Influence of Incorporation Rates of Sulfur on Reduction of the Bicarbonate Injury in Hydroponic Cultivation of 'Ssanta' Strawberry (유황 처리수준이 '싼타' 딸기의 중탄산 피해 경감에 미치는 영향)

  • This research was conducted to determine the influence of incorporation levels of sulfur into a coir dust+pine bark medium (1:1, v/v) on the changes in the bicarbonate ($HCO_3{^-}$) concentrations and pH of soil solution, crop growth, and nutrient uptake of 'Ssanta' strawberry. In the preparing of the mixed medium, sulfur powder was added with the rate of 0 (control), 0.23, 0.45, 0.90, and $1.80g{\cdot}L^{-1}$ and Hoagland nutrient solution containing $240mg{\cdot}L^{-1}$ $HCO_3{^-}$ was supplied during the crop cultivation. The growth measurements and tissue analysis for the determination of nutrient contents were carried out 140 days after solution application and the soil solution analysis was performed every two weeks. As the level of sulfur was elevated, the overall growth of mother plants became better showing that the growth indexes except chlorophyll contents were the lowest in control treatment but the statistical differences were not found among the three treatments of $0.45g{\cdot}L^{-1}$ or higher sulfur. The higher the levels of sulfur incorporation, the higher the occurrence of runners and the growth of daughter plants. The length of the runners and the number of daughter plants occurred per mother plants were higher in the treatments of 0.90 and $1.80g{\cdot}L^{-1}$ than the three treatments of 0, 0.23, and $0.45g{\cdot}L^{-1}$, but the statistical differences were not observed between the 0.90 and $1.80g{\cdot}L^{-1}$ treatments. The rose of pH and $HCO_3{^-}$ concentrations in soil solution of root media continued all the cropping period, but those decreased slightly in the treatments of $0.90g{\cdot}L^{-1}$ or higher. The soil solution concentrations of $K^+$ and $PO_4{^3-}$ in the treatments of 0.90 and $1.80g{\cdot}L^{-1}$ was lower than those in other treatments and the statistical differences in the $Ca^{2+}$ and $Mg^{2+}$ concentrations were not observed among all treatments. The nutrient contents in tissue analyzed in this experiment were the lowest in the control treatments and those increased as incorporation rates of sulfur were elevated. Above results indicated that when 'Ssanta' strawberry is grown hydroponically and the root medium is coir dust and the pine bark (5:5, v/v) mix, the sulfur incorporation rate as pre-planting fertilizer has to be higher than $0.9g{\cdot}L^{-1}$ root medium to reduce the $HCO_3{^-}$ injury.

  • The structure of rapidly solidified Al-Si-based alloys and its relationship to subsequent anodic film growth in near neutral and acid solutions have been investigated. Solidification of the alloys proceeds via pre-dendritic nuclei, associated with rugosity of the casting surface, from which cellular-type growth, comprised of aluminium-rich material surrounded by silicon-containing material, emanates. Observation of ultramicrotomed sections of the alloys and their anodic films reveals the local oxidation of the silicon-rich phase and its incorporation into the anodic alumina film, formed in near neutral solutions. Such incorporation occurs but resultant isolation of the silicon-rich phase is not possible for anodizing in phosphoric acid, and a three-dimensional network of the oxidized silicon-containing phase, with continuing development of porous anodic alumina, is observed.

Photoacoustic Spectroscopic Study on Cobalt Incorporation onto the Surface of Mesoporous Molecular Sieves

  • The incorporation of cobalt into mesoporous molecular sieves MCM-41 and MCM-48 was carried out. Co-PO/MCM41 and Co-PO/MCM48 were prepared using Co(II) acetate solution adjusted to pH = 3.0 with phosphoric acid by the incipient wetness method. Photoacoustic spectroscopy (PAS) was used to study the local environments of Co(II) incorporated into mesopores. The band around 500 nm in PAS of as-prepared Co-PO/MCM41 and Co-PO/MCM48 with Co(II) acetate solution was changed to triplet bands around 600 nm. This could be assigned to the 4 A2(F)-> 4T1(P) transition of Co(II) surrounded tetrahedrally by oxygen ions after calcination. It may be attributable to that the octahedral cobalt species containing phosphate ligands in coordination sphere reacting with framework's silanol groups to be dispersed atomically onto the surface of mesoporous molecular sieves as a tetrahedral species. This is unlike that the Co in Co-Cl/MCM41 and direct-synthesized Co-MCM41 transforms to Co oxide phase upon calcination. Co-PO/MCM41 and Co-PO/MCM48 were stable while treated with water.

Expression of Cinnamic Acid 4-Hydroxylase Chimeric Gene fused with Sesquiterpene Cyclase Promoter from Hot Pepper in Tobacco (고추의 sesquiterpene cyclase promoter-cinnamic acid 4-hydroxylase chimeric gene의 담배에서 발현)

  • Tobacco transformants harboring cinnamic acid 4-hydroxylase gene (C4H) fused with susquiterpene cyclase promoter was developed in order to regulate biosynthesis of phenolic compounds by the expression of the introduced gene. Twenty transformants for each specific promoter were used to analyze the incorporation of the chimeric genes by PCR and Southern blot analysis. PCR products of NPTII(neomycin phosphotransferase) gene (553bp) were detected in the transgenic tobacco plants. The incorporation of the chimeric gene was confirmed in the Southern blot analysis. C4H activity in the transgenic plants was elevated by UV-irradiation and its level was higher compared to that of control plants.

Immunomodulatory Effects of Eckol, a Pure Compound of Ecklonia Cava, on Dendritic Cells

  • Background: Eckol purified from Ecklonia cava, a brown alga has been known to have cytoprotective effects on some cell lines against oxidants and ionizing radiation. However, there is no study about the effects of eckol on immune cells. Methods: Bone marrow (BM)-derived dendritic cells (DCs) were used to demonstrate the immunomodulatory effects of eckol on DCs, such as viability, the expression of surface markers, allogeneic stimulating capacity using MTI, flow cytometric, $^3H$-thymidine incorporation assay. Results: Eckol did protect DCs against cytokine withdrawal-induced apoptosis in a concentration dependent manner based on MTT assay. And also, it increased the expression of MHC class II and CD86 (B7.2) molecules, maturation markers, on the surface of viable DCs gated in FACS analysis. Furthermore, eckol-treated DCs stimulated the proliferation of allogeneic $CD4^+$ T lymphocytes compared to imDCs in $^3H$-thymidine incorporation assay. $CD4^+$ T lymphocytes activated with eckol-treated DCs produced the larger amount of IFN-${\gamma}$ and IL-4 than those cells with imDCs. Conclusion: Taken together, we demonstrate in this study that eckol, a pure compound of Ecklonia cava, may modulate the immune responses through the phenotypic and functional changes of DCs.

Functional Properties of Bifidobacterium longum and Their Incorporation into Cheese Making Process (비피도박테리움 롱검의 기능성과 치즈 제조에 활용)

  • Members of the genus Bifidobacterium are prevalent in the human colon and represent up to 90% of all bacteria in fecal samples of breast-fed infants, and 3~5% of adult fecal microbiota. Bifidobacteria produce organic acids, thus reducing the colon pH to a level inhibitory for pathogenic bacteria. They can also detoxify a number of toxic compounds and adhere to the colon mucosa, thus preventing the adherence of pathogens and induction of colon cancer. Recently, we identified a novel Bifidobacterium longum subsp. longum strain, KACC 91563, in a fecal sample of a Korean neonate, and demonstrated its functional properties. We showed that B. longum KACC 91563 alleviates food allergy through mast cell suppression and produces antioxidative and antihypertensive peptides by casein hydrolysis. Dairy products are considered as an ideal food system for the delivery of probiotic cultures to the human gastrointestinal tract. Cheese affords protection to probiotic microbes during gastric transit due to its relatively high pH, more solid consistency, higher fat content, and higher buffering capacity. Incorporation of B. longum KACC 91563 into cheese making is currently under study.

Distribution of bacterial biomass in the water column of Soyang lake (소양호 수중 생태계에서의 세균 생체물질량의 분포)

  • Microbiological parameters such as bacterial biovolume and biomass in Soyang Reservoir was statistically analyzed with the physico-chemical enviromental factors. Analysis of correlation and multiple regression showed that temperature affects most of microbiological parameters. Variations of total bacterial number, total bacterial biovolume and saprophyte number were highly correlatd with the concentrations of chlorophyll a and pheophytin a. Bacterial production by the $^{3}H$-thymidine incorporation rate was largely affected by Seston. It suggests that microbiological factors such as bacterial biovolume and bacterial biomass were controled by the concentration of seston and distribution of phytoplankton which acts as carbon and energy source for the bacterial community in the water column of Soyang Reservoir.

  • River restoration has recently been performed not only for the improvement of the artificial parts in the past but also for the restoration of abandoned river reaches which were blocked and isolated. For the restoration of abandoned river reaches, it is important to recover the hydraulic and ecological connectivity in the isolated space by longitudinal structures like levees. But because the assessment tools to determine whether the river restoration is performed properly are so rare at present, we aim to provide a tool for assessing ecological connectivity in a target river in this study. In the first step, one-dimensional numerical model for rainfall-runoff and channel routing was developed and then applied to the watershed of the Cheongmi Stream. In this step, a numerical model was developed to assess the restoration of connectivity. The model consists of two parts: one part is to convert the results of one-dimensional channel routing into two-dimensional spatial distribution. The other is to calculate the habitat suitability index according to time steps by using two-dimensional hydraulic features. The model was applied to a restoration area of the Cheongmi Stream. The advantage of this study is that two-dimensional hydraulic analysis can be easily obtained from one-dimensional hydraulic analysis without a complex and time-consuming two-dimensional analysis. HHS (Hydraulic Habitat Suitablility) by sections of target reaches and target species can be easily obtained using the results of this study.

Cytotoxic Activity of the Extracts from Curcuma zedoaria

  • The effect of the hexane extract of Curcuma zedoaria roots and its solvent fractions were investigated on the proliferation of SiHa, SNU-1 and HepG2 cell lines. Among those fractions, final fraction H2-3-1 and H2-3-3 showed cytotoxic effect on SiHa and HepG2 cell lines. The hallmark of apoptosis, DNA fragmentation, also appeared in the final fractions H2-3-1 and H2-3-3 after 24h treatment in SiHa cell line. Furthermore, those fractions were shown to be able to induce cell death in $[^3H]$thymidine incorporation test. These two fractions, H2-3-1 and H2-3-3 were determined as (-)-$\alpha$-curcumene and $\beta$-tumerone respectively by NMR and mass spectrum. From these results, it is speculated that te hexane extract of Curcuma zedoaria is necessary for further studies as a potent inhibitor of the growth of cancer cells.

Behaviour of Uranyl Phosphate Containing Solid Waste During Thermal Treatment for the Purpose of Sentencing and Immobilisation: Preliminary Results

  • Thermal decomposition of the uranyl phosphate mineral phase meta-ankoleite (KUO2PO4·3H2O) has been considered in relation to high temperature thermal sintering for the immobilisation of a uranyl phosphate containing waste. Meta-ankoleite thermal decomposition was studied across the temperature range 25 - 1200℃ under an inert N2 atmosphere at 1 atm. It is shown that the meta-ankoleite mineral phase undergoes a double de-hydration event at 56.90 and 125.85℃. Subsequently, synthetically produced pure meta-ankoleite remains stable until at least 1150℃ exhibiting no apparent phase changes. In contrast, when present in a mixed waste the meta-ankoleite phase is not identifiable after thermal treatment indicating incorporation within the bulk waste either as an amorphous phase and/or as uranium oxide. Visual inspection of the waste post thermal treatment showed evidence of self-sintering owing to the presence of glass former materials, namely, silica (SiO2) and antimony(V) oxide (Sb2O5). Therefore, incorporation of the uranium phase into the waste as part of waste sentencing and immobilisation via high temperature sintering for the purpose of long-term disposal is deemed feasible.
