽력 투여로 HDL-콜레스테롤농도는 대조군에 비하여 상승되었으나 유의적인 효과는 아니었고, 죽력투여로 저하된 HDL-콜레스테롤/총콜레스테롤비는 대조군보다 약 48$\%$가 높아졌으며 동맥경화지수는 약 26$\%$가 낮아졌으나 유의성있는 변화를 나타내지는않았다. 고콜레스테롤식이 급여로 증가된 유리콜레스테를 농도는 고용량투여로 26$\%$가 감소되어 유의성있는 변화를 나타냈으며, 고콜레스테롤식이 급여로 콜레스테릴 에스테르 농도는정상군에 비하여 약 96$\%$이상의 증가를 나타냈으나 죽력 고용량 투여로 대조군에 비하여 32$\%$정도 감소되었다. 고콜레스테롤식이 급여로 간 중 총콜레스테롤량은 정상군에 비하여 유의하게 증가되었고, 죽력 저용량 투여는 증가된 총 콜레스테롤량을 감소시키지 못했으나 고용량 투여로 대조군보다 유의성있게 저하시켰으며, 중성지방량에는 변화가 없었다. 고콜레스테롤식이 급여로 상승된 ALT활성은 죽력투여로대조군에 비하여 유의하게 저하되었으나 ALT 및 ALP활성은 차이는 보이지 않았다. 이상의 실험결과에서 죽력이 in vitro에서 높은 항산화활성과 HMG-Co A reductase 활성을 유의하게 저해시켰고, in vivo에서 고콜레스테롤식이를 급여한 군보다 총콜레스테롤,LDL-콜레스테를 및 중성지질농도 등은 감소시켰으며, HDL-콜레스테롤, 인지질 농도 등은 증가시킴으로써지방간 및 동맥경화의 예방과 치료에 효과적일 것으로 추정되었다.

Effects of Water Extracts from Mulberry Leaves on Hepatic HMG-CoA Reductase and Acyl-CoA-Cholesterol Acyl Transferase Activity in Rats Fed High Cholesterol Diets

  • This study investigated the effects of mulberry leaf extract on lipid metabolism in rats fed a high cholesterol diet. Sprague-Dawley male rats weighing $100{\pm}10g$ were randomly assigned either to one of two normal diet groups, with (NE group) or without (N group) mulberry extract, or one of four high cholesterol groups containing 1% cholesterol and various levels of dietary mulberry leaf extract. The rats fed high cholesterol diets were subdivided into 4 groups according to level of mulberry extract; Mulberry extract free group (HC group), 0.8% mulberry leaf extract group (HCL group), 1.6% mulberry leaf extract (HCM group) and 3.2% mulberry leaf extract (HCH group). The rats were fed their respective diets ad libitum for 4 weeks. The levels of serum triglyceride, total cholesterol and LDL-cholesterol of the HC group were higher than mulberry leaf extract supplemented groups. In contrast, the levels of serum HDL-cholesterol in groups supplemented with mulberry leaf extract were significantly lower than that of HC group. Hepatic total lipids, triglycerides, and cholesterol were significantly higher in the high cholesterol groups compared to those of the normal group, but were lower in the HCL, HCM and HCH groups than in the HC group. HMG-CoA reductase activity was significantly decreased in the HC and HCL groups compared to the normal and NE groups. However, the activities in the HCM and HCH group were similar to that of the normal group. The activity of acyl-CoA-cholesterol acyl transferase (ACAT) was increased in high cholesterol groups compared to the normal group. However, the activity was lower for all of the high cholesterol groups fed mulberry leaf extracts, and was lowest for the highest supplemented group (HCH), with no significantly difference from the normal group. In conclusion, the reduction in serum and hepatic lipid composition by mulberry leaf extract may be due to its modulation of HMG-CoA reductase and ACAT activities.

Corn silk extract improves cholesterol metabolism in C57BL/6J mouse fed high-fat diets

  • BACKGROUNG/OBJECTIVES: Corn silk (CS) extract contains large amounts of maysin, which is a major flavonoid in CS. However, studies regarding the effect of CS extract on cholesterol metabolism is limited. Therefore, the purpose of this study was to determine the effect of CS extract on cholesterol metabolism in C57BL/6J mouse fed high-fat diets. MATERIALS/METHODS: Normal-fat group fed 7% fat diet, high-fat (HF) group fed 25% fat diet, and high-fat with corn silk (HFCS) group were orally administered CS extract (100 mg/kg body weight) daily. Serum and hepatic levels of total lipids, triglycerides, and total cholesterol as well as serum free fatty acid, glucose, and insulin levels were determined. The mRNA expression levels of acyl-CoA: cholesterol acyltransferase (ACAT), cholesterol 7-alpha hydroxylase (CYP7A1), farnesoid X receptor (FXR), lecithin cholesterol acyltransferase (LCAT), low-density lipoprotein receptor, 3-hyroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA reductase), adiponectin, leptin, and tumor necrosis factor ${\alpha}$ were determined. RESULTS: Oral administration of CS extract with HF improved serum glucose and insulin levels as well as attenuated HF-induced fatty liver. CS extracts significantly elevated mRNA expression levels of adipocytokines and reduced mRNA expression levels of HMG-CoA reductase, ACAT, and FXR. The mRNA expression levels of CYP7A1 and LCAT between the HF group and HFCS group were not statistically different. CONCLUSIONS: CS extract supplementation with a high-fat diet improves levels of adipocytokine secretion and glucose homeostasis. CS extract is also effective in decreasing the regulatory pool of hepatic cholesterol, in line with decreased blood and hepatic levels of cholesterol though modulation of mRNA expression levels of HMG-CoA reductase, ACAT, and FXR.

복분자 미숙과와 홍삼 추출물의 콜레스테롤 개선 효과 (Effects of Extracts of Unripe Black Raspberry and Red Ginseng on Cholesterol Synthesis)

  • 본 연구는 복분자 미숙과와 홍삼 물 추출물을 이용하여 인간 간암세포주(HepG2)와 위장관세포주(Caco-2)를 이용하여 콜레스테롤 억제효과 및 그와 연관된 HMG-CoA reductase 활성 억제효과 및 LDL 및 HDL과 관련된 분자기전을 조사하였다. 그 결과 복분자 미숙과와 홍삼 추출물은 모두 콜레스테롤 합성 억제 효과를 보였을 뿐만 아니라 HMG-CoA reductase 활성 억제효과를 보였다. 또한 두 추출물은 단독 투여시 보다 복합 투여시 부가효과를 보였다. 그리고 LDL receptor와 이를 조절하는 SREBP-2를 증가시키고 콜레스테롤 transport 인 ABCA1의 발현을 증가시켜 LDL를 낮추고 HDL를 높이는 효과가 있을 것으로 판단된다. 특히 두 추출물의 복합 투여시 훨씬 높은 시너지 효과를 나타냄을 확인하였다. 이러한 결과는 전통적으로 오랜 기간 사용되어온 천연물인 복분자와 홍삼이 콜레스테롤 예방에 효과적임을 입증한 결과로 관련 대사증후군의 예방에 기여할 것으로 사료된다.

Lovastatin 생합성 유전자를 이용한 lovastatin 생산균주의 탐색 (Screening of lovastatin-producing strains by PCR using lovastatin biosynthesis genes)

  • 본 연구는 Asp. terreus ATCC 20542 변이주로부터 lovastatin 생합성 유전자 중 polyketide 생합성 유전자 등을 이용한 PCR법으로 Aspergillus sp. 이외의 statin계열 물질 생산균주의 탐색법 구축 및 lovastatin 대량생산을 하고자 하였다. Lovastatin 생합성 유전자 중 가장 중요한 유전자인 polyketide synthase gene와 diketide synthase gene로부터 각각의 primer를 제작하여 PCR을 이용한 lovastatin 생산 균주를 탐색하였다. 선발된 7개의 균주의 형태학상의 특성 및 lovastatin 생산성을 검토한 결과 Aspergillus sp. 이외의 Penicillium sp.으로 추정되는 균주를 재선발하여 SJ-2로 명명하였다. 선발된 SJ-2는 액체배양 및 고체배양을 한 후 추출하여 TLC와 HPLC를 통하여 각각의 lovastatin 생산량을 비교, 검토하였다. 또한, SJ-2에 대두를 이용하여 lovastatin 고생산성을 확인한 결과, 대두-전배양체를 $30^{\circ}C$, 1시간동안 열처리하여 접종하여 본배양 15일째에 가장 높은 lovastatin을 생산할 수 있었다. In vitro assay 결과에서는 HMG-CoA reductase에 대한 저해활성도가 75%로 나타났다. 본 연구는 기존의 lovastatin 탐색법으로 널리 알려져 있는 bioassay법이 아닌 lovastatin 생합성 유전자를 이용하여 PCR을 통한 lovastatin 생산균주의 탐색이 신속하고 효과적인 방법으로 사료되었다.

순환기가감방(循環器加減方)이 고지혈증 유발 백서(白鼠)에 미치는 영향 (Effect of Sunhwangigagambang(SHG) on Hyperlipidemia in SD Rats Induced by High Cholesterol Diet)

  • Objective : This study was performed to investigate the effect of Sunhwangigagambang(SHG) on hyperlipidemia in SD rats induced by high cholesterol diet. Method : After treatment with SHG, cytotoxicity, body weight, liver weight, AST, ALT, ALP, total cholesterol, LDL-cholesterol, HDL-cholesterol, triglyceride, glucose, albumin, total protein in serum, malondialdehyde, and gene expression for ACAT and HMG-CoA reductase in hepatic tissue were analyzed. Result : 1. SHG didn't show any cytotoxicity in both human fibroblast cell line and SD rats. 2. SHG significantly inhibited the increase of liver weight by high cholesterol diet compared to the control group. 3. SHG significantly ameliorated the increase of total cholesterol, LDL-cholesterol, and triglyceride and reduction of HDL-cholesterol compared to the control group. 4. SHG significantly reduced glucose level in serum compared to the control group. 5. SHG significantly reduced malondialdehyde in hepatic tissue compared to the control group. 6. SHG significantly down-regulated gene expression of ACAT and HMG-CoA reductase compared to the control group. Conclusion : These results suggest that SHG might be effective in treatment and prevention of hyperlipidemia.
