Chrysin과 emodin에 의한 대장암 세포 항 성장 활성 및 세포사멸 (Anti-proliferative Effects and Apoptosis Induced by Chrysin or Emodin in Human Colorectal HCT116 Cells)

  • 본 연구에서는 시판하는 천연물 library (Selleckchem, L1400)로부터 암세포 항 성장 활성을 보여주는 천연물을 선별하였다. 즉, 인간 대장암 세포주인 HCT116에 50 μM의 각 천연물을 처리한 후 세포 생존율을 측정하였다. 1차 선별과정을 통하여 5종의 천연물(chrysin, diosmetin, emodin, piperlongumine, tanshinone I)을 선별하였다. 5종의 천연물에 의한 NAG-1 단백질의 발현을 확인한 결과 chrysin과 emodin에 의해서 발현이 현저하게 증가하였다. 또한, chrysin과 emodin은 농도의존적으로 세포 생존율을 감소시켰으며, chrysin과 emodin은 항암 단백질인 NAG-1의 발현을 농도 및 시간 의존적으로 증가시켰다. 게다가, chrysin과 emodin 처리에 의해 증가된 PARP cleavage가 NAG-1 siRNA transfection에 의해서 감소됨을 확인함으로써, chrysin과 emodin에 의한 세포사멸과 NAG-1의 발현 증가가 직접적인 관련이 있음을 증명하였다. 따라서, 본 연구결과는 암세포 항 성장 활성을 보여주는 천연물 선별에 대한 기초 데이터를 제공해 주며, chrysin과 emodin에 의한 암세포 항 성장 활성 및 세포사멸의 기전을 이해하는 데 도움을 줄 것으로 판단된다.

L-carnitine에 의한 인간대장암세포주 증식억제 및 산화적손상 기전 규명 (The Anti-Proliferation and Oxidative Damage-Related Mechanism of L-Carnitine in Human Colorectal Cancer Cells)

  • L-carnitine은 라이신과 메티오닌으로 생합성되며 골격근과 심근을 포함한 다양한 동물조직에서 발견된다. L-carnitine이 포함된 식품으로는 양고기, 소고기, 돼지고기 등이 있고 근육발달에 도움을 주며 뼈를 강화하거나 대사작용을 도와주는 기능을 하여 영양 보조제로 많이 섭취하는 것으로 알려져 있다. 최근 L-carnitine은 제 2형 당뇨병, 골다공증, 대사성 신경증후군 등의 다양한 질병의 약물로도 연구 되고 있으며 암에서는 치료 보조제로 개발되어있다. 하지만 대장암에서의 L-carnitine에 대한 효과 및 기전에 대해서는 명확하지 않고 연구된 바가 없기 때문에 본 연구에서 저자들은 L-carnitine의 효능을 인간대장암세포주 HCT116에서 규명하고자 하였다. L-carnitine은 세포 내 활성산소종 (ROS)를 높은 수준으로 증가시켜 세포 증식을 억제하였다. 또한, 세포 증식과 죽음에 관련한 단백질 ERK1/2와 p38을 유의적으로 활성화 시킨다는 것을 입증하였다. 이때, ERK1/2 억제제(PD98059)를 처치하여 ERK1/2의 활성화가 활성산소종 발생 및 세포사멸에 중요하다는 것을 밝혔다. 따라서, 본 연구 결과는 L-carnitine이 대장암세포주의 증식을 억제 할 수 있고 이는 대장암의 치료에 있어 잠재적인 치료 물질이 될 수 있음을 시사하며 이 과정에 관여하는 신호전달기전을 조사하여 항암의 치료기전에서 활성산소종이나 ERK1/2, p38 단백질의 활성화의 중요성을 제시하였다.

감태나무 뿌리 추출물에 의한 대장암세포의 성장억제 및 세포사멸유도 (Anti-proliferative and Apoptotic Activity of Extracts of Lindera glauca Blume root in Human HCT116 Colorectal Cancer Cells)

  • Lindera glauca Blume has been used in Korean traditional medicine to treat the symptoms of paralysis, abdominal pain, speech disorders, extravasations, contusions, and pain caused by rheumatoid arthritis. We investigated the effect of L. glauca Blume extracts on the proliferation of colorectal cancer cells in vitro using HCT116 human colorectal cancer cell lines. We also investigated its mechanism of action. For this purpose, we used the MTT assay, western blotting, DNA fragmentation analysis, and flow cytometry. HCT116 cells were cultured in several concentrations of ethanol extracts of L. glauca Blume root (0, 50, 100 ㎍/mL). In this study, colon cancer cell growth was inhibited by L. glauca Blume root extract in a dose-dependent manner. It was associated with induction of apoptosis as assessed by nuclear fragmentation and cell cycle analysis. Apoptosis was assessed using western blotting for TNF-α, IL-6, NF-κB, Caspase-3, PARP, Bax, Bcl-2, and SIRT1. The extract also dose-dependently upregulated the expression Bax, the pro-apoptotic gene and downregulated the expression of the anti-apoptotic gene Bcl-2. Furthermore, the extract enhanced Caspase-3 activity in a dose-dependent manner. Our findings provide evidence that L. glauca Blume extract may mediate its anti-proliferative effect via the modulation of apoptosis.

Preliminary Research on the Expression, Purification and Function of the Apoptotic Fusion Protein, Sival

  • The objective of the present study was to investigate cloning, expression, and functions of the recombinant protein, Siva1. Siva1 gene was synthesized by RT-PCR from HCT116 cells. Plasmids were cleaved with the restriction endonuclease, BamH1/Sal1 and products were connected to pQE30, which underwent cleavage by BamH1/Sal1. The recombinant plasmid, pQE30-Siva1, was identified after digestion with restriction endonucleases followed by transformation into E. coli M15. Expression of Siva1 was induced by IPTG and identified by SDS-PAGE following purification with affinity chromatography. The results showed that size of Siva1 was 12 kDa, consistent with the molecular weight of the His-Siva1 fusion protein. Functional test demonstrated that Siva1 significantly inhibited the invasion and migration of HCT116 cells. It may thus find clinical application for control of cancers.

Inhibitory Effect of Snake Venom Toxin on Colorectal Cancer HCT116 Cells Growth through Induction of Intrinsic or Extrinsic Apoptosis

  • I investigated whether snake venom toxin(SVT) from Vipera lebetina turanica enhances the apoptosis ability of tumor necrosis factor(TNF)-related apoptosis-inducing ligand(TRAIL) in cancer cells. TRAIL inhibited HCT116 cell growth in a dose-dependent manner. Consistent with cell growth inhibition, the expression of TRAIL receptors; DR4 and DR5 was significantly increased as well as apoptosis related proteins such as cleaved caspase-3, 8, 9 and Bax. However, the expression of survival proteins(eg, cFLIP, survivin, XIAP and Bcl2) was suppressed by the combination treatment of SVT and TRAIL. Pretreatment with the reactive oxygen species(ROS) scavenger N-acetylcysteine reduced the SVT and TRAIL-induced upregulation of DR4 and DR5 expression and expression of the apoptosis related protein such as caspase-3 and-9 as well as cell growth inhibitory effects. The collective results suggest that SVT facilitates TRAIL-induced apoptosis in human colorectal cancer HCT116 cells through up-regulation of the TRAIL receptors; DR4 and DR5 via ROS pathway signals.

들깨 추출물의 항산화 활성과 암세포 기본 특성에 대한 억제 효과 (Antioxidant Activities of Perilla frutescens Britton Seed Extract and Its Inhibitory Effects against Major Characteristics of Cancer Cells)

  • 본 연구에서는 들깨 에탄올 추출물의 항산화 성분 함량 및 활성을 측정하고 들깨 추출물이 암세포의 주요 특징인 성장, colony 형성, 이동, 부착성 등에 미치는 영향을 in vitro 수준에서 분석하고자 하였다. 들깨의 총 폴리페놀 함량과 총 플라보노이드 함량은 각각 222.6 mg gallic acid equivalent/100 g과 285.7 mg quercetin equivalent/100 g으로 측정되었다. 들깨 추출물($87.5{\sim}350{\mu}g/mL$)의 DPPH radical 소거 활성은 24~45%, 철 환원력은 28~62%였다. 한편 $87.5{\sim}350{\mu}g/mL$ 농도의 들깨 추출물은 HCT116 대장암 세포와 H1299 폐암 세포의 성장을 18~94% 억제하는 농도 의존적 활성이 있었다. 또한 $175{\mu}g/mL$ 농도의 들깨 추출물은 HCT116과 H1299 세포에서 모두 colony 형성을 완전히 억제하는 활성이 있었다. 또한 들깨 추출물은 $87.5{\sim}350{\mu}g/mL$ 농도에서 H1299 세포의 이동을 30~37% 억제하였고, 가장 높은 농도인 $350{\mu}g/mL$ 농도에서는 HCT116과 H1299 세포의 부착을 14~16% 억제하였다. 이상의 결과를 통하여 들깨 추출물은 항산화 활성 및 암세포의 주요 특징을 억제하는 활성을 가지는 것으로 생각되며, 향후 이러한 연구 결과가 in vivo 수준에서 재현되는지 여부와 본 활성과 관련된 세부작용기전 또한 규명되어야 할 것으로 생각된다.

Doxorubicin에 의한 내인성 산화질소가 인간 대장암 세포주에서의 세포사멸에 미치는 효과 (Endogenous Nitric Oxide Strengthens Doxorubicin-induced Apoptosis in Human Colorectal Cell Lines)

  • Doxorubicin은 광범위한 암을 치료하는데 사용되는 일반적인 항암제이지만, 내인성 산화질소 생성량과 Doxorubicin의 항암 효과의 상관 관계에 대해서는 아직 명확하게 밝혀지지 않았다. 본 연구에서는 인간 대장암 세포에서 Doxorubicin의 항암 활성에 내인성 산화질소가 미치는 영향을 확인하고자 하였다. HCT116 (p53-WT)과 HT29 (p53-MUT) 세포에서 Doxorubicin 처리에 의해 세포 생존율의 차이를 보였으며, NMA 병행처리는 Doxorubicin의 효과를 감소시켰음을 확인할 수 있었다. 추가 연구를 통해 HCT116과 HT29 세포에서 sub-$G_1$ 기의 세포 빈도와 DNA 단편화의 결과를 통해 내인성 산화질소가 Doxorubicin에 의한 apoptosis를 조절하는 것을 확인하였다. 이러한 결과는 인간 대장암 세포에서 내인성 산화질소와 IAP 발현, p53의 상태에 따른 조절이 Doxorubicin에 의해 유도된다는 것을 보여주며, 이러한 메커니즘은 대장암에서 화학요법의 효율을 향상시키기 위한 전략적인 표적으로 이용할 수 있을 것으로 생각된다.

Chemotherapeutic Candidate Inducing Immunological Death of Human Tumor Cell Lines

  • The immunological death induction by EY-6 on the human tumor cell lines was screened. Human colon carcinoma (HCT15, HCT116), gastric carcinoma (MKN74, SNU668), and myeloma (KMS20, KMS26, KMS34) cells were died by EY-6 treatment with dose-dependent manner. CRT expression, a typical marker for the immunological death, was increased on the EY-6-treated colorectal and gastric cancer cells. Interestingly, the effects on the myeloma cell lines were complicated showing cell line dependent differential modulation. Cytokine secretion from the EY-6 treated tumor cells were dose and cell-dependent. IFN-${\gamma}$ and IL-12 secretion was increased in the treated cells (200% to over 1000% of non-treated control), except HCT116, SNU668 and KMS26 cells which their secretion was declined by EY-6. Data suggest the potential of EY-6 as a new type of immuno-chemotherapeutics inducing tumor-specific cell death. Further studies are planned to confirm the efficacy of EY-6 including in vivo study.

Vinpocetine inhibits the proliferation and induces apoptosis in human colon cancer cells

  • Vinpocetine induces anti-inflammatory effects in various inflammatory diseases via the inhibition of phosphodiesterase type-1-independent nuclear factor-κB signaling pathway and the release of inflammatory cytokines. In this study, we investigated the effect of vinpocetine on the proliferation of colon cancer cells and its underlying molecular mechanisms. Our data showed that vinpocetine inhibits the viability and proliferation of colon cancer cells. Vinpocetine treatment induced cell death in HCT116 cells, which the percentages of sub-G1 phase were significantly increased, and the apoptosis-related genes were regulated after HCT116 cells were treated with vinpocetine. In sum, our findings indicated that vinpocetine could be a therapeutically useful candidate in the treatment of colon cancer.

Calcium Signaling of Dioleoyl Phosphatidic Acid via Endogenous LPA Receptors: A Study Using HCT116 and HT29 Human Colon Cancer Cell Lines

  • In the present study, we have tested the effect of dioleoyl phosphatidic acid (PA) on intracellular $Ca_{2+}$ concentration ($[Ca^{2+}]_{i}$) in two human colon cancer cell lines (HCT116 and HT29). PA and lysophosphatidic acid (LPA), a bioactive lysolipid, increased $[Ca^{2+}]_{i}$ in both HCT116 and HT29 cell lines. Increases of $[Ca^{2+}]_{i}$ by PA and LPA were more robust in HCT116 cells than in HT29 cells. A specific inhibitor of phospholipase C (U73122), however, was not inhibitory to the cell responses. Pertussis toxin, a specific inhibitor of $G_{i/o}$ type G proteins, however, had an inhibitory effect on the responses except for an LPA-induced one in HT29 cells. Ruthenium red, an inhibitor of the ryanodine receptor, was not inhibitory on the responses, however, 2-APB, a specific inhibitor of inositol 1,4,5-trisphosphate receptor, completely inhibited both lipid-induced $Ca^{2+}$ increases in both cell types. Furthermore, by using Ki16425 and VPC32183, two structurally dissimilar specific antagonists for $LPA_{1}/LPA_{3}$ receptors, an involvement of endogenous LPA receptors in the $Ca^{2+}$ responses was observed. Ki16425 completely inhibited the responses but the susceptibility to VPC32183 was different to PA and LPA in the two cell types. Expression levels of five LPA receptors in the HCT116 and HT29 cells were also assessed. Our data support the notion that PA could increase $[Ca^{2+}]_{i}$ in human colon cancer cells, probably via endogenous LPA receptors, G proteins and $IP_{3}$ receptors, thereby suggesting a role of PA as an intercellular lipid mediator.
