The Effects of Ampelopsis Radix on Allergic Inflammation in PMA-stimulated Human Mast Cells (백렴의 알레르기 염증반응에 대한 억제효과)

  • Objectives: In this study, we investigated the inhibitory effects of Ampelopsis Radix methanol(AR-M) extract on allergic inflammation in activated human mast cells and its potential therapeutic or toxic effects. Methods: Ampelopsis Radix(AR) was extracted with 80% methanol. HMC-1 cells, a human mast cell line, were treated with different concentrations of AR-M extract, and then stimulated with PMA plus A23187. The cell toxicity of AR-M extract was determined by MTT assay. The concentrations of $PGE_2$ and cytokines were measured by ELISA. The gene expression of COX-2 and its protein levels were determined by RT-PCR and Western blot. The phosphorylation of ERK MAPK and the NF-${\kappa}B$ activation were determined by Western blot. Results: AR-M extract was significantly inhibited the production of PGE2 and inflammatory cytokines(TNF-${\alpha}$, IL-6 and IL-8) in PMA/A23187-stimulated HMC-1 cells. AR-M extract also attenuated the mRNA expression of COX-2 and its protein induction. Furthermore, AR-M extract attenuated PMA/A23187-induced phophorylation of ERK1/2 MAPK and the NF-${\kappa}B$ p65 subunit translocation into nuclear of HMC-1 cells. AR-M extract significantly decreased PMN A23187-induced release of histamine in a dose-dependent manner. Conclusions: These results indicate that Ampelopsis Radix shows the property of anti-allergic inflammation In vitro through suppressing the production of inflammatory mediators released from mast cells, suggesting have a potential for the treatment of allergic diseases.

  • Objectives : In this study, we investigated anti-allergic effect of Okbyeongpungsan-Hap-Changijasan (KOB01) in allergic rhinitis(AR) experimental animals and mast cells. Methods : The potential anti-allergic effect of KOB01 was investigated in a rat model of compound 48/80-induced systemic anaphylactic shock and a mouse of ovalbumin(OVA)-induced AR, and human mast cell line, HMC-1 culture. Each animals were divided into four groups: normal, control, KOB01-treated(100 and 200 mg/kg) and anti-histamine drug, dosodium cromoglycate (DSCG)-treated(50 mg/kg). Animals were orally treated with KOB01 and DSCG and intraperitoneally injected with compound 48/80($10\;{\mu}g/kg$) or sensitized with 0.1% OVA. The mortality and serum histamine levels were measured in compound 48/80-induced anaphylatic rats. The histological changes in nasal mucosa were investigated in OVA-induced AR mice. Also, mast cell degranulation was observed in compound 48/80-stimulated HMC-1 cells. Results : KOB01 increased mortality and significantly decreased serum histamine levels in compound 48/80-induced anaphylatic rats. The abnormal histological changes such as expansion of grandular cells and hypertrophy of epithelium in nasal mucosa of OVA-induced AR mice was improved by KOB01 treatment nearby a normal group. Therefore, KOB01 inhibited compound 48/80-induced degranulation in HMC-1 cells. Conclusions : These results indicate that KOB01 decrease allergic response through suppressing the mast cell activation in AR and suggest a potential role for KOB01 as a source of anti-allergic agents for use in allergic disorders including of AR.

Effect of Anti-atopic Allergic Reaction in Response to Oriental Herb Extracts (생약재 추출물의 아토피 완화효과)

  • The in vivo and in vitro effects of oriental herb extracts of Cassia obtusifolia, Taraxacum platycarpum and Ulmusmacrocarpa on anti-atopic allergic reaction were evaluated in this study. A mixture of these extracts exhibited more potent anti-allergic activities in human mast cells than those from individual extracts. The herbal mixture significantly inhibited the release of compound 48/80-induced $\beta$-hexosaminidase release in the human mast cell line, HMC-1. The mixture also suppressed the production of PMA and A23187-induced inflammatory cytokines in HMC-1 cells. To further investigate the in vivo effects of the herbal mixture, a Dermatophagoides farinae (DF)-induced atopic dermatitis mouse model was utilized. Oral administration of the herbal mixture significantly decreased the ear thickness and swelling in DF treated NC/Nga mice in a dose dependent manner. Furthermore, serum levels of IgE and interleukin-4 (IL-4) were significantly decreased, whereas interferon-gamma (IFN-$\gamma$) levels were increased in the mixture administrated groups when compared to the control. Taken together, our data indicate the possibility of using a mixture of the oriental herb extract to relieve symptoms of atopic dermatitis.

Effects of Ethanol Extract of Liriope platyphylla on Allergic Inflammation (맥문동(麥門冬) 에탄올 추출물이 알레르기 염증 반응에 미치는 영향)

  • This study investegated the effect of Liriope platyphylla (LP) on allergic reactions and its mechanism of action. We investigated the effect of LP on Evans Blue (EB) extravasation induced by anti-dinitrophenyl (DNP)-IgE in rats. We tested whether the ethanol extract of LP reduced ear skin thickness and historical changes induced by topical application of 2,4-dinitrofluorobenzene (DNFB) to ears of mice. We evaluated compound 48/80-induced release of histamine in rats peritoneal mast cell (RPMCs). We also investigated the regulatory effect of LP on the level of inflammatory mediators in PMACI-induced human mast cell (HMC-1); cytokine IL-6, IL-8, TNF-${\alpha}$ in HMC-1, MAPKs (ERK, JNK and p38) in HMC-1. The ethanol extract of LP (81.3 mg/100 g body weight) significantly inhibited the PCA reaction compared with the control (P < 0.05). However, LP did not prevent topical applications of DNFB-induced ear skin thickening and histological changes. In RPMCs, histamine release induced by compound 48/80 was significantly attenuated by LP at $100{\mu}g/ml$ (P < 0.05). LP extract ($100{\mu}g/ml$) significantly reduced the PMACI-induced IL-6, IL-8, and TNF-${\alpha}$ secretion via inhibition of ERK phosphorylation in HMC-1. In conclusion, the ethanol extract of LP inhibited mast cell-derived, immediate-type allergic reactions, and the result suggest the potential of LP for preventing allergic inflammatory disorders.

  • The high speed machining center(HMC) has been widely applied to manufacture a die and trial product in many machine industry. Because the evaluation for HMC is not sufficiently performed and the efficient cutting conditions can't be selected, a peat loss has been caused in the cost aspect. In this study, the need of preliminary running time and unstable spindle speed is presented by the analysis of acceleration in idling. The Machinability for the TiA1N coated flat end mill and STD11 (H$\sub$R/C60) is evaluated from the trends of tool wear and cutting force according to cutting conditions . The resonance spindle speed is identified through the tool wear and natural frequency test.

  • To apply induction hardening method to a press die having 3-D free surface, the induction hardening tool moves on a press die above 1~2mm gap with constant velocity. Since the induction hardening process requires its own hardening path for each die, a direct teaching method which generates working path directly guided by operators is more suitable than an offline method using CAD/CAM data. Accelerated Degradation Test of Electrolyte Membrane in PEMFC Stack (고분자 전해질 연료전지 스택에서 전해질막의 열화 가속시험)

  • Until a recent day, degradation of PEMFC (Proton Exchange Membrane Fuel Cells) has been mainly studied in unit cell. But operation and degradation of real PEMFC going along in stack instead of unit cell. Therefore in this work, ADT (Accelerated Degradation Test) of PEMFC was done in stack and the result from stack's test was compared with that of unit cell. The polymer electrolyte membrane was degraded by repeated electrochemical and mechanical degradation method among several ADT methods. Current densities of MEA at 0.6V decreased in stack and unit cell, 28.4% and 27.8% respectively after ADT for 312 hours. Hydrogen crossover current densities of membrane increased in stack and unit cell, 16.8% and 15.2% respectively after ADT for 312 hours. The result of ADT in stack was similar that of ADT in unit cell, which showed that ADT method of unit cell was available to the stack.

Decrease of PEMFC Performance by SO2 in Air (공기 중 SO2에 의한 고분자전해질 연료전지의 성능 감소)

  • The effects of $SO_2$ on the performance of proton exchange membrane(PEMFC) were investigated by introduction air containing $SO_2$ into cathode inlet of PEMFC. And the recovery of the cell performance by applying clean air, cycle voltammetry(CV) and high voltage holding following exposure contaminated air was studied. The $SO_2$ concentration range used in the experiments was from 20 ppb to 1.3 ppm. The performance degradation and recovery were measured by constant-current discharging, I-V polarization and electrochemical impedance spectroscopy(EIS). The cell voltage gradually decayed with time and decreased by 17 mV after 200 hours of 20 ppb $SO_2$ injection. The cell performance can be recovered partially by clean air flushing, CV and high voltage holding due to desorption of S from Pt catalyst.

Effect of Electrode Degradation on the Membrane Degradation in PEMFC (PEMFC에서 전극 열화가 전해질 막 열화에 미치는 영향)

  • Until a recent day, degradation of PEMFC MEA (membrane and electrode assembly) has been studied, separated with membrane degradation and electrode degradation, respectively. But membrane and electrode were degraded coincidentally at real PEMFC operation condition. During simultaneous degradation, there was interaction between membrane degradation and electrode degradation. The effect of electrode degradation on membrane degradation was studied in this work. We compared membrane degradation after electrode degradation and membrane degradation without electrode degradation. I-V performance, hydrogen crossover current, fluoride emission rate (FER), impedance and TEM were measured after and before degradation of MEA. Electrode degradation reduced active area of Pt catalyst, and then radical/$H_2O_2$ evolution rate decreased on Pt. Decrease of radical/$H_2O_2$ reduced the velocity of membrane degradation.
