Streptococcus sp. 배양 여과물(HAIRCLETM)의 모발 개선 효과 (Hair Revitalization Effects of Streptococcus sp. Strain Culture Medium Filtrate (HAIRCLETM))

  • Streptococcus sp.는 일반적으로 히알루론산을 생산하는 균주로 알려져 있다. 본 연구에서는 Streptococcus 균주가 배양하면서 만들어내는 여러 대사산물 중 히알루론산 이외의 영양성분에 주목하였다. 히알루론산이 생성되지 않은 조건에서의 Streptococcus sp. 배양 여과물로 모발 실험용 샘플(HAIRCLETM)을 제조하고 모발에 미치는 효과를 확인하였다. 모발의 인장강도는 만능시험기(universal testing machine, UTM)로 측정하였으며, 모발표면의 미세구조는 atomic force microscope (AFM)으로 분석하였다. 이 연구를 통해 Streptococcus sp. 배양 여과물은 손상된 모표피개선, 마찰력증가 및 모발강도의 증가 등의 모발개선 효과가 있는 것으로 확인되었다. 또한 in vitro 모발관련 실험을 통해 Streptococcus sp. 배양 여과물이 모발성장인자인 혈관내피세포 성장인자(vascular endothelial growth factor, VEGF), 간세포성장인자(hepatocyte growth factor, HGF), Wnt10b의 발현 촉진 효과가 있음을 확인 하였으며, 안드로젠 수용체(androgen receptor, AR)와 TGF-β2 발현을 저해시켜 남성형 탈모완화 효과도 있음을 확인 하였다. 또한 산화스트레스에 대해 장벽 관련 인자(INV, CLDN-1)의 발현을 증가시키고, 염증성 인자인 COX-2 발현을 억제시킴으로써 손상된 두피 장벽을 개선하고 유도된 염증을 완화시키는 효능이 있음을 확인하였다.

Muscle Strength and Biochemical Markers as Predictors of Depression in Hemodialysis Patients: A Cross-Sectional Study

  • Patients with chronic renal failure, many of which treated with hemodialysis, present a high prevalence of impaired muscle strength which suggest that muscle mass parameters may be used as markers for changes in muscle in these patients. Measurement of handgrip strength (HGS) is a common, simple, and quick measure of muscle function an indicator of overall muscle strength which has been associated with physical activity and several anthropometric traits. Intercellular adhesion molecule-1 (ICAM-1) and insulin-like growth factor-1 (IGF-1) are biochemical markers associated with inflammatory processes which are a common consequence of dialysis. Additionally, hemodialysis patients frequently present signs of malnutrition and depression. This cross-sectional study aimed to evaluate if muscle and biochemical markers could be used to predict the risk of depression in hemodialysis patients. Several anthropometric parameters, nutrient intake, depression state and the serum levels of ICAM-1 and IGF-1 were determined and Pearson's correlation coefficient and/or Spearman's correlation coefficient were used to test the correlation between them. Our results do not show a correlation between HGF, IGF-1 and ICAM-1 with the depression status of the patients, but mid-arm muscle circumference (MAMC) was statistically and positively correlated with depression. Additionally, ICAM-1 levels were negatively correlated with HGS, MAMC, and IGF-1. Overall, the results of the present study suggest that HGS may be used as an indicator of cardiovascular diseases and MAMC may be a good predictor of the level of depression in hemodialysis patients, although further studies are required.

  • 치수절단술은 유치의 치수치료 방법 중 사용빈도가 높은 시술 중 하나로 치수절단 술식에 사용되는 약제는 치수나 주위조직에 무해하여야 하며, 감염이나 내흡수 등의 부작용이 없어야 한다. 본 연구는 임상에서 유치의 지혈적 치수절단 술식의 약제로 사용되는 ferric sulfate의 유전자 독성을 평가할 목적으로 human gingival fibroblast에 ferric sulfate를 다양한 농도와 접촉시간을 설정한 후 comet assay를 이용하여 유전자 독성을 평가하여 보았다. 그 결과는 다음과 같다. 1. 농도에 따른 세포의 유전자 손상정도의 변화는 ferric sulfate의 농도에 비례하여 유전자 손상이 증가하는 양상을 나타내었다. 2. 농도에 따른 세포의 유전자 손상정도는 0.1mM 이상의 농도에서 대조군과 유의한 차이를 나타내었다(p<0.05). 3. 시간경과에 따른 세포의 유전자 독성의 변화는 대조군과 유의한 차이를 보이지 않았다(p>0.05).

A New Esterase, Belonging to Hormone-Sensitive Lipase Family, Cloned from Rheinheimera sp. Isolated from Industrial Effluent

  • The gene for esterase (rEst1) was isolated from a new species of genus Rheinheimera by functional screening of E. coli cells transformed with the pSMART/HaeIII genomic library. E. coli cells harboring the esterase gene insert could grow and produce clear halo zones on tributyrin agar. The rEst1 ORF consisted of 1,029 bp, corresponding to 342 amino acid residues with a molecular mass of 37 kDa. The signal P program 3.0 revealed the presence of a signal peptide of 25 amino acids. Esterase activity, however, was associated with a homotrimeric form of molecular mass 95 kDa and not with the monomeric form. The deduced amino acid sequence showed only 54% sequence identity with the closest lipase from Cellvibrio japonicus strain Ueda 107. Conserved domain search and multiple sequence alignment revealed the presence of an esterase/ lipase conserved domain consisting of a GXSXG motif, HGGG motif (oxyanion hole) and HGF motif, typical of the class IV hormone sensitive lipase family. On the basis of the sequence comparison with known esterases/ lipases, REst1 represents a new esterase belonging to the class IV family. The purified enzyme worked optimally at $50^{\circ}C$ and pH 8, utilized pNP esters of short chain lengths, and showed best catalytic activity with p-nitrophenyl butyrate ($C_4$), indicating that it was an esterase. The enzyme was completely inhibited by PMSF and DEPC and showed moderate organotolerance.

The roles of growth factors and hormones in the regulation of muscle satellite cells for cultured meat production

  • Cultured meat is a potential sustainable food generated by the in vitro myogenesis of muscle satellite (stem) cells (MSCs). The self-renewal and differentiation properties of MSCs are of primary interest for cultured meat production. MSC proliferation and differentiation are influenced by a variety of growth factors such as insulin-like growth factors (IGF-1 and IGF-2), transforming growth factor beta (TGF-β), fibroblast growth factors (FGF-2 and FGF-21), platelet-derived growth factor (PDGF) and hepatocyte growth factor (HGF) and by hormones like insulin, testosterone, glucocorticoids, and thyroid hormones. In this review, we investigated the roles of growth factors and hormones during cultured meat production because these factors provide signals for MSC growth and structural stability. The aim of this article is to provide the important idea about different growth factors such as FGF (enhance the cell proliferation and differentiation), IGF-1 (increase the number of myoblasts), PDGF (myoblast proliferation), TGF-β1 (muscle repair) and hormones such as insulin (cell survival and growth), testosterone (muscle fiber size), dexamethasone (myoblast proliferation and differentiation), and thyroid hormones (amount and diameter of muscle fibers and determine the usual pattern of fiber distributions) as media components during myogenesis for cultured meat production.

Clinical significance of APOB inactivation in hepatocellular carcinoma

  • Recent findings from The Cancer Genome Atlas project have provided a comprehensive map of genomic alterations that occur in hepatocellular carcinoma (HCC), including unexpected mutations in apolipoprotein B (APOB). We aimed to determine the clinical significance of this non-oncogenetic mutation in HCC. An Apob gene signature was derived from genes that differed between control mice and mice treated with siRNA specific for Apob (1.5-fold difference; P < 0.005). Human gene expression data were collected from four independent HCC cohorts (n = 941). A prediction model was constructed using Bayesian compound covariate prediction, and the robustness of the APOB gene signature was validated in HCC cohorts. The correlation of the APOB signature with previously validated gene signatures was performed, and network analysis was conducted using ingenuity pathway analysis. APOB inactivation was associated with poor prognosis when the APOB gene signature was applied in all human HCC cohorts. Poor prognosis with APOB inactivation was consistently observed through cross-validation with previously reported gene signatures (NCIP A, HS, high-recurrence SNUR, and high RS subtypes). Knowledge-based gene network analysis using genes that differed between low-APOB and high-APOB groups in all four cohorts revealed that low-APOB activity was associated with upregulation of oncogenic and metastatic regulators, such as HGF, MTIF, ERBB2, FOXM1, and CD44, and inhibition of tumor suppressors, such as TP53 and PTEN. In conclusion, APOB inactivation is associated with poor outcome in patients with HCC, and APOB may play a role in regulating multiple genes involved in HCC development.

Long-Duration Three-Dimensional Spheroid Culture Promotes Angiogenic Activities of Adipose-Derived Mesenchymal Stem Cells

  • Mesenchymal stem cells (MSCs) offer significant therapeutic promise for various regenerative therapies. However, MSC-based therapy for injury exhibits low efficacy due to the pathological environment in target tissues and the differences between in vitro and in vivo conditions. To address this issue, we developed adipose-derived MSC spheroids as a novel delivery method to preserve the stem cell microenvironment. MSC spheroids were generated by suspension culture for 3 days, and their sizes increased in a time-dependent manner. After re-attachment of MSC spheroids to the plastic dish, their adhesion capacity and morphology were not altered. MSC spheroids showed enhanced production of hypoxia-induced angiogenic cytokines such as vascular endothelial growth factor (VEGF), stromal cell derived factor (SDF), and hepatocyte growth factor (HGF). In addition, spheroid culture promoted the preservation of extracellular matrix (ECM) components, such as laminin and fibronectin, in a culture time- and spheroid size-dependent manner. Furthermore, phosphorylation of AKT, a cell survival signal, was significantly higher and the expression of pro-apoptotic molecules, poly (ADP ribose) polymerase-1 (PARP-1) and cleaved caspase-3, was markedly lower in the spheroids than in MSCs in monolayers. In the murine hindlimb ischemia model, transplanted MSC spheroids showed better proliferation than MSCs in monolayer. These findings suggest that MSC spheroids promote MSC bioactivities via secretion of angiogenic cytokines, preservation of ECM components, and regulation of apoptotic signals. Therefore, MSC spheroid-based cell therapy may serve as a simple and effective strategy for regenerative medicine.

In vivo와 In vitro 실험에서 가미쌍화탕 및 구성한약재가 마우스의 모발 성장에 미치는 실험적 연구 (Study on the Effect of Gamissanghwa-tang and each Medicinal Plant Extract for the Hair Growth of the Mice using In vivo and In vitro Test)

  • To screen the effective materials for hair loss treatment, the Gamissanghwa-tang extracts were tested. As a result we found that the Gamissanghwa-tang extracts have the hair growth promoting effect. After topical application of each test materials to the back of CS7BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. In the experiments of 5α-reductase type II inhibition assay, Radix Paeoniae Alba, Semen Cuscutae showed effective potential to inhibit the activity of 5α-reductase type II. And hair growth index of the Gamissanghwa-tang extracts ranked as 1.2, especially the hair growth index of Fructus Rubi is highest as 1.8. But there were no plant extracts which have effect on the DNA proliferation of hair dermal papilla cell measured by [³H]thymidine incorporation, the expression of growth factors such as IGF-I, KGF, HGF estimated by RT-PCR and protein synthesis of vibrissae hair follicle measured by [/sup 35/S] cysteine incorporation. Cortex Cinnamomi showed anti-bacterial effect on P. ovale, Radix Paeoniae Alba has the highest radical scavening activity and Radix Glycyrrhizae has the highest effects of NO synthesis. These results suggest that Gamissanghwa-tang can be used as a potent treatment agent for helping hair growth stimulation.

Identification of Heterodera glycines (Tylenchida; Heteroderidae) Using qPCR

  • The soybean cyst nematode, Heterodera glycines, is a major plant-parasitic nematode that has caused important economic losses to Korea's soybean production. Four species of cyst nematodes, H. schachtii, H. glycines, H. trifolii, and H. sojae, all belong to schachtii group are coexist in field soil in Korea. The rapid identification of the nematode is crucial for preventing crop damage and in decision making for controlling this nematode. This study aimed to develop a species-specific primer set for quantitative PCR (qPCR) assay of H. glycines. The specific primer set (HGF1 and HGR1) for H. glycines was designed based on the cytochrome c oxidase subunit I (COI) sequence of mitochondrial DNA. After optimization, it is possible to identify the H. glycines using a qPCR assay with DNA extracted from a single cyst and single second-stage juvenile (J2). The specificity was confirmed by the absence of SYBR fluorescent signals of three other Heterodera species. A serial dilution of DNA extracted from a single cyst was obtained for the sensitivity test. The result showed that the standard curve of the test had a highly significant linearity between DNA concentration and Ct value (R2 = 0.996, slope = -3.49) and that the detection limit concentration of DNA of the primer set was 10 pg of DNA per reaction. Our findings suggested that H. glycines could be distinguished from H. sojae and other Heterodera species when a qPCR assay is used with a specific primer set.

Cripto Enhances Proliferation and Survival of Mesenchymal Stem Cells by Up-Regulating JAK2/STAT3 Pathway in a GRP78-Dependent Manner

  • Cripto is a small glycosylphosphatidylinositol-anchored signaling protein that can detach from the anchored membrane and stimulate proliferation, migration, differentiation, vascularization, and angiogenesis. In the present study, we demonstrated that Cripto positively affected proliferation and survival of mesenchymal stem cells (MSCs) without affecting multipotency. Cripto also increased expression of phosphorylated janus kinase 2 (p-JAK2), phosphorylated signal transducer and activator of transcription 3 (p-STAT3), 78 kDa glucose-regulated protein (GRP78), c-Myc, and cyclin D1. Notably, treatment with an anti-GRP78 antibody blocked these effects. In addition, pretreatment with STAT3 short interfering RNA (siRNA) inhibited the increase in p-JAK2, c-Myc, cyclin D1, and BCL3 levels caused by Cripto and attenuated the pro-survival action of Cripto on MSCs. We also found that incubation with Cripto protected MSCs from apoptosis caused by hypoxia or $H_2O_2$ exposure, and the level of caspase-3 decreased by the Cripto-induced expression of B-cell lymphoma 3-encoded protein (BCL3). These effects were sensitive to down-regulation of BCL3 expression by BCL3 siRNA. Finally, we showed that Cripto enhanced expression levels of vascular endothelial growth factor (VEGF), fibroblast growth factor (FGF), and hepatocyte growth factor (HGF). In summary, our results demonstrated that Cripto activated a novel biochemical cascade that potentiated MSC proliferation and survival. This cascade relied on phosphorylation of JAK2 and STAT3 and was regulated by GRP78. Our findings may facilitate clinical applications of MSCs, as these cells may benefit from positive effects of Cripto on their survival and biological properties.
