Epidermis Proliferative Effect of the Panax ginseng Ginsenoside $Rb_2$

  • Ginseng has been used as a traditional medicine with various therapeutic effects. However, it is still unknown which component of this plant is effective at promoting wound healing. Recently, ginsenoside $Rb_2$ has been reported to improve wound healing. In this study, to investigate the reported wound healing effect of the ginsenoside $Rb_2$, cell morphology and protein factors involved in epidermal formation were evaluated by immunshemical and immunoblotting analysis. $Rb_2$ stimulated epidermal cell proliferation, and the cell showed a 1.5-fold increase in thymidine uptake compared to the control (p<0.05, n=3). Futheremore $Rb_2$, was found to stimulate epidermis formation in a dose-dependent manner in raft culture, and to dose dependently enhance the expressions of protein factors related to cell proliferation, namely, epidermal growth factor and its receptor, fibronectin and its receptor, keratin 5/14, and collagenase 1 (p<0.05, n=3~9). It is believed that ginsenoside $Rb_2$, enhances epidermal cell proliferation by upregulating the expressions of these proliferation-related factors.

  • In recent years, self-healing coatings have been the subject of increasing interest. The ability of such coatings to self-repair local damage caused by external factors is a major factor contributing to their attractiveness. Metals are extensively used in modern society in a range of applications from infrastructure to aircraft to consumer products. The protection of metals, primarily from corrosion has been an active area of materials science for many years. The aim of this review is the demonstration for recent progress achieved in the development of carrier-based self-healing coatings for the protection of metals. This review mainly covers the reports published after 2010. Two main types of carriers for corrosion inhibitors or healing agents-polymer capsules and porous composite inorganic nanoparticles-are described.

농촌 치유관광객의 여가활동 참여, 심리적 자본, 삶의 질의 구조관계 분석 (A Structural Relationships among Leisure Activity, Psychological Capital, and Quality of Life in Rural Healing Tourists)

  • This study aims to examine the structural relationships among leisure activity, psychological capital, and quality of life in rural healing tourists. Data were collected from 407 workers who had experience in rural healing tourism through an online survey. SPSS 28.0 and AMOS 28.0 were used for frequency, confirmatory factor, and structural equation modelling analysis. The results of structural equation modelling indicate participation in leisure activities had a positive effect on psychological capital. Psychological capital had a positive effect on quality of life. The results of this study suggest that leisure activities play an important role in improving workers' psychological capital. Through rural healing tourism, various outdoor activities, cultural activities, and social interaction with people using rural resources can help improve the psychological capital of workers.

피부(皮膚) 창상(創傷) 동물모델에서 흑삼(黑蔘) 열수 추출물 경구 투여의 효과 (Effects of Oral Administered Hot Water Extracts of Korean Black Ginseng on Wound Healing in Mice)

  • Objectives This study aims to evaluate the wound healing effects of oral administered hot water extracts of Korean black ginseng (KBG). Methods 40 C57BL/6 mice were divided into five groups; normal, control, vitamin E 200 mg/kg, KBG 100 mg/kg, KBG 200 mg/kg, each n=8. Skin wounds were made in the back of all mice except normal group using biopsy punches. Wounds were observed on days 7 and 14 after injury. The anti-oxidant and inflammatory protein levels were evaluated using western blotting. Skin tissue was analyzed by hematoxylin & eosin and Masson's trichrome staining method. Results KBG significantly accelerated reducing wound area. KBG significantly decreased myeloperoxidase activity. KBG significantly decreased oxidative stress factors such as NADPH oxidase-4 and p22phox and increased antioxidant enzymes including nuclear factor erythroid 2-related factor2, kelch-like ECH-associated protein-1, heme oxygenase-1, superoxide dismutase, catalase and glutathione peroxidase-1/2. Moreover, KBG significantly decreased inflammation factors including nuclear factor-κB, phosphorylated inhibitor of κBα, cyclooxygenase-2, inducible nitric oxide synthase, tumor necrosis factor-α and interleukin (IL)-6 and increased anti-inflammation cytokine such as IL-4 and IL-10. In addition, KBG significantly increased tight junction proteins including claudin-1, claudin-3, claudin-4. In histopathologic, KBG made the epithelium thin and uniform, and accelerated the remodeling of collagen. Conclusions The results suggest that KBG has healing effects on skin wound in mice by anti-inflammatory and antioxidant activity.

저산소 및 재산소화가 배양된 태아 섬유아세포에 미치는 영향 (Effect of Hypoxia and Reoxygenation on Cultured Human Dermal Fetal Fibroblast)

  • The wound healing process in fetus is quite different form that of adult. Regeneration plays an important role and scarless wound healing is possible in early gestational fetal period. Recently, the various effects of the hypoxia and reoxygenation in the wound healing process have been investigated by many researchers. The hypoxic state is known to alter protein synthesis and gene expression of TGF-${\beta}$, VEGF. The authors hypothesize there may be differences between fetal and adult fibroblast and this difference may play a possible role in the mechanism of scarless fetal wound healing. In this study, we investigated the growth of fibroblast, the amount of collagen deposition, the amount of protein synthesis and gene expression in TGF-${\beta}$(transforming growth factor-${\beta}$), VEGF(vascular endothelial growth factor) under the various hypoxic and reoxygenation conditions. Through these processes, we tried to determine the relationships between scarless fetal wound healing and hypoxic condition. In control group, fetal and adult fibroblasts were cultured under normoxic condition. The experimental groups were allocated into four different groups. The differences in TGF-beta, VEGF under 24, 48, 72 hours were statistically investigated. Compared to adult fibroblast group, there was a statistically significant increase (p<0.01) in the rates of protein synthesis in TGF-beta and VEGF of fetal fibroblast. In this study, these results may reflect the possibility that fetal fibroblast are more susceptible to change in oxygen and has a superior rate of angiogenesis through increased VEGF expression. The possible superiority of angiogenesis in fetal fibroblast may play an important role in scarless wound healing.

The effect of Korean Red Ginseng on full-thickness skin wound healing in rats

  • Background: Panax ginseng is regarded as one of the best compounds for promoting health, and it has been used traditionally as a medicinal herb. Recently, Korean Red Ginseng (RG) has been shown to protect skin from aging and wrinkling; it can also relieve atopic dermatitis and allergy symptoms. This study aimed to evaluate RG's effects on the regeneration of the full-thickness skin wounds in rat. Methods: Full-thickness skin wounds were generated in rats, and then RG was administered either orally or topically. The wound-healing effects of RG were investigated by assessing wound size, mRNA expression patterns of genes related to wound healing, histological staining, and measurements of lipid, moisture, and elasticity in skin tissues. Results: The wound size was smaller, and tissue regeneration rate was faster in the RG-treated group than that in the control group on days 15 and 20 after initiating treatment. On postoperative day 20, skin lipid and moisture content had increased significantly in the RG-treated group. Significant increases in the gene expression levels of transforming growth $factor-{\beta}1$ and vascular endothelial growth factor were found in the RG group during the early stages of wound healing. Matrix metalloproteinase-1 and matrix metalloproteinase-9 showed significant increases in gene expression levels on day 20. Conclusion: The results suggested that RG may promote healing of full-thickness skin wounds in rats. They also provided basic insights into the effects of RG on skin regeneration, supporting its use as a dressing material for wound treatment and its development as a functional food.

Rebalancing SMAD7/SMAD3 Signaling Reduces Adhesion Formation during Flexor Tendon Healing

  • Transforming growth factor-β is a key factor in regulating adhesion formation during tendon healing. We investigated the effectiveness of SMAD family members, SMAD7 and SMAD3, in the TGF-β/Smad signaling during flexor tendon repair. Mouse flexor toe deep tendon rupture anastomosis models were made. On days 3, 7, 14, 21, and 28, the expressions of smad7 and smad3 in flexor tendon tissues were detected by RT-qPCR and western blot. Furthermore, postoperative intraperitoneal injections of SMAD7 agonists or SMAD3 antagonists were given. The degree of tendon healing was evaluated by adhesion testing and biomechanical experiments. Hematoxylin and eosin (HE) staining was used to observe the pathological changes. Immunohistochemistry was used to evaluate the expressions of collagen III, SMAD3, and SMAD7. The mRNA levels of matrix metalloproteinases, Mmp2 and Mmp9, and scleraxis (SCX) in flexor tendon tissue were detected by RT-qPCR. Smad3 expression increased and Smad7 expression decreased in flexor tendon tissue after injury. In addition, the SMAD7 agonist blocked SMAD3 phosphorylation. SMAD7 agonist and SMAD3 antagonist both improved adhesion formation during flexor tendon healing, and decreased the expressions of collagen III, Mmp9, and SCX, while increasing Mmp2 expression. This study provides a possible theoretical basis for the SMAD7-SMAD3 signal cascade during flexor tendon adhesion healing.

Holistic Therapy 프로그램의 개발과 적용한 행복힐링모임 대상자들의 스트레스 변화 (Development of Holistic Therapy Program and Changes in Stress of Happy Healing Group Subjects)

  • 목적: 본 연구의 목적은 새로 개발한 '전인 치유 프로그램'을 행복힐링모임을 통해 수행한 대상자 62명의 스트레스 변화'에 대한 연구를 하는 것이었다. 방법: 전인치유 프로그램을 행복힐링모임에 주당 1회(90분)씩 6회기를 수행하였고, 회기 및단 계별내용을 기록하였다. 결과: 일반인에게 적용할 수 있도록 개발한 전인치유프로그램은 회기별로 '도입-전개-마무리'의 세부적 3단계로 구성되었다. 전인치유요법으로 행복힐링모임 수행 후에 스트레스에 미치는 요인은 사전 평균치에 비해 사후의 평균치는 26.371점으로 유의성이 있게 감소하였다(p < .000). 사회심리적 스트레스의 변화는 사전에서는 34.306점, 사후에는 9.096점으로유의하게감소하였다. 스트레스 대처방식의 변화는 사전에 비해서사후에는 201.661점으로 유의하게 높게 나타났다. 기분 상태 변화는 사전에는 69.451점이었고사후에는 123.983점으로 높게 증가하여 유의한 차이를 보이었다(p < .000). 결론: 전인치유 프로그램을 행복힐링모임에 적용한 대상자에게서는 일반 또는 사회적심리적 스트레스에는 유의하게 감소하였고, 스트레스 대처방식과 기분상태의 변화는 높게 증가하는 경향을 보이었다. 이 결과로 평가할 때에 새 전인 치유요법이 행복힐링모임에 매우 유용한 자연치유적 요법 중의 하나라고 판단되었다.

자연치유와 전인적 의학(Holistic Medicine)으로서의 단식요법(斷食療法)에 관한 고찰 (Consideration on Fasting Cure as Natural Healing and Holistic Medicine)

  • 본 연구에서는 자연치유와 전인적 의학으로서의 단식요법에 대하여 고찰하고자 한다. 연구방법으로는 종교사상에서 나타난 단식의 역사와 의의를 찾아 문헌적으로 고찰하였으며, 단식이 심신치유에 적용된 사례를 연구, 분석하였다. 연구결과 첫째, 단식은 종교사상에서 정신적인 중대한 역할을 함으로 단식행위가 인간의 삶에서 정신적으로 미치는 영향력이 큰 것으로 나타났다. 둘째, 단식은 인간의 심신을 치유하는 수단으로서 오랜 과거로부터 오늘날에까지 이어져 내려오는 자연치유적 의료행위로 중대한 역할을 하고 있는 것으로 나타났다. 결과적으로 단식요법은 자연치유적 방법에 의한 전인적 의학으로서 중요한 가치를 가진다는 점을 알 수 있었다. 본 연구의 시사점은 단식요법을 통해 현대사회에 각종 질병의 요인으로 작용하는 스트레스성 질병과 생활습관성 만성질환에 대하여 전인적 의학 차원에서의 효율적 치유의 방향과 근거를 제시하는데 기초 자료가 될 것으로 사료된다.

소아병동의 치유적 휴게공간에 관한 연구 (A Study on the Healing Rest Space in Pediatric Ward)

  • Purpose: Many researches have clearly begun to address the relationship between people and their physical environments. These represent that children experience traditional hospital environments as unpleasant and scary. More specifically, children in hospitals face many psychological and emotional challenges: isolation, painful procedures, separation from family members, all in unfamiliar settings. For this reason, some have argued that the children's experiences within hospital environments can lead to poorer health outcomes. This study has been started in order to provide basic informations for the planning of Healing rest space in pediatric ward. Methods: Literature reviewes and field surveys to rest space at pediatric ward in Korea and foreign country have been conducted for the data collection. Results: The result of this study can be summarized into eight environment factor for healing rest space in pediatric ward: family, noise, safe, choice opportunity, light, privacy, social support and distract from disease or death. Implications: In the rest space in pediatric ward, it is necessary to consider healing rest space for promoting healing from disease and normal developing of children.
